Abstract
MÉTODOS DE SEQUENCIAMENTO E RECONSTRUÇÃO DE UM ÚNICO POLIPEPTÍDEO. Métodos de sequenciamento de um único polipeptídeo. Os métodos compreendem fornecer uma amostra enriquecida que compreende uma população de polipeptídeos; dividir a amostra enriquecida em duas ou mais subamostras; contatar cada uma de pelo menos duas das suba-mostras com um agente modificador diferente, em que o agente modificador compreende um agente de clivagem, por exemplo, uma exopeptidase, deste modo, gerando fragmentos polipeptídicos que têm uma combinação de padrões de clivagem; e sequenciar, em paralelo, os fragmentos polipeptídicos, deste modo, determinando as sequências de aminoácidos dos fragmentos polipeptídicos. Os fragmentos podem ser alinhados para reconstruir a sequência polipeptídica. Também é fornecido no presente documento um kit que compreende uma pluralidade de moléculas de enriquecimento, por exemplo, anticorpos, aptâmeros ou enzimas, e um dispositivo de preparação de amostras que compreendem códigos de barras e sondas de captura.
