Abstract
La presente invención tiene como objetivo la identificación de nuevas enzimas de biosíntesis de luciferina fúngica, ácidos nucleicos capaces de codificar estas enzimas y proteínas capaces de catalizar ciertas etapas de la biosíntesis de luciferina fúngica. La invención también proporciona la aplicación de ácidos nucleicos para producir dichas enzimas en una célula u organismo. También se proporcionan métodos para la preparación in vitro o in vivo de compuestos químicos idénticos a las luciferinas y preluciferinas fúngicas. También se proporcionan los vectores que comprenden el ácido nucleico descritos en la presente invención. Además, la presente invención proporciona casetes de expresión que comprenden el ácido nucleico de la presente invención y elementos reguladores necesarios para la expresión del ácido nucleico en una célula hospedera seleccionada. Además, también se proporcionan células, líneas celulares estables, organismos transgénicos (por ejemplo, plantas, animales, hongos o microorganismos) que incluyen ácidos nucleicos, vectores o casetes de expresión de la presente invención. La presente invención también proporciona combinaciones de ácidos nucleicos para obtener células, líneas celulares u organismos transgénicos autónomos luminosos. En modalidades preferidas, las células u organismos transgénicos son capaces de producir luciferina fúngica a partir de precursores. En algunas modalidades, las células u organismos transgénicos son capaces de producir preluciferina fúngica a partir de precursores. En algunas modalidades, las células u organismos transgénicos son capaces de bioluminiscencia en presencia de un precursor de luciferina fúngico. En algunas modalidades, las células u organismos transgénicos son capaces de bioluminiscencia autónoma. También se proporcionan combinaciones de proteínas para producir luciferina o sus precursores a partir de compuestos químicos más simples. También se proporciona un kit que contiene ácidos nucleicos, vectores o casetes de expresión de la presente invención para producir células luminosas, líneas celulares u organismos transgénicos.
