Abstract

Se proporcionan métodos para determinar la metilación de ADN. En una modalidad ilustrativa, pero no limitante, el método comprende i) poner en contacto una muestra biológica que comprende un ácido nucleico con un primer material de matriz que comprende una primera columna o filtro donde el material de matriz se une y/o filtra ácidos nucleicos en la muestra y por lo tanto, purifica el ADN; ii) eluir el ADN unido del primer material de la matriz y desnaturalizar el ADN para producir ADN desnaturalizado eluido; iii) calentar el ADN eluido en presencia de iones bisulfito para producir un ácido nucleico desaminado; iv) poner en contacto el ácido nucleico desaminado con un segundo material de matriz que comprende una segunda columna para unir este ácido nucleico desaminado con este segundo material de matriz; v) desulfonar el ácido nucleico desaminado unido y/o simultáneamente eluir y desulfonar el ácido nucleico poniendo en contacto el ácido nucleico desaminado con una solución alcalina para producir un ácido nucleico convertido con bisulfito; vi) eluir este ácido nucleico convertido con bisulfito del segundo material de matriz; y vii) realizar la PCR específica de metilación y/o secuenciación de ácido nucleico, y/o análisis de fusión de alta resolución (HRM) en el ácido nucleico convertido con bisulfito para determinar la metilación del ácido nucleico, en donde al menos los pasos iv) a vi) se realizan en un solo cartucho de reacción.
