Abstract
métodos para determinação da metilação do dna são fornecidos. em uma ilustrativa, mas não limitante modalidade, o método compreende i) colocar uma amostra biológica compreendendo um ácido nucleico, em contato com um primeiro material de matriz compreendendo uma primeira coluna ou filtro, em que o referido material de matriz liga-se e/ou filtra ácidos nucleicos na referida amostra, purificando, assim, o dna; ii) eluir o dna ligado proveniente do primeiro material de matriz e desnaturar o dna para produzir dna desnaturado eluído; iii) aquecer o dna eluído na presença de íons bissulfito para produzir um ácido nucleico desaminado; iv) colocar o referido ácido nucleico desaminado em contato com um segundo material de matriz compreendendo uma segunda coluna para ligar o referido ácido nucleico desaminado ao referido segundo material de matriz; v) dessulfonar o ácido nucleico desaminado ligado e/ou eluir e dessulfonar simultaneamente o ácido nucleico colocando o ácido nucleico desaminado em contato com uma solução alcalina para produzir um ácido nucleico convertido com bissulfito; vi) eluir o referido ácido nucleico convertido com bissulfito do referido segundo material de matriz.
