Abstract

La présente invention concerne un kit pour l’amplification, la détection et/ou la quantification, d’un ou de différents génotypes du virus de l’hépatite virale type b (vhb) présents dans un même ou différents échantillons biologiques, au moyen d’une nouvelle combinaison de set d’amorces ciblant les gènes c, prec, s du virus de l’hépatite b avec un autre set ciblant un adn control interne. L’invention porte sur la combinaison pour la première fois de ces sets d’amorces et sondes pour l’amplification, la détection et/ou la quantification d’une région spécifique au niveau du génome du vhb couplé à la détection du control interne présent naturellement dans le sang humain en utilisant la méthode de pcr en temps réel (qpcr). Cette invention préconise aussi l’utilisation d’un plasmide bactérien, contenant une séquence nucléotidique d’une taille de 1900 paires de base (pb) se situant dans la région conservée des gènes c, pre c, s du vhb, pour la construction d’une gamme étalon (courbe standard) servant à la détection et la quantification fiable du vhb par des nouveaux sets d’amorces et de sondes. En plus de la région spécifique des gènes c, pre c, s, le kit permet la détection d’une région d’une séquence spécifique du gène rpl30 et/ou gapdh qui est utilisé comme contrôle interne (ipc).
