Abstract

본 발명은 식물세포내에서 외래유전자의 발현을 가능하게 하는 벡터에, DPE계 제초제에 대하여 저항성이 있는 Protox의 유전자를 삽입하여 제조한 재조합 발현벡터 및 전기 발현벡터로 식물세포를 형질전환시켜, DPE계 제초제에 대하여 저항성이 있는 Protox를 생성하는 식물체를 제조하는 방법을 제공한다. 본 발명의 방법에 의하여 Protox 유전자가 발현되는 형질전환 식물체를 제조함으로써, DPE계 제초제에 대하여 저항성이 있는 식물체를 개발할 수 있을 것이다.

제초제 저항성 식물체의 제조방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 본 발명의 재조합 발현벡터 pBP14를 제조하는 일련의 과정을 도식화된 그림이다,
제2도는 재조합 발현벡터 pBP14를 BamHI 또는 EcoRI으로 절단한 다음, 0.8% 아가로스 젤 상에서 전기영동한 결과를 나타내는 사진이다,
제3(A)도는 재조합 발현벡터 pBP14을 지니고 있는 아그로박테리움 투메파시엔스 LBA 4404로 형질전환된 담배세포로부터 유도된 슈트를 나타내는 사진이다,
제3(B)도는 형질전환된 담배세포에서 유도된 슈트로부터의 뿌리형성을 나타내는 사진이다.

Claims (7)

  1. DPE(diphenylether)계 제초제에 대하여 저항성이 있는 프로토포리리노겐 옥시데이즈(Protox : protoporphyrinogen oxidase)의 유전자를 식물체에서 발현시키는 재조합 발현벡터.
  2. 제1항에 있어서, 프로토포피리노겐 옥시데이즈는 바실러스 섭틸리스(Bacillus subtilis) 유래인 것을 특징으로 하는 재조합 발현벡터 pBP14.
  3. 제2항의 재조합 발현벡터로 형질전환된 미생물.
  4. 제2항의 재조합 발현벡터 pBP14로 형질전환된 아그로박테리움 투메파시엔스(Agrobacterium tumefaciens) LBA 4404(KCCM-10073).
  5. 제2항의 재조합 발현벡터로 형질전환된 미생물을 식물세포로 도입하고 이를 배양하는 공정을 포함하는, DPE(diphenylether)계 제초제에 대하여 저항성이 있는 프로토포피리노겐 옥시데이즈를 생성하는 식물체의 제조방법.
  6. 제5항에 있어서, 형질전환된 미생물은 아그로박테리움 투메파시엔스(Agrobacterium tumefaciens) LBA 4404(KCCM-10073)인 것을 특징으로 하는 식물체의 제조방법.
  7. 제5항에 있어서, 식물세포는 담배인 것을 특징으로 하는 식물체의 제조방법.
