Abstract
본 발명의 방법은 포유등물 세포에서 인터루킨-5-수용체-의존성 시그날 진달 경로에 대한 조절 효과를 갖는 물질을 스크리닝하는 방법에 관한 것이다.
시험 물질을 기능성 인터루킨-5-수용체를 발현하고 리포터 유전자, 및 여기에 기능적으로 결합된, 인터루킨-5-수용체-의존성 시그날 전달 경로의 제2메신저 물질의 농도 변화에 민강한 조절 서열을 함유하는 제조합 DNA로 형질전환시킨 세포에 적용한다. 인터루킨-5-수용체-의존성 시그날 전달 경로에 특이적이고 잠재적인 약물학적 효과를 갖는 물질은 이 방법에 의해 확인될 수 있다.

Claims (28)
- a) 인터루킨-5 수용체에 커플링된 시그날 전달 징로의 하나 또는 그 이상의 제2메신저 물질의 농도 변화에 반응하는 조절 서열과 리포터 유전자를 함유하는 재조합DNA로 형질전환시켜 리포터 유전자의 발현이 메신저 물질의 농도 변화에 의해 조절되도록 하고, 추가로 b) 조사할 물질에 의해 기능성 인터루킨-5 수용체를 조사할 물질로 발현하는 포유동물 세포를 배양하고, 리포터 유전자 생성물의 농도를 측정하고, 시험 물질에 대해 확인된 활성이 인터루킨-5 수용체 중제된 시그날 전달 경로에 대해 선택적인지 여부를 조사함으로써 인터루킨-5 수용체의 활성화에 의해 개시된 시그날 전달 경로에서의 성분에 대한 물질의 조절 효과를 측정함을 특징으로 하여, 사람 또는 동물 세포에서 수용체-중제된 시그날 전탈 경로에 대한 물질의 조절 효과를 측정하는 방법.
- 제1항에 있어서, 인터루킨-5를 또한 세포에 적용시켜 인터루킨-5 수용체 중제된 시그날 전달 경로에 대한 시험물질의 길항적 효과를 조사함을 특징으로 하는 방법.
- 제1항에 있어서, 기능성 인터루킨-5 수용체를 천연적으로 발현시키는 세포를 사용함을 특징으로 하는 방법.
- 제1항에 있어서, 인터루킨-5 수용체의 β-서브유니트만을 천연적으로 발현하며 인터루킨-5 수용체의 α-서브유니트를 코드화한 서열을 함유하는 재조합 DNA로 형질전환된 세포를 사용하여 세포가 기능성 인터루킨-5 수용체를 발현되도륵 함을 특징으로 하는 방법.
- 제1항에 있어서, 인터루킨-5 수용체의 α-서브유니트를 코드화한 서열 및 β-서브유니트를 코드화한 서열을 함유하는 재조합 DNA로 형질전환시킨 세포를 사용하여 세포가 기능성 인터루킨-5 수용체를 발현되도록 함을 특징으로 하는 방법.
- 제5항에 있어서, α-서브유니트를 코드화한 DNA 서열 및 β-서브유니트를 코드화한 DNA 서열이 두개의 별개의 플라스미드 상에 존재함을 특징으로 하는 방법.
- 제1항 내지 6항 중의 어느 한 항에 있어서, 세포가 기능성 사람 인터루킨-5 수용체를 발현함을 특징으로하는 방법.
- 제1항 내지 6항 중의 어느 한 항에 있어서, 세포가 기능성 뮤린 인터루킨-5 수용체를 발현함을 특징으로 하는 방법.
- 제1항 내지 6항 중의 어느 한 항에 있어서, 세포가 그의 α-서브유니트 및 β-서브유니트가 상이한 종으로부터 유도되는 기능성 인터루킨-5 수용체를 발현항을 특징으로 하는 방법.
- 제9항에 있어서, α-서브유니트가 사람 기원임을 특징으로 하는 방법.
- 제1항 내지 10항 중의 어느 한 항에 있어서, a)에서 정의된 재조합 DNA가 조절 서열로서 c-fos-유전자의 프로모터를 함유함을 특징으로 하는 방법.
- 제1항 내지 10항 중의 어느 한 항에 있어서, a)에서 정의된 재조합 DNA가 조절 서열로서 ICAM-1-유전자의 프로모터를 함유함을 특징으로 하는 방법.
- 제11항 또는 12항에 있어서, a)에서 정의된 재조합 DNA가 리포터 유전자로서 루시페라제 유전자를 함유함을 특징으로 하는 방법.
- 제2항 내지 13항 중의 어느 한 항에 있어서, 기능성 IL-5 수용체 및 다른 성장 인자에 대한 수용체를 발현하며, 이들 수용체들 중의 적어도 하나에 대한 리간드의 존재에 대해 그들의 중식이 의존적인 세포를 수집하고, 한편에서는 단지 인터루킨-5와 함께 배양하고, 다른 한편에서는 시험 물질 및 인터루킨-5와 함께 배양하여 두 세포 배양물의 중식을 비교함으로써, 시현 물질의 길항 활성을 조사함을 특징으로 하는 방법.
- 제1항 내지 13항 중의 어느 한 항에 있어서, 인터루킨-5 수용체 중제된 시그날 전탈 경로에 대한 물질의 조절 효과의 선택성을, 이 물질을 인터루킨-5 수용체 이외의 조절 수용체를 발현이며 a)에서 정의된 재조합 DNA(이의 조절 서열은 또한 조절 수용체에 커플링된 시그날 전달 경로의 제2메신저 물질의 농도 변화에 반응한다)에 의해 형질전환된 포유동물 대조 세포에 대해 동일한 조건하에서 적용시키고, 인터루킨-5 수용체를 발현하는 세포에서 리포터 유전자 생성물의 농도를 대조 세포에서의 농도와 비교함으로써 시험함을 특징으로 하는 방법.
- 제15항에 있어서, 조절 수용체가 인터루킨-3 수용체임을 특징으로 하는 방법.
- 제1항 내지 13항 중의 어느 한 항에 있어서, 추가로 a)에서 정의된 재조합 DNA로 형질전환되고 기능성 인터루킨-5 수용체를 발힌하지 않는 포유동물 세포를 조사할 물질과 함께 동일한 조건하에서 배양하고 리포터유전자 생성물의 농도를 측정함을 특징으로 하는 방법.
- 제1항 내지 17항 중의 어느 한 항에 있어서, 시험 물질이 정해진 조건하에서 정해진 수의 포유동물 세포와 함께 배양한 다수의 물질 중의 하나이며, 리포터 유전자 생성물의 농도를 측정하여, 인터루킨-5 의존성 시그날 전달 경로에 대한 물질의 선택성을 체크하는 스크리닝 분석으로 사용함을 특징으로 하는 방법.
- a) 리포터 유전자, 및 인터루킨-5 수용체에 커플링된 시그날 전탈 경로의 하나 또는 그 이상의 제2메신저 물질의 농도 변화에 반응하는 조절 서열을 함유하는 재조합 DNA에 의해 형질전환되며, b) 인터루킨-5수용체의 α-서브유니트 또는 α-및 β-서브유니트를 코드화한 재조합 DNA로 형질전환되어, 기능성 인터루킨-5 수용체를 발현함을 특징으로 하는 포유동물 세포.
- 제19항에 있어서, 사람 세포임을 특징으로 하는 포유동물 세포.
- 제19항에 있어서, 뮤린 세포임을 특징으로 하는 포유동물 세포.
- 제19항 내지 21항 중의 어느 한 항에 있어서, 천연적으로 인터루킨-5 수용체의 β-서브유니트만을 발현하고 α-서브유니트를 코드화한 서열을 함유하는 재조합 DNA로 형질전환됨을 특징으로 하는 포유동물 세포.
- 제22항에 있어서, α-서브유니트가 다른 종으로부터 유래됨을 특징으로 하는 포유동물 세포.
- 제22항에 있어서, 사람 α-서브유니트를 코드화한 서열을 함유하는 재조합 DNA로 형질전환됨을 특징으로 하는 사람 세포.
- 제22항에 있어서, 뮤린 α-서브유니트를 고드화한 서열을 함유하는 재조합 DNA로 형질전환됨을 특징으로 하는 뮤린 세포.
- 제19항 내지 25항 중의 어느 한 항에 있어서, a)에서 정의된 재조합 DNA가 조절 서열로서 c-fos-유전자의 프로모터를 함유함을 특징으로 하는 포유동물 세포.
- 제19항 내지 25항 중의 어느 한 항에 있어서, a)에서 정의된 재조합 DNA 가 조절 서열로서 ICAM-1-유전자의 프로모터를 함유함을 특징으로 하는 포유동물 세포.
