Abstract

페테틴(Patatins)은 유충의 성장발육을 방해하여, 성숙과 재생을 막으므로써 곤충을 억제한다. 하나이상의 이러한 단백질을 암호하는 유전자들은 식물-기생 미생물들 또는 식물들의 형질전환을 위하여 벡터내로 클론될수 있으며, 이에 의해 곤충만연을 억제하는 방법이 제공된다.

Claims (11)

  1. 곤충에 의한 섭취를 위해 유효량의 살충성 페테틴을 제공하는 것을 포함하는, 식물-섭취 곤충의 만연을 억제하는 방법.
  2. 제1항에 있어서, 상술한 페테틴은, 식물에 적용된 후에 페테틴을 생성하는 식물-기생 미생물에 의하여 제공됨을 특징으로 하는 방법.
  3. 제1항에 있어서, 상술한 페테틴은 식물의 모세포의 사전 유전적 형질전환에 의하여 상술한 식물내에 혼입된 페테틴 유전자의 발현에 의하여 제공됨을 특징으로 하는 방법.
  4. 제3항에 있어서, 상술한 식물은 목화, 옥수수, 토마토 또는 감자인 것을 특징으로 하는 방법.
  5. 다음의 a),b),c)단계를 포함하는, 살충적 유효량의 패테틴을 발현하는, 유전적으로 형질전환되고 곤충에 내성아 있는 식물을 생산하는 방법.
    (i)식물세포내에서 RNA서열이 생산되도록 작용하는 프로모터; (ii)페테틴을 암호하는 구조암호서열: (ii)상술한 식물세포내에서 RNA서열의 3'단말에 폴리아데닐레이트 뉴클레오티드가 첨가되도록 작용하는 3'-비-해독영역을 포함하는 재조합 이중가닥 DNA분자를 식물세포의 게놈내로 삽입하는 단계; 여기서 상술한 프로모터는 상술한 구조암호서열에 대해 이종성이며, 상술한 프로모터는 상술한 구조암호서열과 작동적으로 결합된 다음 상술한 비-해독영역과 작동적으로 결합된다; b)형질전환된 식물세포를 얻는 단계; c)상술한 형질전환된 식물로부터, 살충적으로 유효량의 패테틴을 발현하는 유전학적으로 형질전환된 식물세포를 재생시키는 단계; 여기서 상술한 프로모터는 구조암호서열에 대해 이종성이며, 상술한 식물은 목화, 옥수수, 토마토 및 감자로부터 선택된다.
  6. 제5항에 있어서, 상술한 구조암호서열은 SEQ ID NO:30 또는 SEQ ID NO:31을 포함하는 것을 특징으로 하는 방법.
  7. 제5항에 있어서, 상술한 식물은 옥수수이고, 상술한 구조암호서열은 외떡잎식물에서 향상된 발현을 하도록 합성된 것임을 특징으로 하는 방법.
  8. 제7항에 있어서, 상술한 구조암호서열은 SEQ ID NO:32를 포함하는 것을 특징으로 하는 방법.
  9. 제5항의 방법에 의하여 생산된 식물.
  10. 제9항에 있어서, 게놈내에 하나 이상의 유전자 발현 B.t엔도톡신이 포함된 것을 특징으로 하는 식물.
  11. 제9항의 식물에 의하여 생산된 종자 또는 종자단편.
