Abstract
본 발명은 분자생물학적 1단계 유전자 파괴법(one step gene disruption method)으로 효모 게놈 DNA 상의 knr4 유전자를 불활성화시킴으로써 세포벽을 약화시킨 돌연변이 효모와 이를 이용한 유용 재조합 단백질의 개선된 생산 방법에 관한 것이다.

Claims (6)
1단계 유전자 파괴법(one step gene disruption method)에 의해 knr4 유전자를 불활성화시킨 세포벽이 약화된 돌연변이 효모.
제1항에 있어서, ura3 유전자의 삽입에 의해 knr4 유전자를 불활성화시킨 돌연변이 효모.
제2항에 있어서, 플라스미드 pUC18-knr4-ura3를 사용하여 knr4 유전자를 불활성시킨 돌연변이 효모.
제1항에 있어서, 효모 사카로마이세스 세레비지애 EGY48의 knr4 유전자를 불활성화시킨 돌연변이 효모.
제4항에 있어서, 사카로마이세스 세레비지애 MI01(KCTC 0134BP).
제1항 내지 제5항 중 어느 한 항의 효모를 목적으로 재조합 단백질을 코드하는 유전자를 포함하는 벡터로 형질전환시킨 후 적당한 배지상에서 배양하여 상기 유전자를 발현시킨 다음, 상기 목적 재조합 단백질을 분리 및 정제함을 포함하는 재조합 단백질의 개선된 생산 방법.
