본 발명은 천연 N-말단 서열을 갖는 재조합 아프로티닌 및 재조합 아프로티닌 변형체의 제조 방법에 관한 것으로 재조합 아프로티닌 및 (또는) 재조합 아프로티닌 변형체는 균질하게 프로세싱된, 분비된 단백질로서 존재한다.

Claims (3)

  1. a) 아프로티닌 또는 그 변형체를 위한 유전자, 및 b) 이 유전자의 직접 상류의 리더로서, 신호 펩티다제에 의해 프로세싱되는 프리 서열을 위한 뉴클레오티드 서열을 함유하는, 정확한 N-말단을 갖는 아프로티닌 및 아프로티닌 변형체의 발현 벡터.
  2. 제1항에 있어서, 리더 신호 서열이 프리 알파 인자, PHO1, PHO5 또는 인버타제로부터 유래되는 벡터.
  3. 미생물이 제1 또는 2항에 따른 벡터를 함유하는 것을 특징으로 하는, 미생물을 배양하고 배양 상등액으로부터 아프로티닌 또는 그 변형체를 분리함으로써 정확한 N-말단을 갖는 아프로티닌 및 아프로티닌 변형체를 제조하는 방법.
