Abstract

본 발명은 인돌에 대한 용해도가 매우 높은 수-비혼화성 유기용매와 인돌에 대한 용해도는 비교적 낮지만 효소의 안정성을 증진시키는 수-혼화성 유기용매를 적절히 혼합 사용하여 효소 반응계내의 초기 인돌 농도를 증가시킴으로써 최종 생성물인 L-트립토판의 생성 농도를 증대시키기 위한 개선된 L-트립토판의 제조방법에 관한 것이다.

제1도는 본 발명의 L-트립토판 합성효소원으로 사용된 균주인 이.콜라이(E.coli)의 배양시간에 따른 균체 및 효소 활성을 나타네는 도면이다.

Claims (5)

  1. 트립토판 합성효소를 이용하여 인돌 및 세린으로부터 L-트립토판을 제조하는 방법에 있어서, 수-비혼화성 유기 용매와 효소의 안정성을 증진시키는 수-혼화성 유기용매의 혼합 용매를 효소 반응액에 포함시킴을 특징으로 하는 개선된 L-트립토판의 제조방법.
  2. 제1항에 있어서, 상기 수-혼화성 유기용매가 수-혼화성 알콜 또는 에테르이고, 상기 수-비혼화성 유기용매가 수-비혼화성 알콜, 에테르, 알칸, 에스테르 및 식물성 유지로 이루어진 그룹중에서 선택되는 방법.
  3. 제2항에 있어서, 수-비혼화성 유기용매로는 디에틸렌글리콜 디부틸에테르, 수-혼화성 유기용매로는 폴리에틸렌글리콜을 사용하는 방법.
  4. 제2항에 있어서, 상기 혼합 유기 용매가 총 효소 반응액의 10내지 50부피%의 양으로 사용되는 방법.
  5. 제1항에 있어서, 상기 수-비혼화성 유기 용매와 수-혼화성 유기용매의 혼합비가 1 : 5 내지 5 : 5(v/v)인 방법.
