Abstract

샌드위치 방법의 범주에 속하고, 제1반응물로서 당쇄를 변형시켜서 당쇄-결합물질에 대한 결합성이 상실되게 한 변형물질을 이용하고 제2반응물로서 당복합체의 당쇄에 결합할 수 있는 물질을 이용하는 것을 특징으로 하는 당단백질 및 당지질 등의 당복합체를 측정하는 기본방법이 개시된다.
당복합체의 하나인 류머티양 인자를 측정하는 경우에는 변형 IgG 및 렉틴을 각각 제1반응물 및 제2반응물로서 이용한다.

제1도는 실시예 4에서 행한 각종 환자의 검체 혈청중의 류머티양 인자 측정결과를 나타낸다.

Claims (17)

  1. 하기의 단계들로 이루어지는 샌드위치 방법에 의한 당복합체의 측정방법: (1) 당복합체에 특이적인 결합성을 갖는 물질의 당쇄를 처리하여 당쇄-결합물질에 대한 결합성이 상실되게 한 변형물질(변형 제1반응물)을 검체와 반응시켜서 검체중의 당복합체에 결합시키는 단계; (2) 제2반응물로서 당복합체의 당쇄와의 결합성을 갖는 당쇄-결합물질을 그렇게 결합된 당복합체와 반응시키는 단계; 및 (3)그렇게 결합된 당쇄-결합물질의 양을 결정하는 단계.
  2. 제1항에 있어서, 상기 당쇄-결합물질이 표지된 당쇄-결합물질인 것을 특징으로 하는 방법.
  3. 제1항에 있어서, 상기의 당쇄-결합물질이 렉틴인 것을 특징으로 하는 방법.
  4. 제1항에 있어서, 당쇄-결합물질에 대한 결합성의 상실을 야기하는 상기의 처리가 화학적 처리인 것을 특징으로 하는 방법.
  5. 제4항에 있어서, 상기의 화학적 처리가 과요드산 처리인 것을 특징으로 하는 방법.
  6. 제1항에 있어서, 상기의 변형 제1반응물이 고정화되는 고상을 사용하는 것을 특징으로 하는 방법.
  7. 제1항에 있어서, 상기의 당복합체가 류머티양 인자인 것을 특징으로 하는 방법.
  8. 제1항에 있어서, 상기의 변형 제1반응물은 변형 면역글로불린(IgG)이고, 제2반응물로서 이용되는 당쇄-결합물질은 렉틴이고, 당복합체는 류머티양 인자인 것을 특징으로 하는 방법.
  9. 제8항에 있어서, 상기의 변형 (IgG)는 과요드산으로 처리된 IgG인 것을 특징으로 하는 방법.
  10. 제8항에 있어서, 상기의 렉틴이 표지된 렉틴인 것을 특징으로 하는 방법.
  11. 제8항에 있어서, 상기의 렉틴이 리시누스 콤무니스 아를루티닌(RCA 120), 콘카나발린A(Con A)및 렌즈 쿨리나리스(LCA)로 구성되는 군에서 선택되는 것을 특징으로 하는 방법.
  12. 당복합체에 특이적인 결합성을 갖는 물질의 당쇄를 처리하여 당쇄-결합물질에 대한 결합성이 상실되게 한 변형물질 (변형 제1반응물) 및 당쇄-결합물질을 필수성분으로서 함유하는 당복합체가 측정 시약.
  13. 제12항에 있어서, 당쇄-결합물질에 대한 결합성의 상실을 야기하는 상기의 처리가 화학적 처리인 것을 특징으로 하는 측정시약.
  14. 제13항에 있어서, 상기의 화학적 처리가 과요드산 처리인 것을 특징으로 하는 측정시약.
  15. 제12항에 있어서, 상기의 당쇄-결합물질은 렉틴이고, 변형 제1반응물은 변형 lgG이고, 당복합체는 류머티양 인자인 것을 특징으로 하는 측정시약.
  16. 제15항에 있어서, 상기의 변형 IgG는 과요드산으로 처리된 IgG인 것을 특징으로 하는 측정시약.
  17. 제15항에 있어서, 상기의 렉틴은 리시누스 콤무니스 알글루티닌(RCA 120), 콘카나발린A (ConA) 및 렌즈 쿨리나리스(LCA)로 구성되는 군에서 선택되는 것을 특징으로 하는 측정시약.
