Description

한국형 C형 간염 바이러스의 특이항원인 KHCV UB-코아(CORE) 14 단백질의 정제 방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 한국형 C형 간염 바이러스의 특이항원인 KHCV UB-코아 14 cDNA 절편의 염기서열을 나타낸 것이고,
제2도는 정제 과정의 각 단계에서 얻어지는 조생성물을 SDS-폴리아크릴 아마이드 젤 전기영동결과 및 웨스턴 블롯팅 결과를 나타낸 것이다.

Claims (7)

  1. (가) 재조합 한국형 C형 간염 바이러스의 KHCV코아 14 단백질과 유비퀴틴의 융합 단백질(KHCV UB-코아 14 단백질)을 생성시킨 대장균으로부터 세포를 파괴한 후 불용성 단백질을 제거하고; (나) 우레아로 처리하여 KHCV UB 코아 14 단백질을 해리시키며; (다) 산을 처리하고; (라) 상기에서 얻은 용액을 이온교환 크로마토그래피하고; (마) 상기에서 얻은 분획물을 모아 젤 투과 크로마토그래피시키며; 최종적으로, (바) FPLC-모노(Mono)-S 크로마토그래피시킴을 특징으로 하는 C형 간염 바이러스의 특이항원인 KHCV UB 코아 14 단백질의 정제방법.
  2. 제1항에 있어서, 상기 우레아 용액이 8M 우레아를 포함하는 인산염 완충액임을 특징으로 하는 방법.
  3. 제1항에 있어서, 상기 단계(다)에서 산처리하는 1M 초산을 사용하여 pH4.5 내지 5가 되도록 함을 특징으로 하는 방법.
  4. 제1항에 있어서, 상기 단계(라)에서 이온 교환 크로마토그래피로서 CM-세파로스 컬럼을 사용함을 특징으로 하는 방법.
  5. 제1항에 있어서, 상기 단계 (마)에서 젤 투과 크로마토그래피로서 S-200세파크릴 컬럼을 사용함을 특징으로 하는 방법.
  6. 제1항에 있어서, 상기 단계(마)를 pH6.0 내지 8.0에서 진행시킴을 특징으로 하는 방법.
  7. 제1항에 있어서, 상기 단계(마)에서 용출액으로서 8M 우레아를 포함하는 인산염 완충액을 사용함을 특징으로 하는 방법.
