말토올리고시드 유도체, 이것을 유효 성분으로 함유하는 α-아밀라아제 활성 측정용 시약 및 이것을 사용한 α-아밀라아제 활성의 측정 방법

제1도는 실시예 5에 있어서 α-아밀라아제 활성의 측정에 사용된 검량선의 그래프.

제3도는 실시예 9에 있어서 본 발명의 기질과 대조 기질과의 측정계내에서의 안정성을 나타내는 그래프.

Claims (7)

  1. 아래의 일반 구조식으로 나타내어지는 말토올리고시드 유도체.
    위의 식에서 n은 3∼5의 정수이고, R은 방향족 발색성 그룹이며, X는 〉CHCH2N3또는 〉C=CH2의 그룹이고, Y는 수소 원자, 치환 또는 비치환의 탄화 수소 그룹 또는 알킬-혹은 아릴술포닐 그룹이다.
  2. 제1항에 있어서, 방향족 발색성 그룹 R은 아래의 일반 구조식들로 나타내어지는 것인 말토올리고시드 유도체.
    위의 식에서, R1∼R5는 같거나 상이하며, 수소 원자, 할로겐 원자, 니트로 그룹, 알킬 그룹, 아릴 그룹, 아랄킬 그룹, 아미노 그룹, 술폰산 그룹 또는 카르복실 그룹이고, 또는 R1과 R2혹은 R2와 R3은 함께 결합하여 축합 방향족환을 형성하여도 좋다.
    (위의 식에서 R6은 수소 원자 또는 알킬 그룹이다)
    (위의 식에서 R7은 수소 원자 또는 할로겐 원자이다)
    (위의 식에서 R8∼R15는 같거나 상이하며, 수소 원자, 할로겐 원자, 니트로 그룹, 알킬 그룹, 아릴 그룹, 아릴킬 그룹, 아미노 그룹, 술폰산 그룹 또는 카르복실 그룹이고, R8과 R9혹은 R10과 R11은 함께 결합하여 축합 방향족환을 형성하여도 좋으며, R9및 R10및/또는 R13과 R14는 각각 공통의 산소 원자가 되어 축합 에테르환을 형성하여도 좋고, Z는 질소 원자 또는 N→0이다).
  3. 제1항에 있어서, 2-클로로-4-니트로페닐 65-아지도-65-데옥시-D-말토펜타오시드, 2-클로로-4-니트로페닐 65-아지도-66-데옥시-45-0-메실-D-말토펜타오시드, 2-클로로-4-니트로페닐 55-에노-D-말토펜타오시드, 2-클로로-4-니트로페닐 55-에노-45-0-메실-D-말토펜타오시드, 4-니트로페닐 57-에노-47-0-메톡시메틸-D-말토펜타오시드, 4-니트로페닐 67-아지도-67-데옥시-D-말토헵타오시드, 2, 4-디클로로페닐 67-아지도-67-데옥시-47-0-토실-D-말토헵타오시드, 페놀인도-3′-클로로페닐 65-아지도-65-데옥시-45-0-메틸-D-말토펜타오시드, 4-메틸움벨리페로닐 65-아지도-65-데옥시-D-말토펜타오시드, 레자주리닐 56-에노-D-말토헥사오시드, 루시페리닐 67-아지도-67-데옥시-47-0-알릴-D-말토헵타오시드 또는 페놀인도-3′-클로로페닐 55-에노-45-0-(2-메톡시)에톡시메틸-D-말토펜타오시드인 말토펜타오시드 유도체.
  4. 제1항에 있어서, 2-클로로-4-니트로페닐 65-아지도-65-데옥시-D-말토펜타오시드 또는 2-클로로-4-니트로페닐 55-에노-45-0-메실-D-말토펜타오시드인 말토펜타오시드 유도체.
  5. 제1항에 의한 말토올리고시드 유도체를 유효 성분으로 함유하는 α-아밀라아제 활성 측정용 시약.
  6. α-아밀라아제 함유 시료에 제1항에 의한 말토올리고시드 유도체의 α-아노머와 α-글루코시다아제 및/또는 글루코아밀라아제를 첨가하여 효소 반응을 시켜 유리하는 방향족 발색성 화합물을 정량함을 특징으로 하는 α-아밀라아제의 활성 측정 방법.
  7. α-아밀라아제 함유 시료에 제1항에 의한 말토올리고시드 유도체의 β-아노머 또는 α-아노머와 β-아노머와의 혼합물과 α-글루코시다아제 및/또는 글루코아밀라아제 및 β-글루코시다아제를 첨가하여 효소 반응을 시켜 유리하는 방향족 발색성 화합물을 정량함을 특징으로 하는 α-아밀라아제의 활성 측정 방법.
