제1도는 근사치의 좌우크기, 증폭된 단편의 위치, 및 클론화되고 서열분석된 게놈성 영역의 위치가 도해된 DMD유전자의 개요도이다.
제2도는 산전 진단을 위해 태아의 DNA에서의 결실을 탐지하는데 사용된 PCR반응의 예시도이다.
제3도는 관련이 없는 남자 DMD환자로부터 유래된 림프아세포의 DNA다중 증폭을 나타낸 것이다. A 및 B는 10개 샘플로 된 두세트를 보여준다. 각 DRL#는 알.제이.클레버그 센터프 휴먼제네틱스 다이아그노스틱 리써치 래버러토리(R.J.Kleberg Center for Fuman Genetics Diagnostic Research Laboratory)의 가계수를 의미한다. MW : Hae Ⅲ-절단한 ø X 174 DNA. (-) : 주형 DNA가 반응에 첨가되지 않았음. 증폭된 영역과 유전자상의 영역의 관계는 A의 우측에 나타낸다. 문자는 제1도에서와 상응하는 것이다.

Claims (17)

  1. 다수의 DNA서여을 처리하여 단일 상보본쇄를 형성하고 : 다수의 쌍을 이룬 올리고 뉴크레오티드 프라이머(primer)를 첨가하고(여기에서, 각 쌍은 다른 서열에 의해 특이적이며, 각 쌍의 한쪽 프라이머는 센스(sense)-본쇄내의 서열의 한부분에 실질적으로 상보적이고 각 쌍의 다른쪽 프라이머는 안티-센스(anti-sense) 상보본쇄 내의 동일 서열의 다른 부분에 실질적으로 상보적이다) ; 다수의 프라이머를 이들의 상보적인 서열에 어닐링(annealing)시키고 ; 다수의 어닐링된 프라이머를 각 프라이머의 3´말단으로부터 동시에 연장시켜 각 프라이머 어닐링된 본쇄에 상보적인 연장 생성물을 합성하고(여기에서, 여장 생성물은 이들의 상보본쇄로부터 분리된 후 각 쌍의 다른 프라이머로부터 연장 생성물의 합성을 위한 주형으로서 작용한다) ; 연장 생성물을 이들의 주형으로부터 분리하여 단일 본쇄 분자를 생성하고 ; 단일 본쇄 분자를 상기의 어닐링 단계, 연장 단계 및 분리 단계를 적어도 1회 반복하여 증폭시키고 ; 각각의 상이한 서열로부터 상기의 증폭된 연장 생성물을 동정하는 단계를 포함함을 특징으로 하는, 다수의 DNA서열에서의 결실을 동시에 탐지하는 방법.
  2. 제1항에 있어서, 언급된 서열이 X 및 Y염색체상의 서열의 그룹으로부터 선택되는, 다수의 게놈성 DNA서열에서의 결실을 탐지하는 방법.
  3. 제2항에 있어서, 게놈성 DNA서열이 유전병을 일으키는 결실을 포함하는 X-관련 유전병을 탐지하는 방법.
  4. 제3항에 있어서, 오르니틴 트랜스카바밀라제 결핍증, 하이포크산틴 포스포리보실트랜스퍼라제 결핍중, 스테로이드 설페이타제 결핍증 및 X-관련 근위축증으로 구성된 그룹중에서 선택되는 X-관련 유전병을 탐지하는 방법.
  5. 제4항에 있어서, 다수의 쌍을 이룬 프라이머가 디스트로핀(dystrophin)단백질을 암호화하는 유전자내의 상이한 서열에 상보적인, X-관련 근위축증을 탐지하는 방법.
  6. 제5항에 있어서, 다수의 쌍을 이룬 프라이머가
    로 구성된 그룹중에서 선택되는 방법.
  7. 제3항에 있어서, 게놈성 DNA가 태아의 조직으로부터 유래된 방법.
  8. 제1항에 있어서, 다수의 쌍을 이룬 프라이머가
    로 구성된 그룹중에서 선택되는, 다수의 게놈성 DNA서열에서의 결실을 탐지하는 방법.
