역상 크로마토그래피를 이용한 올리고뉴클레오 타이드의 서열 결정방법

제1도는 올리고뉴클레오타이드의 서열을 결정하는데 사용한 본 발명에 의한 자동화장치의 개략도이다.

Claims (3)

  1. 서열을 결정하고자 하는 올리고 뉴클레오타이드를 고형의 지지체인 담체상의 고정시킨 후, 고정화된 올리고뉴클레오타이드에 맥삼-길버트의 염기 특이적인 용액을 가하는 것을 특징으로 하는 올리고 뉴클레오타이드의 서열 결정 방법.
  2. 제1항의 방법에 있어서, 서열을 결정하는 공정은 서열을 결정하고자 하는 올리고뉴클레오타이드에 맥삼-길버트의 염기 특이적인 용액을 가한후, 생성된 물질을 역상 크로마토그래피의 고형의 지지체 상에 흡착시킨 다음, 이 지지체로 부터 DNA 복합체를 선택적으로 용출시키고, 용출된 물질을 피페리딘 존재하에서 가열한 다음, 가열에 의하여 얻어진 반응 생성물을 역상크로마토그래피의 고형의 지지체상에 흡착시키고, 올리고 뉴클레오타이드 단편을 선택적으로 용출시킨 다음, 이 단편들의 올리고뉴클레오타이드 서열을 분석하는 것을 구성된 것임을 특징으로 하는 방법.
  3. 제1항의 올리고 뉴클레오타이드의 서열을 결정하는데 사용된 장치.
