술지게미 추출물의 산화스트레스로 유도된 세포노화 억제효과를 알아보기 위하여, 하기와 같은 실험을 실시하였다. 인간 섬유아세포(HDFs, Human dermal fibroblasts)를 10% FBS(fetal bovine serum)가 첨가된 DMEM 배지가 들어있는 6-웰 플레이트에 웰 당 1 × 105개로 접종한 후, 5% 농도의 CO2 배양기에서 37℃ 조건으로 24시간 배양하였다. 그 후, 시험물질과 산화스트레스를 유도할 수 있는 과산화수소(H2O2 300 μM)를 함께 FBS가 포함되지 않은 DMEM 배지에 녹인 뒤 처리하였다. EGCG(Epigallocatechin-3-gallate)는 세포노화 억제 양성대조군으로 사용되었다. 72시간 후 세포 배양 상등액을 제거하고, 트립신-EDTA를 처리하여 세포를 떼어낸 다음, Dojindo사의 SPiDER-ßGal으로 세포를 15분 동안 형광염색하고, 유동계수기(flow cytometry, FACS Calibur, Becton Dickinson, San Jose, CA)로 세포를 계수하였다. 노화 관련 β-갈락토시다아제 활성을 나타내는 세포의 비율을 확인하여 세포노화의 변화를 관찰하였다. 그 결과를 도 1에 나타내었다.In order to investigate the inhibitory effect of the sake lees extract on cellular aging induced by oxidative stress, the following experiment was conducted. 술지게미 추출물의 콜라겐 합성 촉진효과를 알아보기 위하여, 하기와 같은 실험을 실시하였다. 인간 섬유아세포를 10% FBS가 첨가된 DMEM 배지가 들어있는 24-웰 플레이트에 웰 당 2 × 104개로 접종한 후, 5% 농도의 CO2 배양기에서 37℃ 조건으로 24시간 배양하였다. 그 후, 시험물질을 FBS가 포함되지 않은 DMEM 배지에 녹인 뒤 처리하였다. TGF-β(Transforming growth factor-β)는 콜라겐 합성 촉진 양성대조군으로 사용되었다. 72시간 후 세포 배양 상등액을 수집하여 샘플을 제조하였다. 상기에서 제조한 샘플의 콜라겐 합성량은 TAKARA사의 콜라겐 측정 키트(MK101, Procollagen Type I C-peptide EIA kit)를 이용하여 프로콜라겐(procollagen) 타입I C-펩타이드(PICP, Type I C-peptide)의 양을 측정하였다. 그 결과를 도 2에 나타내었다.To investigate the effect of sake lees extract on promoting collagen synthesis, the following experiment was conducted. P.C는 엘라스테이즈 활성 저해제로 양성대조군으로 사용되었으며, 시험물질의 엘라스테이즈 활성정도는 96-웰 블랙 플레이트와 Invitrogen사의 엘라스테이즈 활성 측정 키트(EnzChek™ 엘라스테이즈 Assay Kit)를 이용하여 측정하였다. 그 결과를 도 3에 나타내었다.In order to investigate the inhibitory effect of sake lees extract on elastase activity, the following experiment was conducted. 술지게미 추출물의 염증유도물질인 산화질소(nitric oxide) 분비 억제효과를 알아보기 위하여, 하기와 같은 실험을 실시하였다. 마우스 대식세포 (RAW264.7, macrophage)를 10% FBS (fetal bovine serum)가 첨가된 RPMI 배지가 들어있는 24-웰 플레이트에 웰 당 2 × 105개로 접종한 후, 5% 농도의 CO2 배양기에서 37℃ 조건으로 24시간 배양하였다. 그 후, 시험물질을 2% FBS가 첨가된 RPMI 배지에 녹인 뒤 2시간 동안 전처리한 다음, 염증반응을 유도할 수 있는 LPS (Lipopolysaccharide 100 ng/mL)를 각 well에 처리하였다. 1400W는 산화질소 분비 억제 양성대조군으로 사용되었다. 24시간 후 세포 배양 상등액을 수집하여 샘플을 제조하였다. 상기에서 제조한 샘플은 Griess reagent와 반응시켜 산화질소의 안정화된 산화물질인 NO2-(nitrite)를 측정함으로써 생성된 산화질소를 정량하였다. 그 결과를 도 4에 나타내었다.In order to investigate the inhibitory effect of sake lees extract on the secretion of nitric oxide, an inflammation-inducing substance, the following experiment was conducted. The results are shown in Figure 4.

술지게미 추출물의 피지 분비 촉진효과를 알아보기 위하여, 하기와 같은 실험을 실시하였다. 인간 피지세포 (human sebocyte)를 피지세포 배양배지가 들어있는 24-웰 플레이트에 웰 당 1 × 104개로 접종한 후, 5% 농도의 CO2 배양기에서 37℃ 조건으로 24시간 배양하였다. 그 후, 시험물질을 배양배지에 녹인 뒤 처리한 다음 48시간 후 세포 배양 상등액을 제거하고, 4%(v/v) 파라포름알데하이드 용액으로 고정하였다. 팔미트산(palmitic acid)은 피지 분비 촉진 양성대조군으로 사용되었다. 피지세포에서 합성된 피지를 확인하기 위해 Nile red로 상온에서 15분간 염색 한 뒤, 전체 세포 수 확인을 위해 Hoechst 33342로 핵을 염색하여 형광현미경으로 피지 분비 정도를 관찰하였다. 현미경 사진을 ImageJ로 분석하였고, 그 결과를 도 5에 나타내었다.In order to determine the effect of sake lees extract on promoting sebum secretion, the following experiment was conducted. 술지게미 추출물의 미백효과를 알아보기 위하여, 하기와 같은 실험을 실시하였다. 인간 멜라닌세포 (Human melanocyte)를 멜라닌세포 배양배지가 들어있는 6-웰 플레이트에 웰 당 1.5 × 105개로 접종한 후, 5% 농도의 CO2 배양기에서 37℃ 조건으로 24시간 배양하였다. 그 후, 시험물질을 배양배지에 녹인 뒤 처리한 다음 5일 후 세포 배양 상등액을 제거하고, 세포를 Cell Scraper로 수집하였다. Kojic acid는 멜라닌 생성 억제 양성대조군으로 사용되었다. 수집한 세포에 1M 수산화나트륨액을 넣고 세포 내 멜라닌 용액의 흡광도를 450 nm에서 측정하였다. 그 결과를 도 6에 나타내었다.In order to determine the whitening effect of sake lees extract, the following experiment was conducted. 본 발명에 따른 항노화 효과가 우수하다고 판명된 술지게미 추출물이 인체피부에도 안전한지 확인하기 위하여, 술지게미 추출물을 함유한 피부외용제를 제조하고 이에 의한 피부 안전성 검증 실험을 수행하였다.In order to confirm whether the sake lees extract, which was found to have excellent anti-aging effect according to the present invention, is safe for human skin, an external skin preparation containing the sake lees extract was prepared and a skin safety verification experiment was performed.
실시예 1의 술지게미 추출물을 1% 함유한 피부외용제를 하기 표 1의 성분함량으로 제조하였다. 피부 외용제 제조를 위하여, 정제수, 글리세린, 부틸렌글리콜을 혼합하고 70℃에서 용해하였으며(수상파트), 상기 세 성분과 트리메탄올아민을 제외한 나머지 성분을 70℃에서 용해하였다(유상파트). 상기 오일 파트를 수상파트에 첨가하고 호모믹서(일본 Tokushu Kika사)로 교반하여 1차 유화하였고, 여기에 트리메탄올아민을 최종 첨가하였다. 혼합액에 생성된 기포를 제거한 후, 실온으로 냉각시켜 피부 외용제를 제조하였다.An external skin preparation containing 1% of the sake lees extract of Example 1 was prepared with the ingredient contents shown in Table 1 below. 상기와 같이 제조한 각 피부 외용제를 사용하여 건강한 30명의 성인을 대상으로 등 부위에 24시간 폐쇄첩포를 시행하는 것에 의하여 술지게미 추출물이 피부에 자극을 주는지 여부를 측정하였다.We measured whether the sake lees extract irritates the skin by applying an occlusive patch to the back of 30 healthy adults for 24 hours using each skin external preparation prepared as above.
첩포 방법은 Chemotechnique MB Diagnostics AB사의 챔버(IQ Ultimate™)를 이용하였다. 챔버에 상기 각 피부외용제를 20 μL씩 취하여 24시간 동안 폐쇄 첩포를 실시하였다. 첩포 제거 30분 후와 24시간 후에 피부 반응을 관찰하고 평가 기준에 따라 피부 자극의 유무와 정도를 육안평가 하였으며, Frosch&Kligman (1979), CTFA guideline (2007)에 근거하여 표 2의 판정기준에 따라 피부 자극성 평가 지수를 산출한 후, 하기 실험식 1을 이용하여 각 피부외용제의 평균 피부자극 지수를 계산하였으며, Draize 방법을 응용하여 분류한 표 3의 피부 자극 지수를 기준으로 피부 자극도를 평가하였다.The patch method used a chamber (IQ Ultimate™) from Chemotechnique MB Diagnostics AB. 20 μL of each of the above skin external preparations was taken into the chamber and an occlusive patch was applied for 24 hours. 상기 표 4에서 대조군 및 시험군 모두, 자극 반응이 나타나지 않았다. 상기 기재된 식에 따라 계산하면 평균 반응도가 0.00이므로 술지게미 추출물(시험군)을 포함한 피부 외용제는 대조군 피부 외용제와 같이 비자극성 물질로 인체 피부에 안전한 물질로 판정하였다.In Table 4, no stimulation response occurred in both the control and test groups.
