[실험예 2 : 매자기 유산균 발효 추출물의 MMP-1 생성 억제능 측정][Experimental Example 2: Measurement of MMP-1 production inhibition ability of barberry lactic acid bacteria fermentation extract]
본 실험예에서는 실시예 1, 2의 MMP-1 생성 억제 효과를 확인하고자 하였다.In this experimental example, the effect of Examples 1 and 2 on suppressing MMP-1 production was sought to be confirmed.
인간 피부 섬유아 세포(HDFn)를 5×104 cells/well의 농도로 24 well-plate에 분주 한 후 37℃, 5% CO2인큐베이터에서 안정화시켰다. PBS로 2번 헹군 뒤, PBS가 있는 상태에서 UVA를 1 J/cm2로 조사하였다. 2% FBS가 첨가된 배지로 교환과 동시에 실시예 1, 실시예 2 및 실시예5를 농도별로 희석하여 처리하고 양성대조군으로 400 μM adenosine(주름기능성고시원료)을 사용하였다. 48시간 후에 상층액을 수집하고, 남은 세포에 MTT solution을 처리하여 세포독성을 확인하였다. 수집된 상층액을 이용하여 MMP-1 발현량을 확인하기 위해 R&D systems kit (DY901)를 사용하였다. MMP-1 ELISA assay 후 450 nm의 파장에서 흡광도를 측정하여 생성된 MMP-1양을 계산한 후, 대조군과 비교하여 MMP-1 발현 억제율(%)을 계산하였다. 그 결과를 도 3에 나타냈다.Human dermal fibroblasts (HDFn) were distributed in a 24 well-plate at a concentration of 5×10 4 cells/well and stabilized in an incubator at 37°C and 5% CO 2 .
