Abstract

본 발명의 분야

FLS는 효소 분해에 의해 활막 조직으로부터 분리되었다. 요컨대, 미세 수술용 가위로 조직을 1-mm 3블록으로 분리했다. 그런 다음 조직을 효소 분해를 위해 10% FBS가 보충된 DMEM에서 0.1% 콜라게나제 II형 효소와 함께 오비탈(orbital) 진탕 인큐베이터(200 rpm)에서 37℃에서 45분 동안 배양했다.
배양 시간 후, 튜브를 1 내지 2분 동안 격렬하게 와동(vortex)시켜 세포를 방출하고 100 ㎛ 메쉬 스트레이너(strainer)를 사용하여 여과하였다. 여액을 1200 rpm에서 5분 동안 원심분리하고, 세포를 10% FBS 및 100 ㎍/ml 스트렙토마이신 및 100 U/ml 페니실린이 보충된 DMEM에 재현탁하고, 가습 인큐베이터에서 37℃, 5% CO2 하에서 배양하였다. 항비멘틴 항체(PA5-27231, Thermo Fisher Scientific)를 사용한 세포 형태 및 면역세포화학적 염색을 기반으로 한 FLS 식별 후(도 1 및 2), 4 내지 7계대(passage)의 세포를 추가 연구에 사용했다.
실시예 2: WST-1 증식 검정: FLS 증식에 대한 상이한 샘플의 효과를 측정하기 위해 WST-1 검정을 사용하였다. FLS를 96-웰 플레이트에 5 × 103 세포/웰의 밀도로 씨딩하고 5% CO2하에 37℃에서 밤새 배양했다. 그 다음, 세포를 도 3 내지 6에 나타낸 바와 같이 상이한 농도의 샘플(AC3, C3, Cur, BDMC, DMC, BPS)로 72시간 동안 처리하였다. 처리 기간 후, 무혈청 DMEM 배양 배지에 1:10으로 희석된 100 μl WST-1 시약(5015944001, Sigma, USA)을 각 웰에 첨가하고, 37℃, 5% CO2 하에서 2시간 동안 배양하였다. 흡광도는 610 nm의 기준 파장으로 450 nm에서 측정되었다. AC3는 용량 의존적 방식으로 20 ㎍/mL 및 40 ㎍/mL(도 3 및 4)에서 FLS 증식을 더 잘 억제하는 것으로 나타났다. FLS 증식 억제에서 커큐미노이드 유사체에 대한 비교 연구는 20 ㎍/mL에서 BDMC > 커큐민 > DMC(도 5)의 경향을 보인 반면 BPS는 10%만 억제했으며 조합은 보스웰린(Boswellin) PS(BPS) 또는 개별적으로 사용되는 AC3보다 훨씬 효과적이었다(도 6).
실시예 3: 세포 주기 분석: FLS 세포(8 x 104 세포/웰)를 24-웰 플레이트에 씨딩하고 37℃, 5% CO2에서 밤새 배양했다. 24시간 동안 무혈청 배지에서 동기화(synchronization)한 후, 세포를 DMEM 10% FBS에서 24시간 동안 샘플의 존재 유무에 관계없이 처리했다. 처리 기간 후, 세포를 수집하고, 빙냉 PBS에 현탁시켰다. 원심분리 후 펠릿에 70% 빙냉 에탄올을 한 방울씩 적가하여 와동하면서 세포를 고정하고 4℃에서 30분 동안 보관했다. 세포를 어두운 곳에서 37℃에서 30분 동안 프로피듐 요오드(propidium iodide:PI) 용액(50 ㎍/ml)으로 염색했다. DNA 함량은 유세포 분석기(BD FACS Celesta 유세포 분석기)로 분석했다. 커큐미노이드 중 AC3, BDMC는 FLS 복제의 최대 억제를 보였고(도 7, 8, 12 및 14), FLS는 G0/G1 단계에서 정지되었다. 보스웰린 PS(BPS)는 80 ㎍/mL에서만 억제를 나타냈다(도 8, 도 16). 세포 주기 억제에 대한 조합(AC3 + BPS)의 효과는 도 9에 예시된 바와 같이 사용된 개별 샘플(AC3 및 BPS)보다 우수했다.
실시예 4: 염증 마커 측정: FLS 세포(8 x 104 세포/웰)를 24-웰 플레이트에 씨딩하고 37℃, 5% CO2에서 밤새 배양했다. 6시간 및 24시간 동안 DMEM 10% FBS에서 다양한 농도의 샘플 존재 여부에 관계없이 10 ng/ml 재조합 래트 TNF-α(Cat No.400-14, Peprotech, NJ, USA)로 세포를 유도하여 샘플의 항 염증 효과를 평가하였다. 커큐민은 염증 마커(IFN γ, CCL-5, MMP-3 및 카텝신, 표 2)를 억제하여 더 나은 항염증 활성을 보인 반면, BDMC는 TNF-α에서 가장 효과적이었고 가장 중요한 것은 AC3과 BPS의 조합이 가장 효과적이라는 것이다(표 2).
ELISA: 24시간의 배양 기간 후, 배양 상청액을 수집하고 ELISA 방법에 의해 염증 마커 IL6에 대해 평가하였다. 10 ㎍/mL(AC3) 및 40 ㎍/mL(BPS) 농도의 경우, 조합으로 사용했을 때 억제율은 AC3 및 BPS를 개별적으로 사용할 때 3% 및 1%에 비해 20%였다.

RT-PCR 발현 연구: 6시간 배양 후, Trizol 시약®(Ambion, Life Technologies)을 사용하여 제조업체의 지침에 따라 미처리된 세포와 처리된 세포의 총 세포 RNA를 분리한 후 리보뉴클레아제 없는 DNA 분해효소(RNase-free DNase) I 처리(ThermoFisher Scientific)를 수행하여 게놈 DNA를 제거했다. 메신저 RNA 품질 및 농도를 분광광도계(NanoDrop Lite, ThermoFisher Scientific)로 분석했다. 제조업체의 지침에 따라 Revert-aid First Strand cDNA 합성 키트(ThermoFisher Scientific)를 사용하여 총 RNA 1마이크로그램을 cDNA로 역전사하고 사용할 때까지 -80℃에서 보관했다.
이어서 반응 혼합물 20 μl를 내부 대조군으로서 유로핀 인디아(Eurofins India)에서 조달한 특별히 설계된 프라이머를 사용하여 cDNA 증폭을 위한 SYBR 그린 qPCR 마스터 믹스를 사용하여 PCR에 적용하고, 하우스 키핑 유전자 β 액틴(actin)을 각각의 반응과 함께 공동 증폭시켰다(표 2). PCR은 Light cycler 96(Roche Life Science)에서 수행하였으며, 유전자에 대한 PCR 조건은 95℃에서 10분 동안의 초기 변성 후 95℃에서 30초 동안 변성, 60℃에서 30초 동안 프라이머의 어닐링, 72℃에서 30초 동안 연장 및 최종으로 72℃에서 30초 동안 냉각으로 이루어진 35 주기였다(표 1).

실시예 5: 세포자연사 분석 - FLS 세포자연사를 RT-PCR 발현 연구로 평가하였다. 요컨대, FLS 세포(8 x 104 세포/웰)를 24-웰 플레이트에 씨딩하고 37℃, 5% CO2에서 밤새 배양했다. 24시간 동안 무혈청 배지에서 동기화(synchronization)한 후, 세포를 DMEM 10% FBS에서 24시간 동안 샘플의 존재 유무에 관계없이 처리했다. 처리 기간 후, 세포를 수집하고, 총 세포 RNA를 분리하고 앞서 언급한 바와 같이 RT-PCR 분석을 위해 처리하였다(표 3). BCl-2 및 Bax 단백질 계열은 세포자연사의 조절에 중심적인 역할을 한다. Bax/Bcl-2 비율은 세포자연사에 대한 세포 감수성을 결정하는 가변 저항으로 작용할 수 있으며, 세포자연사를 겪고 있는 세포는 Bax/Bcl-2 비율이 더 높다(도 19). 커큐미노이드 중에서 BDMC>DMC>커큐민, AC3 복합체는 대조군과 비교할 때 C3 복합체보다 더 우수하였다(도 19).

실시예 6: 이동-검정- FLS의 이동과 침범은 활막염과 뼈 파괴에 중요한 역할을 한다. FLS는 국소적으로 이동하며 혈류를 통해 먼 부위와 관절을 침범할 수도 있다. RA-FLS는 연골의 매트릭스 분해를 더욱 악화시키는 MMP를 분비하여 궁극적으로 뼈 침식을 초래한다.
FLS의 이동 가능성은 간단한 스크래치 분석 및 FLS 이동에 의한 상처 봉합을 평가하여 연구할 수 있다. 처리되지 않은 FLS는 이동하여 상처를 닫는 반면 처리된 FLS는 봉합이 지연된다(J Immunol March 1, 2014, 192 (5) 2063-2070). FLS 세포(5 × 104세포/웰)를 24-웰 플레이트에 씨딩하고, 37℃, 5% CO2에서 배양하여 약 80 내지 90%의 전면생장(confluence)에 도달한 다음, 200 μl 피펫 팁으로 긁어서 상처를 입혔다. 세포를 PBS로 세척하여 파편 및 부유 세포를 완전히 제거했다. 이어서 세포를 2% FBS를 함유하는 DMEM 배지에서 상이한 농도의 샘플과 함께 배양하였다. 대조군 샘플은 임의의 샘플 없이 세포 및 배양 배지를 함유하였다. Magvision 소프트웨어를 사용하여 단층 간극 폐쇄를 평가하여 세포 이동을 평가했다. 대조군 웰과 비교하여 상처 봉합 억제 백분율을 계산하였다. 각각 15 ㎍/mL의 커큐미노이드 중에서 BDMC(도 21)는 DMC(도 23), 커큐민(도 22)보다 나은 반면 AC3 복합체(도 24)는 C3 복합체(도 25)보다 우수했으며 AC3 + 보스웰린 PS(BPS)(도 29)는 대조군(도 20)에 비해 AC3(도 27) 및 BPS(도 28)보다 우수하였다.
실시예 7: 래트의 관절염 연구
마우스 모델(표 4) 콜라겐-유도된 관절염(CIA)은 널리 연구된 류마티스 관절염의 자가면역 모델이다. 이 모델에서 자가면역 관절염은 완전 프로인트 보조제(Freund's adjuvant)에 유화된 II형 콜라겐(CII)으로 면역화하여 유도된다. 이 동물들은 인간의 자가면역 질병인 류마티스 관절염과 몇 가지 임상적, 조직학적 및 면역학적 특징을 공유하는 자가면역 매개성 다발성 관절염이 발생한다. CII에 대한 면역 반응은 콜라겐 특이적 T 세포의 자극과 면역원(이종 CII) 및 자가항원(마우스 또는 래트 CII) 모두에 특이적인 높은 역가의 항체 생산을 특징으로 한다.

1:1 비율로 불완전 프로인트 보조제(FIA)로 유화될 2형 콜라겐(닭 흉골 연골, Sigma 카탈로그 번호: C9301)을 투여하여 관절염을 유발했다. 0 일째에, 200 μL(200 ㎍/동물)의 콜라겐-FIA 에멀젼을 래트의 꼬리 기저부에 피내 주사하였다(첫 번째 면역화). 부스터 주입을 위해 에멀젼은 위와 동일하게 준비되었고 7일째 콜라겐-FIA 에멀젼 100 μL(100 ㎍/동물)가 주입되었다. 0일부터 20일까지 테스트 샘플을 투여한다.
실시예 8: 관절염 점수: 관절염 점수의 표준화된 방법을 사용하여 네 발 모두의 종창 및 홍반의 정도를 평가했다. 0 - 징후 없음; 1 - 부종이 없는 발적; 2 - 가벼운 부종을 동반한 발적; 3 - 심한 부종을 동반한 발적; 4 - 발적, 심한 부종 및 움직임의 경직. AC3 및 BPS의 조합(도 32)은 각각 100 및 40 mg/kg의 농도에서 효과적이었다. 세레콕시브는 류마티스 관절염 대조군과 함께 양성 대조군으로 사용되었다.
실시예 9: 비장에서 Treg 및 Th17 분석, 비장 세포의 분리: 비장은 림프구 분리를 위해 무균 상태에서 콜라겐 유도 관절염 래트에서 얻었다. 비장은 100 ㎍/ml 스트렙토마이신 및 100 U/ml 페니실린을 함유하는 PBS로 2회 세척하고 미세 수술용 가위로 작은 조각으로 자르고, 주사기의 플런저 말단으로 균질화하고 100 μm 메쉬 스트레이너를 사용하여 여과하였다. 여액을 1200 rpm에서 5분 동안 원심분리하고, PBS로 1 내지 2회 세척한 후, 1x106 세포/ml를 10% FBS가 보충된 RPMI 배지에 현탁시켰다. 불균형은 Th17 세포를 감소시키고 T-세포(Treg)를 증가시킴으로써 발생되며(도 33 내지 35) Th17/Treg의 비율은 3.5에서 RA 대조군과 비교하여 바람직하게는 1, 또는 더 바람직하게는 0 내지 1이다(도 35).
유세포 분석기(FCM) 분석: Th17 및 Treg 세포를 평가하기 위해 분리된 림프구를 20 ng/mL 포르볼 미리스테이트 아세테이트(P8139, Sigma) 및 500 ng/mL 이오노마이신과 함께 제조업체가 제안한 농도로 모넨신(554724, BD)을 함유하는 단백질 수송 억제제인 BD GolgiStop으로 4시간 동안 자극했다.
자극 후, 세포를 수집하고 Th17 및 Treg 개체군을 평가하기 위해 2x105 세포를 다른 튜브에 분취하였다. 분석에 사용된 항체 희석액은 각 제조업체에서 권장한 대로 사용되었다. Th17 개체군을 평가하기 위해 세포를 처음에 FITC 마우스 항-래트 CD3(559975, BD pharmingen) 및 APC 마우스 항-래트 CD4(550057, BD pharmingen) 항체로 염색했다. Treg 개체군을 평가하기 위해 세포를 처음에 APC 마우스 항-래트 CD4(550057, BD pharmingen) 및 BV421 마우스 항-래트 CD25(565608, BD pharmingen) 항체로 염색했다. 각각의 표면 염색 항체를 첨가한 후, 튜브를 어두운 곳에서 4℃에서 30분 동안 배양했다. 표면 염색 후, 제조사의 지시에 따라 고정 및 투과화 용액(BD Cytofix/Cytoperm, 554722)을 사용하여 세포를 고정 및 가동화하고 사포닌 함유 완충액(BD Perm/Wash, 554723)에 재현탁시켰다. 고정 및 투과화 후, Th17 및 Treg 세포를 어두운 곳에서 4℃에서 30분 동안 각각 PE 접합된 항-마우스/래트 IL-17A(12-7177-81, eBioscience) 및 PE-접합된 항-FOXP3(12-5773-80, eBioscience) 항체와 함께 배양하였다. 이어서 세포를 사포닌 함유 완충액으로 2회 세척하고 유세포 분석 전에 염색 완충액에 재현탁시켰다. FCM은 BD FACSCelesta 시스템(BD Biosciences, Franklin Lakes, NJ, USA)에서 수행되었으며 FlowJo 소프트웨어를 사용하여 분석되었다.
실시예 10: 제형: 상기 조성물은 약제학적으로/영양보조적으로(nutraceutically) 허용되는 부형제, 보조제(adjuvants), 희석제, 안정제, 분산성 검, 생체이용률 향상제 또는 담체와 함께 제형화되어 정제, 캡슐, 시럽, 구미(gummies), 산제, 현탁액, 에멀젼(emulsions), 츄어블제(chewables), 캔디(candies) 또는 식용품(eatables)의 형태로 경구 투여된다.
관련 측면에서, 상기 생체이용률 향상제는 피페린(BioPerine®), 퀘르세틴, 마늘 추출물, 생강 추출물 및 나린진을 포함하는 그룹으로부터 선택된다. 또 다른 관련된 측면에서, 상기 안정화제는 로스마린산, 부틸화된 하이드록시아니솔, 부틸화된 하이드록시톨루엔, 메타중아황산 나트륨, 프로필 갈레이트, 시스테인, 아스코르브산 및 토코페롤로 이루어진 그룹으로부터 선택된다. 또 다른 관련 측면에서, 분산성 검은 한천, 알기네이트, 카라기난, 아라비아 검, 구아 검, 로커스트 콩 검, 곤약 검, 잔탄 검 및 펙틴으로 이루어진 그룹으로부터 선택된다.

- 제13항에 있어서, 상기 강화된 BDMC 커큐미노이드 조성물 및 BPS가 1-4:4-1의 비율로 조합되어 존재하는 것인, 조성물.
- 제13항에 있어서, 상기 불균형은 Th17 세포를 감소시키고 T-세포(Treg)를 증가시켜 Th17/Treg 비율을 감소시킴으로써 조절되는 것인, 조성물.
- 제13항에 있어서, 상기 조성물은 안정화제, 생체이용률 향상제 및 항산화제, 약제학적으로 또는 기능식품적으로(nutraceutically) 또는 약용화장품으로(cosmeceutically) 허용되는 부형제 및 향상제를 추가로 포함하고, 정제, 캡슐, 시럽, 구미, 산제, 현탁액, 에멀젼, 츄어블(chewable), 캔디 또는 식용품(eatables)의 형태로 경구 투여되는, 조성물.
- 포유동물에서 류마티스 관절염의 치료적 관리에 사용하기 위한 조성물로서, 상기 조성물은 20% w/w 이상으로 존재하는 강화된 비스데메톡시커큐민(BDMC)을 포함하는, 조성물.
- 제21항에 있어서, 상기 조성물은 20 내지 50% w/w 비스데메톡시커큐민, 10 내지 25% w/w 데메톡시커큐민, 및 30 내지 50% w/w 커큐민을 포함하며, 상기 조성물 중 총 커큐미노이드(curcuminoid)는 20 내지 95% w/w의 범위 내에 있는 것인, 조성물.

- 제21항에 있어서, 상기 조성물은 보스웰리아 세라타(Boswellia serrata)로부터의 보스웰산(B) 및 다당류(PS)를 추가로 포함하고, 상기 보스웰산 및 다당류(BPS)는 각각 35 내지 50% w/w 및 35 내지 45% w/w 범위로 존재하는 것인, 조성물.

- 제21항에 있어서, 상기 강화된 BDMC 커큐미노이드 조성물 및 BPS가 1-4:4-1의 비율로 조합되어 존재하는 것인, 조성물.
- 제21항에 있어서, 상기 포유동물의 류마티스 관절염의 관리는 섬유아세포 유사 활막세포의 증식 및 이동을 억제하고, T-헬퍼 17(Th17), T-세포(Treg)의 불균형을 조절하고, 염증 마커의 수준을 감소시키고, 세포자연사를 촉진하고, 관절염 점수를 개선함으로써 초래되는 것인, 조성물.
- 제21항에 있어서, 상기 조성물은 안정화제, 생체이용률 향상제 및 항산화제, 약제학적으로 또는 기능식품적으로 또는 약용화장품으로 허용되는 부형제 및 향상제를 추가로 포함하고, 정제, 캡슐, 시럽, 구미, 산제, 현탁액, 에멀젼, 츄어블(chewable), 캔디 또는 식용품(eatables)의 형태로 경구 투여되는, 조성물.
