Abstract
상기 조성물은 파골세포의 형성 또는 분화를 억제하고, 파골세포의 아폽토시스를 촉진하는 효과를 가지는 바, 이를 활용하여 파골세포 형성과 관련된 골 질환을 보다 효과적으로 예방 또는 치료할 수 있다.
Description
도 2는 GSK5182의 파골세포 형성 마커에 미치는 영향에 관한 것으로, 도 2A는 GSK5182 처리에 따른 파골세포 분화와 관련된 전사인자의 발현 수준을 측정한 그래프이며 (대조군(control)에 대해 *P < 0.05, **P < 0.001 이다), 도 2B 내지 2D는 이를 웨스턴 블로팅으로 확인한 결과이다.
도 3은 GSK5182의 RANKL 신호전달 케스케이드(cascade)에 대한 영향에 관한 것으로, 도 3A는 MAPKs (JNK, ERK, p38) 인산화 및 전체 형태를 면역 블로팅에 의해 검출한 결과이고, 도 3B는 인산화된 IκBα 항체로 세포 용해물을 면역 블로팅한 결과이며, 도 3C는 GSK5182 처리된 세포에서 IκBα의 면역 블로팅 분석한 결과이다. 또한, 도 3D는 RAW264.7 세포를 NF-κB 루시퍼라제 리포터 단독으로 형질감염한 것이고, 도 3E는 ERRγ 발현 플라스미드와 함께 NF-κB 루시퍼라제로 공동 형질감염시킨 것으로, 루시퍼라제 리포터 분석 시스템(dual-luciferase reporter assay system)으로 측정하였다 (**P < 0.001)
도 4는 GSK5182의 파골세포 세포사멸, 아폽토시스(apoptosis)에 대한 영향에 관한 것으로, 도 4A는 세포를 고정하고 TRAP로 염색한 이미지이며, 파골세포의 세포사멸은 별표(asterisks)로 표시하였다. 도 4B는 파골세포의 생존을 TRAP-양성 MNCs의 백분율로 나타낸 것이고 (**P < 0.001), 도 4C는 파골세포의 세포사멸을 ELISA로 측정한 것이며 (**P < 0.001), 도 4D는 전체 세포 용해물을 cleaved caspase (cas)-3 및 caspase-9에 대한 특이적 항체로 면역 블로팅한 것으로, β-Actin은 로딩 대조군으로 사용되었다.
