Abstract
본 발명은 항종양 아데노바이러스 및 이를 포함하는 항암용 조성물에 관한 것으로, 본 발명의 AR 및 mTOR의 발현을 동시에 억제하는 이중 가닥 shRNA를 코딩하는 발현 카세트와 hTERT 프로모터를 포함하는 아데노바이러스는 체내 면역 반응을 회피하여 암세포에 특이적으로 전달되어 전신 치료 효과가 있으며, 국소 전달이 가능하고, 선택성이 뛰어나며, 최소 침습 치료로도 현저한 항암 효과를 나타내, 다양한 암종에 항암용 조성물 또는 항암보조제로서 유용하게 이용될 수 있다.

암은 전세계적으로 가장 많은 사망자를 내는 질병 중 하나로, 혁신적인 암 치료제의 개발은 이에 대한 치료시 발생되는 의료비를 절감할 수 있음과 동시에 고부가가치를 창출할 수 있다. 또한, 2008년의 통계에 따르면, 기존항암제 내성을 극복 할 수 있는 분자 치료제는 주요 7개국(US, Japan, France, Germany, Italy, Spain, UK)에서 $17.5 billion을 차지했고, 2018년의 경우 약 $45 billion 정도의 market size를 차지하여, 2008년 대비 9.5%의 성장률을 보일 것이라 예측되고 있다. 암의 치료는 수술, 방사선치료, 화학요법, 생물학적 치료로 구분되는데, 이 중에 화학요법은 화학물질로서 암 세포의 증식을 억제하거나 죽이는 치료법으로 항암제에 의하여 나타나는 독성은 상당부분 정상세포에서도 나타나기 때문에 일정 정도의 독성을 나타내며, 항암제가 효과를 나타내다가도 일정 기간의 사용 후에는 효과가 상실되는 내성이 발생하기 때문에 암세포에 선택적으로 작용하고 내성이 생기지 않는 항암제의 개발이 절실하다 (암 정복의 현주소 Biowave 2004. 6(19)). 최근 암에 대한 분자유전정보의 확보를 통해 암의 분자적 특성을 표적으로 한 새로운 항암제의 개발이 진행되고 있으며, 암세포만이 가지고 있는 특징적인 분자적 표적(molecular target)을 겨냥하는 항암제들은 약제 내성이 생기지 않는다는 보고도 있다.
유전자의 발현을 억제하는 기술은 질병치료를 위한 치료제 개발 및 표적 검증에서 중요한 도구이다. 간섭 RNA(RNA interference, 이하 RNAi라고 한다)는 그 역할이 발견된 이후로, 다양한 종류의 포유동물세포(mammalian cell)에서 서열 특이적 mRNA에 작용한다는 사실이 밝혀졌다 (Silence of the transcripts: RNA interference in medicine. J Mol Med (2005) 83: 764773). RNAi는 21-25개의 뉴클레오타이드 크기의 이중나선 구조를 가진 작은 간섭 리보핵산 짧은 간섭 RNA (small interfering RNA, 이하 siRNA라고 한다)이 상보적인 서열을 가지는 전사체(mRNA transcript)에 특이적으로 결합하여 해당 전사체를 분해함으로써 특정 단백질의 발현을 억제하는 현상이다. 세포 내에서는 RNA 이중가닥이 Dicer라는 엔도뉴클라아제(endonuclease)에 의해 프로세싱되어 21 내지 23개의 이중가닥(base pair,bp)의 siRNA로 변환되며, siRNA 는 RISC(RNA-induced silencing complex)에 결합하여 가이드(안티센스) 가닥이 타겟 mRNA를 인식하여 분해하는 과정을 통해 타겟 유전자의 발현을 서열 특이적으로 저해한다 (NUCLEIC-ACID THERAPEUTICS: BASIC PRINCIPLES AND RECENT APPLICATIONS. Nature Reviews Drug Discovery. 2002. 1, 503-514). 베르트랑(Bertrand) 연구진에 따르면 동일한 타겟 유전자에 대한 siRNA가 안티센스 올리고뉴클레오티드(Antisense oligonucleotide, ASO)에 비하여 생체 내/외(in vitro 및 in vivo)에서 mRNA 발현의 저해효과가 뛰어나고, 해당 효과가 오랫동안 지속되는 효과를 포함하는 것으로 밝혀졌다 (Comparison of antisense oligonucleotides and siRNAs in cell culture and in vivo. Biochem. Biophys. Res.Commun. 2002. 296: 1000-1004). siRNA를 포함하는 RNAi 기술 기반 치료제 시장은 향후 세계 시장규모가 2020년경에 총 12조원 이상을 형성하는 것으로 분석되었으며, 해당 기술을 적용할 수 있는 대상이 획기적으로 확대되어 기존의 항체, 화합물 기반 의약품으로 치료하기 어려운 질병을 치료할 수 있는 차세대 유전자 치료기술로 평가되고 있다. 또한 siRNA의 작용 기작은 타겟 mRNA와 상보적으로 결합하여 서열 특이적으로 타겟 유전자의 발현을 조절하기 때문에, 기존의 항체 기반 의약품이나 화학물질(small molecule drug)이 특정한 단백질 표적에 최적화되기까지 오랜 동안의 개발 기간 및 개발 비용이 소요되는 것에 비하여, 적용할 수 있는 대상이 획기적으로 확대될 수 있고, 개발 기간이 단축되면서, 의약화가 불가능한 표적 물질을 포함한 모든 단백질 표적에 대하여 최적화된 리드 화합물을 개발할 수 있다는 장점을 가진다 (Progress Towards in Vivo Use of siRNAs. MOLECULAR THERAPY. 2006 13(4):664-670). 이에, 최근 이 리보핵산 매개 간섭현상이 기존의 화학 합성 의약 개발에서 발생되는 문제의 해결책을 제시하면서 전사체 수준에서 특정 단백질의 발현을 선택적으로 억제하여 각종 질병 치료제, 특히 종양 치료제 개발에 이용하려는 연구가 진행되고 있다. 또한, siRNA 치료제는 기존 항암제와 달리 표적이 명확하여 부작용이 예측 가능하다는 장점이 있으나, 이러한 표적 특이성은 다양한 유전자의 문제에 의해 발생하는 질병인 종양의 경우, 오히려 이러한 표적 특이성은 치료 효과가 높지 않은 원인이 되기도 한다.
본 발명의 목적은 항종양 아데노바이러스를 제공하는 것이다.
아울러, 본 발명의 목적은 암 치료용 조성물을 제공하는 것이다.
상기 과제를 해결하기 위하여, 본 발명은 AR을 표적서열로 하는 염기 서열 및 mTOR를 표적서열로 하는 염기 서열을 포함하는 항종양 아데노바이러스를 제공한다.
아울러, 본 발명은 상기 항종양 아데노바이러스를 포함하는 암 치료용 조성물을 제공한다.
본 발명에 따르면, 본 발명의 AR 및 mTOR의 발현을 동시에 억제하는 이중 가닥 siRNA를 포함하는 shRNA를 코딩하는 발현 카세트와 hTERT 프로모터를 포함하는 아데노바이러스는 체내 면역 반응을 회피하여 암세포에 특이적으로 전달되어 전신 치료 효과가 있으며, 국소 전달이 가능하고, 선택성이 뛰어나며, 최소 침습 치료로도 현저한 항암 효과를 나타내, 다양한 암종에 항암용 조성물 또는 항암보조제로서 유용하게 이용될 수 있다.

상기 암은 대장암, 유방암, 자궁암, 자궁경부암, 난소암, 전립선암, 뇌종양, 두경부암종, 흑색종, 골수종, 백혈병, 림프종, 위암, 폐암, 췌장암, 비소세포성폐암, 간암, 식도암, 소장암, 항문부근암, 나팔관암종, 자궁내막암종, 질암종, 음문암종, 호지킨병, 방광암, 신장암, 수뇨관암, 신장세포암종, 신장골반암종, 골암, 피부암, 두부암, 경부암, 피부흑색종, 안구내흑색종, 내분비선암, 갑상선암, 부갑상선암, 부신암, 연조직육종, 요도암, 음경암, 중추신경계(central nervous system; CNS) 종양, 1차 CNS 림프종, 척수종양, 다형성교모세포종 및 뇌하수체선종으로 구성된 군으로부터 선택되는 어느 하나일 수 있다.
본 발명에 있어서, 용어, "프로모터"란, RNA 중합효소에 대한 결합 부위를 포함하고 프로모터 다운스트림 (downstream) 유전자의 mRNA로의 전사 개시 활성을 가지는, 암호화 영역의 상위(upstream)의 비해독된 핵산 서열을 말한다. 본 발명의 발현 카세트에 있어서, 상기 프로모터는 shRNA의 발현을 개시할 수 있는 어떤 프로모터도 가능하다. 구체적으로, 본 발명의 프로모터로는 모든 시간대에 상시적으로 목적 유전자의 발현을 유도하는 프로모터(constitutive promoter) 또는 특정한 위치, 시기에 목적 유전자의 발현을 유도하는 프로모터 (inducible promoter)를 사용할 수 있으며, 그 예로는 U6 프로모터, H1 프로모터, CMV (cytomegalovirus) 프로모터, SV40 프로모터, CAG 프로모터 (Hitoshi Niwa et al., Gene, 108:193-199, 1991), CaMV 35S 프로모터 (Odell et al.,Nature 313:810-812, 1985), Rsyn7 프로모터 (미국특허출원 제08/991,601호), 라이스 액틴 (rice actin) 프로모터 (McElroy et al., Plant Cell 2:163-171, 1990), 유비퀴틴 프로모터 (Christensen et al., Plant Mol. Biol. 12:619-632, 1989), ALS 프로모터(미국 특허출원 제08/409,297) 등이 있다. 이외에도 미국특허 제5,608,149; 제5,608,144호 제5,604,121호 제5,569,597호 제5,466,785호, 제5,399,680호 제5,268,463호 및 제5,608,142호 등에 개시된 프로모터 등 당업자에게 자명한 공지의 모든 프로모터를 사용할 수 있으며, 이에 제한되는 것은 아니다. 바람직하게는 본 발명의 프로모터는 U6 프로모터, HI 프로모터, CMV 프로모터일 수 있으며, 본 발명의 바람직한 일 구현예에 따르면 U6 프로모터를 사용할 수 있다.
본 발명의 조성물에는 보조제(adjuvant)를 추가로 포함할 수 있다. 상기 보조제는 당해 기술분야에 알려진 것이라면 어느 것이나 제한 없이 사용할 수 있으나, 예를 들어 프로인트(Freund)의 완전 보조제 또는 불완전 보조제를 더 포함하여 그 효과를 증가시킬 수 있다.
본 발명에 따른 조성물은 유효성분을 약학적으로 허용된 담체에 혼입시킨 형태로 제조될 수 있다. 여기서, 약학적으로 허용된 담체는 제약 분야에서 통상 사용되는 담체, 부형제 및 희석제를 포함한다. 본 발명의 조성물에 이용할 수 있는 약학적으로 허용된 담체는 이들로 제한되는 것은 아니지만, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로스, 메틸 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
본 발명의 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀전, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 또는 멸균 주사용액의 형태로 제형화하여 사용될 수 있다.
제제화할 경우에는 통상 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 그러한 고형 제제는 유효성분에 적어도 하나 이상의 부형제, 예를 들면 전분, 칼슘 카르보네이트, 수크로스, 락토오스, 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용될 수 있다. 경구투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 일반적으로 사용되는 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수용성용제, 현탁제, 유제, 동결건조 제제 및 좌제가 포함된다. 비수용성용제, 현탁제로는 프로필렌 글리콜, 폴리에틸렌 글리콜, 올리브유와 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
본 발명에 따른 성물은 개체에 다양한 경로로 투여될 수 있다. 투여의 모든 방식이 예상될 수 있는데, 예를 들면 경구, 정맥, 근육, 피하, 복강내 주사에 의해 투여될 수 있다.
본 발명에 따른 약학 조성물의 투여량은 개체의 연령, 체중, 성별, 신체 상태 등을 고려하여 선택된다. 상기 약학 조성물 중 포함되는 단일 도메인 항체 의 농도는 대상에 따라 다양하게 선택할 수 있음은 자명하며, 바람직하게는 약학 조성물에 0.01 ~ 5,000 ㎍/ml의 농도로 포함되는 것이다. 그 농도가 0.01 ㎍/ml 미만일 경우에는 약학 활성이 나타나지 않을 수 있고, 5,000 ㎍/ml를 초과할 경우에는 인체에 독성을 나타낼 수 있다.
본 발명의 조성물은 암 및 이의 합병증의 예방 또는 치료에 이용될 수 있으며, 항암보조제로도 사용될 수 있다.
또한 본 발명은 약학적으로 유효한 양의 본 발명의 조성물를 개체에 투여하는 단계를 포함하는 암의 예방 및 치료방법을 제공한다.
본 발명의 조성물은 치료적 유효량 또는 약학으로 유효한 양으로 투여한다. 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
하기의 실시예를 통하여 본 발명을 보다 상세하게 설명한다. 그러나 하기 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐 이에 의해 본 발명이 한정되는 것은 아니다.
실시예 1. AR 및 mTOR 이중 표적 siRNA 제작
AR(androgen receptor) 및 mTOR(mammalian target of rapamycin)을 동시에 억제할 수 있는 이중 표적 siRNA(double strand) 세트를 하기 표 1의 서열로 제작하였다 (Bioneer, Daejeon, Korea). 구체적으로, 서열번호 1 및 2로 이루어진 20mer의 siRNA 세트 1은 18mer가 서로 상보적이며, 서열번호 3 및 4로 이루어진 19mer의 siRNA 세트 2는 17mer가 서로 상보적이고, 서열번호 5 및 6으로 이루어진 18mer의 siRNA 세트 3은 16mer가 서로 상보적이며, 서열번호 7 및 8로 이루어진 17mer의 siRNA 세트 4는 15mer가 서로 상보적이고, 서열번호 9 및 10으로 이루어진 19mer의 siRNA 세트 5는 15mer가 서로 상보적이며, 서열번호 11 및 12로 이루어진 18mer의 siRNA 세트 6은 14mer가 서로 상보적이고, 서열번호 13 및 14로 이루어진 17mer의 siRNA 세트 7은 13mer가 서로 상보적이며, 서열번호 15 및 16으로 이루어진 23mer의 siRNA 세트 8은 19mer가 서로 상보적이고, 서열번호 17 및 18로 이루어진 22mer의 siRNA 세트 9는 18mer가 서로 상보적이며, 서열번호 19 및 20으로 이루어진 22mer의 siRNA 세트 10은 18mer가 서로 상보적이고, 서열번호 21 및 22로 이루어진 21mer의 siRNA 세트 11은 17mer가 서로 상보적이며, 서열번호 23 및 24로 이루어진 20mer의 siRNA 세트 12는 16mer가 서로 상보적이고, 및 서열번호 25 및 26으로 이루어진 21mer의 siRNA 세트 13은 17mer가 서로 상보적이다. 하기 표 1의 서열번호 1, 3, 5, 7, 9, 11, 13, 15, 17, 19, 21, 23 또는 25의 siRNA (Antisense AR)가 AR의 mRNA에 상보적으로 결합하며, 서열번호 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24 또는 26의 siRNA (Antisense mTOR)가 mTOR의 mRNA에 상보적으로 결합한다. 따라서, 본 발명의 siRNA 세트 1 내지 13은 AR 및 mTOR 유전자의 발현을 동시에 감소시킨다.

실시예 2. AR 및 mTOR 이중 표적 shRNA 발현 카세트 제작
상기 실시예에서 제작한 siRNA를 세포 내에서 발현할 수 있게 하기 위하여, shRNA를 발현하는 발현 카세트를 제작하였다. 구체적으로, 5'에서 3' 방향으로 상기 표 1의 siRNA 세트의 센스 가닥의 3'에 TTGGATCCAA (TTGGATCCAA 루프) 또는 TTCAAGAGAG (TTCAAGAGAG 루프), 안티센스 가닥 및 TT를 연결하여 shRNA를 코딩하는 DNA 서열을 제작하였다. 대표적으로, 세트 1의 이중 표적 siRNA (서열번호 1 및 2) siRNA 이중 가닥의 서열과 루프 서열을 포함하는 shRNA 발현 카세트들 (TTGGATCCAA 루프 shRNA 및 TTCAAGAGAG 루프 shRNA)을 표 2에 표시하고 (siRNA는 대문자로 추가 서열은 소문자로 표시) 나머지 shRNA들을 서열목록에 기재하였다 (서열번호 28 내지 53). 제작한 shRNA 발현 카세트들을 각각 pE3.1 벡터 (도 1)의 제한효소 PstⅠ 및 EcoRⅤ 절단 위치에 U6 프로모터 (서열번호 27) 이후에 오도록 배치하여, AR 및 mTOR를 표적으로 하는 이중 표적 siRNA를 포함하는 두 종의 shRNA를 세포 내에서 발현하는 재조합 발현 벡터를 제작하였다.

실시예 3. 이중 표적 siRNA 및 shRNA의 유전자 발현 억제 효과 확인
3-1. 이중 표적 siRNA의 유전자 발현 억제 효과
12웰 플레이트에 PC3 세포주, h460 및 A549 세포주를 각각 분주한 뒤, 세포 밀도가 50%가 될 때까지 10% FBS (Hyclone 사)가 첨가된 RPMI 배지 (Hyclone 사)에서 37℃, 5% CO2 조건으로 배양하였다. 그 후, 상기 세포가 배양된 웰에 상기 실시예 1에서 제작한 이중 표적 siRNA 세트 1 내지 13을 웰당 80 pmole씩 3㎕의 리포펙타민(lipofectamine)3000 (Invitrogen, Carlsbad, CA, USA)으로 트랜스펙션하여 AR 및 mTOR를 동시에 낙다운하였다 (세트 1은 PC3 세포주 및 h460에, 세트 2 내지 13은 A549 세포주에). 더불어, 이에 대한 양성 대조군으로서 시판되는 하기 표 3에 기재된 AR에 대한 siRNA와 mTOR에 대한 siRNA를 각각 트랜스펙션하였다. 트랜스펙션 48시간 후, 세포를 파쇄하여 GeneJET RNA Purification Kit (Invitrogen)로 총 RNA를 추출하였다. 추출한 총 RNA를 주형으로 사용하여 RT-PCR 반응을 통해 cDNA로 역전사 한 뒤, q-PCR 반응을 통해 각각의 siRNA와 본 발명의 이중 표적 siRNA 세트 1(si-AT1)에 의한 AR 및 mTOR의 mRNA 발현량을 확인하였다. mRNA 발현량을 확인하기 위해, AR 또는 mTOR에 대한 프라이머 세트와 반응 혼합물[10X reaction Buffer 2 ㎕, HQ Buffer 2 ㎕, dNTP 1.6㎕, Primer(F, R, 10 pmole/ul) 각각 1㎕, Template(500 ng) 2㎕, Taq 0.2㎕, DW 10.2㎕, Total vol. 20㎕]을 이용하였다. PCR 조건으로 [95℃에서 2분, 95℃에서 20초로 30 사이클, 60℃에서 10초 및 72℃에서 30-60초, 72℃에서 5분] 낙다운한 세포 파쇄물에서의 AR 및 mTOR의 mRNA를 cDNA로 변환하였다. 또한, 역전사된 cDNA를 주형으로 이용하여, 반응 혼합물을 [Template(RT-PCR product) 6㎕, Taqman probe 3㎕, 10X reaction Buffer 6㎕, HQ Buffer 6㎕, dNTP 4.8㎕, Taq 0.6㎕, DW 10.2㎕, Total vol.60㎕] 준비하고 qPCR을 수행하였다[95℃에서 10분, 95℃에서 15초 및 로60℃에서 1분당 40사이클]. 참고로, 사용한 프로브는 AR(Thermo, Hs00171172_m1), mTOR(Thermo, Hs00234508_m1), GAPDH(Thermo, Hs02786624_g1)이며, QS3 장비를 이용하여 수행하였다. 모든 반응은 3회씩 반복수행되고 이들의 평균값이 취해졌다. 이렇게 얻은 결과들은 하우스키핑 유전자인 GAPDH의 mRNA 값에 대해 정규화하였다.

그 결과, PC3 세포 및 h460 세포주 모두에서 본 발명의 이중 표적 siRNA 세트 1 및 2 내지 13에 의해 AR 및 mTOR의 발현이 모두 감소하였으며 (도 2 및 3), 감소 정도는 각각의 siRNA에 의한 효과와 유사하거나 더 우수하게 나타났다. 이를 통해, 본 발명의 이중 표적 siRNA가 두 유전자의 발현을 동시에 효과적으로 억제할 수 있음을 알 수 있었다.
실시예 4. 이중 표적 shRNA 코딩 아데노바이러스 제작
hTERT 프로모터 (서열번호 54)-E1A (서열번호 55)-IRES (서열번호 56)-E1B 서열 (서열번호 57) (hTERT-E1A-IRES-E1B 전체 서열: 서열번호 58)을 아데노바이러스 벡터의 SpeⅠ과 ScaⅠ사이에 삽입한 후, U6 프로모터와 상기 실시예에서 제작한 AR 및 mTOR 이중 표적 shRNA 코딩 서열 (U6 프로모터+AR 및 mTOR 이중 표적 shRNA 코딩 서열: 서열번호 59)을 각각 E3 영역(region)에 SpeⅠ사이에 삽입함으로써 hTERT 프로모터 및 이중 표적 shRNA를 발현하고, 감염성이 있는 재조합 아데노바이러스를 제작하였다 (AR 및 mTOR 이중 표적 shRNA 코딩 (발현) 아데노바이러스: CA103) (도 5 참조). 아울러, 대조군으로 hTERT만 삽입한 재조합 아데노바이러스도 제작하였다 (CA10G). 그 후, 제작된 아데노바이러스 벡터의 서열을 분석하여, 이상이 없을 경우 PacⅠ제한효소를 이용하여 바이러스 유전체를 선형화하여 293A 세포에 CaCl2 법을 이용하여 형질도입하여 각 바이러스를 생산하였다.
실시예 5. 이중 표적 shRNA 코딩 아데노바이러스의 유전자 발현 억제 확인
5-1. in vitro
상기 실시예 4에서 제작한 재조합 아데노바이러스 CA103의 표적 유전자 AR 및 mTOR에 대한 발현 억제 효과를 확인하였다. 구체적으로, 인간 전립선암 세포주인 LNcap, C42B 및 22Rv1를 각각 1X105/well로 12 웰 플레이트에 분주한 뒤 1시간 후에 CA10G 및 CA103을 각각의 웰에 2 또는 5 MOI가 되도록 처리하고 72시간 뒤에 RNA prep kit (Takara, 9767A)를 이용하여 RNA prep을 수행하였다. 그 후, Nanodirp을 이용하여 RNA 정량하고 RT 프리믹스 (intron, 25081)를 이용하여 튜브 당 400 ng/20 ul를 넣고 프리믹스(premix) 내용물과 잘 섞어 준 뒤 PCR 기기를 이용하여 45℃에서 1hr 및 95℃에서 5 분간 반응시켜 cDNA을 합성하였다. 합성된 cDNA 2 ul를 주형으로 이용하여 실험군에 해당하는 PCR 혼합물 (총 부피, 20 ul)을 만들었다 (주형 2 ul, 정방향 프라이머 0.5 ul (10 pmole/ul), 역방향 프라이머 0.5 ul (10 pmole/ul), 10 ul 2X master mix(Bioline, BIO-94005) 및 7 ul DW). 만든 PCR 혼합물을 볼텍싱하여 잘 섞고 원심분리한 후 qPCR 기기 (Applied Biosystems, QS3)에서 95℃에서 5분, 95℃에서 10초 및 60℃에서 30초를 40사이클 반응시켰다. 결과는 qPCR 기기에 있는 프로그램을 이용하여 분석하였다.
그 결과, LNcap 세포주에서, hTERT 프로모터와 AR 및 mTOR 이중 표적 shRNA를 코딩 및 발현하는 본 발명의 재조합 아데노바이러스 CA103은 hTERT 프로모터만 포함하는 재조합 아데노바이러스 CA10G에 비해 현저하게 AR 및 mTOR 유전자의 발현을 억제하는 것으로 나타났다 (도 6). 또한, C42B 및 22Rv1 세포주에서도 본 발명의 재조합 아데노바이러스 CA103가 CA10G에 비해 현저하게 AR 및 mTOR 유전자의 발현을 억제하는 것으로 나타났다 (도 7).
5-2. in vivo
balb c nu/nu 마우스에 전립선암 세포주 (22Rv-1)를 피하 이식하여 전립선암 마우스 모델을 구축한 뒤, 상기 실시예 4에서 제작한 재조합 아데노바이러스 CA10G 및 CA103를 각각 종양내 1회 직접 투여 (2×108 pfu/spot, 3 times)하고, 21일 후 종양을 적출하여 종양 내 AR 및 mTOR 유전자의 발현 정도를 웨스턴 블롯 분석 및 IHC 분석으로 확인하였다. 웨스턴 블롯 분석 결과, CA103를 투여한 군에서 대조군 및 CA10G 투여군에 비해 mTOR 및 AR의 발현이 감소한 것으로 나타났으며, IHC 분석 결과에서도 CA103를 투여한 군에서 mTOR 및 AR의 형광 발현이 대조군 및 CA10G 투여군에 비해 70~90% 이상 감소한 것으로 나타나, CA103이 종양 내 표적 유전자인 mTOR 및 AR의 발현을 효과적으로 억제하는 것을 확인하였다 (도 8)
실시예 6. 이중 표적 shRNA 코딩 아데노바이러스의 항암 효과 확인
6-1.
in vitro
상기 실시예 4에서 제작한 재조합 아데노바이러스 CA10G 및 CA103의 암세포 사멸 효과를 비교하였다. 구체적으로, LNcap, C42B 및 22Rv1 세포주를 각각 5X103/well로 96 웰 플레이트에 각각 분주한 뒤 1시간 후에 CA10G 및 CA103을 각각의 웰에 1, 2, 5, 10, 20, 40 또는 50 MOI가 되도록 처리한 후 72시간 뒤에 MTT 시약을 넣고 37℃에서 3시간 동안 인큐베이션하였다. 3시간 후 각 웰에서 배지를 제거하고 DMSO를 100 ul씩 넣은 뒤, 바로 마이크로플레이트 리더기를 사용해 파장 540 nm에서 흡광도를 측정하여 MTT 분석을 수행하였다.
그 결과, LNcap 세포주의 경우 CA10G에 비해 본 발명의 CA103이 현저하게 암세포들을 사멸하는 것을 확인할 수 있었으며 (도 9), C42B 및 22Rv1 세포주의 경우에도 본 발명의 CA103이 현저하게 암세포들을 사멸하는 것을 확인할 수 있었다 (도 10).
6-2.
in vivo
balb c nu/nu 마우스에 전립선암 세포주 (22Rv-1)를 피하 이식하여 전립선암 마우스 모델을 구축한 뒤, 상기 실시예 4에서 제작한 재조합 아데노바이러스 CA10G 및 CA103를 각각 종양내 직접 투여 (2×108 pfu/spot, 3 times)하여 그 생장을 관찰한 결과, 무처리 대조군 (buffer 처리군) 및 벡터 대조군 (CA10G 처리군)에 비해 CA103를 처리한 군에서 종양의 부피 및 무게가 크게 감소하는 것을 확인할 수 있었다 (도 11).

- 인간 텔로미어 프로모터(hTERT); 및
AR(androgen receptor)을 표적서열로 하는 염기 서열 및 mTOR(mammalian target of rapamycin)를 표적서열로 하는 염기 서열을 포함하는 발현 카세트를 포함하는 항종양 아데노바이러스에 있어서,
상기 AR을 표적서열로 하는 염기 서열과 mTOR을 표적서열로 하는 염기서열이 발현되는 경우, 각각 발현된 서열이 부분적 또는 100% 상보적으로 결합하여 이중가닥을 형성하는 것인, 항종양 아데노바이러스. - 제 1항에 있어서, 인간 텔로미어 프로모터는 아데노바이러스의 내재적 유전자와 작동 가능하게 연결된, 항종양 아데노바이러스.
- 제 2항에 있어서, 아데노바이러스의 내재적 유전자는 5'ITR-C1-C2-C3-C4-C5 3'ITR의 구조를 가지며;
상기 C1은 E1A, E1B 또는 E1A-E1B를 포함하고;
상기 C2는 E2B-L1-L2-L3-E2A-L4를 포함하며;
상기 C3는 E3가 포함하지 않거나 E3를 포함하고;
상기 C4는 L5를 포함하며; 및
상기 C5는 E4를 포함하지 않거나 E4를 포함하는, 항종양 아데노바이러스. - 제 1항에 있어서, 발현 카세트가 아데노바이러스의 내재적 유전자의 C3 부위에 위치하는, 항종양 아데노바이러스.
- 제 1항에 있어서, hTERT 프로모터가 아데노바이러스의 내재적 유전자의 E1A 및 E1B와 작동가능하게 연결된, 항종양 아데노바이러스.
- 제 5항에 있어서, E1A 및 E1B 사이에 IRES 서열이 추가로 포함된, 항종양 아데노바이러스.
- 제 1항에 있어서, 발현 카세트는 shRNA를 코딩하는, 항종양 아데노바이러스.
- 제 7항에 있어서, shRNA는 동시에 AR 및 mTOR를 억제하는, 항종양 아데노바이러스.
- 제 1항에 있어서, AR을 표적서열로 하는 염기 서열은 mTOR를 표적서열로 하는 염기 서열의 역상보 서열과 60% 이상의 상보성을 갖는 염기서열을 포함하는, 항종양 아데노바이러스.
- 제 1항에 있어서, mTOR를 표적서열로 하는 염기 서열은 AR을 표적서열로 하는 염기 서열의 역상보 서열과 60% 이상의 상보성을 갖는 염기서열을 포함하는, 항종양 아데노바이러스.
- 제 1항에 있어서, 발현 카세트는 AR을 표적서열로 하는 염기 서열, 헤어핀 구조를 이룰 수 있는 루프 서열, 및 mTOR를 표적서열로 하는 염기 서열을 순차적으로 코딩하는 염기서열을 포함하는, 항종양 아데노바이러스.
- 제 1항에 있어서, 발현 카세트는 U6 프로모터에 의하여 발현이 조절되는, 항종양 아데노바이러스.

- 제 1항에 있어서, 혈청형이 5 타입인, 항종양 아데노바이러스.
- 제 1항에 있어서, 항종양 바이러스는 야생형 아데노바이러스에 비해 종양살상능이 높은, 항종양 아데노바이러스.
