Abstract
본 발명은 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머에 관한 것으로, 이러한 압타머를 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질과 함께 결합한 복합체 등에 응용에 관한 것으로, 본 발명의 압타머 및 그 압타머와 결합한 복합체는 PET(positron emission tomography) 방사선 조영제로 사용하여 알츠하이머병이나 파킨슨병 등을 조기에 진단할 수 있고, BBB를 통과 뇌실질 내에 타겟 물질의 분포를 PET을 통해 조기에 조영할 수 있는 방사선 조영 물질로 사용될 수 있으며, 퇴행성 뇌질환 치료제로 사용될 수 있다.
Description
본 발명은 혈뇌장벽을 통과하는 압타머 및 그것의 응용에 관한 것이다.
알츠하이머병의 경우 뇌실질내 신경세포들 사이에 아밀로이드베타와 TDP-43의 응집이, 파킨슨병은 신경세포내에 Lewy Body(LBs) 라 불리는 단백질의 응집체가 병리적 소견에서 나타나며, LB의 주 성분이 알파시뉴크레인이라 불리는 단백질로 알려져 있다. 이들 타겟 물질의 뇌내 응집은 각 질환의 주요 증세가 나타나기 전부터 일어나는 것으로 알려져 있으며, 이러한 비정상으로 응집된 단백질은 신경세포의 세포사멸을 일으켜 질병을 유발하게 된다.
따라서 뇌질환 관련 타겟 물질의 응집을 조기에 확인하여, 응집을 막는다면 뇌질환 발병률을 획기적으로 줄일 수 있을 것이라 생각된다.
압타머는 "chemical antibody"라고도 불리우는 단일 가닥의 DNA나 RNA (ssDNA 혹은 ssRNA)로, 물질내 염기들의 상보결합을 통해 다양한 모양의 삼차구조를 만들어내어 단백질, 펩타이드 등에 선택적으로 결합하는 물질을 일컫는다. 항체처럼 타겟물질에 대한 선택적 특이성이 높으면서도 사이즈는 1/10정도로 작으며 합성을 통해 생산이 가능하므로 항체처럼 복잡한 세포 배양과 정제 과정을 피할 수 있고, 생화학적 특성을 균일하게 유지 시킬 수 있다는 큰 장점이 있다. 또한 DNA나 RNA는 이미 체내에 존재하는 물질로 독성문제를 해결할 수 있다.
압타머는 뇌질환 타겟 물질에 specific하게 결합할 수 있고, 최근에는 a-syn에 specific하게 결합하는 압타머가 α-synuclein의 응집을 막고 분해를 촉진 시킬 수 있음이 보고되었다 (Yuan Zheng et.al, (2018) Novel DNA Aptamers for Parkinson's Disease Treatment Inhibit α-synuclein Aggregation and Facilitate its Degradation. Molecular Therapy 11:228-242). 그러므로 압타머를 이용하면 타겟 물질에 결합하여 조기 진단에 사용할 수 있을 뿐만 아니라 질병을 예방하고 치료할 수 있어 병을 진단함과 동시에 치료가 가능한 Theragnosis(therapy + diagnosis) 기술로의 활용이 기대된다.
하지만 뇌실질 내에 존재하는 타겟 물질에 결합해서 효능을 나타내기 위해서는 혈액을 통해 투여된 압타머가 BBB를 통과하여야 한다. BBB를 통과하여 뇌실질로 물질을 전달하는 가장 효과적인 방법은 수용체 결합을 통한 transcytosis 이다. Transcytosis를 통해 뇌실질에 물질을 전달한다고 알려져 있는 수용체는 Transferrin receptor, Insulin receptor, Low density lipoprotein receptor 등이 있고, 특히 Transferrin receptor를 타겟하여 뇌실질에 약물을 전달하려는 연구가 많이 진행되고 있다. 그 예로, Transferrin receptor에 결합하는 항체가 BBB를 효과적으로 통과할 수 있음이 보고되었다 (Bell RD, Ehlers MD (2014) Breaching the blood-brain barrier for drug delivery. Neuron 81:1-3). 또한 Transferrin receptor의 세포바깥쪽 부분 (Extracellular domain, ECD)에 결합하는 압타머를 통해 lysosomal 효소를 세포 안으로 효과적으로 전달할 수 있음이 보고되었다 (Chen CH, et.al, (2008) Aptamer-based endocytosis of a lysosomal enzyme. Proc Natl Acad Sci U S A 105:15908-15913).
그렇지만 이러한 receptor들이 뇌혈관에만 존재하는 것이 아니기 때문에 receptor를 타겟으로 하여 뇌실질에 전달되는 약물이 필연적으로 간, 심장, 폐, 신장 등과 같은 조직에 상당량 전달되어 예상치 못한 부작용이 야기될 가능성이 있다. 이와 같은 문제점을 해결하기 위해 뇌혈관만 특이적으로 통과하는 물질을 찾는 연구가 진행되고 있다. 그 예로, 단일사슬 항체 라이브러리를 이용한 스크리닝을 통해 BBB만 선택적으로 통과하는 항체가 보고 되었다 (Angela, et.al, (2014) Identifying blood-brain barrier-selective single-chain antibody fragments. Biotechnol J 9(5):664-74).
본 발명은 상기의 필요성에 의하여 안출된 것으로서 본 발명의 목적은 혈뇌장벽을 통과하는 압타머를 제공하는 것이다.
본 발명의 다른 목적은 신규한 알츠하이머병이나 파킨슨병 등을 조기에 진단하기 위한 PET(positron emission tomography) 방사선 조영제로 제공하는 것이다.
본 발명의 또 다른 목적은 신규한 알츠하이머병이나 파킨슨병 등에 대한 치료제를 제공하는 것이다.
상기의 목적을 달성하기 위하여 본 발명은 서열번호 1 내지 100에 기재된 염기서열로 이루어진 올리고뉴크레오타이드 군으로부터 선택된 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머를 제공한다.
본 발명의 일 구현예에 있어서, 상기 압타머는 서열번호 101의 염기서열을 공통으로 가지는 것이 바람직하나 이에 한정되는 것은 아니다.
또한 본 발명은 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질과 상기 본 발명의 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머를 함께 결합한 복합체를 제공한다.
본 발명의 일 구현예에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질은 뇌 실질내 전달이 어려운 약물, 화학물질, 압타머, siRNA, 안티센스 올리고뉴크레오타이드, miRNA, CpG 올리고뉴크레오타이드, 펩타이 및 단백질로 구성된 군으로부터 선택된 물질인 것이 바람직하나 이에 한정되지 아니한다.
본 발명의 또 다른 구현예에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질과 상기 본 발명의 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머는 링커에 의하여 연결된 것이 바람직하나 이에 한정되지 아니한다.
본 발명의 또 다른 구현예에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질은 방사성 동위원소가 표지된 것이 바람직하나 이에 한정되지 아니한다.
또 본 발명은 상기 본 발명의 복합체를 유효성분으로 포함하는 양전자단층촬영(Positron Emission Tomography; PET)용 조성물을 제공한다.
또한 본 발명은 상기 본 발명의 복합체를 유효성분으로 포함하는 퇴행성 뇌질환 진단용 조성물을 제공한다.
또한 본 발명은 상기 본 발명의 복합체를 유효성분으로 포함하는 퇴행성 뇌질환 치료용 조성물을 제공한다.
본 발명의 일 구현예에 있어서, 상기 퇴행성 뇌질환은 알츠하이머병, 파킨슨병, 또는 헌팅턴 질환인 것이 바람직하나 이에 한정되지 아니한다.
이하 본 발명을 설명한다.
본 발명은 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머와 뇌 안에 존재하는 타겟물질에 선택적으로 결합하는 물질을 함께 결합한 복합체(예를 들어, Bi-specific (dual-specific or chimeric) 압타머)를 제조하여, 이를 positron emission tomography (PET) 방사선 조영제 (Aptamer-based radiotracer)로 사용하여 타겟 물질의 응집을 막아 알츠하이머병이나 파킨슨병 등을 조기에 진단하고 예방 및 치료하는 것이다.
예를 들어, 본 발명은 alpha-synuclein, amyloid beta, tau, SOD1, TDP-43 등 퇴행성 뇌질환에서 응집이 증가하는 다양한 물질들에 선택적으로 결합하여 응집을 억제하는 압타머와 상기 본 발명의 BBB를 통과하는 압타머와 연결하여 구현한 Bi-specific 압타머를 이용하여 BBB를 통과 뇌실질내에 타겟물질의 분포를 PET을 통해 조기에 조영할 수 있고 타겟물질의 응집을 방지하는 Theragnosis 기술로 활용가능한 물질을 개발하는 것에 관한 것이다.
본 발명을 통하여 알 수 있는 바와 같이, 본 발명의 압타머는 혈뇌장벽을 통과하할 수 있으며, 이러한 압타머를 포함하는 복합체는 PET(positron emission tomography) 방사선 조영제로 사용하여 알츠하이머병이나 파킨슨병 등을 조기에 진단할 수 있고, BBB를 통과 뇌실질 내에 타겟 물질의 분포를 PET을 통해 조기에 조영할 수 있는 방사선 조영 물질로 사용될 수 있다. 또한 타겟 물질의 응집을 방지하여 치료제로 사용될 수 있다.

이하 본 발명을 비한정적인 실시예를 통하여 더욱 상세하게 설명한다. 단 하기 실시예는 본 발명을 예시하기 위하여 기재한 것으로서 본 발명의 범위는 하기 실시예에 의하여 제한되는 것으로 해석되지 아니한다.
실시예 1:압타머가 세포내에 발현시킨 alpha-synuclein의 응집을 방지하거나 분해를 촉진하는지 확인
사람 신경모세포종 SH-SY5Y 세포에 α-synuclein을 lenti viral vector를 이용하여 과발현시킨 후에 α-synuclein의 monomer/oligomer에 specific 하게 binding 하는 압타머를 처리한다. 후에 western blot을 통해 α-synuclein의 응집과 분해 정도를 확인하였다.
실시예 2: 인간 혈액 뇌 혈관 세포주(hCMEC/D3)의 ZO-1 발현
In vitro에서 혈뇌장벽 모델을 제작하는데 사용한 hCMEC/D3 세포 주는 인간 뇌혈관 세포로부터 유래된 세포 주로 혈뇌장벽 연구에 가장 많이 사용되는 세포 주로, hCMEC/D3 세포 주를 단일층으로 배양하면서 혈뇌장벽의 중요한 표현형인 밀착 연접이 형성되는지를 밀착 연접 단백질 지표 중 하나인 ZO-1 (초록색)을 면역 형광 염색하여 확인하였고 핵은 Hoechst (파란색)를 이용하여 염색하였다. 그 결과 배양 기간이 길어질 수록 세포들이 서로 접촉하면서 밀착 연접이 잘 형성되는 것을 확인하였다(도 1).
실시예 3:in vitro 인공 BBB model을 제작하여 aptamer selection하여 뇌혈관만 특이적으로 통과하는 aptamer를 발굴하고 특성 연구
인간 뇌혈관세포주인 hCMEC/D3를 이용하여 in vitro 상에서 BBB model을 제작(hCMEC/D3 세포는 Millipore 회사로부터 구입하였다. 배양액은 EBM-2(Lonza)에 5% FBS(gibco), 1.4 μM Hydrocortisone(sigma), 5 μg/ml Ascorbic Acid(sigma), 1 ng/ml bFGF(sigma), 10 mM HEPES(gibco), 1x chemically defined lipid(gibco), P/S(gibco)를 추가하여 사용했다. 배양액의 교환은 2~3일 주기로 수행했다. in vitro 상에서 BBB model을 제작하기 위해 hCMEC/D3의 세포를 transwell (12 well insert, pore size: 0.4μm, corning) 내부에 2.2 x 10 5의 세포를 seeding 해서 6~10일 동안 배양하였다. 이렇게 만들어진 BBB model을 이용하여 DNA aptamer library를 인공모델의 혈관에 해당하는 곳에 처리한 뒤에 DNA pool을 selection하였다. 각 Round 당 한 개의 BBB model을 사용하였음)한 뒤에 DNA aptamer library를 인공모델에서 혈관에 해당하는 곳에 처리하여 hCMEC/D3 세포를 통과한 DNA pool을 selection하였다.
간략하게 기재하면, 도 2에서와 같이, in vitro 혈뇌장벽 모델을 통과하는 압타머를 발굴하기 위해 SELEX를 진행하였다. 이 때 사용한 랜덤 DNA 라이브러리는 총 60-mer이며, 중앙에 30-mer의 랜덤서열과 양 끝에 증폭을 위한 15-mer의 고정된 프라이머 서열로 구성된다(5'-ATGCGGATCCCGCGC-N 30-GCGCGAAGCTTGCGC-3';서열번호 103). 1 라운드에서는 1 nmol의 랜덤 DNA 라이브러리를 혈뇌장벽 모델에 처리하고 37℃에서 60분 반응한 후, 통과한 압타머를 선별하였다.
SELEX 기술은 ssDNA을 95℃에 10분 처리하고 5분 동안 상온에 방치하여 Folding시켜 준비한다. 세포는 PBS로 wash한 뒤에 salmon sperm DNA(10mg/ml)를 포함한 PBS로 10~15분 배양하여 블로킹 과정을 거친다. 그 후, 준비된 ssDNA를 블로킹된 세포에 처리하여 투과된 ssDNA를 페놀/클로로포름/아이소아밀 알코올 (phenol/chloroform/isoamyl alcohol; PCI) 추출법을 이용하여 ssDNA를 선별하였다.). 다음 라운드를 위한 DNA 시료를 얻기 위해 선별한 압타머를 PCR로 증폭하였다.
PCR 증폭은 두단계를 거친다. 이 때 정방향 프라이머는 5'-ATGCGGATCCCGCGC-3'서열(서열번호 104)을 사용했고, 역방향 프라이머는 5'-GCGCAAGCTTCGCGC-3'(서열번호 105) 서열에 biotin을 부착시켜 사용하였다. 일단계는 symmetric PCR을 이용하여 선별된 ssDNA pool을 dsDNA로 만든다.
상기 PCR 조건: Step 1. 95℃ 3min Step 2, 95℃ 10sec, 59℃ 10sec, 72℃ 10sec 11~15 cycle, step3 72℃ 1min이었다.
이 단계는 만들어진 dsDNA를 template로 사용하여 정방향 프라이머와 역방향 프라이머의 비율을 25:1로 하여 asymmetric PCR을 수행한다.
상기 PCR 조건: Step 1. 95℃ 3min Step 2, 95℃ 10sec, 59℃ 10sec, 72℃ 10sec 30cycle, step3 72℃ 1min.
정방향 프라이머의 비율이 높기 때문에 dsDNA보다 ssDNA가 더 많이 만들어지게 된다. 이 때 dsDNA에는 역방향 primer에 부착되어 있는 biotin이 포함되게 된다. SELEX에 사용할 ssDNA를 분리하기 위해서 Streptavidin이 부착되어 있는 magnetic bead를 이용하여 dsDNA를 제거하였다.
2 라운드에서는 처리하는 DNA 시료의 양을 200 pmol로 줄였다. 라운드를 진행할 수록 처리하는 DNA의 시료 양과 반응 시간을 각 50 pmol과 20분으로 줄였다. SELEX의 적합 여부를 판단하기 위해서 4 라운드와 9 라운드에서 모니터링하였다.
PCR 방식을 이용하여 전기 영동방법으로 DNA band의 intensity 측정하는 법으로 확인하였다.
4 라운드를 수행할 때에 100 pmol의 랜덤 DNA 라이브러리와 3 라운드에서 선별하여 증폭시킨 DNA 시료를 각 각의 혈뇌장벽 모델에 처리하고 통과한 DNA의 양을 symmetric PCR을 통해 확인하였다(PCR 증폭 1단계 조건과 동일함) . 그 결과, 3 라운드에서 선별하여 증폭시킨 DNA 시료보다 랜덤 DNA 라이브러리를 처리한 군에서 PCR 산물이 상대적으로 적은 것을 확인하였다. 9 라운드를 진행할 때에는 3 라운드와 8 라운드에서 선별되어 증폭된 DNA 시료를 각 각의 혈뇌장벽 모델에 처리하였다. 통과한 DNA의 양을 PCR로 확인해 보았을 때에 8 라운드에서 선별된 DNA 시료가 더 잘 통과하는 것을 확인하였다. 이를 통해 라운드가 진행될 수록 혈뇌장벽 모델을 보다 더 잘 통과하는 압타머가 선별되었음을 확인하였다(도 3). 9 라운드를 통해 얻은 DNA 시료는 NGS 분석을 실시하였다(NGS 분석은 마크로젠에서 illumina platform타입의 sequencer를 실시함).
NGS 분석한 결과, 상위에 있는 서열 중 대부분이 10-mer 길이의 공통된 서열 (GAGCACGGGG; 서열번호 101)을 포함하고 있음을 확인하였고, 표 1에 기재된 상위의 100개 서열 중 24개의 서열이 상기의 10-mer의 서열을 포함하였다(도 4). 상위 4개의 서열을 이용하여 실험을 진행하였다.
실시예 4:BBB-투과 압타머의 투과율 테스트
서열번호 2 내지 5의 NGS 서열에서 read된 수가 높은 서열(그 서열이 많이 존재하여 그만큼 많이 읽히게 됨) 4개 압타머 서열 (BBB-1, 2, 3, 4)의 3' 부분에 형광 물질인 FAM을 부착하여 각 각의 혈뇌장벽 모델에 처리하고, 1시간 후에 통과한 압타머의 형광 세기를 측정하였다.
이 때 사용한 BBB 모델은 SELEX에 사용한 BBB 모델보다 작은 모델을 사용하였다. 24well insert transwell(pore size : 0.4um, cornig)에 8x10 4의 세포를 7일 동안 배양하여 모델을 제작하였다. 모델의 혈관부분에 해당하는 부분에 각각의 FAM이 부착된 Aptamer를 50pmol 씩 처리하고 1시간 배양하였다. 그 후에 통과한 aptamer의 형광세기와 넣어준 total aptamer의 형광세기를 Synergy H1 (Bioteck)을 이용하여 emission 파장 490nm, detection 파장 520nm에서 측정한 뒤에 통과율을 계산하였다. (통과율 = 통과한 aptamer의 형광세기 / 넣어준 aptamer의 형광세기 X 100) ). 음성대조군 (5'-CGCGCGTCAGGCATTCCTCACAATTCTTCG-3';서열번호 102)과 비교 시, 실험군 전체에서 투과율이 증가하였고 특히 BBB-2, 및 4는 그 증가폭이 크고, 그 중에 BBB-2의 통과율이 가장 높은 것을 확인하였다.
실시예 5:bi- Bi-specific (dual-specific or chimeric) 압타머의 개발
본 발명의 BBB 통과 압타머와 알파시뉴트레인이나 아밀로이드 베타 등의 타겟물질에 대한 압타머를 poly Thymidine linker(T10 ~ T20) 등을 통해 back-to-back으로 연결하여 Bi-specific 압타머를 만든다. 이때 링커에 ribosyUridine 등을 중간에 넣어 transcytosis가 일어난 후 쉽게 끊어져서 타겟 물질에 대한 압타머를 방출하게 디자인할 수 있다.
링커로 연결하면 보통 압타머의 binding affinity가 떨어진다. 따라서 각각의 압타머의 기능이 연결 전과 똑같이 유지될 수 있도록 bi-specific aptamer를 optimization 해야 한다.
BBB 통과 압타머의 sequence를 고정시켜 DNA library를 제작한 다음 알파시뉴트레인이나 아밀로이드 베타 등의 타겟물질에 대한 SELEX를 수행한다.
알파시뉴트레인이나 아밀로이드 베타 등의 타겟물질에 결합하는 압타머의 sequence를 고정시켜 DNA library를 제작한 다음 인공 BBB 모델에 투과를 하는 압타머를 selection 한다.
실시예 6:bi-specific aptamer가 인공 BBB 모델을 통과한 이후에 타겟물질에 결합할 수 있는지 확인
인공 BBB model에서 뇌실질 부분에 해당하는 쪽에 α-synuclein을 overexpression 시킨 SH-SY5Y 세포를 배양킨다. 형광물질로 표지한 bi-specific aptamer를 혈관에 해당하는 쪽에 처리한 후에 SH-SY5Y 세포의 세포질에 형광이 보이는지 확인한다. 또한 α-synuclein을 형광염색하여 aptamer와 얼마나 co-localiztion 되어 있는지 확인한다.

Claims (12)
- 서열번호 1 내지 100에 기재된 염기서열로 이루어진 올리고뉴크레오타이드 군으로부터 선택된 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머.
- 제 1항에 있어서, 상기 압타머는 서열번호 101의 염기서열을 공통으로 가지는 것을 특징으로 하는 압타머.
- 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질과 제1항 또는 제2항 중 하나의 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머를 함께 결합한 복합체.
- 제3항에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질은 뇌 실질내 전달이 어려운 약물, 화학물질, 압타머, siRNA, 안티센스 올리고뉴크레오타이드, miRNA, CpG 올리고뉴크레오타이드, 펩타이 및 단백질로 구성된 군으로부터 선택된 물질인 것을 특징으로 하는 복합체.
- 제3항에 있어서, 뇌 안에 존재하는 타겟 물질은 알파시뉴트레인 타우(tau), 슈퍼옥시드디스뮤타제(SOD)1 또는 아밀로이드 베타인 것을 특징으로 하는 복합체.
- 제3항에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질과 상기 혈뇌장벽 (Blood-Brain Barrier, BBB)을 통과하는 압타머는 링커에 의하여 연결된 것을 특징으로 하는 복합체.
- 제3항에 있어서, 상기 뇌 안에 존재하는 타겟 물질에 선택적으로 결합하는 물질은 방사성 동위원소가 표지된 것을 특징으로 하는 복합체.
- 제3항 내지 제7항 중 어느 한 항의 복합체를 유효성분으로 포함하는 양전자단층촬영(Positron Emission Tomography; PET)용 조성물.
- 제3항 내지 제7항 중 어느 한 항의 복합체를 유효성분으로 포함하는 퇴행성 뇌질환 진단용 조성물.
- 제9항에 있어서, 상기 퇴행성 뇌질환은 알츠하이머병, 파킨슨병, 또는 헌팅턴 질환인 것을 특징으로 하는 조성물.
- 제3항 내지 제7항 중 어느 한 항의 복합체를 유효성분으로 포함하는 퇴행성 뇌질환 치료용 조성물.
- 제11항에 있어서, 상기 퇴행성 뇌질환은 알츠하이머병, 파킨슨병, 또는 헌팅턴 질환인 것을 특징으로 하는 조성물.
