Abstract
본 발명은 레틴알데히드 및 글리실글리신 올레아미드 및 필요한 경우 델타-토코페릴-글루코피라노시드 기반의 국소 조성물, 화장학 및 피부학에서의 피부 노화 방지 용도, 더욱 상세하게는 광-유발 피부 노화 방지 용도에 관한 것이다.
Description
본 발명의 목적은 레틴알데히드 및 글리실글리신 올레아미드 및 필요한 경우 델타-토코페릴-글루코피라노시드 기반의 국소 조성물, 화장학 및 피부학에서의 이의 용도에 관한 것이다.

집합적으로 레티노이드로 칭하는 비타민 A (레티놀) 및 이의 천연 및 합성 유도체는, 특히 배발생, 생식, 시력, 성장, 염증, 분화, 증식 및 세포사멸에서 현저한 영향을 미치는 다종의 물질을 형성한다. 레티노이드의 생물학적 영향은 RAR 타입의 핵내 수용체 (레티노산 수용체) 및 RXR 타입의 수용체 (레티노산의 X 수용체)와의 상호 작용에 의해 조정된다. RAR 수용체의 공지 리간드는 비타민 A의 산성 형태의 트랜스 이성질체이다.
피부는 상당량의 레티노이드, 및 이의 대사 작용에 관여하는 효소를 가진다. 레티노이드는 비타민 A의 천연 또는 합성 유도체이다. 천연 레티노이드로서, 레티놀 및 이의 모든 대사 유도체, 레틴알데히드, all-trans-레티노산, 9-cis-레티노산, 13-cis-레티노산 및 이의 에스테르 예컨대 레티닐 팔미테이트가 언급된다. 합성 레티노이드는 비타민 A에 비하여 단일 효과를 가지는 화합물이 되도록 의도적으로 일부가 변형된 분자이다. 주요 분자는 에트레티네이트, 아시트레틴, 타자로텐, 아다팔렌, 벡사로텐, 모트레티니드이다.

Invest. Dermatol., 115(6): 1054-1058, 2000).
레티노이드 적용은 피부과학에서 두 가지 전략에 따라 수행된다:
피부 질환 치료의 약학적 복용에서, 레티노산 적용은 (0.025% 내지 0.1% 겔 또는 크림) 여드름 치료의 주요 처방이다. 또한 광-피부 노화 및 광선 각화증 치료에 사용된다. 타자로텐은 건선 치료에 처방된다 (0.05% 내지 0.1% 겔). 모트레티니드 (0.1% 크림 또는 로션)은 약한 여드름 치료에 처방된다.
화장 용도로는: 레티닐 팔미테이트 (0.5% 내지 5% 겔), 레티놀 (0.1% 내지 0.6%) 및 레틴알데히드 (0.05%)는 항-노화 처치 및 기능 유지관리에 사용된다.
태양 복사선은 가시광선, 자외선 (UV), 적외선 및 기타 전자기선을 포함한다. 가시광선이 여러 색상으로 분할되는 것과 같이, UVs는 세 가지로 분류된다:
UVAs (315-400 nm)는 지구 표면에 도달하는 주요 UVs 부분이다. 피부를 태우고 조직 침투력으로 깊게 손상을 입힐 수 있다.
UVBs (280-315 nm)는 훨씬 적은 부분이 대지에 도달하지만, 일사 노출에 대한 생물학적 영향에 상당히 기여한다. 발적, 화상, 햇볕에 태움, 안구 병변을 유발한다. 피부 침투력은 낮다.
UVCs (200-280 nm)는 대기권 오존층에 대부분 흡수된다. 피부에 약간만이 침투하지만 안구 병변을 초래할 수 있다.
UVs의 피부에 대한 주요 영향은 홍반, 광노화, 피부암이다. UVs에 노출 후, 표피 및 진피에서 금속단백분해효소 발현이 증가된다. 진피에서 콜라겐 생성을 변경시키는 이들 효소는 주름 발생에 기여한다. 이들 효소 유도는 반복 노출로 유지된다 (Fisher et al., N. Engl. J. UVAs 10 주 조사 후, 임의의 정상 탄성 섬유가 존재하지 않는다.
많은 연구로 만성적 햇빛 노출 및 피부암 발생 소인 간의 연결 관계가 확립되었다. 또한, UVs는 소정의 유전자 예컨대 DNA 수준에서 병변을 제거 또는 복구시키는 p53를 변형시킨다. UVBs 및 UVAs는 인간의 세포 면역에서 동일하게 영향을 주지 않는다. UVAs는 덜 발암성이다. UVBs는 따라서 햇빛 노출에서 주요 발암 인자로 작용한다. 색소는 UVs에 대한 민감성에 관여되는 인자이다. 멜라닌세포의 멜라닌에 의해 흡수되는 UVAs는 피부암 근원일 수 있다. 실제, UVAs는 흑색종 개시 원인이지만, UVBs 또한 전이될 때까지 이의 진행에 관여한다.
레틴알데히드 및 레티노산은 만성적 및/또는 장기간 햇빛 노출로 발생되는 피부 노화, 때로 조숙화에 상당하는 광-피부의 치료에 대한 주요 처방이다. 병변은 받은 선량에 직접 비례한다.

는 올레산 및 글리실글리신 디펩티드의 축합으로 인한 지질-펩티드 또는 아실-펩티드이다.
글리실글리신 올레아미드는 액체 세제 조성물에서 이의 계면활성제 특성이 알려져 있다 (US-4,732,690 및 JP-59 84 994). 특허 EP-1 061 897의 목적은 더욱 일반적으로는 세포 대사 자극에 의한 결합 조직의 단백질 섬유의 당화반응 및 탄력섬유증에 대항할 의도의 조성물 제조에 글리실글리신 올레아미드를 적용하는 것이다.

출원인에 의해 수행된 연구를 통해 각질세포에서 레티노이드 대사작용에 대한 레틴알데히드 및 글리실글리신 올레아미드의 조합의 상승효과 가능성을 보였다.
놀랍게도, 레틴알데히드 및 글리실글리신 올레아미드와 조합되는 델타-토코페놀-글루코피라노시드는 진피 전반에 대한 피부과 활성을 가지고, 피부 노화 및 더욱 상세하게는 햇빛으로-유발된 피부 노화에 대한 매우 현저한 영향을 보인다. 따라서, 3종의 활성제들을 조합하면 광-유발되는 노화 과정에서 발현이 변경되는 많은 유전자들에 대한 매우 광범위한 작용을 가진다.

본 발명에 의한 조성물은 수중유 유화액 (O/W) 또는 유중수 (W/O) 유화액으로 존재한다. 또한 소구체 예를들면 리포솜, 나노캡슐 또는 나노구체로 존재할 수 있다.

조성물은 또한 증점제 또는 유동개질제, 예컨대 예를들면 소수성 개질 에톡시화 비-이온성 우레탄, 증점화 폴리카르복실산 예컨대 아크릴레이트/스테아레스-20 메타크릴레이트의 공중합체, 카보머, 아크릴레이트 및 아크릴레이트의 공중합체 및 가교화 C10-C30 알킬 아크릴레이트; 및 이의 혼합물을 포함한다.
조성물은 상기 조성물 pH 조정을 위한 산 및 염기를 더욱 포함할 수 있다. 염기는 무기 (소다, 칼리, 암모니아) 또는 유기 예컨대 모노-, 디- 또는 트리-에탄올아민, 아미노메틸프로판디올, N-메틸-글루카민, 염기성 아미노산 예를들면 알기닌 및 리신; 및 이의 혼합물이다.

본 발명에 의한 조성물은 연고, 스프레이, 로션, 겔, 폼, 분산액, 세럼, 마스크, 바디 밀크 또는 크림 형태일 수 있다.
본 발명에 의한 조성물은 화장품 및 피부과학에서 주름진 피부 예방 또는 개선에 사용될 수 있고 피부에 견고성 및 촉촉함을 유지시킨다.
본 발명에 의한 조성물은 피부학 또는 화장품에서 국소 제제로 사용될 수 있고, 주름 예방 및/또는 감소 목적으로, 피부가 광-유발된 노화되거나 또는 그렇지 않은 경우라도, 표피 및 진피 세포 활성을 재-개시하여, 피부를 견고하게 하고, 이의 탄력성을 높이고, 피부에서 비타민 A의 생리적 저장을 증가시킨다.
본 발명은 또한 피부 노화, 더욱 상세하게는 광-유발된 피부 노화 방지를 위한 본 발명에 의한 조성물의 화장적 용도에 관한 것이다.

본 발명은 또한 광-유발된 피부 노화 방지를 위한 화장 방법에 관한 것이고 본 발명에 의한 조성물의 피부 적용 단계를 포함한다.
본 발명은 또한 피부에서 비타민 A의 생리적 저장을 증가시키기 위한 화장 방법에 관한 것이고 본 발명에 의한 조성물의 피부 적용 단계를 포함한다.
본 발명은 또한 피부에서 비타민 A의 생리적 저장 (physiological reserves) 결핍과 연관된 피부 질환 치료를 위한 피부과 약물 용도의 본 발명에 의한 조성물에 관한 것이다.
본 발명은 이하 실시예들에 의해 비-제한적으로 설명된다.
실시예 1
국소 레티노이드 사전-처치에 의한 UV 복사선으로 초래된 결핍 예방 (Tran et al., Photochem. Photobiol. 73(4): 425-431, 2001).
연구는 무모 마우스에서 수행되었다. 레티노이드를 국소적으로 7 일 동안 0.05% 적용한 후, UVB 복사선 (1J/cm²)을 조사하였다. 표피 레티노이드를 조사 후 24 h 분석하였다.
결과를 국소 레티노이드로 사전-처치하여 UV 복사선으로 초래된 결핍을 예방이라는 제목으로 도 1에 도시한다.
레틴알데히드 및 레티놀은 표피에 레티노이드를 적용함으로써 광-유발된 표피 레티닐 에스테르 결핍을 방지한다. 레티노산은 임의의 기질 (substrate)을 제공하지 않지만, 레시틴 레티놀 아실전이효소를 유도함으로써 광-유발된 레티노이드 결핍을 방지한다.
결론: 광-유발된 레티노이드 결핍 연구에서 레틴알데히드가 최선의 프로파일을 가진다.
실시예 2
각질세포에서 레티노이드로 초래된 레티닐 에스테르 조절.
각질세포 A431 배양체에서 연구를 수행하였다. 실제, 이들 세포에, 내생적 레티노이드를 측정되지 않았다. 레티노이드를 미리 적용하여 세포 레티노이드를 측정한다. 레티노이드는 HPLC로 측정하였다. 1% 에탄올에 녹인 다양한 조제 또는 조제 조합물에서 24 내지 72 h 동안 배지에서 배양하였다.
각질세포에서 레티노이드 생성을 하기 표에 나타낸다. 이들은 단백질 pmol/mg으로 표현되는 평균 값들이다.

본 실험은 레틴알데히드가 일련의 레티닐 에스테르를 유도한다는 가능성을 확립한다. 레틴알데히드에 의해 유발된 레티닐 에스테르는 글리실글리신 올레아미드에 의해 유의하게 증가하고, 이러한 조합으로 실제적인 상승효과를 보인다. 레티닐 팔미테이트는 세포에 통합된 후, 레티놀로 가수분해되고, 이후 형성된 레티놀은 부분적으로 에스테르화되고, 최종적으로 레틴알데히드보다 세포 레티노이드를 덜 생성한다.
도 2는 배양 48 h 후 각질세포에 유도된 레티닐 에스테르 조절에 대한 레틴알데히드 및 글리실글리신 올레아미드 조합에 의한 상승효과를 보인다. 값들은 레틴알데히드 처리 백분율로 표현된다.
실시예 3
레틴알데히드 및 글리실글리신 올레아미드 및 델타-토코페릴-글루코피라노시드 조합에 의한 UVAs로 조사된 인간 진피 섬유아세포에 대한 효과.
본 연구 목적은 섬유아세포 조사 전 이러한 3종의 활성제들의 보호 특성을 평가하는 것이다.
12-웰 플레이트에 접종되고 배양 배지 중에 24 시간 배양된 정상 인간 진피 섬유아세포에서 연구를 수행하였다.
보호 효과를 보이기 위하여, 배양 배지를 피험 생성물의 혼합물이 함유한 또는 함유하지 않은 (대조) 배지로 교체하고, 세포를 24 시간 배양하였다. 예비-배양 후, 배지를 화합물이 없는 조사 배지로 교체한 후 세포를 UVAs (20 J/cm²)로 조사하거나 조사하지 않았다 (미-조사 대조). 조사 후, 배지를 시험 배지로 교체하고 세포를 48 시간 배양하였다. 모든 실험 조건은 n=3으로 수행하였다. 실험 종료 후, RNA를 추출하고 64종 유전자 발현을 RT-PCR로 측정하였다.
혼합물은 다음으로 이루어진다:
1μM의 레틴알데히드,
1μM의 글리실글리신 올레아미드,
10μM의 델타-토코페릴-글루코피라노시드.
인간 진피 섬유아세포에 대한 UVAs 조사는 명백히 세포외 기질 조합에 관여하는 많은 유전자 발현 및 이러한 소정의 기질 성분 발현을 억제하였다 (도 3). 평행하게, 세포 주기 조절에서 세포외 기질 퇴화에 관여하는 MMP1 및 MMP3 마커의 명백한 자극이 억제되었다. 64종의 시험된 유전자들 중, 25개는 UVs로 조절되었다. 이러한 결과는 예측된 것이고 실험 유효성을 부여하였다.
3종 활성제들의 혼합물 존재에서, 도 3에서 보이는 바와 같이, 조사에 의해 발현이 변하는 13개의 유전자들에서 실질적인 보호 효과가 관찰된다.
놀랍게도, 이들 3종 활성제의 혼합물은 세포 및 세포외 기질 (예를들면 팩실린, 베르시칸)간 상호작용에서 피부 탄성망에 관여하는 유전자 (예를들면 엘라스틴, 피불린 1)및 세포 핵 보호 유전자 (서비빈, 보레알린) 모두에서 보호 효과 가능성을 보인다.
도 3은 이들 3종 활성제들의 혼합물이 조사에 의해 영향을 받은 유전자 발현을 회복시키고 대조군 대비 4종의 유전자들 발현 가능성을 보인다 (엘라스틴, 피불린, 리실 옥시다제-유사 2 및 니도젠 1).
따라서, 이들 3종 활성제들의 혼합물은 조사에 의해 변형을 교정할 뿐 아니라, 진피 보호 효과는, 특히 실제 피부 탄성망 재구성이 관찰되는 피부 탄력성에 관여되는 유전자에 중요하고 이들 3종 활성제들의 혼합물은 피부 노화에 상당히 효과적이다.
실시예 4
UVAs에 조사된 인간 피부 탄성망에 대한 글리실글리신 올레아미드, 레틴알데히드 및 델타-토코페릴-글루코피라노시드의 조합 효과.
연구는 6-웰 플레이트에 접종되고 배양 배지 중 48 시간 배양된 인간 피부 외식편에서 수행되었다. 피부는 UVAs (12 J/cm2)로 조사되거나 조사되지 않았다 (미-조사 대조). 보호 효과를 보이기 위하여, 조사가 끝날 때까지 글리실글리신 올레아미드 0.1%, 레틴알데히드 0.05%, 및 델타-토코페릴-글루코피라노시드 0.05%의 혼합물을 가지거나 가지지 않는(위약 대조)제제를 피부 표면에 (10 mg/cm2) 도포하였다. 24 시간 배양 후, 동일 량의 보호 제제를 다시 피부에 적용하였다. 실험을 24 시간 진행하고 끝난 후 피부를 동결하고 형광 공초점 현미경에서 피부 진피 탄성망을 관찰하기 위하여 면역화학염색 처리하였다. 피브릴린 및 엘라스틴의 발현은 특이 항체로 분석하였다.
UVA 조사로 유발된 탄성 섬유 변경에 대하여 피부를 보호하는 글리실글리신 올레아미드, 레틴알데히드 및 델타-토코페릴-글루코피라노시드 혼합물의 국소 적용에 관한 결과를 도 4에 제시한다. 실제, 피부 외식편에 대한 UVA 조사는 진피에서 피브릴린 및 엘라스틴 발현을 크게 손상시키고 많은 균열로 탄성 섬유망의 실질적인 해체를 유발시킨다. 위약 대조는 피부를 보호하지 않고 UVA-조사 피부와 유사한 탄성망을 보인다. 한편, 3종의 활성제들을 가지는 제제는 UVAs에 의해 영향을 받은 탄성 섬유 구조를 회복시키고 피부 탄력성을 보호한다. 조사되지만 활성제들의 혼합물로 처치된 피부의 탄성망 구조는 비-조사 대조 피부와 동일하다.
따라서 글리실글리신 올레아미드, 레틴알데히드 및 델타-토코페릴-글루코피라노시드의 조합물은 탄성 피부 조직 재구성 및 UVAs에 의한 변형에 대한 피부 보호 가능성을 부여하다. 따라서, 이들 3종 활성제들의 혼합물은 피부 탄력성을 보호하고 피부 광-노화에 효과적으로 작용한다.
전체 결과는 레틴알데히드 및 글리실글리신 올레아미드의 조합물이 시너지 효과로 피부에서 비타민 A의 생리적 저장을 계속 증가시키는 가능성을 제공한다. 레틴알데히드, 글리실글리신 올레아미드 및 델타-토코페릴-글루코피라노시드의 조합물은 진피에서 다수의 목표물에 상승효과적으로 작용하여 효과적으로 피부 노화에 작용하는 가능성을 제공한다.

- 활성 성분으로서 레틴알데히드 및 글리실글리신 올레아미드 및 적어도 하나의 화장학적 또는 피부학적으로 수용 가능한 담체를 포함하는 화장 또는 피부과 조성물.
- 제1항에 있어서, 델타-토코페릴-글루코피라노시드를 더욱 포함하는, 조성물.
- 제1항 또는 제2항에 있어서, 조성물 총 중량 기준으로 0.01 내지 1중량%의 레틴알데히드를 포함하는, 조성물.
- 제1항 내지 제3항 중 어느 하나의 항에 있어서, 조성물 총 중량 기준으로 0.0001 내지 20중량%의 글리실글리신 올레아미드를 포함하는, 조성물.
- 제2항 내지 제4항 중 어느 하나의 항에 있어서, 조성물 총 중량 기준으로 0.001 내지 10중량%의 델타-토코페릴-글루코피라노시드를 포함하는, 조성물.
