발명의 개요

발명의 상세한 설명
I. 정의
달리 정의되지 않는 한, 본 명세서에 사용된 기술 용어 및 과학 용어는 해당 분야의 숙련자가 통상적으로 이해하고 있는 것과 동일한 의미를 갖는다.
본 명세서에서 사용되는 용어 "항염증"은 감염, 질환 및/또는 외상에 의해 유발되는 조직 염증을 방해, 예방 및/또는 감소시키는 능력을 말한다. 본 발명의 항염증 화합물을 이러한 능력을 갖는다.
본 명세서에서 사용되는 용어 "항미생물"은 미생물(박테리아, 바이러스, 효모, 곰팡이, 포자, 원생동물, 기생충 등을 포함하나 이들에 한정되지 않음)을 죽이거나, 이의 증식을 억제하고/거나 예방하거나, 또는 미생물 감염을 약화시키고/거나 근절시키는 능력을 말한다.

. 방법 및 제제
본 발명의 제제는 N-할로겐화 아미노산 및 항염증 화합물을 포함한다. 본 발명의 N-할로겐화 아미노산은 다음 일반식을 갖는다:

항염증 화합물의 또 다른 예는 항알러지 및 항가려움증 제제, 이를 테면 올로파타딘, 케토티펜, 네도크로밀 소듐, 크로몰린 소듐 및 항히스타민을 포함하나 이들에 한정되지 않는 비만 세포 스태빌라이저이다.
본 발명의 특정 방법 및 제제는 이들의 항미생물 특성을 개선시키기 위하여 상 이동제(phase transfer agent)와 함께 N-할로겐화 아미노산의 사용을 포함한다. 본 명세서에 그 전체가 참조로 인용되는 미국 가특허출원 제60/915,291호[2007년 5월 1일, 발명의 명칭: "N-HALOGENATED AMINO ACID FORMULATIONS"]에는 이러한 N-할로겐화 아미노산 제제가 개시되었다.

. 적용
본 발명은 감염 조직으로 고통받는 대상, 특히 임의의 감염성 박테리아 또는 곰팡이로 감염된 포유동물 대상 및 더욱 바람직하게는 인간 포유류를 치료하는 것에 관한 것이다. 본 발명의 방법에 따라 치료되거나 예방될 수 있는 다른 박테리아 감염 및 원생동물 감염 및 이 감염과 관련된 질병은 문헌 [J. P. . 약제 및 제제
A. 용법
"약제학적 유효량"이라는 문구는 이미 업계에 알려진 용어이며, 본 발명의 약제에 도입되는 경우 임의의 의학적 치료에 적용가능한 합당한 이익/위험 비로 일부 원하는 효과를 생성하는 화합물의 양을 말한다. 유효량은 치료되는 질환 또는 감염 요인, 염증의 양, 투여되는 특정 제제 또는 감염 요인 또는 질환의 경중도와 같은 인자에 따라 달라질 수 있다.
"약제학적으로 허용되는"이라는 문구는 이미 업계에 알려진 용어이며, 해당 분야의 숙련자에 의해 결정된 합당한 이익/위험 비에 비례하여, 과도한 독성, 자극, 알러지 반응 또는 다른 문제나 합병증 없이 인간과 동물의 조직에 접촉하여 사용하기에 적합한 제제, 폴리머 및 다른 물질 및/또는 제형을 말한다.

B. 제제

특정 구체예에서, 제제는 포유동물 눈에 적용하기에 적합하다. 예를 들어, 안과적 투여를 위하여, 제제는 용액, 현탁액, 겔, 또는 연고일 수 있다.
부비동 및 호흡기 감염 적용에 사용하기 위하여, 네뷸라이저 또는 해당 분야의 숙련자에게 잘 알려진 다른 장치를 사용하는 에어로졸 제제에 적합한 제제가 사용될 수 있다.

미생물 감염과 관련된 조직 염증을 감소시키기 위하여 최소량의 활성 성분이 사용되도록 제제의 항염증 활성이 최대화되어야 한다. 특정 제제 중에 항염증 화합물의 양은 해당 분야의 숙련자에 의해 결정될 수 있다. 필요한 농도는 선택된 특정 항염증 화합물, 항염증 활성을 갖는 다른 성분의 존재 혹은 부재 및 제제 중에 함유된 항염증 제제의 작용에 따라 달라질 것이다. 허용되는 안정성과 독성을 유지하면서 원하는 항염증 활성을 얻는 데 필요한 농도는 본 명세서에서 "유효량"으로 언급된다.
본 발명의 제제를 포함하는 성분의 농도가 변할 수 있음이 고려된다. 비제한적인 면에서, 퍼센트는 총 제제의 중량 또는 부피로 계산될 수 있다. 해당 분야의 숙련자는 농도가 주어진 제제 중에 성분의 첨가, 치환 및/또는 제거에 따라 변할 수 있음을 이해할 것이다.
특정 구체예에서, 제제가 적용되거나 시행될 조직에 맞는 생리 pH를 갖는 국소 제제 (특히 상기 기술된 국소 안과용 제제)가 바람직하다.
본 발명의 특정 제제는 2개 파트 시스템으로 투여될 수 있다. 예를 들어, 사용자가 제제를 투여할 준비가 될 때까지 N-할로겐화 아미노산은 제제의 한 파트에 존재할 수 있으며, 제제의 하나 이상의 성분은 분리 용기 또는 동일한 용기의 다른 부분으로 분리된다. 투여 순간 또는 그 전에, 2개의 파트는 사용자에 의해 혼합될 수 있다. 2개 파트 시스템은 결합 시에 제제의 하나 이상의 성분이 안정성 문제를 가지는 경우 유용할 수 있다. 또한, 2개 파트 시스템은 특정 구체예에서, 코/부비동 분무 투여 시스템의 부분으로 이용될 수 있다.
C. 투여 경로
본 명세서에 기술된 방법에서, N-할로겐화 아미노산 및 항염증 화합물을 포함하는 제제의 약제학적 유효량을 대상에 투여하는 것은 해당 분야의 숙련자에게 알려진 임의의 방법에 의할 수 있다.
예를 들어, 제제는 국부, 국소, 피부내, 병변내, 비내, 피하, 경구, 흡입에 의해, 주사에 의해, 표적 세포를 직접적으로 베이딩 (bathing)하는 국부화 관류에 의하여, 카테터를 통해 또는 세척을 통해 투여될 수 있다.
특정 구체예에서, 제제는 안구 표면에 국소적으로 투여된다. 안과적 투여에 있어서, 국소, 결막하, 안구주위, 안구뒤, 서브테논 (subtenon), 안구내, 망막하, 공막부근 뒤 및 맥락막위 투여를 포함하는 눈에 대한 모든 국부 경로가 사용될 수 있는 것으로 고려된다.
다양한 귀 투여 기술도 고려된다. 특정 구체예에서, 제제는 이도에 직접 전달될 수 있다 (예: 국소적 귀 점적액 또는 연고; 귀 내 또는 귀 근처에 이식된 서방형 장치). 국부 투여에는 제제를 위한 귀 근육내, 고실내 및 달팽이관내 주사 투여 경로가 포함된다. 또한, 본 발명의 특정 제제가 귀내 삽입물 또는 이식 장치로 제제화될 수 있음이 고려된다. 예를 들어, 제제의 전달은 문헌 [Amer. J. Otology, Vol. 16: 158-163, 1995; Ear Nose Throat, Vol. 76:674-678, 1997; Otolarngol Head Neck Surg, Vol. 120:649-655, 1999]에 기술된 바와 같이 고실에 대하여 내시경 보조(고막 절개를 위한 레이저-보조 내시경 포함) 주사로 실행될 수 있다. 국부 투여는 또한 소 이관 카테터의 수단에 의하여 귀인두관을 통한 주사 또는 주입 및 고막절개술 절개를 통하여 장착한 내재 카테터의 사용을 통하여 미세 (EMG 기록) 바늘을 사용하는 고막을 통한 주사에 의해 실행될 수 있다. 또한, 제제는 숙련된 임상의의 적절한 재량 및 주의와 함께, 중이/내이의 윈도우 막(window membrane) 또는 근처 구조에 대하여, 제제에 침지된 겔폼(gelform) 또는 유사 흡수제 및 점착 제품의 장착으로 내이에 투여될 수 있다.
부비동 조직 감염, 코 감염, 상기도 감염, 폐/하기도 감염, 식도 감염 및 다양한 합병증의 치료를 위해 본 명세서에 기술된 제제의 투여는 해당 분야의 숙련자에게 알려진 많은 방법을 통한 것일 수 있다. 하기도 감염을 위한 바람직한 투여는 네뷸라이저 또는 기타 유사 장치의 사용에 의한 에어로졸 형성을 통한 것일 것이다. 부비동 감염 치료용 제제는 점적액 형태 (종종, 귀 제제는 부비동 감염의 치료를 위하여 사용될 수 있음) 또는 에어로졸 형태로 투여될 수 있다. 식도 감염은 액체 또는 에어로졸 제제의 투여에 의해 치료될 수 있다.
본 발명의 제제의 다른 투여 방법은 피부 패치를 통한 것, 폐내, 비내, 최적의 방식으로 제제화된 리포좀을 통한 것 및 서방형 데포 제제를 통한 것일 수 있다. 다양한 장치가 예를 들어, 특히 인간 대상의 내이를 치료 및/또는 진단하는 데 사용하도록 설계된 다기능성 장치를 제공하는 미국 특허 제5,476,446호에 예시된 바와 같거나, 카테터를 통하여 감염된 귀 구획에 제제를 전달하는 데 사용될 수 있다. 또한, 본 목적을 위한 다른 장치에 대해서는 미국 특허 제6,653,279호를 참고한다.
V.
실시예
다음 실시예는 본 발명의 선택된 구체예를 추가로 설명하기 위하여 기술한 것이다. 이들 제제에(및 본 발명의 다른 구체예에), 증점제(viscosity agent), 이를 테면 하이드록시프로필메틸셀룰로오스 (HPMC), 소듐 카복시메틸셀룰로오스 (NaCMC), 하이드록시에틸셀룰로오스 (HEC), 잔탄 검 등이 도입될 수 있다. 본 발명의 구체예에 따른 제제는 예를 들어, 고체, 액체, 반고체 등과 같은 다양한 물질 상태로 제조될 수 있다.

실시예

N-할로겐화 아미노산 제제의 항미생물 활성을 표준 미생물 분석으로 평가하였다. 이 평가의 결과를 하기의 표 1 및 2에 요약하였다. 평가를 위하여, 박테리아 및 곰팡이 분리물을 신선한 세포의 공급원으로서 적절한 아가 배지에서 밤새 배양하였다. 이들 신선한 세포의 현탁액을 대략 1 × 108 cfu/mL로 염수 중에 준비하였다. 이들 현탁액을 시험 제제(2,2-디메틸-N,N-디클로로타우린 및 대조군 용액의 다양한 용액)에 직접 가하였다. 시험 제제 용액 중의 세포의 초기 농도는 대략 1 × 106 cfu/mL이었다. 실온에서 60분 이하 동안 시험 제제에 대한 미생물의 노출을 수행하였다. 선택된 시간에, 분취량(aliquot)을 취해 4 ℃에서 인산 완충 염수로 희석하였다. 일련의 희석과 Milliflex 카세트 상의 여과 후에 생존성을 결정하였다.

이상과 같이 본 발명 및 이의 구체예를 상세히 기술하였다. 그러나 본 발명의 범위는 본 상세한 설명에 기술된 임의의 공정, 제조, 물질의 조성물, 화합물, 수단, 방법 및/또는 단계의 특정 구체예에 한정되지 않는다. 본 발명의 목적 및/ 또는 본질적인 특성에서 벗어나지 않고, 다양한 변형, 치환 및 변이가 개시된 물질에 가해질 수 있다. 따라서, 해당 분야의 숙련자는 본 명세서에 기술된 구체예와 실질적으로 동일한 결과를 얻거나 실질적으로 동일한 기능을 수행하는 이후의 변형, 치환 및/또는 변이가 본 발명의 관련 구체예에 따라 사용될 수 있는 것임을 본 개시물로부터 용이하게 알 것이다. 따라서, 하기의 청구 범위는 이들의 범위 내에 본 명세서에 개시된 공정, 제조, 물질의 조성물, 화합물, 수단, 방법 및/또는 단계에 대한 변형, 치환 및 변이를 포함하도록 의도된다.
Claims (14)
- N-할로겐화 아미노산 및 항염증 화합물을 포함하는 항미생물 활성 제제.
- 제 1항에 있어서, 항염증 화합물이 NSAID, 항알러지제 및 이들의 배합물로 구성된 군으로부터 선택되는 제제.
- 제 1항에 있어서, N-할로겐화 아미노산이 클로로타우린인 제제.
- 제 3항에 있어서, 클로로타우린이 소듐 2,2-디메틸-N,N-디클로로타우린인 제제.
- 제 3항에 있어서, N-할로겐화 아미노산이 제제의 성분과 이온쌍을 형성하는 제제.
- 감염 조직을 N-할로겐화 아미노산 및 항염증 화합물을 포함하는 제제의 약제학적 유효량과 접촉시키는 것을 포함하는 조직 감염의 치료 방법.
- 제 6항에 있어서, 항염증 화합물이 NSAID, 항알러지제 및 이들의 배합물로 구성된 군으로부터 선택되는 방법.
- 제 6항에 있어서, N-할로겐화 아미노산이 클로로타우린인 방법.
- 제 8항에 있어서, 클로로타우린이 소듐 2,2-디메틸-N,N-디클로로타우린인 방법.
- 제 6항에 있어서, 감염 조직이 안구, 귀, 코, 부비동 또는 피부 조직인 방법.
