Claims (15)
- 세포내에서 다수의 세포 성분을 확인하는 방법에 있어서,세포 샘플을 적어도 하나의 항체와 반응시키며, 여기서 각각의 항체는 특정한 세포 성분과 결합하고 특정한 형광 신호를 발생시키는 단계;상기 세포 샘플을 하나 이상의 핵산 탐침을 사용해서 동소 보합에 의해 처리하며, 여기서 각각의 핵산 탐침은 상기 세포내에서 타겟 핵산 시퀀스와 보합하도록 구성되고 특정한 형광 신호를 발생시키는 단계;상기 반응되고 처리된 세포 샘플의 하나 이상의 이미지를 발생시키는 단계; 및상기 이미지(들)에서 상기 항체 및 상기 핵산 탐침 모두에 대응하는 형광 신호를 검출 및 분석하는 단계를 포함하는 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 제 1항에 있어서,상기 세포 샘플이 혈액 샘플인 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 제 2항에 있어서,상기 혈액 샘플이 말초 혈액 샘플인 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 제 3항에 있어서,상기 혈액 샘플이 임신한 여자의 것인 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 제 1항에 있어서,대조구와 비교된 상기 형광 신호를 정량화하는 단계를 더 포함하는 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 제 1항에 있어서,상기 하나 이상의 핵산 탐침이 상기 세포 샘플의 X 및/또는 Y 염색체에 보합하도록 구성되는 것을 특징으로 하는 세포내에서 다수의 세포 성분을 확인하는 방법.
- 적어도 하나의 타겟 핵산에 의해 형성된 유전자 상태가 세포 샘플에 있는 지를 검출하기 위해 컴퓨터 시스템을 동작시키는 방법에 있어서,핵산에 타겟되는 보합된 형광 물질로 라벨된 탐침 및 관심있는 세포의 비-핵산 성분에 지향된 형광 면역염색을 갖는 고정된 샘플을 이미징하는 단계 - 상기 탐침 및 면역염색의 형광 라벨이 다름 - 와;상기 샘플로부터 형광을 검출하는 단계와;상기 면역염색 및 상기 탐침으로부터의 형광을 표시하는 관심있는 물체수를 결정하는 단계와;상기 면역염색 및 상기 탐침 모두로부터의 형광을 보여주는 세포의 통계적인 기대 빈도로부터 유전자 상태가 있는 지를 결정하는 단계를 포함하는 것을 특징으로 하는 적어도 하나의 타겟 핵산에 의해 형성된 유전자 상태가 세포 샘플에 있는 지를 검출하기 위해 컴퓨터 시스템을 동작시키는 방법.
- 제 7항에 있어서,상기 핵산 탐침이 상기 세포 샘플의 X 및/또는 Y 염색체에 보합되도록 구성되는 것을 특징으로 하는 적어도 하나의 타겟 핵산에 의해 형성된 유전자 상태가 세포 샘플에 있는 지를 검출하기 위해 컴퓨터 시스템을 동작시키는 방법.
- 제 7항에 있어서,상기 면역염색이 태아 헤모글로빈에 결합하는 것을 특징으로 하는 적어도 하나의 타겟 핵산에 의해 형성된 유전자 상태가 세포 샘플에 있는 지를 검출하기 위해 컴퓨터 시스템을 동작시키는 방법.
- 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법에 있어서,상기 샘플을 관심있는 세포의 비-핵산 성분에 지향된 형광 면역염색으로 처리하는 단계와;상기 샘플을 관심있는 핵산 시퀀스에 지향된 형광 핵산 탐침으로 처리하는 단계와;상기 형광 면역염색 및 상기 형광 핵산 탐침으로부터 형광 신호를 검출하도록 작동 구성된, 컴퓨터화된 마이크로스코픽 시각 시스템을 사용해서 세포 샘플의 일부를 커버하는 광학 필드를 관찰하는 단계와;상기 형광 신호 검출에 의해 관심있는 핵산 시퀀스를 갖는 관심있는 세포를 확인하는 단계를 포함하는 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
- 제 10항에 있어서,상기 관심있는 세포가 태아 세포인 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
- 제 11항에 있어서,상기 태아 세포가 모계 혈액에서 인출되는 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
- 제 10항에 있어서,상기 핵산 탐침이 X 및/또는 Y 염색체 DNA 시퀀스를 포함하는 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
- 제 10항에 있어서,상기 컴퓨터화된 시각 시스템이 하나의 대물 렌즈를 사용해서 상기 면역염색 및 상기 핵산 탐침으로부터 형광 신호를 얻는 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
- 제 10항에 있어서,관심있는 핵산 시퀀스를 갖는 것으로 확인된 관심있는 세포의 수를 토대로 시험 진단을 자동 발생시키는 단계를 더 포함하는 것을 특징으로 하는 자연적으로 존재하는 태아 세포 농도를 함유한 모계 혈액의 샘플을 준비하는 방법.
