Abstract
Description
Claims (12)
- 다음 단계를 포함하는, 포유류에 이식하기 위한 세포의 생산 방법:(a) 불멸화(immortalized) 되지 않는 골수 줄기 세포를 공급하는 단계;(b) 상기 세포를 혈관 평활근 세포, 내피 세포, 심장 외막 세포, 지방세포, 파골 세포, 골 모세포, 심장 섬유아세포, 대식 세포, 신경 세포 전구체, 신경세포, 성상세포, 골 근육 세포, 평활근 세포, 췌장 세포 전구체, 췌장 β-세포, 또는 간세포로 분화시키도록 유도하는 조건 하에서 상기 골수 줄기 세포를 배양하는 단계;(c) 단계 (b)의 세포의 분화 상태를 모니터링하는 단계; 및(d) 상기 세포의 10% 내지 100%가, 상기 세포 타입에 특이적인 단백질의 검출 가능한 양을 발현할 때 단계 (b)의 세포를 수집하는 단계.
- 제 1 항에 있어서,(e) 단계 (d)로부터의 상기 세포를 상기 포유류로 이식하는 단계를 더욱 포함하는 방법.
- 제 1 항에 있어서, 상기 골수 줄기 세포는 상기 포유류로부터 유래되는 방법.
- 제 1 항에 있어서, 상기 포유류는 인간인 방법.
- 제 1 항에 있어서, 상기 수집 단계 (d)는 단계 (b)의 상기 세포의 50% 내지 80%가 상기 세포 타입으로 분화될 때 수행되는 방법.
- 제 1 항에 있어서, 상기 모니터링 단계 (c)는, 단계 (b)의 상기 골수 줄기 세포와 공동 배양된 자가 골수 줄기 세포의 분화를 평가하는 것을 포함하고, 여기에서 상기 자가 골수 줄기 세포의 분화 상태는 단계 (b)의 상기 골수 줄기 세포의 분화 상태와 상관되는 방법.
- 제 1 항에 있어서, 상기 모니터링 단계 (c)는, 단계 (b)의 상기 골수 줄기 세포와 공동 배양된 동형 또는 이형 골수 줄기 세포의 분화를 평가하는 것을 포함하고, 여기에서 상기 동형 또는 이형 골수 줄기 세포의 분화 상태는 단계 (b)의 상기 골수 줄기 세포의 분화 상태와 상관되는 방법.

- 제 8 항에 있어서, 단계 (b)와 (c)의 사이에, 단계 (b)의 세포의 분화 상태를 모니터링하는 단계를 더욱 포함하는 방법.
- 포유류로 이식할 때 혈관 평활근 세포로 분화되도록 유도된 세포의 생산 방법으로서, 다음 단계를 포함하는 방법:(a) 불멸화(immortalized) 되지 않는 줄기 세포의 집합체를 공급하는 단계;(b) 상기 세포를 혈관 평활근 세포 전구체로 분화시키도록 유도하는 조건 하에서 단계 (a)의 상기 세포를 배양하는 단계; 및(c) 세포의 10% 내지 100%가 혈관 평활근 세포 전구체일 때 단계 (b)의 세포를 수집하는 단계로서, 여기에서 상기 세포는 포유류로 이식했을 때 혈관 평활근 세포로 분화되도록 유도된다.
- 제 10 항에 있어서, 단계 (b)와 (c)의 사이에, 단계 (b)의 세포의 분화 상태를 모니터링하는 단계를 더욱 포함하는 방법.
- 다음을 포함하는 약제학적 조성물:(a) 심근 세포 또는 심근 세포 전구체;(b) 내피 세포 또는 내피 세포 전구체; 및(c) 혈관 평활근 세포 또는 혈관 평활근 세포 전구체, 여기에서 상기 세포는 인간에 도입시 심장 기능을 개선시키기 위해 충분한 양이다.
