Description

비염 치료를 위한 Η1 길항제 및 안전한 스테로이드제의 용도{USE OF AN H1 ANTAGONIST AND A SAFE STEROID TO TREAT RHINITIS}
본 발명은 코 이상, 특히 비염을 치료하기 위한 안전한 스테로이드제와 배합된 H1 길항제/항알레르기제의 용도에 관한 것이다.
알레르기 비염은 경구용 항히스타민제 및/또는 경구용 스테로이드제 요법으로 치료되어 왔다. 전형적으로 전신 치료에서는 필요한 치료 부위에서 유효한 농도를 얻기 위해서 투여하는 약제 화합물의 농도를 더욱 높일 필요가 있다. 항히스타민 화합물은 졸음에서 나타나는 중추 신경계(CNS) 활성을 갖는 것으로 공지되어 있다. 또한 점막 건조에서 나타나는 항콜린성 활성을 가질 수 있다. 경구 또는 비강내 투여되는 스테로이드제 요법은 또한 부실부전증, 심혈관 이상, 및 면역억제와 같은 심각한 전신계성 부작용을 일으킬 수 있다. 알레르기성 소아 환자에서 성장 지연이 특히 주요 관심사이다.
비강내 배합 요법이 공지되어 있다. 예를 들면, WO 97/01337에는 비염 치료를 위한 국소 비강용 항히스타민제 및 국소 비강용 스테로이드제의 배합물이 기술되어 있다. WO 97/46243에는 스테로이드제 및 항히스타민제를 포함하는 비강용 스프레이가 기술되어 있다. 이 공개문헌은 안전한 스테로이드제의 용도에 대하여는 기술하거나 제안하고 있지 않다. 또한 다른 활성 성분(예: Pharmacobel사의 Cortinasal; Dupa사의 Neovvine; Rontag사의 Nicorin; Smithkline Beecham상의 Rinosular; Cusi사의 Rinocusi; 및 Meider사의 Comfonin)을 포함하는, 스테로이드제 및 항히스타민제 모두를 포함하는 비강내 제품도 존재한다.
비염을 치료하기 위한 안전한 스테로이드제와 배합된 H1 길항제/항알레르기제의 용도에 대하여 공지된 바 없다.
발명의 요약
본 발명은 비염을 치료하기 위한 H1 길항제/항알레르기제 및 안전한 스테로이드제 배합물의 비강내 조성물에 관한 것이다. 포유동물에서 조성물을 사용하는 방법 또한 주시된다.
발명의 바람직한 일면
본 발명은 알레르기 비염과 관련된 재채기 및 비루를 치료하기 위한 H1 길항제/항알레르기제 조성물을 포함한다. 본 조성물은 또한 안전한 스테로이드제를 포함하고, 본 명세서에서 사용되는 바, 용어 "안전한 스테로이드제"는 유발된 울혈과 함께 에오시노필 및 뉴트로필 관련 염증을 치료하지만 전신 생체이용성은 결여되어 있거나 전신 흡수후 급속하게 불활성화되는 스테로이드제를 언급한다.
본 발명에 따라 유용한 H1 길항제/항알레르기제는 유효한 화합물, 제한하는 것은 아니지만, 에메다스틴, 로라타딘, 5-[2-[4-비스(4-플루오로페닐)하이드록시메틸-1-피페리디닐]에틸]-3-메틸]-2-옥사졸리디논 에탄디오에이트), 데스로라타딘, 아젤라스틴, 올로파타딘, 레보카바스틴, 에피나스틴, 및 케토티펜을 포함한다.
본 명세서에서 사용될 수 있는 안전한 스테로이드제는 안전한 스테로이드제의 정의를 충족시키는 한, 리멕솔론 및 로테프레드놀로 제한하지 않고 어느 글루코코르티코이드를 포함한다.
H1 길항제/항알레르기제 및 안전한 스테로이드제(화합물)은 코로 전달되기 위하여 다양한 형태의 비강내 제제으로 혼입될 수 있다. 예를 들면, 비강내 제제는 방부제, 예로서, 벤즈알코늄, EDTA, 및 트로메타민; 점성 조절제, 예로서, 하이드록시 프로필 메틸 셀룰로오스(HPMC) 및 관련제; 독성 조절제, 예로서, 염화나트륨(NaCl); 습윤제/계면활성제, 예로서, 티록사폴 또는 폴리소르베이트 80; pH 조절제, 및 물을 포함할 수 있다.
화합물은 바람직하게 pH 약 6.0 내지 8.0의 비강내 현탁제 또는 액제로 제형화된다. H1 길항제/항알레르기제는 통상 이들 제제중에 0.01% 내지 0.5중량%의 양으로, 바람직하게 0.02% 내지 0.1% 중량%의 양으로 포함될 것이다. 안전한 스테로이드제는 통상 이들 제제중에 0.05% 내지 1.5중량%의 양으로, 바람직하게 0.1% 내지 1.0% 중량%의 양으로 포함될 것이다. 따라서, 비강내 전달을 위해서는 본 분야의 기술자의 일반적인 처방에 따라 1일 2회까지 1 내지 2 스프레이의 이들 제제를 콧구멍에 투여할 것이다.
본 발명의 바람직한 조성물은 리멕솔론(0.1%)과 올로파타딘(0.1%) 및 리멕솔론(0.1%)과 에메다스틴 0.05%을 포함한다.
하기 실시예는 본 발명의 조성물의 일례이며, 이로써 제한되지 않는다.

Claims (7)

  1. H1 길항제/항알레르기제 및 안전한 스테로이드제를 포함하는 약제학적 유효량의 조성물을 투여하는 것을 포함하는, 포유동물에서 비염을 치료하는 방법.
  2. 제 1항에 있어서, 조성물이 에메다스틴, 로라타딘, 5-[2-[4-비스(4-플루오로페닐)하이드록시메틸-1-피페리디닐]에틸]-3-메틸]-2-옥사졸리디논 에탄디오에이트) , 데스로라타딘, 아젤라스틴, 올로파타딘, 레보카바스틴, 에피나스틴, 및 케토티펜으로 구성된 그룹으로부터 선택되는 H1 길항제/항알레르기제를 포함하는 방법.
  3. 제 1항에 있어서, 조성물이 리멕솔론 및 로테프레드놀로 구성된 그룹으로부터 선택되는 안전한 스테로이드제를 포함하는 방법.
  4. 제 2항에 있어서, 조성물이 에메다스틴, 올로파타딘, 및 데스로라타딘으로 구성된 그룹으로부터 선택되는 H1 길항제/항알레르기제를 포함하는 방법.
  5. 에메다스틴 및 리멕솔론을 포함하는 약제학적 유효량의 조성물을 투여하는 것을 포함하는, 포유동물에서 비염을 치료하는 방법.
  6. 올로파타딘 및 리멕솔론을 포함하는 약제학적 유효량의 조성물을 투여하는 것을 포함하는, 포유동물에서 비염을 치료하는 방법.
  7. 데스로라타딘 및 리멕솔론을 포함하는 약제학적 유효량의 조성물을 투여하는 것을 포함하는, 포유동물에서 비염을 치료하는 방법.
