Claims (6)
- 신호서열(signal peptide)을 포함하는 락토페린 유전자가 알콜 옥시다아제(alcohol oxidase) 프로모터(promoter)의 조절을 받도록 재조합 발현벡터를 제조하는 단계;상기 재조합 발현벡터를 메탄올자화효모(Methylotrophic yeast)인피치아 패스토리스(Pichia pastoris)에 형질 전환시켜 재조합 효모를 얻는 단계; 및상기 재조합 효모를 발효배양하는 단계로 이루어짐을 특징으로 하는피치아 패스토리스를 이용하여 인간 락토페린을 생산하는 방법.
- 제1항에 있어서, 상기 재조합 발현벡터가 pDBThLf-3KS임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 재조합 효모가피치아 패스토리스SM5-3KS(KCTC 10409BP)임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 재조합 효모의 발효배양은 BMGY(1%의 효모 추출액, 2%의 펩톤, 100㎖의 인산칼륨 완충액(pH 6.0), 1.34%의 YNB, 4×10-5%의 비오틴(Biotin), 1%의 글리세롤/ℓ) 배지에서 종배양한 후, 동일 배지에서 본배양을 수행함을 특징으로 하는 방법.
- 제1항의 방법에 의해 제조된 인간 락토페린.
- 피치아 패스토리스SM5-3KS(KCTC 10409BP).
