Abstract
Description
Claims (16)
- (a) 리포터 유전자(reporter gene)에 기능적으로 연계된 신호-반응성 인자를 포함하는 인디케이터 세포;(b) 인캡슐화(encapsulating) 물질; 및(c) 투과성 멤브레인을 포함하고,상기 인디케이터 세포는 상기 인캡슐화 물질 내에 인캡슐화되고 상기 인캡슐화 물질 및 상기 인디케이터 세포는 상기 투과성 멤브레인 내에 함유되는, 인캡슐화된(encapsulated) 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 인디케이터 세포는 배아 줄기 세포 또는 골수 줄기 세포를 포함하는 인캡슐화된 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 인디케이터 세포는 인간 세포를 포함하는 인캡슐화된 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 인디케이터 세포는 마우스 세포 또는 돼지 세포를 포함하는 인캡슐화된 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 리포터 유전자는 β-갈락토시다제, 녹색 형광 단백질, 또는 루시퍼라제를 코딩하는 인캡슐화된 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 인캡슐화 물질은 알기네이트, 콜라겐, 젤라틴, 키토산, 폴리락트산, 폴리글리콜산, 또는 폴리락타이드/글리콜라이드 공중합체를 포함하는 인캡슐화된 세포 인디케이터 시스템.
- 제 1 항에 있어서, 상기 투과성 멤브레인은 폴리에틸렌 테레프탈레이트 멤브레인, 나일론 메쉬, 다공성 나일론 멤브레인, 또는 다공성 폴리테트라플루오로에틸렌(PTFE/테플론)을 포함하는 인캡슐화된 세포 인디케이터 시스템.
- 다음 단계를 포함하는, 배양 중인 세포의 분화 상태를 결정하는 방법:(a) (ⅰ) 리포터 유전자(reporter gene)에 기능적으로 연계된 신호-반응성 인자를 포함하는 인디케이터 세포;(ⅱ) 인캡슐화(encapsulating) 물질; 및(ⅲ) 투과성 멤브레인을 포함하고, 상기 인디케이터 세포는 상기 인캡슐화 물질 내에 인캡슐화되고 상기 인캡슐화 물질 및 상기 인디케이터 세포는 상기 투과성 멤브레인 내에 함유되는 인캡슐화된(encapsulated) 세포 인디케이터 시스템 및 세포의 군집을 제공하는데, 여기에서 상기 리포터 유전자의 발현은 상기 세포의 군집의 분화 상태와 관련되고; 그리고(b) 상기 세포의 군집의 생존능력의 실질적 손실을 야기하지 않는 방법을 사용하여, 상기 세포가 상기 투과성 멤브레인 내에 함유되는 동안 상기 리포터 유전자의 발현을 측정하는데, 여기에서 상기 리포터 유전자의 상기 발현 레벨은 상기 세포의 군집의 분화 상태의 지표인 단계.
- 제 8 항에 있어서, 상기 인디케이터 세포는 배아 줄기 세포 또는 골수 줄기 세포를 포함하는 방법.
- 제 8 항에 있어서, 상기 인디케이터 세포는 인간 세포를 포함하는 방법.
- 제 8 항에 있어서, 상기 인디케이터 세포는 마우스 세포 또는 돼지 세포를 포함하는 방법.
- 제 8 항에 있어서, 상기 리포터 유전자는 β-갈락토시다제, 녹색 형광 단백질, 또는 루시퍼라제를 코딩하는 방법.

- 다음 단계를 포함하는, 환자의 심장 조직으로 이식하기 위한 골수 줄기 세포를 제조하는 방법:(a) (ⅰ) 리포터 유전자(reporter gene)에 기능적으로 연계된 신호-반응성 인자를 포함하는 인디케이터 세포;(ⅱ) 인캡슐화(encapsulating) 물질; 및(ⅲ) 투과성 멤브레인을 포함하고, 상기 인디케이터 세포는 상기 인캡슐화 물질 내에 인캡슐화되고 상기 인캡슐화 물질 및 상기 인디케이터 세포는 상기 투과성 멤브레인 내에 함유되는 인캡슐화된(encapsulated) 세포 인디케이터 시스템 및 상기 환자로부터 유래된 골수 줄기 세포의 군집을 제공하는데, 여기에서 상기 리포터 유전자의 발현은 상기 골수 줄기 세포의 분화 상태와 관련되고;(b) 상기 골수 줄기 세포가 심장 세포로 분화되도록 유도하는 조건 하에서 상기 인캡슐화된 세포 인디케이터 시스템 및 상기 골수 줄기 세포의 군집을 배양하고;(c) 상기 세포의 군집의 생존능력의 실질적 손실을 야기하지 않는 방법을 사용하여, 상기 세포가 상기 투과성 멤브레인 내에 함유되는 동안 상기 리포터 유전자의 발현을 측정하고; 그리고(d) 상기 리포터 유전자의 발현이, 상기 골수 줄기 세포의 적어도 일부가 심장 세포로 분화되도록 유도되는 것을 나타낼 때, 상기 골수 줄기 세포를 수집하는 단계.
- 제 15 항에 있어서, 상기 투과성 멤브레인은 투명 또는 반투명인 방법.
