Abstract
본 발명은 비독성이지만 소화관에서 일반 순환계로 이동할 수 있는 변형 보툴리눔 독소를 이용하여 일반 순환계로 항원 또는 치료제를 전달하는 조성물 및 방법에 관한 것이다.
Description
클로스트리듐 속의 신경독소는 공지된 강력한 단백 독소이다. Clostridium tetani (파상풍 독소)에 의해서 생산되는 신경독소는 개방된 상처에 의해서 인간에서 발병한다. 그러나 적어도 선진 국가에서는 파상풍에 의한 감염은 안전하고 효과적이며 값싼 백신이 보급됨으로 인하여 더 이상 주요한 공중 보건의 문제가 아니다. 이 백신은 기본적으로 발효조에서 성장한 C. tetani에서 유래된 포르말린으로 불활성화된 배양 상청액이다.
유기물인 Clostridium botulinum, Clostridium butyricum 및 Clostridium baratii에 의해서 생산되는 보툴리누스 신경독소(BoNT)는 보툴리누스 질병과 관련된 강력한 병인학적 물질이다(Simpson, L. Annu. Rev. Pharmacol. Toxicol. 1986 26:427-453). 인간은 가끔 상처에 의해 보툴리누스 중독이 발생하기는 하지만, 보통은 식중독에 의해서 이 신경독소에 노출된다. 보툴리눔 독소 중독을 예방하기 위해서 파상풍 백신과 유사한 백신이 개발되어 왔다. 그러나 보툴리눔 독소는 7가지의 다른 혈청형이 존재하기 때문에, 불활성화된 독소로 완벽한 예방을 하는 것은 7가지의 독특한 백신을 만들어서 이들을 혼합하여 투여하는 것에 의해서만 가능하다. 현재, 상기 7가지 혈청형 중 5가지만이 보툴리누스 독소 백신에서 존재한다. 또한, 혈청형의 일부는 배양액에서 고도의 독소를 생산하지 않는 균주로 이루어져있다. 따라서, 완전한 활성의 독소를 취급하는 것과 관련된 높은 위험성 때문에 백신을 위한 독소의 성장, 정제, 및 불활성화의 과정은 시간이 소모되고 비용이 들게 된다(Clayton 등, Infection and Immunity 1995, 63(7):2738-2742). 이때, 백신은 1차 실험 용도로 Center of Disease Control에 의해서만 얻을 수 있다.
통상적으로 보툴리누스 감염은 독소에 의한 감염된 식품을 섭취하거나, 소화관에서 독소를 생산할 수 있는 유기물에 의해 오염된 식품을 섭취하여 발생된다. 경로와는 상관없이 보툴리눔 독소는 약 150 kDa의 분자량을 가지는 상대적으로 비독성인 단일 사슬의 폴리펩티드로 합성된다. 완전한 중독이 되기 위해서는 분자는 프로테아제에 의해 분해되어 H 사슬(Heavy chain: 약 100,000 달톤)이 디설피드 결합에 의해 L 사슬(Light chain: 약 50,000 달톤)에 연결되는 디체인 구조를 만드는 번역후(posttranslational) 처리를 거쳐야만 한다. 상기 디체인 분자는 생물학적 활성에 작용하는 홀로톡신(holotoxin)이다. BoNT는 소화관에서 일반 순환계(림프 및 혈액)로 이동하여 독소 작용의 표적 자리인 콜린계 신경 말단에 작용을 미친다. 이 독소는 신경에 침입하여 아연-의존성 엔도프로테아제로 작용하여 토세포작용(exocytosis)에 필수적인 폴리펩티드를 분해한다(Montecucco, C. and Schiavo, G. Mol. Microbiol. 1994, 13:1-8). 이들 폴리펩티드의 분해는 전달물질의 분비를 방해하고 마비를 초래한다.
독소의 H 사슬은 콜린계 신경 말단의 외부에서 내부로 독소를 결합시키고 이동시키는데 하는데 필수적인 것으로 알려져 있다. 또한 독소의 L 사슬은 콜린계 신경 말단을 중독시키는 독소의 능력에 작용하는 아연 의존성의 엔도프로테아제 활성을 가지는 것으로 알려져 있다(Neimann 등, Behring Inst. Mitt. 1991 89:153-162). 따라서, Clostridium botulinum 유래의 비독성 50 kDa 카르복시기 단말의 분획을 포함하는 보툴리누스 독성에 대한 백신은 공개되어 있다. LaPenotiere 등, Toxicon 1995 33(10): 1383-6 및 Clayton 등, Infection and Immunity 1995 63(7):2738-2742. 또한, 고도로 선택된 신경독소 및 파상풍 독소가 중추 신경계에 약물, 호르몬, 효소, 또는 항바이러스 물질을 전달하는데 작용할 수 있는 비독성의 치료학적 수단으로 전환될 수 있다는 것이 제안되어 있다.
본 발명은 경구투여용 백신 및 치료제를 변형 보툴리눔 독소에 의해 전신으로 전달하는 조성물 및 방법에 관한 것이다. 상기 독소는 소화관 벽을 통과하여 이동하는 능력을 유지하지만 비독성으로 변성된다.

다음의 실시예는 본 발명을 설명하기 위해서 제공된 것으로 본 발명은 제한하고자 하는 것은 아니다.
본 발명의 목적은 소화관에서 일반 순환계로 이동할 수 있는 능력을 유지하지만 비독성인 변형 보툴리눔 독소를 제공하고자 하는 것이다. 상기 변형 보툴리누스 독성은 보툴리누스 독성을 포함하는 항원 펩티드의 경구용 백신 및 기타 치료제를 경구투여하여 일반 순환계를 전달시키기 위한 경구용 백신으로 사용될 수 있다.
오늘날 약품에 대한 주요한 도전 중 하나는 경구 경로에 의해서 투여될 수 있는 약제를 개발하는 것이다. 전신의 면역을 유발시키는 경구용 펩티드 백신의 개발은 특히 해결하기 어려운 것으로 알려져 있다. 경구용 펩티드 백신의 개발과 관련된 어려움은 인간의 소화관에서 발견되는 낮은 pH 조건 및 단백질 분해 효소에 노출됨에 의해 분해되고; 병을 유발하는 약제의 항원 도메인이 너무 커서 소화관의 관강(lumen)에서 일반 순환계로 비특이적 확산을 할 수가 없고; 또한 소화관에서 수용체에 결합하여 일반 순환계로 능동 수송할 수 있는 펩티드 백신을 구상할 수 없다는 점을 포함한다. 이러한 어려움에도 불구하고, 상당한 노력이 경구용 백신을 위한 연구에 바쳐지고 있다. 예를 들면 감자 또는 바나나와 같은 유전공학적 식품을 대규모의 백신화를 위한 벡터로 사용하는 접근이 최근에 제안되었다. 그러나 항원 펩티드를 섭취하고자 하는 식품에 유전공학 처리하는 것은 이러한 문제를 해결하지는 못한다. 따라서 신뢰성이 있고 계속하여 항원 펩티드 또는 기타 치료제를 소화관에서 일반 순환계로 이동시킬 수 있는 약물 전달 담체가 요구다.
본 발명은 보툴리눔 독소 및 기타 치료제를 포함하는 항원 펩티드를 일반 순환계로 전달하는 경구용 전달 담체로 사용할 수 있는 변형 보툴리눔 독소를 제공한다. 보툴리눔 독소가 단일막에서 성장한 극성화된 소화관 세포의 점막 측에 있는 혈청특이적인(serospecific) 수용체에 결합하여 소화관에서 일반 순환계로 이동한다는 것을 발견하게 되었다. 결합된 독소는 세포를 통과하여 능동적으로 이동하고 그대로 전달되고 단일막의 융모막 측에서 변형되지 않는다. Clostridium에 의해서 분비되는 보툴리눔 독소를 포함하는 단백질의 비공유 복합체의 성분인 적혈구 응집소(hemagglutinin)와 같은 보조 단백질이 소화관 벽을 통과하는 독소의 결합 및 이동을 조절할 수 있다는 것이 제안되었다. 그러나 재조합 형태의 홀로톡신으로 실시된 실험에서는 보툴리눔 독소 자체가 소화관 세포의 표면에서 수용체를 인식하는 결합 도메인을 소유한다는 것을 제안하였다. 또한, 소화관에서 일반 순환계로 이동하는 단백질의 능력은 변성시키지 않고 독소를 비독화하기 위해서 독소의 L 사슬에 대해서 변성이 이루어질 수 있다는 것을 설명하고 있다. 따라서 본 발명의 목적에 의해서 ″변형 보툴리눔 독소″는 소화관에서 일반 순환계로 이동하는 능력을 유지하지만 비독성인 보툴리눔 독소를 의미한다. 보툴리눔 독소를 비독화하는 변성은 L 사슬의 아미노산 서열에 대한 돌연변이 및 L 사슬 또는 이의 일부의 결실을 포함한다. 바람직한 실시예에서 L 사슬의 기질 결합 모티프 또는 아연 결합 모티프에 대해서 돌연변이를 실시하는 것이다. 본 발명에 목적에 의해서 ″비독성″은 콜린계 신경말단을 변형 보툴리눔 독소에 노출시키는 것으로 신경 말단에서 전달물질의 분비가 봉쇄되고 마비가 초래되지는 않는다는 것을 의미한다. 보툴리눔 독소를 비독성으로 변성됨에 의해 독소가 소화관에서 일반 순환계로 이동할 수 있는 능력에 대한 작용은 본 명세서에서 제공되는 교시에 따라 일상적으로 실행할 수 있으므로, 당업자는 본 발명의 변형 보툴리눔 독소를 동정할 수 있을 것이다. 변형 보툴리눔 독소의 정의 내에 포함되는 것은 보툴리눔 독소 이외의 단백질에 대한 선택된 항원 또는 치료제를 추가로 포함할 수 있는 보툴리눔 독소이다.
예를 들면, 조성물은 홀로톡신의 L 사슬의 아연 결합 모티프가 불활성화된 보툴리누스 신경독소를 포함하여 제조된다. 변형된 독소는 홀로톡신이 신경 점막의 봉쇄를 초래하는 능력을 보유하지는 않기 때문이 비독성이지만, L 사슬에 대한 변형이 소화관의 관강을 통과하여 일반 순환계로 이동하는 독소의 분자의 능력에 대해서는 부정적인 작용을 하지 않는다. 바람직한 실시예에서 L 사슬의 아연 결합 모티프를 포함하는 적어도 3가지의 아미노산이 변형된다. 상세하게는 천연 서열의 His(229 자리), Glu(230 자리), 및 His(233 자리)의 아미노산은 SEQ ID NO:1에서와 같이 Gly, Thr, 및 Asn의 아미노산으로 각각 치환되었다. 변형 보툴리눔 독소를 암호화하는 핵산 서열은 SEQ ID NO:2에서 나타나 있다. 이들 아미노산의 치환은 촉매인 아연 및 기타 2가의 양이온과 결합하는 홀로톡신의 능력을 제거하였다.
실험은 단락되지 않거나 또는 단일 사슬의 보툴리눔 독소가 결합하고 소화관의 벽을 이동한다는 것을 도시하며 실행되었다. 따라서 본 발명의 변형 보툴리눔 독소는 단락 자리가 제거된 조성물도 포함한다.
본 발명의 변형 보툴리눔 독소의 생리적 활성은 생체내 독성 실험, 마우스 횡경막의 신경-반횡경막 제제에 대한 실험실내 활성 및 시냅토솜 조산물 제제에서 효소 활성을 통해서 결정되었다. 이들 실험에서, 변형 보툴리눔 독소는 본 명세서에서 변형된 재조합 물질 또는 변형 rBoNT/C로 칭하여 지는 것으로 위치 조절(site-directed) 돌연변이 유발인자를 사용하여 보툴리누스 독소 혈청형 C로부터 엔도프로테아제 활성에 필수적인 홀로톡신의 L 사슬의 아연 결합 모티프를 불활성화시켜서 생성되었다. 그러나 효소를 이용하여 펩티드를 절단하고 결과의 단편을 교차연결하는 당업자에 의해서 일반적으로 실행되는 생화학 기술을 포함하는 다른 펩티드 합성 방법도 사용될 수 있지만 이것으로 제한되는 것은 아니다. 또한, 보툴리누스 혈청형의 구조적 및 기능적 유사성에 따라 당업자는 보툴리누스 혈청형 C 이외의 혈청형으로부터 변형 보툴리눔 독소를 일상적으로 제조할 수 있다. 예를 들면 보툴리눔 독소의 모든 혈청형은 약 150,000의 분자량을 가진 비교적 불활성의 전구체로 합성된다. 각 경우에, 전구체는 프로테아제에 의해 ″단락(nick)″되어 L 사슬(50,000 kDa)과 디설피드 결합에 의해 연결된 H 사슬(100,000 kDa)을 가지는 디체인 분자를 생산한다. 보툴리눔 독소의 모든 혈청형은 H 사슬은 주로 독소를 콜린계 신경 말단에 향하도록 하는 조직-표적 도메인으로 작용하고, L 사슬은 신경 말단의 내부에서 작용하여 전달물질 분비를 방지하는 작용을 하여 콜린계 신경 말단에서 전달물질 분비를 선별적으로 방지하는 작용을 한다. 모든 혈청형의 L 사슬은 아연 의존성의 메탈로엔도프로테아제로 작용하여 전달물질을 분비하는데 필수적인 일군의 폴리펩티드의 하나 이상을 분해하는 것이다. 모든 혈청형에는 효소적 활성에 필수적인 His-Glu-X-X-His(SEQ ID NO: 3)의 아연 결합 모티프가 있다. 상기 결합 모티프를 변형하는 것은 반드시 효소의 활성의 손실을 유발한다. 또한, 아연 결합 모티프의 영역에 해당하는 각 혈청형의 일부에 대한 핵산 및 아미노산 서열의 배열은 아연 결합 모티프에 인접하고 이를 포함하는 영역에서 고도로 동일한 서열을 나타내는 것이다. 따라서 보툴리누스 혈청형 C를 사용한 실험은 전체 혈청형을 대표할 수 있는 것이다.
변형 rBoNT/C 홀로톡신의 생체내 독성 실험은 16주 동안의 모니터링 기간동안 높은 함량(동물당 10㎍을 복강내로 투여)을 투여한 경우에서도 마우스에서 급성 독성이 발생되지 않았다는 것을 나타내었다. 모든 실험 동물에서 현저한 신경독성 또는 분명한 다른 유해한 효과는 관찰되지 않았다. 반대로 100 ng 천연의 BoNT/C를 복강내로 투여한 마우스는 주사후 2 내지 2.5시간 내에 사망하였다.
변형 BoNT/C 홀로톡신의 독성은 마우스의 횡경막 신경-반횡경막 제제의 실험실내 실험에서 천연의 BoNT/C의 독성과 비교되었다. 횡경막 신경-반횡경막 제제에 변형 보툴리누스 독성을 첨가하는 것은 신경근육의 마비(1×10-10M; n=4)를 유발하지 않는 다는 것이 밝혀졌다. 반대로 천연의 BoNT/C의 첨가 (1×10-12M; n=8)는 전달물질의 분비 방지에 의한 마비를 유발하였다(평균±S.E.M. = 152±17 분).
이들 변형 보툴리눔 독소의 면역 반응을 일으키는 능력에 대해서는 경구 투여(p.o.), 피하주사(s.c.)후에 실험을 실시하였다. 임뮤노블롯 분석에 의해서 나타난 바에 의하면, 변형 rBoNT/C 홀로톡신 경구 및 피하주사에 의한 투여 모두에서 전신에서 항원의 생산을 유발하였다. 따라서 본 발명의 변형 보툴리눔 독소는 소화관에서 생존 잔류하는 능력을 유지하여 소화관의 외부로 능동 이동되었다. 또한 이러한 사실은 소량의 무관한 단백질을 포함하는 변형 보툴리눔 독소의 비-동종의 제제를 피하내로 투여하는 것으로 이들 무관한 단백질에 대해서 면역 반응을 유발시킬 수 있다는 것을 발견하여 검증되었다. 그러나 경구 투여는 변형 보툴리눔 독소에 대해서만 항체를 생산하였다.
변형 보툴리눔 독소의 경구 및 피하내의 투여에 의해서 발생된 항원의 예방 효과는 혈청의 중화 실험 및 생체내 독성 실험에서 설명되었다. 투여경로와는 상관없이 변형 보툴리눔 독소로 면역화된 동물에서 얻은 혈청은 과량(∼10,000 LD50)의 천연 BoNT/C를 불활성화시켰다. 유사하게, 생체내의 독성 실험에서 경구 또는 피하내의 투여 경로를 통해 변형 보툴리눔 독소로 면역을 실시하는 것은 천연의 독소의 이후 주사의 작용을 현저하게 감소시킨다. 경구 투여 경로에 의해서 변형 보툴리눔 독소가 제공된 동물은 적어도 3달 동안 혈청에서 탐지가능한 항원을 보유하였다. 또한 변형 보툴리누스 독성을 경구 투여 또는 피하내 투여로 수여받은 동물은 3회의 예방접종후에 3개월동안 천연의 BoNT/C 시도에 대해서 보호되었다.
따라서 이들 실험의 결과에서는 비독성이지만 소화관에서 일반 순환계로 이동하고 보호 항체를 유발시키는 능력을 유지하는 변형 보툴리눔 독소가 본 명세서에서 제공된 교시에 다라서 이루어질 수 있다는 것을 설명하고 있다. 또한, 본 발명의 변형 보툴리눔 독소를 포함하는 조성물은 동물에서 보툴리누스에 대한 경구용 백신으로 효과적인 것이다.
또한, 본 발명의 변형 보툴리누스 독성이 소화관에서 일반 순환계로 이동할 수 있는 능력을 유지하고 있기 때문에, 이들 변형 보툴리눔 독소는 보툴리눔 독소 이외의 단백질에 대한 항원 및 치료제를 일반 순환계로 전달하기 위한 경구투여를 위한 전달 담체로 사용될 수 있다. 변형 보툴리눔 독소를 경구용 백신을 위한 캐리어로 이용할 수 있는 여러 가지 방법에 있다. 예를 들면 L 사슬의 아연 결합 모티프의 불활성화하는 것이 소화관의 외부로 이동하는 독소의 능력에 대해 부정적인 효과를 미치지 않기 때문에, 천연 보툴리눔 독소의 아연 결합 모티프는 다른 단백질의 선택된 항원, 즉 보툴리누스 이외의 단백질과 교체되어서 다른 단백질에 대한 경구용 백신을 생산할 수 있다. 또한 단백질 화학 및 분자 생물학의 잘 공지된 기술을 이용하여 선택된 항원 또는 이의 일부를 변형 보툴리눔 독소에 부착시킬 수도 있다. 결과의 변형 보툴리눔 독소는 비독성일 뿐만 아니라 소화관에서 일반 순환계로 이동할 수 있는 능력을 유지할 수도 있으므로 선택되는 항원은 경구투여되는 경우 일반 순환계에 도달하여 상기 단백질에 대한 전신의 면역 반응을 유발시킬 수 있다. 변형 보툴리눔 독소와 함께 경구투여할 수 있는 백신의 예로는 Bacille Calmette-Guerin, 콜레라, 디프테리아, B형 간염, 홍역, 뇌수막염, 유행성 이하선염, 백일해, 역병, 폴리오, 광견병, 풍진, 파상풍, 장피푸스, 및 황열병의 백신을 포함하나 이것으로 한정되는 것은 아니다. 상기 경구용 백신은 독립적으로 또는 DTP(디프테리아, 파상풍, 백일해)와 같이 혼합하여 투여될 수 있다. 상기와 같은 경구용 백신의 전달능력은 의료인이 확보되지 않은 지역에서는 매우 중요하다. 또한, 본 발명의 경구용 백신은 숙달된 인력의 감소 외에도 주사에 사용된 실린지와 관련된 비용을 감소시킬 수 있고 및/또는 사용된 실리지의 폐기와 관련된 문제를 감소시킬 수 있기 때문에 경제적인 이점을 제공할 수 있다.
본 발명의 경구용 백신은 살균된 생리식염수, 0.1% 젤라틴을 포함한 살균된 식염수, 또는 1.0 ㎎/㎖의 소의 혈청 알부민을 포함한 살균된 식염수와 같은 제약학적으로 수용가능한 캐리어 내에 있는 변형 보툴리눔 독소를 포함하는 것이 바람직하다. 또한, 본 발명의 변형 보툴리눔 독소를 식물에 유전공학처리하여 감자 또는 바나나와 같은 식물에 의해 제조된 식품을 광범위한 백신화의 벡터로 이용할 수 있게 된다. 외래 펩티드를 발현하도록 식물을 유전공학처리하는 방법은 예를 들면 1996년 6월 6일 출원된 PCT/US96/09558호에 잘 개시되어 있다.
본 발명의 변형 보툴리눔 독소는 키메릭 경구용 치료제를 구성하는데 유용하다. 본 발명의 실시예에서, 치료제는 변형 보툴리눔 독소에 연결되어서 다음의 2가지의 군의 경구투여용 물질을 제공할 수 있다: (1) 생물학적으로 안정한 결합을 가진 신규한 약제, 및 (2) 생물학적으로 또는 화학적으로 불안정한 결합을 가진 접합된 프로드러그(prodrug)로, 혈액에 도달하면 캐리어와 해리됨. 키메릭 치료방법의 예는 Lautenslager, G.T. 및 Simpson, L.L., ″Chimeric Molecules Constructed with Endogenous Substance″, Advances in Molecular and Cell Biology, Vol 9, pp 233-262, JAI Press, Inc.(1994)에 개시되어 있다. 예를 들면 치료용 펩티드는 변형 보툴리눔 독소에 부착하여 치환체의 특성을 가진 제제를 만들어서 경구투여될 수 있다. 이러한 경우의 예로는 경구 투여된 혈전분해제의 생성을 들 수 있다. P-selectin과 조직 플라스미노겐 활성 인자(TPA)를 혼합하여 이루어진 용융 단백질은 혈전용해 작용을 발현하고 트롬비에 표적을 한다. 이 키메라는 혈류에 도입되어야만 한다. 그러나 분자 생물학 또는 단백질 화학을 이용하여 '1차' 키메릭 분자는 본 발명의 변형 보툴리눔 독소에 부착될 수 있어서 경구투여에 의해서 일반 순환계에 전달되는 부가적인 장점을 가지는 고도의 키메라를 만드는 것이 가능하다. 다른 예는 경구투여된 항-신생물 형성제(anti-neoplastic drug)를 구성하는데 있다. 하나의 분자의 세포독성 특성을 이용하는 여러 가지 항신생물 형성 약제는 독소에 대해 특이적으로 작용하는 다른 분자의 일부를 용융시킨다. 최근의 예로는 상피 성장 인자(EGF)에 용융된 Pseudomonas exotoxin(PE)의 아미노 말단을 사용하여, EGF-수용체-생산된 암세포에 대한 세포독성제로 사용될 수 있는 키메라 EGF-PE를 생산한다. 상기 키메라를 본 발명의 변형 보툴리누스에 결합하는 것으로 경구투여가능한 고도의 키메라를 형성시킬 수가 있다.
백신 또는 다른 치료제용 캐리어로서 변형 보툴리눔 독소를 사용하는 일반적인 개념은 한가지를 제외하는 인간과 인간외의 동물에 대해서 동일한 것이다. 보툴리눔 독소의 모든 혈청형은 모든 종에서 약물을 위한 캐리어로 동등하게 효과적이지는 않을 것이다. 임상적 실험에서는 인간은 혈청형 A, B, 및 E의 작용에 특히 민감하다는 것을 제안한다. 이것은 이들 3가지 혈청형이 위장관에서 흡수되는 효율과 관련될 것이다. 따라서 혈청형 A, B, 및 E는 인간을 위한 치료제의 바람직한 캐리어가 될 것이다.
반대로, 인간 이외의 대부분의 동물은 혈청형 C에 대해서 특히 민감하다. 이것은 수의학적 약제에서는 인간이외의 동물의 치료제를 위한 바람직한 캐리어는 혈청형 C가 될 것이라는 것을 제안한다. 변형 보툴리눔 독소와 함께 경구투여할 수 있는 동물용 백신의 예로는 아데노바이러스 제2형, Bordetella bronchispetica, 보툴리누스, 칼리시바이러스, Chlamydia psittaci, Clostridium Perfringens C형과 같은 클로스트리듐속의 질병, 코로나바이러스, 디스템버, 말의 뇌척수염, Escherichia coli, 고양이과의 전염성 복막염, 고양이과의 백혈병 바이러스, 고양이과의 범백혈구감소증, 간염, 렙토스피라병, 파라인플루엔자 바이러스, 파보바이러스, 광견병, 비기관염 바이러스, 파상풍을 포함하나 이것을 제한되는 것은 아니다.
New England Biolab(Beverly, MA)로부터 제한 효소 및 DNA 변형 효소를 구입하였다. 발현 벡터 Pqe-30 및 니켈-나이트릴로트리아세트산(Ni-NTA) 아가로스는 QIAGEN(Chatsworth, CA)으로부터 구입하였다. 6×His 친화성 표지에 대하여 특이적인 모노클론 항체(mAb) 또한 QIAGEN으로부터 구입하였다. 안티-신탁신 mAb는 SIGMA(St. Louis, MO)로부터 구입하고 항-BoNT/C 항체는 Centers for Disease Control(CDC, Atlanta, GA)로부터 구입하였다. BoNT/C DNA의 EcoRI 단편을 함유하는 플라스미드 pCL8 및 pCH3은 이미 Kimura et al. BBRC 1990 171:1304-1311에 기재되어 있는 것들이다.
실시예 1
rBoNT/C 홀로톡신 합성용 발현 벡터 제작
도 1은 천연 보툴리눔 독소의 개략도이다. 도 2는 변형 보툴리눔 독소인 rBoNT/C의 개략도이다. 변형 보툴리눔 독소, rBoNT/C에 대한 핵산 및 단백질 서열 각각을 SEQ ID NO: 2 및 SEQ ID NO: 1에 나타내었다.
DNA 단편 분리법, DNA 폴리머라제 I의 크레노 단편(Klenow fragment)을 사용하는 돌출 말단(overhanging end)의 수복 방법, 및 T4 리가제를 사용하는 연결법(ligation)이 당업자들에게 공지되어 있으며, 예를 들어 이러한 내용은 Sambrook 등의 Molecular Cloning: A Laboratory Mannual, 2nd ed.(1989, cold Spring Harbor Laboratory Press, Cold Spring Harbor, NY)에 기재되어 있다. 모든 클로닝 단계 및 발현은 pREP4 리프레서 플라스미드를 함유하는 Escherichia coli 균주 M-15(QIAGEN)에서 수행되었다.
PCR을 사용하여 만들어진 3개의 개별적인 독소 단편으로부터 재조합 변형 보툴리눔 독소를 코딩하는 유전자를 조립하여 벡터 pQE-30에 연결시켜 플라스미드 pQE-TC1을 얻었다. 먼저, pBot C2가 생산되는 순차적인 2 단계를 통해 플라스미드 pCL8로부터 BoNT/C의 아미노 말단 부분을 코딩하는 DNA 단편(단편 I 및 Ⅱ)을 증폭시켰다. 하기의 올리고 뉴클레오타이드 프라이머 쌍을 사용하여 DNA 단편 I(nt 4-689)을 증폭시켰다:

상기 정방향 프라이머는 BoNT/C 서열과 상보적이었으며 1632 위치에 내재성 EcoRI 자리를 함유하였다. 역방향 프라이머 내에 SacI 제한 효소 자리를 개시 코돈의 인접 다운스트림에 삽입하였다. 증폭된 단편 Ⅲ을 EcoRI 및 SacI을 사용하여 분해한 후, EcoRI 및 SacI으로 분해된 플라스미드 pQE-30 내에 개별적으로 클로닝하여 플라스미드 pBot C3을 만들었다. 끝으로, 플라스미드 pBot C2로부터 유래된 EcoRI-EcoRI 단편을 EcoRI 분해된 소의 내장의 알칼리성 포스페이타제에 의하여 탈인산화된 플라스미드 pBot C3으로 삽입하여 플라스미드 pQE-TC1을 만들고 전체 크기의 변형 보툴리눔 독소를 코딩하는 DNA를 제작하였다. DNA 시퀀싱을 통해 모든 PCR 단편을 재분석하였다.
아연 결합 부분을 코딩하는 DNA 분절 내의 KpnI 제한 효소 자리를 조작하여 올리고뉴클레오타이드 프라이머를 설계하였다. 이러한 DNA 분절 내 에 KpnI 제한 효소 자리를 만들면 아연 결합에 필수적인 3 아미노산의 돌연 변이(His229→Gly; Glu230→Thr; 및 His233→Asn)가 가능하게 하며 정상 타입 BoNT/C DNA를 예비 클로닝하지 않고도 변형 보툴리눔 독소를 코딩하는 DNA를 복구할 수 있다. 플라스미드 pQE-TC1으로부터 합성된 재조합 변형 보툴리눔 독소는 아미노 말단에 11개의 부가 아미노산, 즉 Arg-Gly-Ser-His-His-His-His-His-His-Gly-Ser(SEQ ID NO: 10)를 함유하였다.
실시예 2
중성 독소 발현 최적화
변형 rBoNT/C를 코딩하는 구조 유전자 앞에 위치하는 pQE-30 벡터의 서열을 변형시키는데 PCR을 사용하였다. 5'-말단에 10개의 부가 뉴클레오타이드를 함유하는 새로운 정방향 프라이머 5'-CGGTACCATGCCAATAACAATTAACAACTTT-3'(SEQ ID NO: 11) 및 BoNT/C 서열의 892 위치에 BglⅡ 제한 효소 자리를 포함하는 새로운 역방향 프라이머 5'-AGCTATAGATCTATAATAATCCAA-3'(밑줄 친 부분의 뉴클레오타이드는 BglⅡ 제한 효소 자리임)(Kimura et al., Biochem. Biophys. Res. Comm. 1990 171:1304-1311)를 사용하여 rBoNT/C의 아미노 말단 부분을 코딩하는 DNA 단편을 재증폭하였다. 증폭된 단편을 T4 폴리머라제로 처리하고 BglⅡ을 사용하여 절단한 후, 크레노우에 의하여 채워진 pQE-TC1의 BamHI 자리와 BglⅡ 자리 사이에 삽입하여 플라스미드 pQE-TC2를 얻었다.
실시예 3
변형 rBoNT/C 홀로톡신의 발현 및 정제
배양물을 37℃ Lennox L 배양액 내에서 0.6 내지 0.8 A600으로 흔들어 주며 배양하였다. 이소프로필-β-D-티오갈락토피라노사이드를 최종 농도가 1.0 mM이 되도록 첨가하고 5시간 동안 추가 배양하였다. 1 리터의 유도 배양물로부터 얻어진 박테리아를 4℃에서 원심 분리하여 수확하고 300 mM NaCl을 함유하는 pH 7.4의 50 mM 소듐 포스페이트 완충 용액 20 ㎖로 재현탁하였다. 얼음 상에서 Model 60 Sonic Dismembrator(Fisher Scientific, Malvern, PA)를 사용하여 각각 75%의 동력으로 1분씩 2번의 펄스를 가하여 초음파 처리하여 세포 현탁액을 용균시켰다. 용균물을 4℃에서 20,000×g로 30분 동안 원심 분리하였다. 맑은 상청액을 1 ㎖의 조밀한(packed) Ni-NTA 수지와 혼합하고 4℃ 반응기에서 1시간 동안 배양하여 25 ㎖의 칼럼에 넣었다. 30 부피의 세척 완충 용액(50 mM 소듐 포스페이트, pH 6.0, 300 mM NaCl, 25 mM 이미다졸)을 사용하여 칼럼을 세척하였다. 용리 완충 용액(50 mM 소듐 포스페이트, pH 4.5, 300 mM NaCl)을 사용하여 결합 단백질을 용리시켰다. 소듐 도데실설페이트 폴리아크릴아마이드 겔(SDS-PAGE) 상에서 정제 단백질을 분석하였다.
실시예 4
임뮤노블롯 분석(Immunoblot Analysis)
세포 추출물을 임뮤노블롯 분석하여 플라스미드 pQE-TC1으로부터 재조합 변형 보툴리눔 독소의 발현을 조종하는 E. coli의 능력을 조사하였다. 웨스턴 블로팅(Western blotting) 분석용 단백질을 Laemmli, U.K. Nature 1970 22:680-685에 기재된 방법에 따라 10% 폴리아크릴아마아드 겔 상에서 분리하여 나이트로셀룰로스로 옮겨 6×His 친화성 표지를 함유하는 면역 반응성 단백질을 검출하였다. 항-6×His 친화성 표지 mAb 1:2000 희석액 또는 항-BoNT/C 항체를 37℃에서 1시간 동안 1차 항체로 배양하였다. 제조자의 지시(ECL; Amersham Corp., Arlington Heights, IL)에 따라 강화 화학발광체(chemiluminescence)를 사용하여 막을 전개시켰다. IPTG를 사용하여 재조합 단백질 합성을 유도하고 용해된 세포 분액(aliquot)을 SDS-PAGE 상에서 전개시켰다.
항-6×His 표지 또는 항-BoNT/C 항체를 사용하여 웨스턴 블롯 분석한 결과 발현 정도가 매우 낮게 나타났다. 또한, pQE-30 벡터 및 설계된 pQE-TC2의 BamHI 자리에 클로닝된 중성 독소 DNA로부터 유래된 4개의 사이토신 뉴클레오타이드를 함유하지 않는 새로운 플라스미드를 제작하였다. 항-6×His 표지 항체를 사용하여 웨스턴 블롯 분석한 결과 pQE-TC2가 변형 rBoNT/C 홀로톡신의 합성을 조종에 있어 보다 효과적인 것으로 나타났다. 사실, 1 내지 2 mg의 변형 rBoNT/C 홀로톡신은 1ℓ의 Lennox 배양액으로부터 정제될 수 있었다.
변형 rBoNT/C 홀로톡신을 가시적으로 분해하지 않고 가용성 형태로 합성하였다. 그러나, Clostridium botulinum과 달리, E. coli는 변형 rBoNT/C 홀로톡신을 효과적으로 절단하지 못하였다. 변형 rBoNT/C 홀로톡신 내에서는 쿠마시 염색 또는 웨스턴 블롯팅에 의하여 극소량의 L-사슬만이 검출 가능하였다. 그러나, 변형 rBoNT/C 홀로톡신은 고정된 TPCK-트립신(Pierce, Rockford, IL)에 의하여 효과적으로 절단되었으며 정확한 분자량의 H 사슬 및 L 사슬을 생산하였다. pQE-TC2로부터 합성된 변형 rBoNT/C 홀로톡신은 아미노 말단에 14개의 부가 아미노산(Arg-Gly-Ser-His-His-His-His-His-His-Gly-Ser-Gly-Thr(SEQ ID NO: 13))을 함유하였다. 이러한 14 아미노산 분절 내의 6×His 서열은 합성된 단백질의 정제 및 검출에 사용되었다. 이러한 방법으로 생성된 재조합 단백질은 6×His 친화성 표지를 사용하는 Ni-NAT 수지 상에서의 친화성 크로마토그래피를 사용하여 정제하였다. pH(용리 완충 용액 pH 4.5)를 낮춰 특이적으로 결합된 단백질을 용리시켜 SDS-PAGE 상에서 분석하였다. 친화성 수지로부터 용리된 단백질을 분석하여 독소가 80 내지 90%의 순도(homogeneity)로 정제될 수 있음을 발견하였다. 본 명세서에 기재된 모든 실험에 정제된 변형 재조합 BoNT/C 또는 변형 rBoNT/C를 사용하였다.
실시예 5
재조합 단백질의 생물학적 분석
하기 실시예에 기재된 바와 같이 생체내 독성 테스트, 생쥐의 횡경막 신경-반격막(phrenic nerve-hemidiaphragm) 제조에 대한 시험관내 활성, 및 천연 시냅토솜(synaptosome) 제조 시의 효소 활성을 사용하여 정제된 재조합 단백질의 생물학적 활성을 분석하였다.
A. 생체내 독성 테스트
변형 rBoNT/C 홀로톡신의 독성을 테트하였다. 히스티딘 친화성 수지로부터 용리 정제된 변형 rBoNT/C 홀로톡신을 1 mg/㎖의 BSA를 포함하는 PBS로 희석하여 생쥐에 복강내(i.p.) 주사하였다. 100 ㎕의 PBS-BSA 분액 내의 rBoNT/C 홀로톡신을 25 g의 평균 중량을 가지는 동물 당 10㎍의 농도로 투여하였다. 전체 16주 동안 동물들을 관찰하여 비특이적 독성을 배제하였다.
B. 시험관내 독성 테스트
Simpson et al. J. Pharmacol. Exp. Ther. 1990 254:98-103에 기재된 방법을 사용하여 생쥐의 횡경막 신경 반격막 제조에 대한 독성을 생물학적으로 분석하였다. 조직을 절제하여 95%의 O2및 5%의 CO2를 포함하며 35℃를 유지하는 생리 완충 용액으로 현탁하였다. 생리 용액은 하기 조성물을 포함한다(mM 단위):NaCl, 137; KCl, 5; CaCl2, 1.8; MgSO4, 1.0; NaHCO3, 24; NaH2PO4, 1.0; D-글루코스, 11; 및 젤라틴, 0.01%. 횡경막 신경을 연속적으로 자극하고(1.0 Hz; 0.1 내지 0.3 밀리초 동안 지속) 근경련을 기록하였다. 신경원성 자극에 대한 근경련 반응 시의 50%로 환산하여 독소에 의하여 유도되는 마비 증상을 측정하였다.
C. 기질 절단
Rosahl et al. Cell 1993 75:661-670에 기재된 방법에 따라 시냅토솜(1 mg/㎖)을 제조하였다. 37℃ Tris-완충 염수(TBS) 또는 10 mM의 디티오트리톨(dithiothreitol)을 함유하는 TBS 내 변형 rBoNT/C 홀로톡신(100 nM)의 존재 하에서 시냅토솜을 90분간 배양하였다. 대조 실험 시, 천연 BoNT/C의 존재 하 및 부재 하에서 시냅토좀 막을 배양하였다. 15% SDS-PAGE 상에서 단백질을 분리하고 나이트로셀룰로스로 옮겨 안티-신탁신(anti-syntaxin) mAb를 사용하여 면역 반응성 단백질을 검출하였다.
실시예 6
변형 rBoNT/C 홀로톡신으로 면역 접종된 생쥐 내에서의
혈청 항체 반응
대조 실험에서는 대략 25 g 중량의 스위스-웹스터 여자 생쥐(Ace Animals, Boyertown, PA)에 rBoNT/C 홀로톡신 또는 TBS를 s.c. 또는 p.o. 중 하나로 면역 접종하여 혈청 면역 반응을 일으키는 이러한 펩타이드의 활성을 측정하였다.
A. 면역 접종 및 샘플 수집
s.c. 주입 시, 각 동물에는 0.1 ㎖의 용리 완충 용액 내의 2 ㎍ 단백질이 투여되었다. 경구 투여 시에는 각각의 동물에 위장내 주입 니들(intragastric feeding needle)을 통하여 0.2 ㎖의 용리 완충 용액 내의 4 ㎍의 단백질을 투여하였다. 당일에 생쥐를 면역 접종하고 14, 28, 및 42일째에 예방 접종하였다. 면역 접종 후 21, 35, 및 49일째에 동일하게 면역 접종된 생쥐로부터 혈청 샘플을 수집하여 혼합하였다. 혈청 수집을 위하여 이소푸란 마취 상태에서 헤파린화 모세관을 사용하여 레트로-오비탈 망상 조직(retro-obital plexus)에서 생쥐의 혈액을 채취하였다.
B. 항체 제조용 혈청 분석
절단되지 않은 변형 보툴리눔 독소에 대한 면역 반응성을 임뮤노블롯 분석하여 면역 접종된 생쥐 또는 대조 생쥐로부터 얻어진 혈청의 항체를 분석하였다. SDS-PAGE를 사용하여 재조합 항원(변형 보툴리눔 독소; 0.1 ㎍/lane)을 분리하여 나이트로스셀룰로스 막으로 옮겼다. 막을 5%의 탈지 분유 TBS로 차단하고 스트립으로 분획하여 다양한 혈청 샘플을 사용해 면역 반응성 단백질을 검출하였다. 하룻밤 동안(18시간) 실온에서 1:1000 희석 혈청을 사용하여 1차 배양하였다. 2차 호스래디쉬 퍼옥시다제-표지된 항-생쥐 IgG는 실온에서 1시간 동안 1:10,000 희석액에서 배양되었다. 광범위하게 세척한 후, ECL(Amersham)을 사용하여 막을 전개시켰다.
실시예 7
면역 접종된 생쥐로부터 유래된 혈청의 중화 활성
다양한 혈청 샘플의 천연 BoNT/C 중화 활성을 실험으로 측정하였다. 다음과 같은 3개의 상이한 혈청원을 테스트하였다:
1) 비면역 혈청;
2) 변형 rBoNT/C 홀로톡신 p.o.가 취해진 동물로부터 얻어진 혈청; 및
3) 변형 rBoNT/C 홀로톡신 s.c.가 취해진 동물로부터 얻어진 혈청.
37℃에서 1시간 동안 10 ㎕의 예비 면역 혈청 또는 면역 혈청 또는 PBS-BSA를 사용하여 천연 BoNT/C(10 ㎕, 100 ng)를 배양하였다. 이어서, 배양 혼합물을 1 mg/㎖의 BSA를 포함하는 80 ㎕의 PBS로 희석하여 i.p. 주사하였다. 48시간 동안 생쥐를 관찰하여 다양한 혼합물의 잔류 독성을 측정하였다.
실시예 8
천연 BoNT/C 감염에 대한 생쥐의 방어
3차 예방 접종 후 3달째에 rBoNT/C로 면역 접종된 생쥐 한 마리 당 100 ng의 천연 BoNT/C를 i.p. 주사하였다. 5일 동안 감염된 동물의 생존을 관찰하였다.

본 발명은 특정한 실시예를 참고로 하여 기재되었으나, 당업자는 본 발명의 내용 및 범위를 벗어나지 않는 범위에서 본 발명의 다른 실시예 또는 변형이 가능함을 이해할 수 있을 것이다. 첨부된 청구항은 이러한 모든 실시예 및 이에 해당하는 변형을 포함하는 것으로 해석된다.

- 소화관에서 일반 순환계로 이동가능하며 비독성으로 변성된 보툴리눔 독소를 포함하는 변형 보툴리눔 독소.
- 제1항에 있어서,선별된 항원을 추가로 포함하는 변형 보툴리눔 독소.
- 제1항에 있어서,치료제를 추가로 포함하는 변형 보툴리눔 독소.
- 제1항의 변형 보툴리눔 독소 및 제약학적으로 수용가능한 담체를 포함하는 경구용 보툴리누스 중독 백신.
- 제2항의 변형 보툴리눔 독소 및 제약학적으로 수용가능한 담체를 포함하는 선별된 항원에 대한 경구용 백신.
- 제3항의 변형 보툴루눔 독소를 동물에 투여하는 단계를 포함하는 동물에서 치료제의 경구 전달 방법.
