This culture was performed at 30°C and 250 rpm in 50 mL of SCD (Leu-) medium with 10 g/L of glucose and 10 g/L of xylose in a 250 mL flask. In batch culture, a pre-culture was prepared by culturing at 30°C and 250 rpm in 3 ml of YP medium containing 20 g/L of glucose. Cells cultured overnight were harvested and inoculated into fresh YP medium at an initial OD 600 of ~1.0. This culture was performed at 30°C and 250 rpm in 50 mL of YP medium with 10 g/L of glucose and 10 g/L of xylose in a 250 mL flask.

A Rezex ROA-Organic Acid H+ column (Phenomenex, Torrance, USA) was used with 5 mM sulfuric acid at a flow rate of 0.6 mL/min at 60 °C. After harvesting the culture medium, it was centrifuged at 13,000 rpm for 5 minutes. Then, the supernatant was obtained and filtered using a 0.22 μm syringe filter to measure the concentration of metabolites.
MAA 분석MAA analysis

The concentration of MAA was measured using high-performance liquid chromatography (HPLC, SCL-40, Shimadzu, Kyoto, Japan) equipped with a variable wavelength detector (VWD) at 344 nm. 100% Solvent A (Solvent A: 0.1 M triethylammonium acetate in water, pH 7.0) for 3 min, rising from Solvent A to 50% Solvent B (Solvent B: acetonitrile) for 12 min, 2 min. The composition for producing shinorine according to claim 1, wherein the microorganism is a microorganism of the genus Saccharomyces or Escherichia genus.
The method of claim 1, wherein the microorganism is further transformed with a gene encoding one or more proteins selected from the group consisting of dihydroquinate synthase, O-methyltransferase, and ATP-grasp ligase, for producing shinorine. Composition.
The method of claim 4, wherein the gene encoding the dihydroquinate synthase includes the base sequence of SEQ ID NO: 2, the gene encoding the O-methyltransferase includes the base sequence of SEQ ID NO: 3, and the ATP - A composition for producing shinorine, wherein the gene encoding the graft ligase includes the base sequence of SEQ ID NO: 4.

The method of claim 6, wherein the gene encoding the xylose reductase includes the base sequence of SEQ ID NO: 5 or SEQ ID NO: 6, and the gene encoding the xylitol dehydrogenase includes the base sequence of SEQ ID NO: 7 , A composition for producing shinorine, wherein the gene encoding the xylulokinase includes the base sequence of SEQ ID NO: 8.
The composition for producing shinorine according to claim 4, wherein the microorganism has a TAL1 (transaldolase) gene removed, deleted, attenuated, or inactivated.
A method for producing shinorine comprising the step of reacting mycosporine glycine with the composition of any one of claims 1 to 8.
The method of claim 9, further comprising the step of separating or purifying shinorine from the reaction solution.
A method for producing shinorine comprising the step of reacting xylose or glucose with the microorganism of any one of claims 4 to 8, a culture or pulverized product of the microorganism.
The method of claim 11, further comprising the step of separating or purifying shinorine from the reaction solution.
