Abstract
본 발명은 신규한 크로마논 또는 이의 고리열림 형태인 페닐프로페논 백본 화합물 및 이를 유효성분으로 포함하는 다발성골수종의 예방 또는 치료용 조성물에 관한 것이다. 본 발명의 화합물은 다양한 다발성골수종 세포주에 대해 현저한 사멸효과를 보일 뿐 아니라 다발성골수종 및 골수이형성증후군 치료에 널리 사용되고 있는 상용화된 면역조절제인 레날리도마이드(lenalidomide)를 상회하는 인 비보 치료항암 효과를 보인다. 아울러, 본 발명의 화합물은 탈리도마이드(thalidomide)나 이의 유사체인 레날리도마이드와 병용 투여시 현저한 상승 효과를 나타내어 난치성 질환인 다발성골수종에 대한 효율적인 치료제 또는 치료 보조제 조성물로 유용하게 이용될 수 있다.
Description
본 발명은 크로마논 또는 이의 고리열림 형태인 페닐프로페논 백본을 가지는 신규 화합물 및 이를 포함하는 다발성골수종을 비롯한 골수암의 예방 또는 치료용 조성물에 관한 것이다.
다발성 골수종은 백혈병, 림프종과 함께 대표적인 혈액종양으로 B림프구의 성숙형태인 형질세포가 증식하는 혈액암이다. 평균 65세 이상의 고령에서 발병하며, 우리나라를 비롯한 동아시아에서는 비교적 낮은 발병률을 보였으나, 2000년대 이후 공해, 환경호르몬 노출 증가 및 고령화 등으로 그 발병율이 가장 급격하게 증가 중인 암종의 하나이다. 다발성골수종은 멜팔란(melphalan), 프레니손(prednisone) 요법을 근간으로 치료가 시도되어 왔으나 면역조절제인 탈리도마이드(thalidomide) 및 레날리도마이드(lenalidomide)와 프로테오좀 억제제인 보르테조밉(bortezomib) 계열의 항암제가 임상 현장에서 널리 이용되고 있다. 그러나, 이들 약물은 정상 조혈모세포를 파괴하거나 이의 기능을 억제하여 범혈구감소증 외에도 말초신경염, 색전/혈전증, 이치성 원발암의 발병 등 심각한 부작용을 야기한다. 뿐만 아니라 다발성골수종은 약물 내성을 증가와 함께 사망률 또한 크게 증가하고 있다.
이에, 다발성골수종의 성공적 치료와 환자의 생존율 개선을 위해서는 상기 언급한 부작용이 획기적으로 경감된 신규한 항암제의 개발이 요구되고 있다. 따라서, 본 발명은 다발성골수종세포에 대한 현저한 사멸효과를 보이는 신규한 저분자 화합물을 발굴하고 이의 항암 효과를 세포주와 동물 실험을 통해 다각적으로 확인하는 한편, 정상인 공여자 유래 말초혈액 조혈모세포군에 대한 유의한 독성이 나타나지 않음을 관찰함으로써 난치성 혈액암인 다발성골수종에 대한 효율적인 치료 조성물을 제공하고자 하였다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 다발성골수종을 비롯하여 형질세포(plasma cell)의 비정상적인 증식을 원인으로 하는 난치성 혈액암에 대한 효과적이면서도 부작용 없는 저분자 화합물 치료제를 개발하기 위하여 예의 연구 노력하였다. 그 결과, 상기 화학식 1로 표시되는 크로마논 또는 페닐프로페논 화합물이 다양한 다발성골수종 세포주에 대해 선택적인 사멸효과를 보이고 인 비보에서 상용화된 약물을 상회하는 현저한 치료 효과를 보인다는 사실을 발견함으로써, 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 하기 화학식 1로 표시되는 신규한 크로마논 또는 페닐프로페논 화합물 및 이를 유효성분으로 포함하는 다발성골수종의 예방 또는 치료용 조성물을 제공하는 데 있다
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 하기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용 가능한 염을 제공한다:

상기 화학식에서, R1은 C1-C3 알킬이고; R2는 수소 또는 (n은 0 내지 2의 정수)이며; R3은 수소이고 R4는 (m은 0 내지 2의 정수이고, A1은 비치환되거나 하이드록시, 할로겐, C1-C3 알킬, 니트로 또는 로 치환된 C6-C10 아릴 또는 5원 내지 10원고리의 헤테로아릴)이거나, 또는 R3와 R4는 서로 결합하여 비치환되거나 하이드록시, 할로겐 또는 C1-C3 알킬로 치환된 C6-C10 아릴 또는 5원 내지 10원고리의 헤테로아릴로 치환된 6원고리의 헤테로사이클로알킬 또는 헤테로사이클로알켄을 형성한다.

본 명세서에서 용어 “알킬”은 직쇄 또는 분쇄의 포화 탄화수소기를 의미하며, 예를 들어, 메틸, 에틸, 프로필, 이소프로필 등을 포함한다. C1-C3 알킬은 탄소수 1 내지 3의 알킬 유니트를 가지는 알킬기를 의미하며, C1-C3 알킬이 치환된 경우 치환체의 탄소수는 포함되지 않은 것이다.
본 명세서에서 용어“할로겐”은 할로겐족 원소를 나타내며, 예컨대, 플루오로, 클로로, 브롬 및 요오도를 포함한다. 구체적으로, 본 발명의 할로겐은 브롬이다.
본 명세서에서 용어“헤테로아릴”은 헤테로원자로서 고리 내에 산소, 황 또는 질소를 포함하는 헤테로사이클릭 방향족기를 의미한다. 헤테로원자의 개수는 1-3이며, 구체적으로는 1-2이다. “5원 내지 10원 고리의 헤테로아릴”은 탄소 및 헤테로 원자를 모두 포함하여 고리를 이루는 원자의 수가 5개-10개인 헤테로아릴을 의미하며, 고리는 단일고리(monocycle) 또는 2개 고리(bicycle)일 수 있다.
본 발명의 구체적인 구현예에 따르면, 상기 헤테로아릴은 티오펜, 퓨란, 벤조퓨란 및 퀴놀린으로 구성된 군으로부터 선택된다.
본 발명의 구체적인 구현예에 따르면, 상기 n은 1이고 m은 0이다.
본 발명의 구체적인 구현예에 따르면, 상기 헤테로사이클로알킬은 옥사논(oxanone)이고, 상기 헤테로사이클로알켄은 디하이드로피라논(dihydropyranone)이다.

본 명세서에서 용어“약제학적으로 허용되는 염”은 약학적으로 허용되는 무기산, 유기산, 또는 염기로부터 유도된 염을 포함한다. 적합한 산의 예로는 염산, 브롬산, 황산, 질산, 과염소산, 푸마르산, 말레산, 인산, 글리콜산, 락트산, 살리실산, 숙신산, 톨루엔-p-설폰산, 타르타르산, 아세트산, 트리플루로초산, 시트르산, 메탄설폰산, 포름산, 벤조산, 말론산, 나프탈렌-2-설폰산, 벤젠설폰산 등을 들 수 있다. 적합한 염기로부터 유도된 염은 나트륨 등의 알칼리 금속, 마그네슘 등의 알칼리 토금속, 및 암모늄 등을 포함할 수 있다.
본 발명의 다른 양태에 따르면, 본 발명은 상술한 본 발명의 화합물 또는 이의 약제학적으로 허용 가능한 염을 유효성분으로 포함하는 암의 예방 또는 치료용 조성물을 제공한다.
본 명세서에서 용어“예방”은 질환 또는 질병을 보유하고 있다고 진단된 적은 없으나, 이러한 질환 또는 질병에 걸릴 가능성이 있는 대상체에서 질환 또는 질병의 발생을 억제하는 것을 의미한다.
본 명세서에서 용어“치료”는 (a) 질환, 질병 또는 증상의 발전의 억제; (b) 질환, 질병 또는 증상의 경감; 또는 (c) 질환, 질병 또는 증상을 제거하는 것을 의미한다. 본 발명의 조성물은 종양 세포주의 증식을 억제하고 세포 사멸을 유도하며 종양의 병변부를 감소시킴으로써 종양으로 인한 증상의 발전을 억제하거나, 이를 제거하거나 또는 경감시키는 역할을 한다. 따라서, 본 발명의 조성물은 그 자체로 이들 질환 치료의 조성물이 될 수도 있고, 혹은 항암 효과를 가지는 다른 약리성분과 함께 투여되어 상기 질환에 대한 치료 보조제로 적용될 수도 있다. 이에, 본 명세서에서 용어“치료”또는“치료제”는“치료 보조”또는“치료 보조제”의 의미를 포함한다.
본 명세서에서 용어“투여”본 발명의 조성물의 치료적 유효량을 대상체에 직접적으로 투여함으로써 대상체의 체내에서 동일한 양이 형성되도록 하는 것을 말한다.
본 발명에서 용어“치료적 유효량”은 본 발명의 조성물을 투여하고자 하는 개체에게 치료적 또는 예방적 효과를 제공하기에 충분한 정도로 함유된 조성물의 함량을 의미하며, 이에“예방적 유효량”을 포함하는 의미이다.
본 명세서에서 용어“대상체”는 제한 없이 인간, 마우스, 래트, 기니아 피그, 개, 고양이, 말, 소, 돼지, 원숭이, 침팬지, 비비 또는 붉은털 원숭이를 포함한다. 구체적으로는, 본 발명의 대상체는 인간이다.
본 발며의 구체적인 구현예에 따르면, 본 발명의 조성물로 예방 또는 치료하고자 하는 암은 혈액암이다.
본 명세서에서 용어“혈액암”은 혈액의 구성 성분인 백혈구, 적혈구, 혈소판; 혈액이 생성되는 골수; 또는 면역 체계를 구성하는 림프구, 림프절, 림프관 등의 림프계에 발생한 악성 종양을 의미한다.
보다 구체적으로는 본 발명의 조성물로 예방 또는 치료하고자 하는 혈액암은 골수암이며, 가장 구체적으로는 다발성 골수종(Multiple myeloma)이다.
본 명세서에서 용어“다발성골수종”은 골수에서 면역체계를 담당하는 백혈구의 한 종류인 형질세포(Plasma Cell)가 비정상적으로 분화, 증식하여 발생하는 혈액암을 의미한다.
본 발명의 구체적인 구현예에 따르면, 본 발명의 조성물은 하기 화학식 34로 표시되는 화합물 또는 이의 약학적으로 허용 가능한 염을 추가적으로 포함한다:

보다 구체적으로는 상기 B1은 -NH2이고 B2는 수소이다. 옥텟 규칙(octet rule)에 의할 때, B2가 수소인 경우 가 단일결합임은 자명하다. B1은 -NH2이고 B2는 수소인 화학식 7 화합물은 레날리도마이드(C13H13N3O3, 3-(4-아미노-1-옥소이소인돌린-2-일)피페리딘-2,6-디온에탈리도마이드)이다.
레날리도마이드(lenalidomide)는 다발성골수종 및 골수이형성증후군 치료에 널리 사용되고 있는 면역조절제로서, 혈관신생억제 및 항염증 활성을 가진다. 본 발명에 따르면, 본 발명의 화합물은 다발성골수종에 대한 대표적인 상용화 치료제인 레날리도마이드와 비교하여 동등이상의 치료 효과를 발휘할 뿐 아니라, 레날리도마이드와 병용 투여시 현저한 상승효과(synergistic effect)를 나타내었다. 이에, 본 발명의 화합물은 그 자체로 단독 항암제로 적용될 수 있음은 물론, 레날리도마이드 또는 유사 면역조절제의 효율적인 치료 보조제로 유용하게 이용될 수 있다.
본 발명의 또 다른 구체적인 구현예에 따르면, 상기 B1은 수소이고 B2는 산소이다. 옥텟 규칙(octet rule)에 의할 때, B2가 산소인 경우 가 이중결합임은 자명하다. B1은 수소이고 B2는 산소인 화학식 7 화합물은 탈리도마이드(C13H10N2O4, 2-(2,6-디옥소피페리딘-3-일)-1H-이소인돌-1,3(2H)-디온)이다.
탈리도마이드(thalidomide)는 레날리도마이드의 원형이 되는 유사체로서 역시 2005년 미국에서 레날리도마이드가 승인되기 전까지 다발성골수종의 치료에 오랜기간 사용되어 왔다.
본 발명의 조성물이 약제학적 조성물로 제조되는 경우, 본 발명의 약제학적 조성물은 약제학적으로 허용되는 담체를 포함한다. 본 발명의 약제학적 조성물에 포함되는 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물은 다양한 투약 루트를 통해 투여될 수 있으며, 구체적으로는 비경구로 투여될 수 있고, 보다 구체적으로는 경구, 정맥, 동맥, 피하, 복강, 피내, 근육내, 뇌실내, 척추강내, 흡입, 비강, 관절강내 또는 국소 투여될 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 본 발명의 약제학적 조성물의 바람직한 투여량은 성인 기준으로 0.0001-100 ㎎/kg 범위 내이다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액, 시럽제 또는 유화액 형태이거나 엑스제, 산제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 또 다른 양태에 따르면, 본 발명은 전술한 본 발명의 산화 세륨 나노 복합체 또는 이를 유효성분으로 포함하는 약제학적 조성물을 대상체에 투여하는 단계를 포함하는 염증 또는 자가면역 질환의 예방 또는 치료 방법을 제공한다. 본 발명에서 이용되는 산화 세륨 나노 복합체 및 이를 통해 예방 또는 치료될 수 있는 염증 또는 자가면역 질환에 대해서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
본 발명의 또 다른 양태에 따르면, 본 발명은 상술한 본 발명의 화합물 또는 이의 약제학적으로 허용되는 염을 유효성분으로 포함하는 하기 화학식 34로 표시되는 화합물 또는 이의 약학적으로 허용 가능한 염과의 병용 투여용 항암 조성물을제공한다:

본 발명에서 이용되는 화학식 34 화합물에 및 이를 통해 예방 또는 치료될 수 있는 암종에 대해서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
상술한 바와 같이, 본 발명의 화합물은 레날리도마이드와 병용 투여시 현저한 상승효과(synergistic effect)를 나타내었으므로, 항암 효과를 극대화하고 환자의 생존률을 개선시키기 위해 본 발명의 화합물을 레날리도마이드를 비롯한 프탈이미드계 화합물과 병용투여할 수 있다.
병용투여는 본 발명의 화학식 1 화합물과 화학식 7 화합물이 모두 포함된 단일 제형을 투여함으로써 수행되거나, 또는 각 화합물이 개별적으로 포함된 별개의 제형이 동시에 투여되거나 또는 임의의 순서로 적절한 시간 차를 두고 순차적으로 투여됨으로써 수행될 수 있다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:

(ⅰ) 화합물 B의 합성
2,4,6-트리하이드록시아세토페논(화합물 A) 25.0g(0.135 mol) 및 N,N-디이소프로필에틸아민(DIPEA) 87.129g(0.674 mol)을 MC 1100 ml에 녹였. 용액을 0℃로 냉각시킨 뒤, 클로로메틸메틸에테르(MOMCl) 43.418g(0.539 mol)을 천천히 첨가 후 실온에서 2시간 교반하였다. 얻어진 생성물을 실리카겔 컬럼크로마토그래피에 의해 분리 정제(EA:Hex = 1:4 부피비)하여 1-(2-하이드록시-4,6-bis(메톡시메톡시)페닐)에탄-1-온 (화합물 B) 29.6g(수율 85.7%)를 얻었다.
(ⅱ) 화합물 C의 합성
얻어진 화합물 B 29.6g(0.116 mol)를 아세톤 1200ml에 녹인 후, 프레닐 브로마이드 28.69g(0.193 mol) 및 K2CO3 63.857g(0.462 mol)을 첨가하였다. 플라스크 내부를 Ar 치환하고 8시간 가열 환류하였다. 얻어진 생성물을 실리카겔 컬럼 크로마토그래피에 의해 분리정제(EA:Hex = 1:4 부피비)하여 1-(2,4-bis(메톡시메톡시)-6-((3-메틸부트-2-엔-1-일)옥시)페닐)에탄-1-온(화합물 C) 36.4g(수율 97.1%)를 얻었다.
(ⅲ) 화합물 D의 합성
얻어진 화합물 C 36.4g(0.112 mol)에 N,N-디에틸아닐린 205.4g(1.694 mol)을 첨가하여 플라스크 내부를 Ar 치환하였다. 220℃에서 1.5시간 동안 교반한다. 10% HCl 수용액과 EA를 이용하여 추출하고 MgSO4를 이용하여 수분을 제거한다. 얻어진 생성물을 실리카 겔 컬럼 크로마토그래피에 의해 분리 정제(EA:Hex = 1:4 부피비)하여 1-(6-하이드록시-2,4-bis(메톡시메톡시)-3-(3-메틸부트-2-엔-1-일)페닐)에탄-1-온 (화합물 D) 25.2 g (수율 69.1 %)를 얻었다.
(ⅳ) 화합물 E의 합성
얻어진 화합물 D 25.2g(77.689 mmol)를 DMF 240 ml 중에서 메틸 아이오다이드 27.6 g (0.194 mol)과 K2CO3 21.474 g (0.155 mol)을 첨가하고 플라스크 내부를 Ar 치환한다. 이후 실온에서 24 시간 교반한다. 얻어진 생성물을 물과 EA를 이용하여 추출하고 MgSO4를 이용하여 수분을 제거하였다. 얻어진 생성물을 실리카 겔 컬럼 크로마토그래피에 의해 분리정제(EA:Hex = 1:4 부피비)하여 1-(6-메톡시-2,4- bis(메톡시메톡시)-3-(3-메틸부트-2-엔-1-일)페닐)에탄-1-온(화합물 E) 24.605g (수율 93.6 %)를 얻었다.

얻어진 화합물 B 10.9g(42.537 mmol)를 DMF 131ml 중에서 메틸 아이오다이드 18.1g(0.128 mol)과 K2CO3 17.636g(0.128 mol)을 첨가하고 플라스크 내부를 Ar 치환한다. 이후 실온에서 24시간 교반한다. 얻어진 생성물을 물과 EA를 이용하여 추출하고 MgSO4를 이용하여 수분을 제거한다. 얻어진 생성물을 실리카 겔 컬럼 크로마토그래피에 의해 분리 정제(EA:Hex = 1:4 부피비)하여 1-(2-메톡시-4,6-bis(메톡시메톡시)페닐)에탄-1-온(화합물 C’) 8.930g(수율 77.7 %)를 얻었다.

(ⅰ) 화합물 F0의 합성
1-(6-메톡시-2,4-bis(메톡시메톡시)-3-(3-메틸부트-2-엔-1-일)페닐)에탄-1-온(화합물 E) 500.0mg(1.478 mmol) 및 4-(메톡시메톡시)벤즈알데하이드 270.1mg (1.625 mmol)을 에탄올 8.6ml에 녹였다. 용액을 0℃로 냉각시킨 뒤, KOH 4.145g (73.877 mmol)과 H2O 5.3 ml를 첨가 후 플라스크 내부를 Ar 치환한다. 이후 실온에서 24 시간 교반한다. 생성된 화합물을 EA로 추출한 후, 실리카 겔 컬럼 크로마토그래피에 의해 분리 정제(EA:Hex = 1:3 부피비)하여 (E)-1-(6-메톡시-2,4-bis(메톡시메톡시)-3-(3-메틸부트-2-엔-1-일)페닐)-3-(4-(메톡시메톡시)페닐)프롭-2-엔-1-온(화합물 F0) 443.7mg(수율 61.7%)를 얻었다.
(ⅱ) XN의 합성
얻어진 화합물 F0 443.7 mg (0.912 mmol)를 메탄올 110 ml에 녹인 후, 6N HCl 수용액을 12.1 ml를 첨가한다. 플라스크 내부를 Ar 치환하고 24 시간동안 실온에서 교반한다. 얻어진 생성물을 EA로 추출한 후, 실리카 겔 컬럼 크로마토그래피에 의해 분리 정제(EA:Hex = 1:2 부피비)하여 (E)-1-(2,4-디하이드록시-6-메톡시-3-(3-메틸부트-2-엔-1-일)페닐)-3-(4-하이드록시페닐)프롭-2-엔-1-온(XN) 321.0mg (수율 99.3%)를 얻었다.

(ⅲ) 화합물 F1의 합성
4-브로모벤즈알데하이드 328.1 mg (1.773 mmol)을 합성예 3의 (ⅰ)을 참고하여 합성을 진행하여, (E)-3-(4-브로모페닐)-1-(6-메톡시-2,4-bis(메톡시메톡시)-3-(3-메틸부트-2-엔-1-일)페닐)프롭-2-엔-1-온(화합물 F1) 654.5mg(수율 87.6 %)을 얻었다.
(ⅳ) 화합물 1의 합성
얻어진 화합물 F1 654.5 mg (1.295 mmol)을 합성예 3의 (ⅱ)를 참고하여 합성을 진행하여, ((E)-3-(4-브로모페닐)-1-(2,4-디하이드록시-6-메톡시-3-(3-메틸부트-2-엔-1-일)페닐)프롭-2-엔-1-온(화합물 1) 91.7mg(수율 17.0%)을 얻었다.

벤조퓨란-2-카브알데하이드 324.43 mg (2.220 mmol)을 합성예 6의 (ⅰ)을 참고하여 합성을 진행하여, (E)-3-(벤조퓨란-2-일)-1-(2-메톡시-4,6-bis(메톡시메톡시)페닐)프롭-2-엔-1-온 (화합물 D’4) 536.8 mg (수율 72.8 %)을 얻었다.
(ⅹ) 화합물 19의 합성
얻어진 화합물 D’4 536.8 mg (1.347 mmol)을 합성예 6의 (ⅱ)를 참고하여 합성을 진행하여, (E)-3-(벤조퓨란-2-일)-1-(2,4-디하이드록시-6-메톡시페닐)프롭-2-엔-1-온 (화합물 19) 410.9 mg (수율 98.3 %)을 얻었다.

얻어진 화합물 D’5 276.1 mg (0.579 mmol)을 합성예 6의 (ⅱ)를 참고하여 합성을 진행하여, (E)-1-(2,4-디하이드록시-6-메톡시페닐)-3-(4-이소프로필페닐)프롭-2-엔-1-온 (화합물 20) 265.1 mg (수율 54.8 %)을 얻었다.

실험예
실험방법
인간 다발성골수종 세포주인 U266(ATCC CCL-155), IM-9(ATCC CCL-159), RPMI 8226(ATCC TBI-196) 및 ARH-77(ATCC CRL-1621) 세포주를 ATCC(American Type Culture Collection, Rockville, MD, USA)에서 구입하였다. 세포는 RPMI1640 배양액(Gibco Laboratories)에 10% 열 불활성화 우태아혈청(Gibco Laboratories), 1% 페니실린/스트렙토마이신(Thermo Fisher Scientific)을 첨가하여 사용하였다. 세포는 5% CO2, 37℃ 조건의 항온배양기에서 배양시켰으며 3-4일 간격으로 계대 배양하였다.
실험동물
총 12 마리의 수컷 NSG(NOD scid gamma) 마우스를 사용하였다. 실험동물은 사육상자 당 5마리씩 온도 20-24℃, 습도 40-60%, 명/암주기 12시간으로 조절되는 전남대학교 의생명과학융합센터 준특정병원균부재(Semi-specific pathogen free; Semi-SPF) 동물 실험사에서 사육하였다. 멸균수와 사료를 제한없이 공급하였고, 모든 실험절차는 동물 보호법(Animal welfare act)을 준수하였으며, 전남대학교 의생명과학융합센터 동물실험윤리위원회의 승인 하에 수행하였다(승인번호: CNU IACUC-H-2020-36).
세포생존율 분석
KBB-NX 계열 합성유도물질은 DMSO(Dimethyl Sulfoxide)에 100 mM의 stock으로 만들어 -20℃에 보관하여 배지에 희석하여 사용하였다. 다발성골수종 세포주인 U266, IM-9, RPMI 8226와 ARH-77에 25μM, 50μM, 100μM 동일한 농도의 KBB-NX 계열 화합물을 처리하여 100μL의 배양액에 부유하여 96-웰 플레이트(5x105 cells/well)에 분주한 뒤 24시간 동안 배양하였다. 그 이후 Cell viability Assay Kit (EZ-CyTox, Daeil Lab Service CO., Seoul, Korea)를 이용하여 배양배지에 EZ-CyTox 10μL를 섞어 1시간 동안 5% CO2, 37℃ 조건의 암실에서 반응시켰다.
EZ-CYTOX는 세포의 탈수소효소(Dehydrogenase)와 반응하여 주황색의 수용성 포르마잔(formazan)을 생성함으로써 450nm의 흡광도에서 VERSA Max Micro 플레이트 리더(Molecular Devices, Sunnyvale, CA, USA)를 이용하여 측정하였다. 시험 화합물을 처리한 세포에서 처리되지 않은 세포에 대한 생존세포 백분율 계산식은 다음과 같다: *세포생존율(%)=[OD (KBB-NX 처리 세포)-OD (Blank)] / [OD(세포)- OD(Blank)] x 100
세포사멸분석
KBB-NX를 48시간 처리한 다발성골수종 세포 U266, IM-9, RPMI8226, ARH-77(3 x 106 cells/mL)를 1 x PBS(phosphate-buffered saline, pH 7.4)로 수차례 수세 후 세포가 부유된 1 x PBS 500μL(1x106 cells/mL)에 Annexin V-FITC 5μL와 PI(propidium iodine) 5μL(Sigma-Aldrich, St. Louis, MO) 첨가 후 실온 암실에서 30분간 염색하였다. 이후 1X binding buffer 500μL를 추가하여 FACSCalibur(Becton Dickinson, Franklin Lakes, NJ, USA)로 유세포 검사를 진행 후 CellQuest 소프트웨어(Becton Dickinson) 프로그램을 통해 분석하였다.
세포주기 분석
KBB-NX-16을 48시간 처리한 다발골수종 세포 IM-9(3 x 106 cells/mL)를 수집하고, 1 x PBS로 수세한 뒤, 70% 에탄올로 24시간 고정시켰다. 다음으로 1 x PBS로 수차례 수세 후 세포가 부유된 1 x PBS 500μL(1 x 106 cells/mL)에 0.1% RNase A, PI 25 ㎍/mL를 처리하여 실온 암실에서 30분간 반응시켰다. 이후 FACS Calibur와 Cell Quest software을 이용하여 세포주기를 분석하였다.
세포내 활성 산소 측정
KBB-NX-20을 48시간 처리한 다발골수종 세포 IM-9(3 x 106 cells/mL)를 수집한 후, DCFH-DA(20μM)(Sigma-Aldrich) 용액으로 부유하였다. 이를 37℃에서 30분간 반응시킨 후 유세포분석기로 분석하였다.
웨스턴 블롯 분석
KBB-NX-16과 KBB-NX-20을 50μM를 처리한 다발골수종 세포(1 x 107 cells/ mL)에서 단백질의 추출을 위하여 RIPA 용액(50 mM Tris-HCl, pH7.4, 150 mM NaCl, 1% Triton X-100, 0.1% SDS, 1 mM EDTA, 1 mM Na3VO4, 1 mM NaF, 프로테아제 억제제 칵테일) 50 μL을 첨가한 후 4℃에서 30분간 반응시켰다. 그 후 4℃에서 20분 동안 15,000g에서 원심분리하여 상층액의 단백질을 수집한 후 Bradford Protein assay kit(BioRad, Hercules, CA)를 사용하여 단백질 농도를 정량하였다. 정량 후 25 μg 단백질을 10-12% SDS-PAGE시행 후 300 mA에서 2시간 동안 PVDF 막에 전이시켰다. 단백질이 전이된 막에 5% Non-fat dry milk를 넣은 TBS-T를 이용하여 블로킹한후 일차 항체인 항-PHB2 및 항-β-actin 항체(Cell signaling technology, Danvers, MA)를 4℃에서 18-24시간 반응시켰다. 반응 후 TBS-T로 10분 간격 3회 세척 후 이차 항체와 1시간 반응시킨 후 다시 한번 TBS-T로 10분 간격 3회 세척하였다. 세척 후 막을 ECL 용액(Amersham, USA)로 1분간 반응 하여 감광시키고 밴드의 크기를 비교하여 단백질의 발현 유무와 발현량을 정량화하였다.
세포사멸 관련 단백질 분석
세포사멸과 관련한 단백질 발현은 human apoptosis array kit (R&D systems, MN, USA)를 사용하였다. 35개의 아폽토시스와 관련한 항체가 붙어 있는 니트로셀룰로스 막을 1시간 동안 상온에 블로킹시킨 후, 추출한 단백질 200μg을 4℃에서 12시간 반응시켰다. 반응 후, 1시간 동안 검출 항체 칵테일을 처리하였으며, 스트렙타비딘-HRP를 30분 반응시켜, Chemi Reagent로 가시화하였다.
다발성골수종 이식모델의 구축
마우스 꼬리 정맥에 RPMI8226-GFP-FLUC 세포 주사
6-11 주령 NSG 마우스 12마리를 이용하여 100μL의 1XPBS에 부유된 1 x 107 RPMI8226-GFP-FLUC 세포주(GenTargetInc., CA, USA)를 꼬리 정맥에 주입하였다.
다발성골수종 발생 여부 관찰
마우스에 RPMI8226-GFP-FLUC 세포를 주입하고, 8일 후 생체내 영상 분석장비인 IVIS(In vivo imaging system)(NightOWL Ⅱ LB983 NC100, Berthold, Germany)를 이용하여 암 생성 여부를 확인하였다.
다발성골수종 마우스 모델에 KBB-NX 계열 합성화합물 투여
다발성골수종 마우스 모델에서 KBB-NX-2의 효과를 알아보기 위해 RPMI8226- GFP-FLUC 세포주를 주입하고 2주 후 5mg/kg의 KBB-NX-2를 일주일에 2일 간격으로 3회(3 cycle) 꼬리 정맥 내로 투여하였다. 대조군은 동일 양의 PBS를 투여하였고, 양성대조군은 동일한 양의 레날리도마이드(Sigma-Aldrich)를 투여하였다.
초기 간, 신장 및 심장 독성평가
KBB-NX 계열 합성화합물 중에 초기 독성평가로 KBB-NX-16과 KBB-NX-20을 대상으로 랫드의 대퇴정맥에 5mg/kg 용량을 정맥주사하여 3시간 후, 채혈하여 간독성(표지자 ALT), 신장독성(BUN, 크레아티닌) 및 심장독성 (CK-MB, 미오글로빈) 표지자를 검사하여 평가하였다.
실험결과
도 1 내지 26에서 보는 바와 같이, 본 발명의 화합물은 인간 다발성골수종 세포주인 U266, IM9, RPMI 8226 및 ARH-77에 대해 현저한 사멸효과를 보임을 다각적인 인 비트로 실험을 통해 확인할 수 있었다.
본 발명의 화합물이 다발성 골수종 세포를 사멸시키는 작용 기전(Mode of Action)은 도 27에 나타난 바와 같다. 즉, KBB-NX 화합물은 세포내 PHB2 단백질(미토콘드리아의 내막에 주로 위치)과 결합하여 PHB2 단백 기능을 무력화시켜 세포내 활성산소 발생을 증가시키고 미토콘드리아 기능변이를 야기한다. PHB2에 대한 길항작용을 하는 KBB-NX는 세포 내 ROS를 증가시켜 세포의 대사 스크레스를 초래함으로써 병적 보상기전이 작동하여 세포내 미토콘드리아 과증식을 유도, 과도한 ROS가 세포내에 생성되어 ROS 매개 세포고사가 촉진된다. 세포 내에서 증가된 ROS는 세포고사 경로의 주요 인자인 pERK 및 pJNK를 증가시켜 세포고사를 촉진시키고(proapoptotic effect), SubG1 단계에서 세포 주기를 정지시킨다(antiproliferative effect). KBB-NX 화합물 처리에 의해 PHB2 단백질 감소 - ROS 증가 매개 세포고사가 유도됨을 단백질 어레이를 이용한 분석을 통해 규명하였다(도 27b).
한편, 다발골수종 세포주(RPMI8226)에 EF1a 프로모터 부위에 렌티바이러스 벡터를 이용하여 퓨로마이신 내성 유전자와 루시퍼라제-GFP 유전자를 삽입하여 마우스 내의 MM 세포의 동태를 생체내 생물 발광영상 분석기법으로 파악하고 모니터링하였다(도 28). 생체 내 모니터링이 가능한 상기의 유전자삽입 다발골수종 세포주를 6-11주령의 복합면역결핍 NSG마우스의 꼬리정맥을 통하여 1 x 107/ml 주입하여 다발골수종 마우스모델 구축하였다. 그 결과, 본 발명의 화합물을 투여하지 않은 군에서는 다발골수종세포 이식 후 50일 째 모두 사망한 반면, KBB-NX-2와 레날리도마이드 치료군에서는 각각 64일까지 생존함으로써 KBB-NX 화합물의 다발성골수종 치료 효과를 인 비보에서도 명확히 확인하였다(도 29).
아울러, KBB-NX-16과 KBB-NX-20을 비롯한 본 발명의 화합물들은 초기 급성독성평가에서 유의한 간독성, 신장독성 및 심장독성이 관찰되지 않았다(도 30).
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
