Abstract
본 발명은 생체기반 유기체를 이용한 자기구동 마이크로로봇에 관한 것으로서, 더욱 상세하게는 해체 온도(Td) 이상에서 분리되는 치료용 세포 및 자성 나노입자를 포함하는 마이크로로봇 및 이를 이용한 치료 시스템에 관한 것이다.
Description
본 발명은 생체기반 유기체를 이용한 자기구동 마이크로로봇에 관한 것으로서, 더욱 상세하게는 해체 온도(Td) 이상에서 분리되는 치료용 세포 및 자성 나노입자를 포함하는 마이크로로봇 및 이를 이용한 치료 시스템에 관한 것이다.
종래 질병의 치료를 위해서 약물을 복용하거나 체내에 직접 주입하는 방식이 사용되고 있는데, 이러한 방식은 치료 약물이 병변 부위로 정확하게 도달하지 못하여 치료 효율이 떨어질 뿐만 아니라, 치료 후의 통증 및 흉터를 유발하는 문제가 있다.
이러한 통증 및 흉터를 최소화하고 치료 효율을 향상시키기 위해 최소 침습을 통한 자기구동 마이크로로봇에 대한 연구가 제안되었으나, 치료 후 자기구동 마이크로로봇 내 자성 나노입자가 체내에 남아있게 되어 잠재적으로 세포 독성을 가지며, 완벽한 자연 배출이 보장되지 않는다.
또한, 상기 자기구동 마이크로로봇은 외부 자기장을 통해 병변 부위로 빠른 약물 전달이 가능하지만, 정확한 타겟팅이 쉽지 않아 정상세포까지 약물이 영향을 미쳐 부작용을 일으킬 수 있고, 모세혈관 등의 국부 영역에서는 이미징이 불가능하여 적용이 힘든 문제점을 가진다.
이에, 최근 박테리아 등의 생체기반 유기체 또는 면역세포를 이용한 치료용 마이크로로봇에 대한 연구가 활발히 진행되고 있다. 상기 박테리아 및 면역세포는 병변 부위, 구체적으로 고형암에 화학주성을 가짐에 따라 암을 진단하거나 약물을 담지하여 타겟팅이 가능한 마이크로로봇으로 사용될 수 있다.
구체적인 일예로, 표면에 박테리아가 부착된 치료용 마이크로로봇은 박테리아의 뛰어난 운동성 및 화학주성으로 인해 마이크로로봇이 병변 부위로 이동함에 따라 질병의 치료가 가능하다.
그러나, 이러한 박테리아 기반의 치료용 마이크로로봇을 혈관 주사 투입하는 경우, 박테리아의 추진력이 약해 혈류를 거슬러 이동하기 힘들어 실제 병변 부위로의 타겟팅이 매우 어렵다. 또한, 혈액 내에서 순환되어 병변 부위로 우연히 도착하는 박테리아 기반 마이크로로봇의 양이 최초 투여량 대비 극히 소량이어서 치료 효과가 미미하다. 뿐만 아니라, 박테리아 및 면역세포 자체의 면역 반응 및 안정성 문제로 인해 정상세포까지 영향을 미칠 가능성이 존재한다.
이에 따라, 자기구동에 의한 매크로 타겟팅 및 생체기반 유기체를 이용한 국부 타겟팅을 통한 병변 부위로의 빠르고 정확한 약물 전달이 가능한 동시에, 치료 후에는 자성 나노입자가 체내에 체류하지 않고 완전히 제거될 수 있는, 새로운 마이크로로봇의 개발이 요구되고 있다.
본 발명의 목적은 자기장 구동 및 치료용 세포의 화학주성을 통해 특정 영역에 정확하고 빠른 표적화가 가능한 마이크로로봇을 제공하는 것이다.
본 발명의 다른 목적은 외부 열 발생 장치에 의해 마이크로로봇이 가열되어, 해체 온도(Td) 이상에서 치료용 세포의 선택적인 분리가 가능한 마이크로로봇을 제공하는 것이다.
본 발명의 다른 목적은 자성 나노입자의 회수가 가능하여 인체에 무해하며 안정성이 우수한 마이크로로봇을 제공하는 것이다.
본 발명의 다른 목적은 상기 마이크로로봇을 이용한 자기장 구동 치료 시스템을 제공하는 것이다.
상기 목적을 달성하기 위한 본 발명에 따른 마이크로로봇은 치료용 세포 및 자성 나노입자를 포함하며, 상기 자성 나노입자는 상기 치료용 세포의 표면과 리간드-수용체 결합을 통해 결합되며, 상기 리간드-수용체 결합은 해체 온도(Td) 이상의 온도에서 분리되는 것을 특징으로 한다.
본 발명의 일 양태에 있어서, 상기 치료용 세포는 약물 또는 약물 담지체를 더 포함하는 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 해체 온도(Td)는 인체의 체온 이상인 것을 특징으로 하는 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 리간드 및 수용체에서 선택되는 어느 하나는 단백질 또는 펩타이드를 포함하는 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 치료용 세포는 박테리아 또는 면역세포를 포함하는 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 해체 온도(Td)는 외부 근적외선 또는 외부 교류 자기장의 인가시 자성 나노입자의 발열에 의해 도달되는 온도인 것을 특징으로 하는 것일 수 있다.
본 발명의 또 다른 양태는 자기장 구동 시스템; 마이크로로봇 및 환부를 이미징하기 위한 영상 촬영부; 상기 이미징을 실시간으로 검출하는 검출기; 및 상기 자기장 구동시스템의 제어를 위한 통합제어 모듈;을 포함하고, 상기 마이크로로봇은 생체 내에 투여되어 치료용 세포를 병변 부위에 표적할 수 있는 제1항에 따른 마이크로로봇을 포함하는 자기장 구동 마이크로로봇 치료 시스템이다.
본 발명의 일 양태에 있어서, 상기 자기장 구동 시스템은 근적외선 조사부 또는 교류 자기장 입력부를 더 포함하는 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 영상 촬영부는 양방향 X-ray 또는 MRI 시스템인 것일 수 있다.
본 발명의 일 양태에 있어서, 상기 마이크로로봇은 자기장 구동 시스템에 의해 병변 부위로 이동되고, 병변 부위로 이동 후 근적외선 또는 교류 자기장을 가하여 마이크로로봇의 온도를 리간드-수용체 결합의 해체 온도(Td) 이상으로 가열하며, 치료용 세포로부터 분리된 자성 나노입자는 자기장 구동 시스템에 의해 병변 부위로부터 이송되어 제거되는 것을 특징으로 하는 것일 수 있다.
본 발명에 따른 마이크로로봇은 외부 자기장을 통해 병변 부위로 빠르게 이동이 가능한 동시에 국부 영역에서 치료용 세포의 화학주성에 의한 정확한 표적화가 가능한 장점이 있다.
또한 본 발명에 따른 마이크로로봇은 외부 열 발생 장치에 따른 발열에 의해 해체 온도(Td) 이상에서 분리가 가능하여, 타겟 부위에서 치료용 세포 분리를 통한 약물의 선택적 방출이 가능한 장점이 있다.
또한 본 발명에 따른 마이크로로봇은 분리된 자성 나노입자의 회수가 용이한 장점이 있다.
또한 본 발명에 따른 마이크로로봇은 독성의 유발이 없고 타겟이 되는 부위외의 다른 조직에 대한 부작용을 방지하여 치료 효과를 극대화할 수 있는 장점이 있다.

본 발명에 따른 마이크로로봇은 치료용 세포 및 자성 나노입자를 포함하는 것으로, 상기 자성 나노입자는 치료용 세포의 표면과 리간드-수용체 결합을 통해 결합되며, 상기 리간드-수용체 결합은 해체 온도(decomposition temperature, Td) 이상의 온도에서 분리되는 것을 특징으로 한다.
상기 치료용 세포는 병변에 대한 화학주성을 가지는 것으로, 병변 부위에 대한 마이크로로봇의 자발적인 이동성을 부여할 수 있다.
본 발명의 일 양태에 따른 치료용 세포는 면역 세포 및 박테리아를 포함하는 것일 수 있으며, 이들은 대장암, 유방암 등을 포함한 일반적인 고형암(solid cancer) 및 허혈성 질환의 병변 부위에 대한 화학주성을 가짐에 따라 마이크로로봇의 암세포, 심장 및 뇌의 병변 부위에 대한 우수한 표적지향성을 부여할 수 있으나, 상기 질병 및 치료용 세포로 제한되는 것은 아니다.
상기 면역 세포 종류로서 예를 들면, 매크로파지(Macrophage), 수지상세포(dendritic cell), 자연 살해 세포(natural killer cell) 및 T 세포 등을 들 수 있으며, 상기 박테리아의 종류로서 예를 들면, 살모넬라(Salmonella typhimurium), 대장균(Escherichia coli), 쉬겔라(Shigella), 비피도박테리움(Bifidobacterium), 예르시니아(Yersinia), 리스테리아(Listeria), 마이코플라즈마(Mycoplasma), 스트렙토코커스(Streptococcus) 및 클로스트리디움(Chlostridium) 등을 들 수 있으나, 이에 제한되는 것은 아니다.
바람직하게는 편모를 이용하여 이동 가능한 박테리아를 치료용 세포로 사용하여 마이크로로봇의 운동성 및 추진력을 더욱 향상시킬 수 있다.
또한, 상기 박테리아는 화학주성에 따른 질병의 표적능을 가질 뿐만 아니라, 병변 부위로 이동하여 질병을 치료할 수 있는 치료능을 자체적으로 나타낼 수 있다. 구체적으로, 박테리아는 심근경색, 뇌경색, 고형암 등과 같은 허혈성 지역을 갖는 질환에 대해 추적 및 치료능을 가지고 있다. 또한, 유전자 조작을 통해 치료능을 가지는 박테리아로 조작하는 것도 가능하며, 일 예를 들면, 살모넬라(S. typhiumrium) 균주를 유전자 조작을 통해 박테리아 내의 구아노신 5'-2인산-3'-2인산(guanosine 5'-diphosphate-3'-diphosphate, ppGpp)의 합성을 결손시켜 박테리아 자체의 독성을 줄이는 한편, 세포 용해소 A (Cytolysin A)의 세포 독성 물질을 낼 수 있는 유전자를 삽입하여 암세포를 죽이는 효과도 낼 수 있도록 조작이 가능하다.
또한, 상기 박테리아는 운동성을 극대화시키거나, 약독화(attenuation)함으로써 인체 내 면역 반응을 피하거나, 과도한 증식에 따른 부작용을 방지하고자 자살 유전자를 삽입한 것을 사용할 수 있다.
구체적으로, 박테리아는 인체 감염 시, 면역세포들에 의해 공격을 받아 대부분 제거되나, 유전자 조작된 약독화 박테리아를 사용하는 경우, 면역세포나 면역반응들로부터의 공격을 회피할 수 있다.
또한, 박테리아가 인체 내에서 과도하게 증식하였을 경우, 이로 인한 부작용이 나타나므로 개체수의 조절 및 제거가 반드시 고려되어야 하는데, 유전자 조작을 통해 자살유전자에 관여하는 유전자를 인위적으로 삽입함으로써 박테리아의 자체 사멸 또는 외부에서 원하는 시점에 인위적으로 박테리아의 제거를 조절할 수 있다.
아울러, 박테리아는 아라비노스(Arabinose) 등에 의해 유도되는 배드 프로모터(pBad promoter) 시스템의 증가에 의한 사멸유전자(세포 용해소 A)를 분비하여 주변의 암세포를 제거하도록 유전자 조작된 것을 사용하는 것일 수 있으며, 이 경우 항생제에 의해 용이하게 제거가 가능하다.
이를 통해, 체내 면역세포 및 면역 반응으로부터의 공격, 과도한 증식으로 인한 부작용 없이 화학주성을 통해 병변 부위로 마이크로로봇을 보다 효과적으로 이동시킬 수 있다.
또한, 상기 박테리아는 질병의 진단 및 질병 치료 정도를 모니터링 하기 위한 표지 유전자 또는 단백질을 더 포함하는 것일 수 있으며, 상기 표지 유전자 또는 단백질로서 예를 들면 발광단백질, 형광단백질, 양전자단층촬영을 위한 마커 단백질, 아비딘 또는 수용체를 암호화하는 유전자 등을 들 수 있으나, 이에 제한되는 것은 아니다. 구체적인 일 예로, 녹색을 나타내는 녹색 형광 단백질(green fluorescent protein, GFP)와 같은 단백질에 관련된 유전자를 삽입한 박테리아를 사용, 형광 현미경을 통해 박테리아의 이동성을 확인할 수 있다. 또한 루시퍼레이즈(luciferase)는 루시페린(luciferin)을 산화시키면서 빛을 내는 단백질로서, 상기 단백질에 관련된 유전자를 삽입한 박테리아는 동물 실험(in vivo) 내에서 박테리아의 위치를 추적할 수 있다. 이를 통해, 인체 외부에서 형광 또는 발광효소를 발현하는 박테리아의 추적이 가능하므로 이동경로를 파악할 수 있으며, 특정 병변 부위로의 이동 정도에 관한 추적이 가능하다.
본 발명의 일 양태에 따른 자성 나노입자는 상기 치료용 세포의 표면에 결합하여 외부 자기장을 통한 마이크로로봇의 이동을 가능하게 하는 것으로, 공지의 외부 전자기 구동 시스템(external magnetic actuation system)을 이용하여 마이크로로봇을 3차원적으로 이송시킬 수 있다.
종래 전자기 구동 시스템을 이용한 치료용 마이크로로봇의 경우, 병변 부위 근처로의 매크로 타겟팅은 가능하나, 모세혈관 등의 국부 영역에서는 이미징이 불가하고 전자기 구동 시스템에 의해 정확한 타겟팅은 어려운 문제가 있었다. 그러나, 본 발명의 마이크로로봇은 상기 전자기 구동 시스템을 이용한 즉각적인 병변 부위의 위치 이동 이후에, 치료용 세포의 화학주성에 의한 정밀한 병변 타겟팅이 가능하여 병변 부위로 마이크로로봇의 정밀한 표적화가 이루어질 수 있다.
뿐만 아니라, 해체 온도(Td) 이상에서 상기 치료용 세포 및 자성 나노입자가 분리됨에 따라, 표적화 후 체액이나 조직에 잔류하는 자성 나노입자를 자기장 구동을 통해 국소 부위로 고농도로 집중화시킬 수 있으며, 카테터 또는 주사기 바늘을 통해 물리적으로 완전히 제거할 수 있다. 또는 자성을 가진 카테터 또는 주사기 바늘을 병변 부위에 삽입하고 자성을 가진 카테터 또는 주사기 바늘 부위로 자성 나노입자를 집중화시킨 후 체내로부터 완전히 제거함에 따라, 잔류하는 자성 나노입자에 의한 유해성을 유발하지 않는다.
상기 자성 나노입자의 종류로서 구체적인 예를 들면, 철, 니켈, 코발트, 산화철(Fe2O3, Fe3O4), 페라이트(CoFe2O4, CuFe2O4, MgFe2O4, ZnFe2O4, NiFe2O4, MnFe2O4), MnBi, MnSb, Y3Fe5O12, CrO2, MnAs, Gd, Dy, EuO 및 철합금(PePt, CoPt) 등을 들 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 자성 나노입자의 평균입경은 1 내지 1,000 nm, 구체적으로 2 내지 500 nm, 더욱 구체적으로 5 내지 200 nm인 것일 수 있으나, 이에 제한되는 것은 아니다. 상기 범위의 평균입경을 가지는 자성 나노입자는 병변 부위에서 입자의 탈락 없이 안정적으로 머무를 수 있으며, 마이크로로봇의 약물 방출 이후 자기장 구동을 통한 자성 나노입자의 제거 시, 카테터 또는 주사기 바늘 부위로 이동이 용이하고 카테터 또는 주사기 바늘의 관을 통한 체내로부터의 제거 또한 효과적이다.
상기 자성 나노입자의 함량은 마이크로로봇 총 중량의 10 내지 80 중량%, 바람직하게 30 내지 70 중량%일 수 있으며, 더욱 바람직하게 40 내지 60 중량%일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 자성 나노입자는 도 1에 도시된 바와 같이, 치료용 세포 표면을 둘러한 형태로 존재함에 따라 마이크로로봇이 다수의 자성 나노입자를 포함함에도 불구하고 하나의 입자로 작용할 수 있어, 외부 자기장의 인가 시 자기장의 방향에 따라 즉각적 위치 이동이 용이하고 병변 부위로 보다 빠르고 정확하게 표적화할 수 있다.
본 발명의 일 양태에 따른 자성 나노입자 및 치료용 세포는 리간드-수용체 결합을 통해 결합된 것으로서, 상기 리간드 및 수용체에서 선택되는 어느 하나는 단백질 또는 펩타이드인 것일 수 있으나, 이에 제한되는 것은 아니다.
구체적인 예를 들어, 자성 나노입자는 단백질 수용체로 코팅된 것일 수 있으며, 상기 단백질 수용체에 특이적으로 결합하는 리간드를 표면에 가지는 치료용 세포를 함께 사용하는 것일 수 있다. 이를 통해, 단백질 수용체 및 리간드가 콘쥬게이트(conjugate) 결합될 수 있으며, 이를 통해 치료용 세포 및 자성 나노입자가 결합된 마이크로로봇으로 기능할 수 있다.
상기 단백질 수용체 및 리간드는 서로 상호작용하여 콘쥬게이트를 형성할 수 있고, 국부적으로 가열하여 마이크로로봇의 주변 온도가 해체 온도(Td) 이상이 될 경우 서로 물리적으로 분리될 수 있는 조합이라면 제한받지 않고 사용될 수 있다. 하나의 바람직한 일 양태로는 단백질 수용체 및 리간드는 서로 콘쥬게이트를 형성하되, 상기 리간드가 병변 부위에 존재하는 단백질 수용체와 서로 콘쥬게이트를 형성할 수 있는 리간드라면 치료용 세포와 자성 나노입자의 분리 이후 치료용 세포의 화학주성 및 리간드와 병변 부위의 단백질 수용체의 높은 결합력에 의해 보다 정확한 표적화가 가능할 수 있다.
상기 단백질 수용체로서 보다 구체적인 예를 들면, 스트렙트아비딘(streptavidin) 및 아비딘(avidin) 등을 들 수 있으며, 상기 리간드로서 바이오틴(biotin) 및 바이오티닐화(biotinylated) 펩타이드 등을 들 수 있다.
바람직한 다른 일 양태에 따른 상기 수용체 및 리간드로는, 스트렙트아비딘 및 바이오틴을 사용하여 스트렙트아비딘이 코팅된 자성 나노입자 및 표면에 바이오틴이 결합된 치료용 세포가 리간드-수용체 결합된 마이크로로봇일 수 있다.
이 때, 상기 스트렙트아비딘이 코팅된 자성 나노입자로 MagCellect사의 SA-SPION가 예시될 수 있으며, 바이오틴이 결합된 박테리아는 박테리아 균주와 바이오틴을 함께 배양하여 제조된 것일 수 있으나, 이에 제한되는 것은 아니다.
상기와 같이 스트렙트아비딘-바이오틴 결합된 마이크로로봇의 경우, 바이오틴이 생물학적 활성의 손실 없이 스트렙트아비딘과 용이하게 콘쥬게이트 결합할 수 있으며, 매우 안정적인 구조를 나타낸다. 상기 스트렙트아비딘-바이오틴 결합된 마이크로로봇을 해체 온도(Td) 이상으로 가열함으로써 스트렙트아비딘-바이오틴 결합이 끊어질 수 있으며, 이에 따라 분리된 치료용 세포는 화학주성에 의해 정확한 표적 부위에 도달할 수 있으며, 자성 나노입자는 자기장 구동을 통해 병변 부위로부터 이송된 후, 국소 부위로 고농도 집중화시켜 카테터 또는 주사기 바늘을 통해 물리적으로 완전히 제거될 수 있다.
바람직한 일 양태에 따른 다른 구체적인 예로는 단백질 수용체로서 엽산 수용체 단백질인 글리코실포스파티딜이노시톨이 결합된 단백질(glycosylphosphatidyinositol-linked protein)과 리간드로서 엽산의 조합이 있을 수 있다. 보다 상세하게 설명하면, 자성 나노입자의 표면은 엽산 수용체 단백질로 코팅되어 있으며, 치료용 세포의 표면에는 엽산을 리간드로 포함함으로써 엽산 수용체 단백질과 엽산이 콘쥬게이트 결합되어 치료용 세포 및 자성 나노입자가 결합된 마이크로로봇으로 기능할 수 있다. 상기 엽산-엽산 수용체 단백질 결합 역시 해체 온도(Td) 이상으로 가열함으로써 상기 결합이 끊어질 수 있으며, 이에 따라 분리된 치료용 세포는 화학주성에 의해 정확한 표적 부위에 도달할 수 있으며, 치료용 세포의 표면이 엽산을 포함함으로써 특히 암세포에 대해 우수한 표적성을 나타낼 수 있다.
본 발명의 일 양태에서, 상기 치료용 세포는 약물 또는 약물 담지체를 더 포함하는 것일 수 있으며, 구체적으로 상기 약물은 치료용 세포 내에 담지되거나, 치료용 세포 표면에 화학적으로 결합된 것일 수 있으나, 이에 제한되는 것은 아니다.
약물의 종류로서 구체적인 예를 들면, 저분자 합성 화합물, 저분자 천연 화합물, 펩타이드, 단백질, 항체, 치료용 DNA 및 SiRNA 등을 들 수 있으며, 표적 질병에 따라 항암제 및 허혈성 질환 치료제 등을 사용할 수 있으며, 표적 질병에 따라 다양한 약물이 사용될 수 있음은 공지이므로, 전술한 바와 같은 약물로 제한되는 것은 아니다.
또한 상기 약물 담지체는 약물을 함유하는 생분해성 나노입자일 수 있으며, 구체적으로, 생분해성을 갖는 천연 고분자(natural polymer), 천연 고분자로부터 변형된 고분자(modified natural polymer) 및 합성 고분자(synthetic polymer)에서 선택되는 어느 하나 또는 둘 이상의 혼합물을 사용하는 것일 수 있다. 예를 들어 키토산(chitosan), 히알루론산(hyaluronic acid), 펙틴(pectin) 및 알긴산 (alginic acid) 등의 천연 고분자, 글리콜 키토산(glycol chitosan), 카르복시메틸 셀룰로오스(carboxylmethyl cellulose, CMC), 히드록시에틸 셀룰로오스(hydroxyethyl cellulose), 히드록시프로필 메틸 셀룰로오스(hyroxypropyl methyl cellulose, HPMC), 메틸셀룰로오스(methyl cellulose) 및 젤라틴(gelatin) 등의 변형 고분자, 폴리(베타-드록시에틸 메타아크릴레이트)(poly(β-hydroxyethyl methacrylate), PHEMA), 폴리비닐알코올(polyvinyl alcohol, PVA), 폴리에틸렌 글리콜(polyethylene gylcol, PEG), PLGA(poly(lactic-co-glycolic acid)), PGA(polyglycolic acid), PLA(polylactic acid), 폴리하이드록시알칸산(polyhydroxyalkanoate, PHA) 및 폴리카프로락톤(poly(ε-caprolactone)) 등의 합성 고분자 등을 들 수 있으나, 이에 제한되는 것은 아니다.
구체적인 일 예로, 생분해성 고분자인 PLGA를 약물 담지체로 사용하는 경우, 이를 포함하는 마이크로로봇을 외부 자기장을 통해 병변 부위로 근처로 빠르게 이동시킨 후, 도 2와 같이 해체 온도(Td) 이상으로 마이크로로봇을 가열시킴으로써 리간드(biotin)-수용체(streptavidin) 결합이 끊어질 수 있으며, 이에 따라 분리된 치료용 세포는 화학주성에 의해 정확한 표적 부위에 도달할 수 있다.
이 때, 치료용 세포 내 존재하는 PLGA는 표적 부위에서 락트산 및 글리콜산으로 생분해될 수 있으며, 상기 락트산 및 글리콜산은 삼카르복실산회로(tricarboxylic acid cycle, TCA)에 의해 체내에서 이산화탄소와 물로 분해될 수 있다. 이러한 약물 담지체의 분해 과정에서, 담지된 약물이 서서히 방출됨에 따라 단시간 내 과도한 약물의 분비로 인한 독성에 대한 부작용을 최소화 하고 일정한 기간 동안 약물 전달이 가능하게 하여 치료 효과를 현저히 향상시킬 수 있다.
또한, 앞서 분리되어 병변 부위 부근에 잔류하는 자성 나노입자는 자기장 구동을 통해 국소 부위로 고농도 집중화시켜 카테터 또는 주사기 바늘을 통해 물리적으로 완전히 제거할 수 있으며, 이에 따라, 별도의 유해한 물질이 체내에 남지 않아 부작용을 유발하지 않는다.
본 발명의 일 양태에서, 해체 온도(Td)는 인체의 체온 이상의 온도에서 분리되는 것일 수 있으며, 구체적으로 35 내지 60℃의 온도 범위일 수 있고, 보다 구체적으로 45 내지 60℃의 온도 범위에서 리간드-수용체 결합이 끊어지며 치료용 세포 및 자성 나노입자로 분리되는 것일 수 있다.
이는 구체적으로, 경구, 경피, 직장, 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관내 주사로 마이크로로봇을 체내에 투입하고 외부 자기장을 이용하여 병변 부위로 이동시킨 후, 상기 온도 범위에서 마이크로로봇의 리간드-수용체 결합이 끊어짐에 따라, 분리된 치료용 세포가 화학주성에 의해 병변으로 정확히 도달될 수 있다.
상기 체온 이상의 해체 온도(Td)는 외부 자극 장치, 구체적으로 마이크로로봇에 열을 발생시키는 장치를 통해 도달할 수 있으며, 구체적인 예를 들어 교류 자기장(alternating magnetic field, AMF) 등의 자기장 또는 근적외선(near infrared ray, NIR) 등의 광을 마이크로로봇에 인가하여 상기 범위의 해체 온도(Td)에 도달하는 것일 수 있으나, 이에 제한되는 것은 아니다. 바람직하게는 외부 교류 자기장의 인가를 통한 마이크로로봇 내 자성 나노입자의 자기 유도 발열에 의해 상기 범위의 해체 온도(Td) 도달되는 것일 수 있으나, 이에 제한되는 것은 아니다.
보다 구체적으로, 도 3에 도시된 바와 같이 외부 자기장을 통해 마이크로로봇을 병변 부위로 이동시킨 후 외부에서 교류 자기장을 마이크로로봇에 인가할 경우, 자성 나노입자가 자기 유도 발열될 수 있다. 이를 통해 해체 온도(Td) 이상으로 가열된 마이크로로봇은 리간드(biotin)-수용체(streptavidin) 결합이 끊어지면서 치료용 세포 및 자성 나노입자로 각각 분리될 수 있다.
또는, 상기 외부 자기장을 통해 병변 부위로 이동된 마이크로로봇의 외부에서 근적외선을 인가할 경우 자성 나노입자가 광 발열될 수 있으며, 이를 통해 해체 온도(Td) 이상으로 가열된 마이크로로봇의 리간드(biotin)-수용체(streptavidin) 결합이 끊어지면서 치료용 세포 및 자성 나노입자로 분리될 수 있다.
이를 통해, 치료용 세포는 화학주성에 의해 정확한 표적 부위로 이동하여 자체 치료 효과를 발현하거나, 담지된 약물 및 약물 담지체가 방출되어 치료 효과를 나타낼 수 있으며, 잔류하는 자성 나노입자는 앞서 설명한 바와 같이 체내로부터 물리적으로 완전히 제거될 수 있다.
상기의 교류 자기장 및 근적외선을 인가하는 장치는 당업계에 공지된 것을 사용할 수 있으므로 상세한 설명은 생략한다.
본 발명의 일 양태에 따른 마이크로로봇은 구, 캡슐, 다면체 등의 다양한 형상을 가질 수 있으며, 바람직하게는 구형일 수 있다. 또한, 평균직경은 1 내지 3,000 μm, 구체적으로 1 내지 1,000 ㎛인 것일 수 있으나, 이에 제한되는 것은 아니다. 상기 범위의 크기를 가지는 마이크로로봇은 주사기를 이용하여 체내에 용이하게 투입할 수 있어 바람직하다.
또한, 마이크로로봇의 총 부피 중 치료용 세포가 차지하는 부피는 10 내지 90 부피%일 수 있고, 구체적으로 20 내지 80 부피%일 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명은 마이크로로봇의 제조방법을 제공한다.
본 발명에 따른 마이크로로봇은 각각 리간드가 도입된 치료용 세포 및 수용체가 도입된 자성 나노입자를 사용하여 리간드-수용체 결합을 통한 마이크로로봇을 제조할 수 있다. 이 때, 상기 치료용 세포 및 자성 나노입자의 혼합비는 1:1 내지 5:1 중량비일 수 있으며, 리간드가 도입된 치료용 세포가 배양된 배지에 수용체가 도입된 자성 나노입자를 투입하여, 10 분 내지 1 시간 동안 반응시켜 리간드-수용체 결합을 유도하는 것일 수 있으나, 상기 방법으로 제한되는 것은 아니다.
또한, 상기 치료용 세포, 자성 나노입자, 리간드 및 수용체의 종류는 앞서 상술한 바와 같다. 상기 치료용 세포는 약물 또는 약물 담지체를 더 포함하는 것일 수 있으며, 상기 약물의 함량은 마이크로로봇 총 중량에 대하여 0.01 내지 10 중량%, 구체적으로 0.1 내지 1 중량%일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 자기장 구동 마이크로로봇 치료 시스템은 자기장 구동 시스템; 마이크로로봇 및 환부를 이미징하기 위한 영상 촬영부; 상기 이미징을 실시간으로 검출하는 검출기; 및 상기 자기장 구동시스템의 제어를 위한 통합제어 모듈;을 포함하고, 상기 마이크로로봇은 생체 내에 투여되어 치료용 세포를 병변 부위에 표적할 수 있는 마이크로로봇을 포함한다.
도 4는 본 발명의 일 실시예에 따른 자기장 구동 마이크로로봇 치료 시스템(100)을 개략적으로 나타낸 구성도이다. 도시한 바와 같이, 상부에는 X-ray 등을 환자의 환부에 조사하는 영상 촬영부(120)가 구성되어 있고 하부에는 상기 X-선을 이미지로 검출하는 검출기(130)가 구성되어 있으며 중앙부에는 환부에 투여된 마이크로로봇을 환부에 정확하게 위치시키기 위한 자기장 구동 시스템(110) 및 이의 제어를 위한 통합제어 모듈(140)이 구성되어 있다.
이 때, 상기 영상 촬영부는 양방향 X-ray 또는 MRI(magnetic resonance imaging) 시스템일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 자기장 구동 시스템은 외부 자극부(150), 구체적으로 근적외선(NIR) 조사부 또는 교류 자기장(AMF) 입력부를 더 포함하는 것일 수 있다. 상기 근적외선 조사부는 병변 부위에 표적시킨 마이크로로봇에 근적외선을 가함으로써, 마이크로로봇에 포함된 자성 나노입자의 유도 발열을 통해 온도 상승을 유발하여, 해체 온도(Td) 이상에서의 마이크로로봇의 분리를 보다 용이하게 수행할 수 있다. 분리 후 체류하는 자성 나노입자는 카테터 또는 주사기 바늘을 통해 체내로부터 완전히 제거할 수 있다.
또한, 상기 교류 자기장 입력부는 병변 부위에 표적시킨 마이크로로봇에 외부 교류 자기장을 가함으로써, 마이크로로봇에 포함된 자성 나노입자의 자기 유도 발열을 통해 해체 온도(Td) 이상으로 마이크로로봇을 가열시킬 수 있으며, 이를 통해 마이크로로봇의 분리를 유도할 수 있다. 이 후 체류하는 자성 나노입자는 상기와 동일한 방법으로 체내에서 완전히 제거할 수 있다.
또한, 본 발명에 일 양태에서, 자기장 구동 마이크로로봇 치료 시스템(100)은 암 치료 및 허혈성 질환 치료 등에 적용될 수 있으며, 보다 구체적으로, 주사기(150)을 이용하여 병변 부위(160)에 본 발명의 마이크로로봇을 투여한 후, 영상 촬영부(120) 및 검출기(130)를 통하여 실시간으로 획득한 3차원 이미지를 따라 마이크로로봇에 포함된 자성 나노입자의 위치와 병변 부위의 위치를 파악한다. 이어서, 자기장 구동 시스템(110) 및 통합제어 모듈(140)을 이용한 자기장 조절을 통해 상기 마이크로로봇을 표적 부위에 위치시킬 수 있으며, 이 후, 외부 자극부(150)를 통해 해체 온도(Td) 이상으로 가열된 마이크로로봇이 분리됨에 따라 치료용 세포가 화학주성에 의해 정확한 병변 부위로 이동하여 약물을 방출함으로써, 신속하고 정확한 타겟팅이 가능하여 치료 효율을 현저히 향상시킬 수 있다.
도 5는 도 4의 자기장 구동 마이크로로봇 치료 시스템(100)을 보다 구체적으로 도시하고 있는 것으로, 근적외선 조사부를 포함하는 자기장 구동 마이크로로봇 치료 시스템 장치를 촬영한 사진이다. 상기와 같은 시스템을 통하여 마이크로로봇은 자기장 구동 시스템에 의해 병변 부위로 이동될 수 있으며, 병변 부위로 이동 후에는 근적외선 또는 교류 자기장을 가하여 마이크로로봇의 온도를 리간드-수용체 결합의 해체 온도(Td) 이상으로 가열할 수 있다. 이에 따라, 마이크로로봇 내 치료용 세포 및 자성 나노입자가 분리될 수 있으며, 상기 치료용 세포는 정확한 병변 부위로 도달하여 치료 효과를 발현하며, 자성 나노입자는 자기장 구동 시스템에 의해 병변 부위로부터 이송되어 체외로 완전히 제거할 수 있다.
상기 자기장 구동 마이크로로봇 치료 시스템의 일 실시예에 따라 마이크로로봇이 적용된 치료방법은 도 6과 같은 흐름도를 가질 수 있다. 도 6에 도시된 바와 같이, 먼저 마이크로로봇을 제작한다(S100). 이어서, 상기 마이크로로봇을 주사기에 넣고 도 5와 같은 자기장 구동 마이크로로봇 치료 시스템이 준비된 상태에서 표적 부위에 국부 주사한다(S200). 이어서, 자기장 구동 시스템 및 통합제어 모듈을 이용하여 마이크로로봇을 병변 부위로 표적화한다(S300). 이어서, 근적외선 조사부, 교류 자기장 입력부와 같은 외부 장치를 이용하여 마이크로로봇을 해체 온도(Td) 이상으로 가열하여 리간드-수용체 결합을 해체하여 치료용 세포 및 자성 나노입자를 분리한다(S400). 이어서, 분리된 치료용 세포가 화학주성을 통해 정확한 표적 부위로 이동하여 치료 효과를 부여한다(S500). 이어서, 자기장 구동 시스템 및 통합제어 모듈을 이용한 자기장 조절을 통해 앞서 분리된 자성 나노입자를 고농도로 집중화시킨 후, 주사기 바늘 등을 이용하여 자성 나노입자를 회수한다(S600). 상기 (S600) 단계는 (S400) 단계 이후에 (S500) 단계와 동시에 수행할 수도 있으며 단계적으로 이어서 수행할 수도 있다.
상술한 바와 같은 흐름도를 통해 자기구동 마이크로로봇에 의해 정확한 병변 부위로의 표적화를 구현할 수 있을 뿐만 아니라, 치료 효과를 나타내는 약물의 방출 또는 세포의 전달 후 자성 나노입자를 실질적으로 완전하게 제거할 수 있는 장점을 가진다. 통상적으로 자성 나노입자를 포함하는 의약전달체를 체내에 투여할 경우, 자성 나노입자는 생분해되지 않고 완벽한 자연 배출이 보장되지 않아 체내에 그대로 잔류하게 됨으로써 잠재적 독성을 유발하게 된다. 그러나 자성 나노입자를 마이크로로봇으로부터 해체한 후 자기장 구동에 의해 자성 나노입자를 고농도로 국소 부위로 집중화한 후 실질적으로 완전하게 제거함으로써 잠재적 독성 문제를 근본적으로 해결할 수 있는 점에서 현저한 효과를 가진다.
또한, 본 발명은 상기 자기장 구동 마이크로로봇을 이용한 질병의 치료방법을 제공한다.
본 발명이 일 양태에 따른 마이크로로봇을 이용한 질병의 치료방법은 구체적으로, 상기 마이크로로봇이 담지된 주사기 등을 이용하여 환자 체내로 마이크로로봇을 투입한 후, 외부 자기장을 이용하여 병변 부위로 마이크로로봇을 빠르게 이동시킬 수 있다.
이어서, 근적외선 또는 교류 자기장을 가하여 마이크로로봇을 해체 온도(Td) 이상으로 가열하여 리간드-수용체 결합을 끊음으로써, 마이크로로봇 내 치료용 세포 및 자성 나노입자를 분리시킬 수 있다.
이를 통해 분리된 치료용 세포가 화학주성에 의해 병변 부위로 정확히 도달되어 병변 부위에만 표적화하여 선택적으로 치료효과를 나타낼 수 있으며, 치료용 세포가 약물을 더 포함함으로써 상기 치료용 세포 내 담지된 약물이 서서히 방출됨으로써, 정상세포에 대한 부작용 없이 병변 부위에서만 우수한 치료 효과를 나타낼 수 있다.
또한, 분리된 자성 나노입자는 병변 부위에 카테터 또는 주사기 바늘을 삽입하거나, 자성을 가진 카테터 또는 주사기 바늘로 자성 나노입자를 국소 부위로 고농도로 집중시킨 후, 체내로부터 완전히 회수하여 잔류하는 자성 나노입자로 인한 독성을 원천적으로 차단하여 치료효과를 더욱 향상시킬 수 있다.
이하 실시예 및 비교예를 바탕으로 본 발명을 더욱 상세히 설명한다. 다만 하기 실시예 및 비교예는 본 발명을 더욱 상세히 설명하기 위한 하나의 예시일 뿐, 본 발명이 하기 실시예 및 비교예에 의해 제한되는 것은 아니다.
[실시예 1] 마이크로로봇의 제조
본 실험에 이용된 약독화 S. typhimurium 균주(ΔppGpp), SHJ2037 (relA::cat, spoT::kn)은 본 발명자들의 종전 논문(Two-dimensionallocomotionofamicrorobotwithanovelstationary electromagnetic actuation system. Smart Mater. Struct. 18, 115017, 2009)에 기재된 것을 사용하였다. 구체적으로는 relA 유전자 및 spoT 유전자를 결손시켜 ppGpp(Guanosine 5'-diphosphate 3'-diphosphate) 합성능이 결여된 약독화 S. typhimurium 균주(ΔppGpp) SHJ2037는 기탁번호 KCTC 10787bP로 국제기탁기관인 한국생명공학연구원내 유전자은행에 기탁되어 있다. 50 μg/ml 카나마이신(Sigma)을 포함하는 LB 배지(Difco Laboratories)에서 격렬하게 공기공급을 하면서 37℃에서 Salmonella를 배양하였다.
이어서, 배양된 S. typhimurium 균주(ΔppGpp) 3 X 108 CFU/mL의 막 단백질을 500 μg의 바이오틴(biotin, EZ-Link NHS-LC-Biotin, Thermo Scientific, Rockford, IL)을 37℃에서 1시간동안 배양하여, 바이오틴이 표면에 결합된 S. typhimurium 균주(ΔppGpp)인 치료용 세포를 얻었다. 상기 바이오틴은 박테리아의 세포 표면에 많이 존재하는 OmpA와 결합하는 특징을 가지고 있다.
이어서, 상기의 바이오틴 결합된 치료용 세포와 자성 나노입자로써 평균입경이 150 nm이고 스트렙타비딘(streptavidin)이 코팅된 산화철인 MagCellect사의 SA-SPION을 3:1의 중량비로 37℃에서 30분 동안 박테리아 배지에서 반응시켜, 리간드(biotin)-수용체(streptavidin) 결합이 유도된 마이크로로봇을 제조하였다.
이상과 같이 본 발명에서는 특정된 사항들과 한정된 실시예 및 도면에 의해 설명되었으나 이는 본 발명의 보다 전반적인 이해를 돕기 위해서 제공된 것일 뿐, 본 발명은 상기의 실시예에 한정되는 것은 아니며, 본 발명이 속하는 분야에서 통상의 지식을 가진 자라면 이러한 기재로부터 다양한 수정 및 변형이 가능하다.
본 발명의 사상은 설명된 실시예에 국한되어 정해져서는 아니되며, 후술하는 특허청구범위뿐 아니라 이 특허청구범위와 균등하거나 등가적 변형이 있는 모든 것들은 본 발명 사상의 범주에 속한다고 할 것이다.

상기 영상 촬영부는 양방향 X-ray 또는 MRI 시스템인, 자기장 구동 마이크로로봇 치료 시스템. - 제7항에 있어서,
상기 마이크로로봇은 자기장 구동 시스템에 의해 병변 부위로 이동되고, 병변 부위로 이동 후 근적외선 또는 교류 자기장을 가하여 마이크로로봇의 온도를 리간드-수용체 결합의 해체 온도(Td) 이상으로 가열하며, 치료용 세포로부터 분리된 자성 나노입자는 자기장 구동 시스템에 의해 병변 부위로부터 이송되어 제거되는 것을 특징으로 하는, 자기장 구동 마이크로로봇 치료 시스템.
