Abstract
Description
도 2a 및 도 2b는 본 발명의 일 실시예에 따른, 대장암 환자 유래 오가노이드를 기반으로 한 약물 반응성 스코어 (Drug Response Score; DRS) 예측 모델의 모식도를 나타낸 도이다.
도 3은 본 발명의 일 실시예에 따른, 예측 정확도를 확인하기 위한 라소 회귀 곡선(Lasso regression)을 나타낸 도이다.
도 4a 및 도 4b는 본 발명의 일 실시예에 따른, 3 개의 CpG 프로브를 통해 무작위로 선택된 40 개의 트레이닝 세트의 DRS 분포를 정량화한 결과를 나타낸 도이다.
도 5는 본 발명의 일 실시예에 따른, 10 개의 테스트 세트로 DRS 분포를 정량화하여 3 개의 CpG 프로브를 통한 약물 반응성 예측 모델을 검증한 결과를 나타낸 도이다.

Claims (25)
- (a) 목적하는 개체로부터 얻어진 생물학적 시료에 대하여 서열번호 1 내지 3으로 표시되는 프로브를 이용하여 DNA 메틸화 수준을 측정하는 단계; 및
(b) 측정된 DNA 메틸화 수준을 이용하여 약물 반응성 스코어(Drug Response Score; DRS)를 계산하는 단계;를 포함하는 대장암 환자의 5-플루오로우라실(5-Fluorouracil; 5-FU)에 대한 치료 반응성을 예측하는 방법. - 제 1항에 있어서,
상기 DNA 메틸화 수준은 서열번호 4 내지 11 중 적어도 하나로 표시되는 프로브를 추가로 이용하여 측정되는 것인, 방법. - 제 1항에 있어서,
상기 DNA 메틸화 수준은 서열번호 4 내지 14 중 적어도 하나로 표시되는 프로브를 추가로 이용하여 측정되는 것인, 방법. - 제 1항에 있어서,
상기 DNA 메틸화 수준은 서열번호 4 내지 18 중 적어도 하나로 표시되는 프로브를 추가로 이용하여 측정되는 것인, 방법. - 제 1항 내지 제 4항 중 어느 한 항에 있어서,
상기 방법은 측정된 DNA 메틸화 수준으로부터 베타 값(β value)을 얻는 단계를 추가로 포함하는, 방법. - 제 5항에 있어서,
상기 약물 반응성 스코어(DRS)를 계산하는 단계는 베타 값에 가중치 계수(coefficient) 값을 곱하거나; 상기 프로브가 2개 이상인 경우 각 베타 값에 가중치 계수 값을 곱한 뒤 이들을 합산하여 수행되는 것인, 방법. - 제 6항에 있어서,
상기 가중치 계수는 -0.40 내지 2.05의 유리수인, 방법. - 제 8항에 있어서,
