FAAH 효소는 지방산 아미드 예컨대 아난다미드(N-아라키도노일에탄올아민), N-올레오일에탄올아민, N-팔미토일에탄올아민 및 올레아미드를 분해한다. 본 발명의 제6 측면에서, N-메틸사이클로펜틸아민의 산염의 합성 공정이 제공되며, 상기 공정은 사이클로펜틸아민과 클로로포름산염 또는 디-tert-부틸 카보네이트와의 반응을 포함하여 사이클로펜틸카바메이트를 형성하고, 사이클로펜틸카바메이트의 환원 공정 및 N-메틸사이클로펜틸아민의 산염으로의 산성화 공정이 수반된다.
제6 측면의 공정은 N-메틸사이클로펜틸아민의 산염, 식 A 화합물의 제조에서 중요한 중간체를 효율적으로 제조하는데 사용될 수 있다. 본 공정은 높은 수율과 우수한 양질의 제품을 제공한다. 바람직한 산염은 HCl 염이다. 그러나, 다른 무기 및 유기 산염 또한 사용 가능하다.

실험에 사용된 동물은 Interfauna Iberica(스페인)로부터 얻은 수컷 NMRI 마우스(무게 27-44 g)였다. 마우스는 제어된 환경 조건(12시간 명/암 주기 및 실내 온도 22±1℃) 하에서, 케이지 당 5를 유지하였다. 음식 및 수돗물은 자유식으로 공급되었고 모든 실험은 낮 시간 동안 수행되었다.
동물에 30 mg/kg 또는 3 mg/kg의 본 발명의 화합물 또는 비교 화합물이 경구(8 ml/kg; 0.5% 카복시메틸셀룰로오스(CMC)에 현탁 또는 물에 용해된 화합물) 또는 동물 사양 스테인레스 스틸 커브 니들(Perfectum, U.S.A.)을 사용하여 비히클(컨트롤)을 통해 투여되었다. 희생 15분 전에 동물을 복강내 투여된 펜토바르비탈 60 mg/kg으로 마취하였다. 간 조각, 왼쪽 폐엽 및 소뇌 없이 뇌를 제거하여 막 버퍼(3 mM MgCl2, 1 mM EDTA, 50 mM Tris HCl pH 7.4)를 함유하는 플라스틱 바이알에 넣었다. 조직은 분석할 때까지 -30℃에서 저장되었다.
동물은 18 시간 초과의 시간 시점을 제외하고 항상 화합물 투여 전 하룻밤 동안 금식되었고, 음식은 투여 날 아침에 제거되고 화합물은 같은 날 오후에 투여되었다. 이후 동물은 물만 주어지고 다른 것은 아무것도 제공되지 않았다.

대사 안정성 분석
시험 화합물의 안정성이 NADPH 존재 및 부재 하의 MLM(마우스 간 마이크로솜) 또는 HLM(인간 간 마이크로솜)에서 수행되었다.

3.1 물질 및 방법
a) 시약 및 용액

b) 조직 준비
Wistar rats로부터 얻은 냉동된 뇌를 Potter-Elvejhem(500 rpm에서 8 스트로크)을 이용하여 20 ml 1 mM MgCl2, 20 mM HEPES(4-(2-하이드록시에틸)-1-피페라진에탄설포닉 애시드) pH 7.0에서 균질화하였다. 균질 현탁액은 36000g, 4℃에서 20분간 원심분리하였다(Beckman, 70Ti 로터). 펠렛은 동일한 버퍼 15 ml에 재현탁시키고 동일한 조건으로 원심분리하였다. 펠렛은 동일한 버퍼 15 ml에 재현탁시키고 36000g, 4℃에서 20분간 원심분리한 후에 37℃에서 15분간 인큐베이션하였다. 이후 각 펠렛을 10 ml 3mM MgCl2, 1 mM EDTA(에틸렌디아민테트라아세트산), 50 mM Tris(2-아미노-2-하이드록시메틸-프로판-1,3-디올) pH 7.4에 재현탁하고 BSA(소 혈청 알부민)(50-250 ㎍/ml)의 표준 곡선을 이용하여 BioRad Protein Assay(BioRad)로 단백질을 결정하였다.

0℃에서 THF(20ml) 중의 사이클로펜틸아민(3 ml. 30.3 mmol) 용액에 3M 소듐 하이드록사이드(15.15 ml, 45.5 mmol) 및 에틸 클로로포르메이트(3.47 ml, 36.4 mmol)을 각각 첨가하였다. 생성된 두 개의 상을 갖는 혼합물을 상온에서 4시간 동안 교반하였다. 반응 혼합물은 MTBE(30mL)와 암모늄 하이드록사이드(5 mL)로 희석하였다. 생성된 혼합물을 상온에서 10분간 교반한 다음 분리시켰다. 유기 층을 물, 0.5M HCl로 세척, Na2SO4 상에서 건조, 여과하였다. 여과액은 감압 하에서 농축하였다. 에틸 사이클로펜틸카바메이트(4.35 g)가 95% 수율로 무색 오일로서 수득되었고 추가 정제 없이 다음 단계에서 사용되었다.

질소 하의 상온에서 THF(20mL) 중의 LAH(2,414 g, 63,6 mmol, 5eq) 현탁액에 20분에 걸쳐 THF(8mL) 중의 에틸 사이클로펜틸 카바메이트(2 g, 12,72 mmol) 용액을 첨가하였다. 노트: 가스 방출. 적하 깔때기를 THF(2mL)로 세정하였다. 반응 혼합물을 6시간 동안 65℃(내부 온도, 환류)로 가열하였다. 현탁액을 0℃(얼음물 욕조)로 냉각시켰다. 현탁액을 MTBE(30mL)로 희석하였다. 현탁액에 2.4 mL 물(강한 가스 방출 및 발열 반응이 관찰됨)을 적하하고, 3.6 mL 10% NaOH(잘 이루어진 교반이 필수적임)를 적하하고 마지막으로 7.2 mL 물을 적하하였다. 생성된 슬러리는 상온으로 가온하고 상온에서 30분간 교반하였다. 백색 현탁액에 MgSO4(10g)을 첨가하였다. 생성된 슬러리를 10분간 교반한 다음, 여과하였다. 고체를 MTBE(20mL)로 세척하였다.
결합된 여과액에 진한 HCl(1,272 ml, 15,27 mmol, 1.2eq)을 첨가하였다. 생성된 혼합물을 상온에서 하룻밤 동안 교반한 다음 농축 건조시켰다. 잔류물은 프로판-2-올(20mL)에 용해시킨 다음 2 vol(4mL)으로 농축하였다. 이후 생성된 용액에 MTBE(12mL)를 첨가하였다. 백색 결정 고체가 분쇄되었다. 슬러리를 상온에서 1시간 동안 교반한 다음 고체를 수집하고, MTBE(4mL)로 세척하고, 50℃에서 진공 오븐 하에서 4시간 동안 건조시켰다. 백색 침상(884mg)의 제1 수확물이 수득되었다. 결합된 모액 및 세척액을 농축 건조시켰다. 이소프로필 아세테이트(iPrOAc)를 잔류물에 첨가하고, 백색 결정이 나타나기 시작했다. iPrOAc를 더 첨가하고 고체 일부가 플라스크 벽면 상에 껍질이 되었다. 일부 DCM을 첨가하고 맑은 고체가 수득되었다. DCM을 제거하고 백색 고체를 크래쉬(crash)하고, 여과 및 iPrOAc로 세척하였다. 백색 결정 고체를 50℃에서 진공 오븐 하에서 4시간 동안 건조시켰다. 백색 침상(547mg)의 제2 수확물이 수득되었다. N-메틸 사이클로펜틸아민 하이드로클로라이드가 83% 몰 수율로 백색 침상으로 수득되었다.

65℃에서 메탄올 중의 수소 압력 하에서 Pd/C 및 트리에틸아민의 촉매량의 존재 하에서 사이클로펜타논 및 N-메틸아민 하이드로클로라이드의 사용이 가장 좋은 결과를 제공하는 것을 확인하였다. 이러한 조건 하에서, N-메틸사이클로펜틸아민 하이드로클로라이드는 49% 수율로 백색 고체로서 분리되었다.
시약 당량 및 팔라듐의 소스(source)가 수율 및 생성물의 품질을 개선하기 위해 시험되었다(메틸아민 하이드로클로라이드의 제거). 다소 과량의 메틸 아민 하이드로클로라이드(1.1 eq)와 Pd/C(JM, 5R39 페이스트)를 사용하는 것은 수율을 69%까지 개선 가능하게 하였다.
메틸 아민 하이드로클로라이드의 제거는 소듐 카보네이트의 존재 하에서 N-메틸사이클로펜틸아민 하이드로클로라이드를 디클로로메탄에 현탁하고 증류함으로써 가능하다. 최종 생성물에서 메틸아민은 검출되지 않는다.
프로토콜 설명
탄소 상 팔라듐 5%에, 5R 39 페이스트(0.75g, 0.176 mmol, 0.001eq), MeOH(105 ml), 메틸아민 하이드로클로라이드(13.24g, 196 mmol), 사이클로펜타논(15,77 ml, 178 mmol) 및 마지막으로 트리에틸아민(0.621 ml, 4.46 mmol)을 순차적으로 첨가하였다. 생성된 슬러리를 오토클레이브에 넣고 5 bar 수소로 충전하였다. 오토클레이브를 65℃에서 가열하고 하룻밤 동안 교반하였다. 반응 혼합물을 천천히 냉각시켰고 TLC(용리액 PE/에틸 아세테이트 8:2, dip permanganate)는 출발 물질을 나타내지 않았다. 블랙 슬러리는 셀라이트를 통해 여과 및 MeOH(10mL)로 세척되었다. 메탄올을 제거하고 이소프로판올(60mL)로 대체하였다. 용액은 2 vol으로 농축하고 이소프로필 에테르(60ml)를 첨가하였다. 생성된 혼합물은 상온에서 교반하였다. 백색 고체가 관찰되었고 이후 슬러리를 1시간 동안 0℃에서 교반한 다음, 여과하였다. 고체를 이소프로필 에테르/프로판-2-올 9:1(30mL)로 세척하고, 진공 오븐 하에서 하룻밤 동안 건조시켰다. N-메틸사이클로펜틸아민 HCl의 백색 결정 고체(16.9g, 69.5% 수율)를 수득하였다.
Claims (13)
- 제 1항에 있어서,
상기 화합물은 식 A 또는 그의 약학적으로 허용 가능한 염의 구조를 갖는 것인, 화합물.
- 질환의 치료 또는 예방용 약학적 조성물로서,
상기 조성물은 제 1항 또는 제 2항에 따른 화합물을 유효성분으로 포함하고,
상기 조성물은 하나 이상의 약학적으로 허용 가능한 부형제를 더 포함하고,
상기 질환은 식욕 장애, 비만, 대사성 장애, 악액질, 식욕 부진, 통증, 염증, 신경 독성, 신경 외상, 뇌졸중, 다발성 경화증, 척수 손상, 파킨슨병, L-도파 유도성 운동 장애, 헌팅턴병, 투렛 증후군, 지연성 운동 장애, 근육 긴장 이상, 근위축성 측삭경화증, 알츠하이머병, 간질, 정신 분열증, 불안, 우울증, 불면증, 구역질, 구토, 알콜 장애, 약물 중독, 고혈압, 순환계 쇼크, 심근 재관류 손상, 동맥 경화증, 천식, 눈 질환, 암, 염증성 장 질환, 급성 및 만성 간 질환, 관절염 및 골다공증으로 이루어진 군에서 선택되는 1 이상인 것인, 질환의 치료 또는 예방용 약학적 조성물.
- 제 3항에 있어서,
하나 이상의 추가 활성 약학 성분(active pharmaceutical ingredients)을 더 포함하는, 약학적 조성물.
- 제 4항에 있어서,
상기 하나 이상의 추가 활성 약학 성분은 아난다미드, 올레오일 에탄올아미드 또는 팔미토일 에탄올아미드인 것인, 약학적 조성물.
- 제 3항에 있어서,
상기 질환은 눈 질환인 것인, 약학적 조성물.
- 제 6항에 있어서,
상기 눈 질환은 고안압증, 녹내장 및 망막 병증으로 이루어진 군에서 선택되는 1 이상인 것인, 약학적 조성물.
- 제 3항에 있어서,
상기 약물은 아편제, 니코틴, 코카인, 알코올 및 정신 자극제로 이루어진 군에서 선택되는 1 이상인 것인, 약학적 조성물.
- 제 3항에 있어서,
상기 조성물은 질환의 진전 또는 증상이 FAAH 효소의 기질과 관련된 질환의 예방 또는 치료에 사용되는 것인, 약학적 조성물.
- 제 9항에 있어서,
상기 질환은 엔도카나비노이드 시스템(endocannabinoid system)과 관련된 질환인 것인, 약학적 조성물.
- 제 3항에 있어서,
상기 간 질환은 간염 및 간경변으로부터 선택된 것인, 약학적 조성물.
- 제 1항 또는 제 2항에 따른 화합물의 합성 방법으로,
상기 합성 방법은 N-메틸사이클로펜틸아민의 산염의 합성 공정을 포함하고, 상기 공정은 사이클로펜틸아민과 클로로포름산염 또는 디-tert-부틸 카보네이트와의 반응을 포함하여 사이클로펜틸카바메이트를 형성하고, 사이클로펜틸카바메이트의 환원 공정 및 N-메틸사이클로펜틸아민의 산염으로의 산성화 공정이 수반되는 합성 방법.
- 제 1항 또는 제 2항에 따른 화합물의 합성 방법으로,
상기 합성 방법은 N-메틸사이클로펜틸아민의 산염의 합성 공정을 포함하고, 상기 공정은 메틸아민의 산염의 존재 하에서의 사이클로펜탄온의 환원적 아민화 공정을 포함하는 합성 방법.
