Abstract
본 발명에 따르면, 바이오마커들이 미분화된 줄기세포들 뿐만 아니라 Passage 증가, 단순 배양에 따른 유전자 발현 변화를 나타내는 특성이 변화된 세포들과 구별되어 부갑상선 조직 세포로의 분화를 확인할 수 있게 하고 이를 통해 분화의 유도 뿐만 아니라 부갑상선 호르몬을 생산하고 제어 가능한 줄기 세포 치료제를 개발하는데 활용될 수 있다.
Description
도 2는 부갑상선 조직 세포로의 분화를 확인하여 주는 알려진 바이오마커인 PTH, CHGA, CASR의 발현을 RT-PCR을 통해 확인한 결과를 나타내는 도이다.
도 3은 부갑상선 조직 세포로의 분화에 의해 제조된 분화 세포의 mRNA발현을 마이크로어레이로 분석한 Heatmap 결과를 나타내는 도이다.

- SLC7A14 (solute carrier family 7, member 14), AMOT (angiomotin), FLG2 (fibrinogen-like 2), JUNB (jun B proto-oncogene), HAPLN1 (hyaluronan and proteoglycan link protein 1) 및 INHBA (inhibin beta A)로 이루어진 군으로부터 선택되는 어느 하나 이상의 mRNA 또는 단백질의 수준을 측정하는 제제를 포함하는, 중간엽 줄기세포로부터 부갑상선 조직 세포로의 분화를 탐지하기 위한 바이오마커 검출용 조성물로서,
상기 mRNA 수준을 측정하는 제제가 상기 유전자의 mRNA에 특이적인 프라이머 쌍 또는 프로브인 검출용 조성물. - 삭제
- 제1항에 있어서, 상기 중간엽 줄기세포는 인간 유래인 것을 특징으로 하는 검출용 조성물.
- 제3항에 있어서, 상기 중간엽 줄기세포는 인간의 편도 유래 중간엽 줄기세포인 검출용 조성물.

(b) 상기 핵산 시료 중, SLC7A14 (solute carrier family 7, member 14), AMOT (angiomotin), FLG2 (fibrinogen-like 2), JUNB (jun B proto-oncogene), HAPLN1 (hyaluronan and proteoglycan link protein 1) 및 INHBA (inhibin beta A)로 이루어진 군으로부터 선택되는 어느 하나 이상의 유전자의 발현량을 측정하는 단계; 및
(c) 상기 단계 (b)의 SLC7A14 (solute carrier family 7, member 14), AMOT (angiomotin), FLG2 (fibrinogen-like 2) 및 JUNB (jun B proto-oncogene)로 이루어진 군으로부터 선택되는 하나 이상의 유전자의 발현량이 감소하거나, HAPLN1 (hyaluronan and proteoglycan link protein 1) 또는 INHBA (inhibin beta A) 유전자의 발현량이 증가할 때, 중간엽 줄기세포가 부갑상선 조직 세포로 분화된 것으로 판정하는 단계를 포함하는, 중간엽 줄기세포로부터 부갑상선 조직 세포로의 분화를 탐지하는 방법. - 제10항에 있어서, 상기 단계 (b)에서 유전자 발현량 측정은 상기 마커 유전자에 특이적인 프라이머를 이용하여 부갑상선 조직 세포로 분화시킨 중간엽 줄기세포로부터 얻은 핵산 시료와 반응시켜 역전사 중합효소연쇄반응(RT-PCR) 또는 실시간 중합효소연쇄반응(Real time PCR)을 수행하는 것인, 중간엽 줄기세포로부터 부갑상선 조직 세포로의 분화를 탐지하는 방법.
