Abstract
본 발명은 보론산 화합물이 화학적으로 결합된 글루코오스 감응성 히알루론산 나노입자 및 이를 포함하는 조성물에 관한 것이다.
본 발명에 따른 나노입자를 이용하는 경우, 독성 문제를 가지는 기존의 조영제 및 항암제의 사용을 배제한 채, 암세포 특이적인 생물학적 기전을 이용하여 암을 진단 및 치료할 수 있다.

Description
본 발명은 보론산 화합물이 히알루론산에 화학적으로 결합된 글루코오스 감응성 나노입자에 관한 것이다.
당 감응성 특징을 가지는 재료를 이용한 당뇨병 치료에 관한 연구들이 진행되어 왔다. 페닐보론산을 이용한 고분자 합성체들은 현재까지 당뇨병 치료에 많이 쓰여지고 있고, 상기 합성체들을 약물전달체로써 활용하는 방법들이 개발되고 있다. 또한 생체적합형 고분자 나노입자를 이용한 암 치료 연구가 활발히 진행되고 있다. 그러나 대부분의 연구는 암세포뿐 아니라 정상세포도 파괴하는 부작용을 가진다.
잠재적 독성을 가진 조영제 및 항암제를 생체친화적 재료를 통해 암 특이적으로 전달하고자 하는 연구가 활발히 진행되고 있다. 최근에는 진단과 치료를 동시에 가능케하는 융합 기술의 하나로 '테라그노시스' 연구 패러다임이 각광받고 있다[Ryu J et al., "Tumor-targeting multi-functional nanoparticles for theragnosis: New paradigm for cancer therapy", Advanced Drug Delivery Reviews 64 : 1447-1458183-192(July 4 2012)]. 암세포의 경우 정상 세포에 비해 비효율적으로 에너지가 생산되는 와버그 효과(Warburg Effect)를 나타낸다. 이러한 비정상적인 작용은 암 세포내 고농도의 포도당 이입을 야기한다[Reuben JS, "Glucose metabolism and cancer", Current Opinion in Cell Biology 18 : 598-608 (OCT 12, 2006); Robert AG et al., "Why do cancer have high aerobic glycolisys", Nature Review 4 : 891-899 (NOV 2004)]. 이러한 암세포만의 독특한 생물학전 기전에 입각하여 암을 진단한 예는 보고되어 왔다. 그러나, 세포 대사 조절을 통해 암의 진단 및 치료를 동시에 적용한 예는 아직 보고되지 않았다.
한편, 소수성을 띄는 페닐아날린 계열 화합물은 포도당과 특이적으로 결합하여 소수성 특징을 잃게 됨이 보고되었다[Shull B et al., "P-Boronophenylalanine complexes with fructose and related carbohydrates and polyols", US Patent 6,169,076 B1 (Jan 2 2001)]. 이러한 현상을 당뇨병 치료에 적용한 예는 보고되었다[Kataoka, K, et al. "Totally synthetic polymer gels responding to external glucose concentration: Their preparation and application to on-off regulation of insulin release." Journal of the American Chemical Society 120 : 12694-12695 (NOV 1998)]. 그러나, 상기 현상을 암 진단 및 치료에 적용한 예는 전무하다.
질환 진단 및 치료를 위한 종래기술로서 표적지향형 프로브, 조영제, 및 치료제가 포함된 생체친화적 전달체가 많이 이용되고 있다. 특히 암의 진단 및 동시 치료가 가능한 고분자 나노입자가 다수 개발되었다. 하지만, 이에 사용되는 대부분의 조영제는 현재까지 신장 독성과 방사선 노출 같은 잠재적 독성 문제가 끊임없이 문제시 되고 있다. 또한 불가항력적인 항암제의 비특이적 전달에 따른 위험성은, 항암제 치료가 아직까지 임상에서 부수적인 치료 방법으로 국한되는 주된 원인이 되고 있다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 독성 문제를 가지는 기존의 조영제 및 항암제의 사용을 배제하고, 암세포 특이적인 생물학적 기전을 이용한 암 특이적 진단 및/또는 치료용 조성물을 개발하고자 연구 노력하였다.
그 결과 히알루론산에 보론산 화합물이 결합된 글루코오스 감응성 나노입자가 증진된 투과 및 유지 효과(Enhanced Permeability and Retention(EPR) effect)에 의해 암 조직, 특히 간암 조직에 선택적으로 전달하는 것을 발견하였다. 또한, 상기 보론산 화합물은 글루코오스와 결합하여 암 조직을 영상화하고, 암세포 내 에너지 결핍을 유도함을 규명함으로써 본 발명을 완성하게 되었다.
특히 히알루론산은 암세포의 CD44 수용체와 결합하는 특징을 가지므로, 이를 이용하여 나노입자의 암 특이적 전달이 가능하다.
따라서, 본 발명의 목적은 글루코오스 감응성 나노입자를 제공하는데 있다.
본 발명의 다른 목적은 간암의 진단, 치료, 또는 진단 동시 치료용 조성물을 제공하는데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명은 하기 화학식 1로 표시되는 반복단위의 카르복실기에 보론산 화합물이 결합된 히알루론산-보론산 반복단위를 포함하는 히알루론산 복합체를 제공한다.

또한, 본 발명은 전술한 히알루론산 복합체의 자가-집합(self-aggregated)체로서, 소수성 코어를 갖는 구형입자(spherical particle)인 글루코오스 감응성 나노입자를 제공한다.
또한, 본 발명은 히알루론산과 보론산 화합물을 반응시켜 히알루론산 복합체를 제조하는 단계를 포함하는 히알루론산 복합체의 제조방법을 제공한다.
또한, 본 발명은 전술한 히알루론산 복합체를 수용액 상에 분산하여, 상기 히알루론산 복합체를 자가-집합(self-aggregated)시켜, 소수성 코어를 갖는 구형입자(spherical particle)를 제조하는 단계를 포함하는 글루코오스 감응성 나노입자의 제조방법을 제공한다.
또한, 본 발명은 전술한 글루코오스 감응성 나노입자를 포함하는 간암 진단 또는 치료용 약제학적 조성물을 제공한다.

본 발명자들은 독성 문제를 가지는 기존의 조영제 및 항암제 사용을 배제한 채, 암세포 특이적인 생물학적 기전을 이용한 암 특이적 진단, 치료 또는 진단 동시 치료용 조성물을 개발하고자 연구 노력하였다.
그 결과 히알루론산에 보론산 화합물이 결합된 나노입자를 이용하는 경우, 암 조직에 증진된 투과 및 유지 효과(Enhanced Permeability and Retention(EPR) effect)에 의해 선택적으로 전달되고, 글루코오스와 결합하여 암 조직을 영상화하며, 암세포 내 에너지 결핍을 유도함을 규명하였다.
이하, 본 발명을 보다 상세하게 설명한다.
본 발명은 하기 화학식 1로 표시되는 반복단위의 카르복실기에 보론산 화합물이 결합된 히알루론산-보론산 반복단위를 포함하는 히알루론산 복합체에 관한 것이다.

상기 화학식 1로 표시되는 반복단위는 화학식 1의 반복단위라할 수 있다.
본 발명에서 '반복단위(repeat unit)'란, 중합체(고분자, polymer)에서 최소의 반복되는 구성 단위를 의미한다. 예를 들어, 선상 중합체에서 반복단위를 M이라 표현할 경우, 중합체의 구조는 -[M]n-과 같이 표현할 수 있다.
본 발명의 히알루론산 중합체는 반복단위로서 히알루론산-보론산 반복단위를 포함한다. 본 발명에서는 상기 히알루론산-보론산 반복단위와 함께 화학식 1로 표시되는 히알루론산 반복단위를 함께 포함할 수 있다. 상기 두 종류의 반복단위는 규칙적인 순서로 배열되거나 또는 불규칙하게 배열될 수 있다.
상기 히알루론산-보론산 반복단위의 함량은 히알루론산 복합체 100 몰부에 대하여 15 내지 55 몰부, 18 내지 40 몰부 또는 20 내지 35 몰부일 수 있다.
본 발명의 히알루론산-보론산 반복단위에서 보론산 화합물로 페닐보론산을 사용할 수 있으며, 상기 페닐보론산으로 N-(4-페닐보로닉)숙시남산, 3-카르복시벤젠벤조익산, 4-카로복시피리딘-3-벤조익산 및 (3-아미노메틸페닐)벤조익산 클로라이드로 이루어진 그룹으로부터 선택된 하나 이상을 사용할 수 있다.
상기 보론산 화합물은 아미드 결합(amide bond)을 통해 히알루론산 반복단위에 결합될 수 있다. 구체적으로, 히알루론산의 카르복실기는 보론산 화합물의 아민기와 반응하여 아미드 결합을 형성할 수 있다.
이 경우, 보론산 화합물로는 (3-아미노메틸페닐)벤조익산 클로라이드를 사용할 수 있다.
또한, 상기 보론산 화합물은 링커로서 디아민 화합물을 매개로 하여 아미드 결합(amide bond)을 통해 히알루론산 반복단위에 결합될 수 있다. 구체적으로, 히알루론산의 카르복실기는 디아민 화합물의 아민기와 아미드 결합을 형성하며, 상기 디아민 화합물 중 결합을 형성하지 않은 아민기는 보론산 화합물의 카르복실기와 아미드 결합을 형성할 수 있다.
이 경우, 보론산 화합물로는 N-(4-페닐보로닉)숙시남산, 3-카르복시벤젠벤조익산 및 4-카로복시피리딘-3-벤조익산으로 이루어진 그룹으로부터 선택된 하나 이상을 사용할 수 있다.
또한, 상기 디아민 화합물로는 에틸렌디아민, 부틸렌디아민, 헥사메틸렌디아민, 펜타에틸렌헥사아민 또는 1,5-디아미노-2-메틸펜탄을 사용할 수 있다.
본 발명의 히알루론산 복합체의 분자량은 50 내지 500 kDa 또는 200 내지 300 kDa일 수 있다.
본 발명의 히알루론산 복합체에서 히알루론산 반복단위와 히알루론산-보론산 반복단위의 히알루론산 부분은 친수성 성질을 가지며, 히알루론산-보론산 반복단위의 보론산 부분은 소수성 성질을 가진다. 즉, 상기 히알루론산 복합체는 친수성부 및 소수성부를 모두 포함하는 양친매성 고분자이다.
또한, 본 발명은 전술한 히알루론산 복합체의 자가-집합(self-aggregated)체에 관한 것이다. 상기 히알루론산 복합체는 자가-집합하여, 소수성 코어를 가지는 구형입자(spherical particle)를 형성한다. 이때, 구형이란 원형 및 타원형을 포함할 수 있다. 상기 구형입자는 나노입자라 표현할 수 있으며, 상기 나노입자는 글루코오스 감응성을 가지므로 글루코오스 감응성 나노입자라 표현할 수 있다.
본 발명의 히알루론산 복합체는 양친매성 성질을 가지는데, 소수성 성질을 가지는 보론산 부분이 다수의 내부 코어를 형성하여 안정한 구형입자를 형성할 수 있다. 이러한 구형상은 체내의 일반적인 글루코오스 환경에서는 입자 구조가 안정하게 유지되며, 암 조직이 가지는 특이적인 고농도 글루코오스 환경에서는 입자 구조가 유지되지 않는다. 따라서, 암 세포 내로 이입된 글루코오스 감응성 나노입자는 글루코오스를 포집하여 효과적으로 호기성 해당작용(aerobic glycolysis)을 억제시키고, 암 성장을 억제할 수 있다. 상기 글루코오스 감응성 나노입자의 글루코오스 포집을 통한 해당작용 억제 기전은, 정상 세포에 직접적으로 타격을 주지 않는다. 이는 종래 항암 약물 또는 조영제의 전달체로서의 역할에 국한되던 나노입자의 잠재적 독성 문제를 해결할 수 있다.
본 발명에 따른 글루코오스 감응성 나노입자의 평균 입경은 특별히 제한되지 않는다. 예를 들어, 상기 입경은 200 내지 400 nm, 또는 200 내지 300 nm일 수 있다. 일반적으로 히알루론산-보론산 반복단위의 함량이 많아질수록 팩킹 효과에 의해 나노입자의 평균 입경이 작아지는 경향을 가진다.
본 발명에 따른 글루코오스 감응성 나노입자는 구형입자에 화학적으로 표지되거나, 소수성 코어의 내부에 물리적으로 로딩되는 형광 신호 물질을 추가로 포함할 수 있다. 상기 형광 신호 물질을 사용하여 나노입자를 다양한 암의 표적 진단에 이용할 수 있다.
상기 화학적 표지는 나노입자에 형광 신호 물질이 화학적으로 결합(예를들어, 공유결합)하여 도입되는 것을 의미하며, 화학적 표지의 경우에는 형광 신호 물질은 소수성 코어의 내부에 형성될 필요가 없다.
상기 형광 신호 물질로는 예를 들어, Ce6(Chlorine 6), Cyanine 염료 시리즈(Cy3, Cy5, 또는 Cy5.5), 플루오로세인과 그 유도체, 로다민과 그 유도체, 루시퍼 엘로우, B-파이토에리쓰린, 9-아크리딘이소티오시아네이트, 루시퍼 옐로우 VS, 4-아세트아미도-4'-이소티오-시아나토스틸벤-2,2'-다이설폰산, 7-다이에틸아미노-3-(4'-이소티오시아토페닐)-4-메틸쿠마린, 석시니미딜-파이렌부티레이트, 4-아세트아미도-4'-이소티오시아나토스틸벤-2,2'- 다이설폰산 유도체, LC™-Red 640, LC™-Red 705, 알렉사 염료 시리즈(Alexa dye series), 리사민, 이소티오시아네이트, 에리쓰로신 이소티오시아네이트, 다이에틸렌트리아민 펜타아세테이트, 1-다이메틸아미노나프틸-5-설포네이트, 1-아닐리노-8-나프탈렌 설포네이트, 2-p-토우이디닐-6-나프탈렌설포네이트, 3-페닐-7-이소시아나토쿠마린, 9-이소티오시아나토아크리딘, 아크리딘 오렌지, N-(p-(2-벤족사조일릴)페닐)멜레이미드, 벤족사디아졸, 스틸벤 및 파이렌 등 형광 유기 물질 및 형광 무기 반도체 나노입자(양자점)를 사용할 수 있다.
또한, 본 발명은 전술한 히알루론산 복합체의 제조 방법에 관한 것이다.
상기 히알루론산 복합체는 히알루론산과 보론산 화합물을 반응시키는 단계를 통해 제조될 수 있다.
히알루론산은 천연고분자로써 생체적합성이 뛰어나고, 세포외 기질에 자연적으로 존재하는 히알루론산과 독성이 낮으며, 면역원성이 낮다. 또한, 상기 히알루론산은 암 세포의 CD44-수용체와 결합하는 특성을 가진다. 상기 특성을 이용하여 상기 나노입자의 생체 내 암 특이적인 전달이 가능하다.
상기 히알루론산의 분자량은 50 내지 500 kDa, 또는 200 내지 300 kDa일 수 있다. 상기 히알루론산의 분자량은 제조되는 히알루론산 중합체의 사슬의 길이와 연관이 있으며, 이는 글루코오스 감응성 나노입자의 직경에 영향을 준다. 본 발명에서 히알루론산의 분자량은 상기 관점에서 선택될 수 있다.
상기 히알루론산은 히알루론산의 염의 형태로 사용할 수 있다. 염으로는 소듐 히알루로네이트를 사용할 수 있다. 본 발명의 실시예에서는 히알루론산 및 히알루론산 염을 통칭하여 HA라 표현할 수 있다.
본 발명에서는 상기 히알루론산과 보론산 화합물을 반응시키기 전에 히알루론산과 디아민 화합물을 반응시켜 히알루론산을 개질하는 단계를 추가로 수행할 수 있다.
이하, 개질된 히알루론산을 개질 히알루론산 또는 히알루론산 유도체라 표현할 수 있다.
상기 디아민 화합물로는 전술한 성분을 사용할 수 있다.
상기 디아민 화합물은 두개의 아민기를 가지는데, 하나의 아민기는 히알루론산과 결합을 형성하며, 다른 하나의 아민기는 후술할 보론산 화합물과 결합을 형성할 수 있다. 즉, 디아민 화합물은 히알루론산과 보론산 화합물의 결합을 위한 링커로서의 역할을 수행할 수 있다.
구체적으로, 상기 단계에서는 히알루론산과 디아민 화합물이 반응하여 상기 히알루론산에 아민기(-NH2)가 형성된다. 상기 반응은 EDC/NHS 반응(Bartczak. D, Kanaras A.G, Preparation of peptide-functionalized gold nanoparticles using one pot EDC/Sulfo-NHS coupling. Langmuir 2011;27:10119-10123)에 의해 수행될 수 있다. 상기 히알루론산에의 아민기의 형성은 히알루론산의 반복단위, 즉 D-글루쿠론산 및 D-N-아세틸글루코사민이 글리코시딕 결합에 의해 연결된 반복단위 모두에 형성되는 것은 아니다.
본 발명에서 히알루론산 또는 개질 히알루론산은 보론산 화합물과 반응하여 히알루론산 복합체를 형성한다. 상기 히알루론산 복합체는 전술한 바와 같이 상기 화학식 1의 반복단위의 카르복실기에 보론산 화합물이 결합된 히알루론산-보론산 반복단위를 포함한다. 또한, 본 발명의 복합체는 상기 히알루론산-보론산 반복단위와 함께 히알루론산 반복단위를 포함할 수 있다.
본 발명에서 보론산 화합물은 도 1에 나타난 바와 같이, 글루코오스와 결합을 형성한다. 이를 통해 글루코오스의 해당작용을 억제시키고, 암 성장을 억제할 수 있다.
본 발명의 보론산 화합물은 전술한 종류를 사용할 수 있다.
일 구체예에서 보론산 화합물은 하기 구조를 가질 수 있다.

상기 화학식 2에서 디하이드록시보로닐기와 아마이드기는 서로 오르쏘(o), 메타(m), 또는 파라(p)에 위치할 수 있고, 바람직하게는 파라에 위치할 수 있다. 또한, n은 1 내지 5의 정수, 또는 2 내지 3의 정수일 수 있다.
본 발명의 실시예에서는 보론산 화합물로 N-(4-페닐보로닉)숙시남산을 사용할 수 있다. 다만, 이는 본 발명의 일 구체예일뿐이며, 본 발명인 나노입자의 글루코오스 포집 능력은 도 1에 나타낸 바와 같이 보론산기의 존재 여부에 의하여 결정되는 것으로, 반드시 N-(4-페닐보로닉)숙시남산이 사용될 필요는 없다.
본 발명에서는 히알루론산 또는 개질 히알루론산 및 보론산 화합물의 반응을 통해 결합이 형성된다. 이러한 결합은 히알루론산 또는 개질 히알루론산과 보론산 화합물 사이의 화학적 결합을 의미한다. 상기 결합은 카복실기와 아민기 사이에 이루어지는 아미드 결합일 수 있다.
본 발명에서 히알루론산 또는 개질 히알루론산에 결합되는 보론산 화합물의 치환도(DS)는 15 내지 55, 18 내지 40, 또는 20 내지 35일 수 있다. 상기 보론산 화합물의 치환도는 히알루론산의 반복단위, 즉 D-글루쿠론산 및 D-N-아세틸글루코사민이 글리코시딕 결합에 의해 연결된 반복단위 100 유닛 당 보론산 화합물이 결합된 반복단위의 수로 정의된다. 15 이상의 치환도를 갖는 경우 제조되는 복합체의 구조적 안정성을 유지할 수 있으며, 55를 초과하는 치환도를 갖는 복합체는 나노입자 형성에 있어 어려움이 따르며 효율적이지 못하다. 상기 치환도에 의해 제조되는 히알루론산 복합체에서 히알루론산-보론산 반복단위의 함량은 히알루론산 복합체 100 몰부에 대하여 15 내지 55 몰부를 가지게 된다.
또한, 본 발명에서는 히알루론산 복합체가 자가-결합하여 구형입자를 형성하는 단계를 포함한다.
상기 히알루론산 복합체를 수용액, 예를 들어 증류수 내에 분산시키면, 상기 복합체는 자발적으로 자가-집합(self-aggregated)되어 다수의 소수성 코어를 갖는 구형입자(spherical particle)를 형성한다. 본 발명의 히알루론산 복합체는 양친매성의 성질을 갖는데, 소수성의 성질을 갖는 보론산 부분이 다수의 내부 코어를 형성하며 안정한 구형입자를 형성할 수 있다. 이러한 구형상은 체내의 일반적인 글루코오스 농도 환경에서는 그 형상을 유지하며, 암 조직이 가지는 특이적인 고농도 글루코오스 환경에서 감응성을 보이게 된다.
또한, 본 발명에서는 글루코오스 감응성 나노입자에 형광 신호 물질을 표지하거나, 소수성 코어에 상기 형광 신호 물질을 로딩하는 단계를 추가로 포함할 수 있다.
상기 형광 신호 물질로는 전술한 종류를 제한없이 사용할 수 있다. 또한, 형광 신호 물질의 표지 또는 로딩은 당업계의 일반적인 방법을 사용하여 수행할 수 있다.
또한, 본 발명은 글루코오스 감응성 나노입자를 포함하는 조성물에 관한 것이다.
상기 조성물은 간암의 진단 또는 치료용으로 사용할 수 있으며, 진단 동시 치료용으로 사용할 수 있다.
본 발명의 조성물은 글루코오스 감응성에 의해 암 세포의 호기성 해당 대사(Glycolytic metabolism)를 억제한다. 암 세포 내로 이입된 글루코오스 감응성 나노입자는 글루코오스를 포집하여 효과적으로 이들의 해당작용을 억제시키고, 암 성장을 억제할 수 있게 한다. 특히, 본 발명의 조성물에 의한 해당 작용 억제는 세포에 직접적으로 타격을 주지 않는다.
본 발명의 조성물은 포도당 감응성에 의해 암 조직 특이적 진단이 가능하다. 본 발명의 조성물은 암 조직에 증진된 투과 및 유지 효과(Enhanced Permeability and Retention(EPR) effect)에 의해 선택적으로 전달되고, 글루코오스 감응성을 기반으로 글루코오스와 결합하여 암 조직을 영상화할 수 있다. 본 발명의 조성물은 종래 공지된 또는 앞으로 개발될 체내 고분자 거동을 관찰할 수 있는 모든 방법을 통하여 암세포를 관찰하는 것을 가능케 한다.
일 구체예에서, 형광 표지된 글루코오스 감응성 나노입자와 소광제(BHQ-3, Black hole quencher-3)가 결합된 글루코오스 비감응성 컨쥬게이트의 혼합입자를 제조하고, 이를 암 세포 내로 전달시킨다. 채내에서 글루코오스 감응성 나노입자는 글루코오스와 선택적으로 결합하고 상기 나노입자가 분해됨에 따라, 소광제 영역에서 벗어나 퀀칭되었던 형광 신호가 복원되는 원리를 통하여 암 특이적 영상화가 가능하다. 상기 진단 방법은 이에 제한되지 않으며, 다른 이미징 방법들을 제한없이 사용할 수 있다.
본 발명의 조성물은 유효성분으로서의 글루코오스 감응성 나노입자 외에 약제학적으로 허용되는 담체를 추가로 포함할 수 있다. 상기 담체로는 제제시에 통상적으로 이용되는 것으로, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, DMSO, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 사용할 수 있다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다.
본 발명의 약제학적 조성물은 비경구 투여가 바람직하고, 예를 들어, 정맥내 투여, 복강내 투여, 근육내 투여, 피하투여 또는 국부 투여를 이용하여 투여할 수 있다.
본 발명의 일 구현예에 따르면, 본 발명의 조성물은 간 조직에 대한 국소적 투여 또는 전신적 투여에 의해 체내 주입된다. 본 발명인 조성물의 유효성분인 글루코오스 감응성 나노입자는 암세포에 대한 표적지향성을 갖고, 특히 간 특이적 전달의 특성을 가지므로, 간 조직에 국소 투여 하거나 전신적 투여를 통하여 간암의 진단 및 치료가 가능하다. 본 발명의 간암은 일반적인 간세포암뿐만 아니라, 간내 담도암, 다른 조직으로부터 전이되어 간 조직에서 발생된 악성 종양과 같이 간 조직 내에 위치한 모든 악성 종양을 포함한다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 질병 증상의 정도, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하며, 보통으로 숙련된 의사는 목적하는 치료에 효과적인 투여량을 용이하게 결정 및 처방할 수 있다. 한편, 본 발명의 조성물의 투여량은 바람직하게는 1일 당 0.001-1000 mg/kg(체중)이다.
본 발명의 약제학적 조성물은 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화 됨으로써 단위 용량 형태로 제조되거나 또는 다용량 용기내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.

소듐 히알루로네이트(Sodium hyaluronate, HA)(MW = 200 kDa)는 Lifecore(USA)로부터 구입하였다. N-(4-페닐보로닉)숙시남산(N-(4-Phenylboronic) succinamic acid, PBA), 에틸렌디아민(ethylenediamine), 1-에틸-3-(디에틸아미노프로필)-카보이미드(1-ethyl-3-(dinethylaminopropyl) carbodiimide, EDC), 산화중수소(deuterium oxide, D2O), 디메틸 술폭시드(dimethyl sulfoxide, DMSO), deuterated dimethyl sulfoxide (DMSO-d6), 글루코오스(glucose), 자일로오스(xylose), 2-데옥시-D- 글루코오스(2-deoxy-D-glucose) 및 3-(4,5-디메틸티아졸-2-일)-2,5-디페닐 테트라졸륨 브로마이드(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium boromide, MTT)는 시그마-알드리히사(Sigma Aldrich)(USA)로부터 구입하였다.
인산염완충식염수(Phosphate buffered saline, PBS), 둘베코수정이글배지(Dulbecco's modified Eagle's medium, DMEM), 페니실린-스트렙토미신(penicillin-streptomycin), 트립신-EDTA(trypsin-EDTA) 및 소태아혈청(fetal bovine serum, FBS)은 Gibco로부터 구입하였다. N-하이드록시술포숙신이미드(N-hydroxysulfosuccinimide, sulfo-NHS)는 Thermo Scientific로부터 구입하였다. EZ-Cytox는 Dogen(Korea)로부터 구입하였고, 염산 독소루비신(doxorubicin hydrochloride)은 LC Laboratories(USA)로부터 구입하였다. Flamma 675 carboxylic acid은 BioActs(Korea)로부터 구입하였다. 물은 증류하였고, 밀리-Q 시스템을 이용하여 탈이온화하였다. 면역결핍 쥐(Immunodeficient mice)(BALB/C nude, male, 5 week-old)는 Orient Bio Inc로부터 구입하였다.

1. 글루코오스
감응성
나노입자 제조
카보디이미드 화학공법(carbodiimide chemistry)을 통해 히알루론산(HA)의 백본(backbone)에 PBA를 공유결합(covalently conjugated)시켜 글루코오스 감응성 나노입자를 제조하였다. 이하, 글루코오스 감응성 나노입자는 HA-PBA 나노입자 또는 HAPBA 나노입자라 표현할 수 있다.
본 발명에서는 목적하는 치환도(DS)에 따라 각 성분들의 함량을 용이하게 조절할 수 있다.
(1) 히알루론산 개질 (히알루로네이트-에틸렌디아민 결합체 (Hyaluronate-ethylenediamine conjugate) 제조)
소듐 히알루로네이트를 MES 완충용액(pH 6.5)에 용해시켜, 1%(w/w) 농도의 용액을 제조하고, 에틸렌디아민을 첨가한 후, EDC 및 술포-NHS를 용액에 첨가하였다. 하룻밤동안 상온(room temperature)에서 반응시킨 후, 3일 동안 증류수로 투석하여 잔류 시약(residual reagents)을 제거하였다. 투석 후, 용액을 동결건조 하였다. 상기에 의해 히알루론산에 아민기가 도입되었다.
(2) 히알루론산 복합체 합성
(1)에서 제조된 개질 히알루론산이 용해된 MES 완충용액(1% w/v) 및 PBA가 용해된 메탄올(1% w/v)을 혼합하였다. 상기 혼합액에 EDC 및 술포-NHS를 첨가하였다. 반응 후, 혼합물을 투석 백(MWCO = 3500)으로 로드하고, 물 및 메탄올의 혼합액으로 4일 동안 투석하였다. 투석 후 용액을 동결건조하였다.

에틸렌디아민은 카보디이미드 화학공법에 의해 HA의 카르복시기에 화학적으로 결합된다. PBA는 히알루로네이트-에틸렌디아민 결합체(conjugate)에 화학적으로 도입된다.
(3) 글루코오스

(2)에서 제조된 히알루론산 복합체를 증류에 용해시켰다.
히알루론산 복합체의 농도에 따라, 농도가 0.08 mg/ml 이상에서 상기 히알루론산 복합체는 자발적으로 조립되어 글루코오스 감응성 나노입자(HA-PBA 나노입자)를 형성한다.
이하, HA-PBA 나노입자에서 HA에 PBA가 결합되는 치환도(DS)에 따라, 치환도가 20일 경우 HA-PBA 20(HAPBA 20)로, 치환도가 33일 경우 HA-PBA 33(HAPBA 33)으로 표현하였다.
실험예
1. 히알루론산 복합체의 형성 확인
히알루론산 복합체의 형성 유무를 FTIR과 1H-NMR을 통하여 확인 하였다.
HA와 PBA의 결합(conjugation)은 500-2200 cm-1의 범위에서 FTIR spectroscopy(Nicolet 6700, Thermoscientific)에 의해 측정되었다.
도 3은 히알루론산 복합체의 FTIR 분석 결과를 나타낸다. 상기 도 3에서 (a)는 HA, (b)는 히알루로네이트-에틸렌디아민 결합체, (c)는 치환도가 히알루론산 복합체의 분석 결과를 나타낸다.
상기 도 3에 나타난 바와 같이, (b) 및 (c)는 1730 cm-1에서의 C=O stretch의 피크가 감소하는 확인할 수 있다. 이는 결합에 의해 HA의 카르복실기를 소모했기 때문이다. 1560 cm-1에서의 흡수 피크는 아민 결합이 형성에 해당하는 N-H 결합을 나타낸다.
HA와 PBA의 결합(conjugation)은 또한 1H-NMR spectroscopy를 통해 확인할 수 있다.
도 4는 (a) 히알루론산, (b) 보론산 화합물 그리고 (c) 히알루론산 복합체의 1H-NMR 분석 결과를 나타낸다.
HA의 아세틸 양성자의 피크는 1.9 ppm에서 나타났다. 사카라이드 양성자의 피크는 3.5-3.9 ppm에서 관찰되었다. 7.4-7.8 ppm의 피크는 HA-PBA 결합을 나타낸다. 이 결과는 PBA가 HA에 성공적으로 결합되었음을 나타낸다.
DS 값은 히알루론산에 결합되는 보론산 화합물의 비율을 변화시킴으로 조절할 수 있다. 실제 DS 값은 20 내지 33의 범위였다.
실험예
2. 나노입자의 크기 및 형태 확인
HA에 결합된 PBA의 치환도(DS)를 원소 분석기(FLASH EA1112)를 이용하여 분석하였다. HA-PBA 나노입자(1 mg/ml)의 입경은 동적 광 산란(Dynamic light scattering, DLS)(Nano ZS, Malvern instrument)에 의해 측정되었다. 또한, HA-PBA 나노입자의 형태(morphology)는 원자력현미경(atomic force microscopy, AFM)에 의해 관찰되었다. HA-PBA 나노입자는 실리콘 웨이퍼의 표면에 놓았으며, 이미지 촬영 전에 2시간 동안 건조시켰다.
상기 측정 결과를 하기 표 1에 나타내었다.

자가 조립된 HA-PBA 나노입자는 수성 조건에서 제조되었다. 상기 나노입자의 평균 직경은 치환도(DS)가 20 및 33일 때, 각각 257 nm 및 223 nm였다.
도 5는 합성된 나노입자의 형태를 확인하기 위한 AFM 이미지를 나타낸다.
상기 도 5를 통해 HA-PBA 나노입자가 구형을 지니는 것을 확인할 수 있다.
실험예 3. 히알루론산 복합체의 임계 마이셀 농도(critical micelle concentration, CMC) 측정
(1) 방법
히알루론산 복합체의 임계 마이셀 농도(CMC)는 형광 분광법으로 분석하였다.
피렌(pyrene)을 테트라하이드로퓨란(tetrahydrofuran, THF)에 용해시킨 후 증류수에 희석하고(12 × 10-7 M), 상이한 농도를 가지는 히알루론산 복합체 용액에 첨가하였다. 상기 히알루론산 복합체 용액의 농도는 1.0×10-5 mg/ml에서부터 1.0×10 mg/ml였다. 피렌 여기 스펙트라(pyrene excitation spectra)를 얻기위하여, 방출(emission) 및 여기(excitation)를 위한 슬릿 너비(slit widths)를 각각 5 nm 및 2.5 nm로 조절하였다. CMC는 농도에 따른 390 내지 378 nm의 파장에서 강도비(intensity ratio)의 플롯으로부터 결정되었다.
또한, HA-PBA 나노입자의 글루코오스 감응성(responsiveness)을 결정하기위해, 글루코오스(0 내지 10 mg/ml)의 존재하에서 나노입자의 크기 및 형광 강도의 변화를 측정하였다.
(2) 결과
도 6은 히알루론산 복합체의 CMC(임계 마이셀 농도)를 측정한 결과를 나타낸다.
본 발명에서는 HA-PBA 나노입자에 포집된 피렌의 강도비(I390/I378)를 측정하여 CMC를 계산하었다. 피렌은 주변의 친수성 및 소수성 환경에 의해 서로 다른 광물성(photophysical characteristics)을 가진다.
히알루론산 복합체가 마이셀을 형성하는 최소 농도를 구하기 위하여, 히알루론산 복합체의 농도에 따른 형광 강도 비율(fluorescence intensity ratio) 값을 측정하였다. 측정 결과 치환도가 20일 경우 CMC 값은 0.08 mg/ml였다. 즉, 히알루론산 복합체는 0.08 mg/ml 이상의 농도에서 마이셀, 즉 나노입자를 형성한다는 것을 확인할 수 있다(도 6(a)).
또한, 치환도가 20인 HA-PBA 나노입자(1mg/ml)를 사용하여 다양한 글루코오스 농도 조건에서 여기 스펙트럼을 측정하였다.
글루코오스의 농도는 1.0×10-5 mg/ml에서부터 1.0×10 mg/ml로 변화시키면서 측정하였다. 글루코오스-감응성 HA-PBA 나노입자에 대한 피렌의 형광 강도 비율은 1.48 mg/ml에서 감소하였다. 이는 HA에 결합된 PBA와 글루코오스의 디올 사이의 상호작으로인해 자가-조립 구조가 붕괴되었음을 시사한다. 즉, 1.48 mg/ml 초과의 농도에서 나노입자의 마이셀 형태가 붕괴되는 것을 확인할 수 있다(도 6 (b)).
또한, 합성된 나노입자가 다른 단당류와 특이적인 감응성을 가지는지 확인하기 위하여 자일로오스(Xylose) 존재하에서 CMC를 측정하였다.
도 7은 다양한 다양한 농도의 자일로오스의 존재하에서 HA-PBA 나노입자(1 mg/ml)의 피렌 여기 스펙트라를 측정한 결과를 나타낸다.
자일로오스는 HA-PBA 나노입자의 안정성에 영향을 미치지 않았다. 이는, HA-PBA 나노입자는 정상 글루코오스 농도(1 mg/ml)에서 안정성과 구조를 유지할 수 있음을 의미한다. 또한, 본 발명의 시스템은 다른 단당류(monosaccharides)에 의해 변경될 수 없음을 의미한다.
일반적으로, 평균 혈중 당 농도는 1 mg/ml이다. 본 발명의 히알루론산 복합체는 1.48 mg/ml 초과의 글루코오스 농도에서 상기 나노입자(자가-조립 구조)가 붕괴하므로, 이하 약 1.5 mg/ml 미만의 글루코오스 농도를 저농도 글루코오스로, 1.5 mg/ml 이상의 글루코오스 농도를 고농도 글루코오스로 표현하였다. 실시예에서 저농도 글루코오스는 1 mg/ml, 고농도 글루코오스는 4.5 mg/ml를 의미한다.
실험예 4. 나노입자의 독성 평가
(1) 방법
히알루론산, 보론산 화합물 재료가 독성을 갖고 있는지 확인하기 위하여, HepG2(Hepatocellular carcinoma), HCT 116(colorectal carcinoma) 세포를 사용하여 MTT 분석을 통해 세포독성(세포 생존력)을 평가하였다.
HepG2 및 HCT 116 세포를 각각 DMEM(10% FBS, 1% penicillin streptomycin) 및 RPMI 1640 media(10% FBS, 1% penicillin streptomycin)에서 배양하였고, 세포 독성 시험에 사용하였다.
세포를 96-웰 조직 배양 플레이트(96-well tissue culture plate)에 5×103 cells/well로 씨딩(seeding)하고, 치환도가 20인 HA-PBA 나노입자(농도: 0.1 또는 0.5 mg/ml)를 처리한 후, 37℃에서 5% CO2 조건하에서 24시간 동안 인큐베이션하였다.
인큐베이션 후, 세포를 PBS로 세번 세척하고, 1 mg/ml MTT 용액을 처리하였다. 4 시간 동안 인큐베이션 후, 포르마잔 결정(formazan crystals)을 DMSO에 용해시키고, UV/VIS 분광 광도계(spectrophotometer)(SpectraMax M2e, Molecular Devices)를 사용하여 540 nm에서의 흡광도를 측정하였다.
(2) 결과
고농도 글루코오스 배지(글루코오스 농도: 4.5 mg/ml)에 분산된 다양한 농도의 HA-PBA 나노입자(치환도 20)로 처리한 HepG2 및 HCT 116 세포의 in vitro 세포 독성을 평가하였다. 독성 평가시, HA-PBA 나노입자는 고농도 글루코오스 농도에서 구조를 유지하지 않기 때문에 고농도 글루코오스 조건에서 MTT 분석을 수행하였다.
도 8은 (a) HepG2 세포 및 (b) HCT 116 세포에 대한 HA-PBA 나노입자의 in vitro 세포 독성 측정 결과를 나타낸다.
도 8에 나타난 바와 같이, 0.1 mg/ml, 0.5 mg/ml 농도의 HA-PBA 나노입자 존재하에서 세포의 생존능력은 변화가 거의 없는 것으로 나타났다. 이를 통해 나노입자의 독성이 낮다는 것을 확인할 수 있다.
실험예 5. 나노입자의 해당작용 저해( glycolysis inhibition) 효과 검증(in vitro)
(1) 방법
해당 작용에 의한 세포내 ATP 함량 및 젖산(lactate) 생성은 in vitro에서 조사하였다.
HepG2 세포(5 × 105 cells/well)를 6 웰-조직 배양 플레이트에서 24 시간 동안 배양하였다. 배양된 세포를 저농도 글루코오스 배지에 분산시킨 치환도가 20인 HA-PBA 나노입자(1 mg/ml) 또는 2-데옥시-D-글루코오스(2DG)(0.25 mg/ml)로 12 시간 동안 처리하였다.
2-데옥시-D-글루코오스는 해당작용 억제제로 사용되었다. ATP 함량은 ATP colorimetric/fluorometric assay kit (Biovision)를 사용하여 측정하였다. 젖산 분비(Lactate secretion)는 EnzyChromTM L-lactate assay kit (Bioassay)을 이용하여 정량화하였다. 세포 운명(cell fate)에서 HA-PBA 나노입자에 의한 해당작용 억제 효과는 flow cytometer (BD FACS Caliber)를 사용하여 조사하였다.
간략하게, HepG2 세포는 HA-PBA 나노입자 또는 2DG로 12 시간 동안 처리되었다. Annexin-V-FITC 항체 및 프로피디움 요오드화물(PI)은 각각 세포 사멸(apoptotic) 및 세포 괴사(necrotic cell death)의 마커로서 사용되었다. HepG2 세포 1 × 104 cells/well를 24-웰 조직 배양 플레이트에 씨딩하고, 37°C에서 5% CO2 조건에서 24 시간 동안 인큐베이션하였다. 세포를 PBS로 세척하고, 치환도가 20인 HA-PBA 나노입자(1 mg/ml)를 포함하는 저농도 글루코오스 배지 또는 고농도 글루코오스 배지를 8 시간 동안 처리하였다. 또한, HA-PBA 나노입자를 처리하기 전에, 세포를 저농도 글루코오스 배지에 용해된 히알루로네이트(2.5 mg/ml)로 전처리 하였다. 배지는 매일 신선한 것으로 변경하였다. 미리 결정된 시점에서, 세포를 2 시간 동안 EZ-Cytox으로 처리하고, UV/VIS spectrophotometer을 사용하여 450 nm 에서 흡광도를 측정하였다. 배양 3일 이내에 세포 수의 변화로부터 세포 성장 속도를 계산하였다.
(2) 결과
HepG2 세포에서 HA-PBA 나노입자에 대한 해당작용 저해 효과를 조사하기 위해, ATP 및 젖산(lactate) 함량 변화를 정량화하였다. HA-PBA 나노입자가 HepG2 세포에 전달되었을 때, 세포내 ATP 함량은 2-데옥시-D-글루코오스(2DG) 만큼 감소하였다(도 9(b) 참조). 2DG는 일반적으로 글루코오스에서 글루코오스-6-PO4로의 통로를 차단하여 해당 분해에서 ATP 생성을 억제하므로, 해당 억제제로서 사용된다(도 9 (a) 참조).
또한, 도 10은 HepG2 세포의 in vitro 젖산 분비를 나타낸다. 상기 도 10에 나타난 바와 같이, HA-PBA 나노입자가 HepG2 세포에 전달되었을 때, 젖산의 분비는 2-데옥시-D-글루코오스(2DG) 만큼 감소하였다.
이 발견은 HA-PBA 나노입자가 호기성 해당작용의 억제에 효율적임을 나타낸다.
HA-PBA 나노입자에 의한 해당 작용 억제가 세포 사멸(apoptotic) 및 괴사(necrotic)에 의해 세포의 사멸(death)에 영향을 주는지 여부를 유세포 분석법(flow cytometric analysis)으로 조사하였다.
Annexin V은 포스파티딜세린(phosphatidylserine)에 친화성을 가지는 Ca2+ 의존성 인지질 결합 단백질이다. 포스파티딜세린은 세포막의 외부 leaflet에 노출되는 민감한 프로브로 사용되며, 사멸된 세포를 검출하는데 사용된다. 한편, propidium iodide은 누설된(leaky) 괴사 세포의 DNA 만을 염색한다. 점도표 결과 및 강도의 백분율을 도 11에 나타내었다.
대조군 및 2DG 그룹과 비교하여, HA-PBA 나노입자 처리 그룹에서 더 많은 사멸 세포가 발견되었다. 세포사멸/in-late apoptotic HepG2 세포의 비는 대조군 및 2DG 처리 세포에서 각각 6.76%/5.77% 및 18.81%/4.65%이었다. 그러나, HA-PBA 나노입자 처리 그룹에서 세포사멸 및 in-late apoptotic HepG2 세포의 수는 22.83% 및 19.55%이었다. 이는 HA-PBA 나노입자가 해당 작용을 억제할 뿐만 아니라, 간세포 암종 세포에서 세포사멸을 유도한다는 것을 나타낸다.
HA-PBA 나노입자에 의한 세포 성장의 억제를 확인하기 위하여, 상기 나노입자의 세포내 이입 및 HepG2 세포의 성장 저해를 평가하였다. 세포를 HA-PBA 나노입자로 8 시간 처리하고, 저농도 글루코오스 배지 또는 고농도 글루코오스 배지에서 3일 동안 배양하였다. 또한, HA-PBA 나노입자의 세포내함입(internalization)이 CD44 수용체에 의해 매개되었는지 테스트하기위해 저농도 글루코오스 배지에 용해된 가용성 HA로 세포를 전처리 하였다.
도 12는 저농도 글루코오스 배지, 고농도 글루코오스 배지, HA-PBA 나노입자가 포함된 저농도 글루코오스 배지, 나노입자가 포함된 고농도 글루코오스 배지 및 HA로 전처리되며 HA-PBA 나노입자가 포함된 저농도 글루코오스 배지로 매일 처리된 HepG2 세포의 성장 결과를 나타낸다.
세포의 성장 속도는 고농도 글루코오스 배지로 처리된 세포에서 증가하였다. 이는 고농도 글루코오스 조건에서 HA-PBA 나노입자의 분해가 일어나므로 세포 성장 억제 효능이 관찰되지 않은 것이다. 그러나, 저농도 글루코오스 배지에서는 세포 성장속도가 감소하여다. 이는 저농도 글루코오스 조건에서 나노입자는 그 형태를 유지하며, 상기 나노입자가 호기성 해당작용의 억제를 유도하는 것으로 예측된다.
고농도 글루코오스 배지에 HA-PBA 나노입자를 처리하기 전에, CD 44 수용체를 차단하기 위해 가용성 HA로 CD44-양성 HepG2 세포를 전처리했을 때, 세포 성장 속도는 나노입자에 의해 방해받지 않았다. 이는, HA-PBA 나노입자가 정상 글루코오스 수준(저농도 글루코오스 수준)의 배지에서 구조를 유지할 수 있고, 암세포에서 해당 작용을 억제하는 세포의 CD 44 수용체와 결합할 수 있음을 나타낸다.
실험예 6. 세포 이입(in vitro)
(1) 방법
HepG2 및 NIH3T3에서의 글루코오스-감응성(responsive) HA-PBA 나노입자의 수용체매개세포내섭취(Receptor-mediated endocytosis)는 공초점 주사 레이져 현미경(confocal laser scanning microscopy)(Olympus, FV1200)을 사용하여 측정하였다.
Cy5.5 결합 HA-PBA 나노입자(치환도 20)를 먼저 제조하였다.
HA-PBA 나노입자 및 Cy5.5-NHS를 증류수에 용해시켰다(Cy5.5/polymer = 0.01 w/w). 혼합물을 하룻밤동안(avoiding light) 반응시켰다. 용액을 4일 동안 투석하고 동결건조시켰다.
HepG2 세포 및 NIH3T3 세포 1 × 104 cells/well를 12-웰 비-조직 배양 플레이트에 씨딩하고, 24 시간 동안 인큐베이트하였다. 세포를 Cy5.5 결합 HA-PBA 나노입자 (1 mg/ml)로 처리하였다. 4 시간 후, 배지를 제거하고, PBS로 3회 세척하였다. 세포를 4% 포름알데하이드로 10 분간 고정시킨 후, 4’,6’-diamino-2-phenylindole(DAPI, Vectashield)를 포함하는 봉입재(mounting medium)를 사용하여 슬라이드 글래스에 장착하였다.
CD44 수용체매개세포내섭취를 평가하기 위하여, 나노입자를 첨가하기 전에 세포를 저농도 글루코오스 배지(2.5 mg/ml)에 용해된 히알루로네이트 용액(2.5 mg/ml)으로 2 시간 전처리하였다.
Cy5.5 결합 HA-PBA 나노입자 및 lysotracker green DND-26 (Invitrogen)는 HepG2 및 NIH3T3에 의해 글루코오스-감응성 HA-PBA 나노입자의 세포내 이입을 모니터링하기 위해 사용되었다.
HepG2 세포 또는 NIH3T3 세포 1 × 104 cells/well는 12-웰 비-조직 배양 플레이트에 씨딩하고, 사용 전 24 시간 인큐베이트하였다. Cy5.5 결합 HA-PBA 나노입자(1 mg/ml)가 플레이트에 첨가하고 4 시간 동안 인큐베이트 하였다. 그 후, lysotracker green DND-26(100 nM)이 플레이트에 첨가하고 2 시간 동안 인큐베이트하였다. 배지를 제거하고, PBS로 3회 세척하였다. 세포를 4% 포름알데하이드로 10 분간 고정시킨 후, 4’,6’-diamino-2-phenylindole(DAPI, Vectashield)를 포함하는 봉입재(mounting medium)를 사용하여 슬라이드 글래스에 장착하였다. 그리고, 형광 현미경(fluorescence microscopy) (Nikon, TE2000-E)에 의해 그 형태를 관찰하였다.
(2) 결과
세포에 의한 HA-PBA 나노입자의 수용체-매개 엔도시토시스(endocytosis)를 조사하였다.
CD44-양성 세포(HepG2) 또는 CD44-음성 세포(NIH3T3)를 Cy5.5-결합 HA-PBA 나노입자에 처리하여 CD44-의존 엔도시토시스를 조사하였다.
도 13은 저농도 글루코오스 배지 또는 고농도 글루코오스 배지에서 Cy5.5 결합 HA-PBA 나노입자가 처리된 (a) HepG2 및 (b) NIH3T3 세포의 공초점 현미경 이미지를 나타낸다.
상기 도 13에 나타난 바와 같이, 가용성 HA-처리군에 비해 저농도 글루코오스 배지로 배양된 HepG2 세포에서 Cy5.5의 신호가 높게 검출된 것을 확인할 수 있다. 대조적으로, 고농도 글루코오스 배지에서 배양된 HepG2 및 CD44-음성 NIH3T3 세포는 유의한 신호가 관찰되지 않았다. 이러한 결과는 HA-PBA 나노입자가 고농도 글루코오스 조건 하 및 CD44 음성 세포에 의해서 잡히지 않는다는 것을 시사한다. 또한, 용해성 HA로 전 처리하면 세포 내 CD44 수용체가 차단되어 세포 흡수가 감소되었다.
HA-PBA 나노 입자는 CD44 양성 세포에 우수한 결합 친화력을 가지며, 수용체-매개 엔도시토시스는 일차 세포 흡수 경로가 될 수 있다.
또한, lysotracker green DND-26은 HA-PBA 나노입자의 세포내 이입을 추적하는데 사용되었다.
lysotracker green은 산성 세포기관(organelles)에 높은 선택성을 가지며, 추적에 자주 사용된다. 형광 현미경에 나타난 바와 같이, Cy5.5-결합 HA-PBA 나노입자의 신호는 고농도 글루코오스 조건 또는 CD44-음성 세포와 비교하여 저농도 글루코오스 조건에서 배양된 HepG2 세포에서 높게 검출되었다. 이는 HA-PBA 나노입자가 endocytic pathway를 통해 HepG2 세포에 이입된 것을 나타낸다.
실시예 7. 종양-표적 능력(In vivo)
(1) 방법
종양-보유 마우스 모델(tumor-bearing mouse model)에서, HA-PBA 나노입자의 생체 내 분포(biodistribution)를 조사하기 위해, 누드마우스(athymic nude mice)(20g, 5 주령, Orient lab animal)에 마취제(Zoletil(35 mg/kg)/Rompun(2 mg/kg))를 투여한 다음, HepG2 세포 현탁액(5 × 106 cells /mouse) 을 마우스의 왼쪽 옆구리에 접종하였다.
14일 후, Cy5.5 결합 HA-PBA 나노입자를 10 mg/kg의 용량으로 마우스의 꼬리 정맥 내로 주사하였다. 주사 24 시간째에, 마우스를 희생시키고, 주요 기관 및 종양을 추출하고, Kodak image station (Kodak Image Station 4000 MM, Kodak)을 사용하여 관찰하였다. (n=3)

(2) 결과
도 15는 in vivo에서 (a) HA-PBA 나노입자의 생체분포 및 (b) 종양 보유 마우스 모델에서 종양 특이성을 정량화한 결과를 낸다.
Cy5.5 결합 HA-PBA 나노입자의 가장 강력한 신호는 정상 장기와 비교하여 종양 부위에서 검출되었다. 이는 EPR(enhanced permeation and retention) 효과뿐만 아니라 리셉터-매개 엔도시토시스에 의해 종양 부위에서 축적된 것으로 예측된다. HARE(hyaluronan receptor for endocytosis) 및 세망내피계(RES)를 발현하는 간 시누소이드 내피세포(liver sinusoidal endothelial cells)에 의한 HA-PBA 나노입자의 세포내 이입에 의해 간에서 신호가 검출되었다. 이러한 결과는 HA-PBA 나노입자가 종양 부위에 축적되어 암 치료에 대한 잠재력을 나타낼 수 있음을 나타낸다.
실험예 8. in vivo 치료 효과 평가(간암 성장 저해 효과)
(1) 방법
HA-PBA 나노입자의 치료 효과를 평가하기 위해, 실험예 7의 종양-보유 마우스 모델(tumor-bearing mouse model)을 준비하였다.
HA-PBA 나노입자(20 mg/kg), 파클리탁셀(paclitaxel)(10 mg/kg) 및 PBS를 종양 보유 마우스의 꼬리 정맥에 주사하였다(1주일 동안 5회 주사). 종양 부피의 변화를 2주 동안 모니터링하였다(n=7). 마우스로부터 종양 조직을 추출하고, optimal cutting temperature(OCT) 화합물에 넣었다. 종양 조직을 얇게 썰어(12 μm 두께), 헤마톡실린-에오신(hematoxylin and eosin, H&E) 및 DeadEndTM colorimetric TUNEL 시스템(Promega, USA)으로 염색하였다.
(2) 결과
*마우스의 종양 부피가 약 90 mm3에 도달하였을 때 HA-PBA 나노입자, 파클리탁셀 및 PBS를 주사하였다.
도 16은 in vivo에서 (a) HA-PBA 나노입자의 치료 효능 결과 및 (b) 종양 조직의 H & E 및 TUNEL 염색 결과를 나타낸다.
종양 부피 및 체중을 17일 동안 모니터링 한 결과, HA-PBA 나노입자 및 파클리탁셀을 처리한 그룹은 PBS 군과 비교하여 종양 성장률이 억제되었다. 모든 그룹에서 체중 감소는 관찰되지 않았다.
이 결과는 HA-PBA 나노입자가 원하지 않는 부위에서 부작용 없으며, 우수한 치료 효과를 가짐을 나타낸다. 이 향상된 치료 효능은 HA-PBA 나노입자의 종양-표적화 능력에 기인한다.
또한, 종양 조직의 조직학적 이미지는 HA-PBA 나노입자의 우수한 치료 효능을 나타낸다. HA-PBA 나노입자로 처리된 마우스에서 다수의 사멸 세포가 관찰되었다. 이러한 향상된 치료 효능은 글루코오스-반응성 나노입자가 종양 부위에서 해당 작용을 막음으로서 종양 세포의 증식을 억제함을 나타낸다.

히알루론산-보론산 반복단위의 함량은 히알루론산 복합체 100 몰부에 대하여 15 내지 55 몰부인 히알루론산 복합체.
- 제 1 항에 있어서,
히알루론산-보론산 반복단위에서 보론산 화합물은 아미드 결합(amide bond)을 통해 히알루론산 반복단위에 결합되는 히알루론산 복합체.

보론산 화합물은 페닐보론산인 히알루론산 복합체.
- 제 4 항에 있어서,
페닐보론산은 N-(4-페닐보로닉)숙시남산, 3-카르복시벤젠벤조익산, 4-카로복시피리딘-3-벤조익산 및 (3-아미노메틸페닐)벤조익산 클로라이드로 이루어진 그룹으로부터 선택된 하나 이상인 히알루론산 복합체.
