Abstract
일 양상에 따른 중간엽줄기세포, AgRP 또는 그의 활성 단편을 포함하는 조성물은 유비퀴틴 프로테아좀의 활성을 증가시켜 유비퀴틴 프로테아좀 활성 감소와 관련된 질환, 예를 들면, 퇴행성 뇌질환의 예방, 치료, 또는 개선에 유용하게 사용될 수 있는 효과가 있다.
Description
중간엽 줄기세포 또는 AgRP를 포함하는 유비퀴틴 프로테아좀 활성 증가를 통한 퇴행성 뇌질환 치료제에 관한 것이다.
퇴행성 뇌질환은 나이가 들어감에 따라 발생하는 퇴행성 질환 중에서도 뇌에서 발생하는 질환을 의미하며, 주요 증상과 침범되는 뇌부위를 고려해 구분할 수 있는데, 알츠하이머 질환이나 파킨슨 질환 등이 포함된다. 퇴행성 뇌질환은 노화에 따른 신경퇴화와 유전적 및 환경적 요인들로 인해 단백질이 응집돼 신경세포가 사멸해 야기되는 것으로 알려져 있지만, 아직 정확한 원인은 밝혀지지 않아 원인 규명을 위한 기초 연구가 활발히 진행되고 있는 상황이다.
한편, 미국의 어윈 로즈와 이스라엘의 아론 치카노베르, 아브람 헤르슈코 박사는 유비퀴틴(ubiquitin)이라는 분자가 붙어있는 단백질을 유비퀴틴 프로테아좀 시스템 (Ubiquitin-proteasome system; UPS)이 선택적으로 분해하는 과정을 밝혀내 2004년 노벨 화학상을 수상 하였다. 유비퀴틴 프로테아좀 시스템은 진핵 세포에서 불량 단백질 제거, 세포 분열, 손상된 DNA 수리와 같은 생체 과정에 관여한다. 퇴행성 뇌질환과 같은 질병에서 유비퀴틴 프로테아좀 시스템이 이상이 생기게 되면 독성 단백질이 생체 내 축적이 되는 현상을 일으키게 된다. 특히, 퇴행성 뇌질환인 알츠하이머병에서, 아밀로이드 단백질이 유비퀴틴 프로테아좀 시스템에 활성을 억제하고, 알츠하이머병 동물 모델에서 뇌의 프로테아좀 활성이 감소된다는 것이 보고된바 있다(비특허 문헌 1 내지 6 참조).
이에 본 발명자들은, 뇌의 유비퀴틴 프로테아좀 활성을 조절하는 물질을 찾기 위해 노력한 결과, 중간엽줄기세포가 신경 세포의 프로테아좀 활성을 증가시킨다는 것을 확인하였고, 프로테아좀 활성 조절 단백질인 AgRP(Agouti related peptide)을 분비한다는 것을 동정함으로써, 본 발명을 완성하였다.

용어 "치료"는 질환, 장애 또는 병태, 또는 그의 하나 이상의 증상의 경감, 진행 억제 또는 예방을 지칭하거나, 그를 포함하며, "유효성분" 또는 "약제학적 유효량"은 질환, 장애 또는 병태, 또는 그의 하나 이상의 증상의 경감, 진행 억제 또는 예방에 충분한 본원에서 제공되는 발명을 실시하는 과정에서 이용되는 조성물의 임의의 양을 의미할 수 있다.

Clin. Invest., 58: 699, 1976)의 문헌들에 개시되어 있다. 상기 중간엽줄기세포는 AgRP(Agouti-related peptide) 또는 그의 활성 단편을 분비하거나, AgRP 또는 그의 활성 단편을 분비하도록 유전적으로 조작된 것일 수 있다.

이하 하기 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예. 인간 중간엽줄기세포를 이용한 퇴행성 뇌질환 치료 효능을 갖는 단백질의 스크리닝 및 그에 의해 동정된 단백질의 활성 분석
1. 인간 중간엽줄기세포를 이용한 퇴행성 뇌질환 치료 효능을 갖는 단백질 스크리닝
1.1. 인간 제대 중간엽줄기세포의 신경 세포에 대한 활성 확인
인간 제대 중간엽줄기세포(Wharton's Jelly-MSC: WJ-MSCs)가 신경 세포의 유비퀴틴 프로테아좀의 활성을 증가시킨다는 것을 확인하기 위해 하기와 같이 실험을 수행하였다.
인간 제대 중간엽줄기세포는 본원에서 승인한 IRB (IRB# 2015-09-023-003)의 기준에 따라 삼성서울병원 산부인과와의 협업 연구를 통하여 탯줄을 확보한 후 기존에 보고된 프로토콜에 따라 중간엽줄기세포 분리 진행을 하였다(Choi, M, Lee, HS, Naidansaren, P, Kim, HK, O, E, Cha, JH, et al. (2013). Proangiogenic features of Wharton's jelly-derived mesenchymal stromal/stem cells and their ability to form functional vessels. Int J Biochem Cell Biol 45: 560-570). 구체적으로, 탯줄을 3-4 cm 길이 자른 다음, 조직을 잘게 자르고 콜라게나아제 용액(Gibco, USA)을 이용하여 세포외기질을 분해하기 위해 60-90 분 동안 처리 해준 다음, 0.25% 트립신 (Gibco, USA)을 추가하여 30분 동안, 37°C에서 분해를 더 유도하였다. 이후에, 소태아혈청(FBS; Biowest, USA)을 첨가하여, 1000g에서 10분 동안 원심분리를 이용하여 세포를 얻고, 둘베코 변형 이글 배지(Dulbecco Modified Eagle Medium) (DMEM; Biowest, USA) 에 20% FBS와 1% 페니실린-스트렙토마이신(Gibco,USA)이 첨가된 배지를 이용하여 37 ℃, 5% CO2 환경에서 세포를 배양하였고, passage 5 에서 6 이된 WJ-MSC를 실험에 사용하였다.
신경 세포는 SH-SY5Y(ATCC, USA)를 사용하였다. 먼저, 6 웰 플레이트(BD Falcon, USA)에서 SH-SY5Y (ATCC, USA)세포가 90% confluency가 되었을 때, transwell insert (BD Falcon, USA)를 각 웰에 삽입하여, insert chamber에 인간 제대 중간엽줄기세포(1X105/1mL)를 분주하여 혈청이 첨가 되지 않은 Minimal Essential Medium alpha 1X (MEMα1x; Gibco-Invitrogen, USA)에서 24시간 동안, 37 ℃, 5% CO2 환경에서, 공동 배양을 하였다.
이후에, 인간 제대 중간엽줄기세포가 신경 세포의 유비퀴틴 프로테아좀의 활성을 증가시키는지 확인하기 위해, 세포에서의 프로테아좀의 주요 분해 활성인 키모트립신 유사 활성, 유비퀴틴 컨쥬게이트 단백질의 양, 및 형광 기질이 도입된 신경세포에서의 형광 세기를 측정하였다. 대조군으로는 중간엽줄기세포와 공동배양하지 않은 신경세포를 사용하였다.
구체적으로, 수집된 세포를 레틱 버퍼(retic buffer) (30 mM Tris (pH 7.8), 5mM MgCl2, 5mM KCl, 0.5mM DTT, 2 mM ATP)를 이용하여 초음파 처리(ultra-sonication)를 한 이후에, 5 μg의 단백질과 키모트립신 유사 형광기질인 Suc-Leu-Leu-Val-Tyr-AMC (Enzo, Republic of Korea)을 200 μM (최종 농도: 100 μM)으로 96 웰 블랙/클리어 플레이트(black/clear plate) (BD Falcon, USA)에 넣고, 37 ℃에서 4시간까지 배양하였다. 반응은 에탄올을 첨가하여 정시시켰다. 키모트립신 유사 형광기질 활성화는 플루오로미터(fluorometer) (Glomax, Promega, USA)를 사용하여 측정을 수행하였고, 그 결과를 도 1a에 나타내었다.
프로테아좀 시스템으로 분해되는 유비퀴틴 컨쥬게이트 단백질의 양의 측정은 웨스턴 블랏팅을 통해 확인하였다. 웨스턴 블랏팅은 먼저, 레틱 버퍼(retic buffer)로 초음파 처리한 단백질을 우레아 버퍼(urea buffer) 와 1:1로 혼합하였다. 이후에, 원심분리를 하여, 상층액을 모아서 Bradford assay (Bio-rad, USA)를 이용하여 단백질 정량을 하였다. 각 샘플에서 10 μg에 단백질을 겔에 로딩한 후, SDS를 이용하여 전기영동을 수행하였다. 단백질을 니트로셀룰로오스 막으로 트랜스퍼시킨 이후에, 5% 탈지유(BD Difco, USA)에 1% TBST (Tris-buffered saline with 0.1% Tween 20) 이 첨가된 용액에 상온에서 1시간 동안 배양하였다. 이후에, 유비퀴틴 컨쥬게이트 (Ub, 1:1000; Santa Cruz, USA) 와 베타 액틴(1:5000; Santa Cruz, USA) 항체를 처리하였고, 4 ℃°C에서 하룻밤동안 배양하였다. 이후, 이차 항체(goat anti-mouse IgG-HRP; Ab frontier, USA)를 한 시간 동안 상온에서 처리하고, 블랏들은 ECL (Advansta, USA) 를 이용하여 발달시켰고, 그 결과를 도 1b에 나타내었다.
형광기질이 도입된 신경세포에서의 형광 세기의 측정은 GFP가 붙어 있는 프로테아좀의 기질은 GFP-degron을 신경 세포에 리포펙타민 2000 (Life Technologies, USA) 형질전환 방법으로 과발현하고, 공초좀 현미경 (LSM 700; Carl Zeiss AG, Jena, Germany)으로 관찰한 후 형광 세기를 Image-J (NIH, USA) 분석 프로그램을 이용하여, 측정하였고 그 결과를 도 1c에 나타내었다.
그 결과, 도 1에 나타낸 바와 같이, 공동 배양을 한 신경 세포에서 프로테아좀의 주요 분해 활성인 키모트립신 유사 활성이 공동 배양을 하지 않은 신경 세포 보다 높고, 유비퀴틴 컨쥬게이트 단백질의 양이 공동 배양을 한 신경 세포에서 감소하였으며, 공동 배양을 한 신경 세포에서 공동 배양을 하지 않은 신경 세포 대비 형광 세기가 줄어드는 것을 확인하였다. 상기의 결과로, 인간 제대 중간엽줄기세포를 신경 세포와 공동 배양시, 신경 세포의 프로테아좀 활성이 높아지는 것을 알 수 있다.
1.2. 인간 제대 중간엽줄기세포로부터 분비되는 단백질의 동정
인간 제대 중간엽줄기세포를 신경 세포와 공동 배양하였을 시, 분비되는 단백질을 동정하기 위해 하기의 실험을 수행하였다.
구체적으로 인간 제대 중간엽줄기세포로부터 분비되는 단백질을 확인하기 위해 레이바이오 사이토킨 어세이(Raybio cytokine assay) 및 정량적 실시간 PCR(real time quantitative reverse transcription PCR)을 수행하였다.
레이바이오 사이토킨 어세이를 하기 위해 공동배양을 한 후 배지를 제거하고, 센트리콘(centricon) (Millipore, USA)을 사용하여 농축하였다. 농축한 배지 샘플들은 509 분비단백질 분석이 가능한 RayBio® Human Cytokine Antibody Arrays - Biotin-Label Based G Series (Raybiotech, USA)를 이용하여 분석을 수행하였다. WJ-MSC 공동배양을 통해 10배 분비가 증가된 단백질들을 하기 수학식 1로 분석을 하였고, 그 결과를 도 2a에 나타내었다.

정량적 실시간 PCR은 먼저 배양한 세포에서 TRIzol (Life Technologies, USA) 를 이용하여 RNA 추출을 하였고, SuperScriptTM II Reverse Transcriptase (Invitrogen, USA)를 사용하여 cDNA 합성을 하였다. PCR를 진행 하기 위해 다음과 같은 프라이머들을 제작 및 사용을 하였다;
AGRP Forward (FOR): AGT CAC GTG TGG CCC TTC AT(서열번호 1)
AGRP Reverse (REV): TCC GGG ATT CTT GCC TAG AG(서열번호 2)
GAPDH FOR: CGA GAT CCC TCC AAA ATC AA(서열번호 3)
GAPDH REV: CCT TCT CCA TGG TGG TGA A(서열번호 4).
Step ONE Plus system (AB, USA) 과 2X Power SYBR Green Master Mix (AB, USA)를 사용하여 PCR을 수행하였고, 그 조건은 다음과 같았다: 95 ℃ 10 분, 95 ℃ 15초, 59℃ 30초(40 cycles). AGRP 발현 분석은 Livak 과 Schmittgen의 2-△△CT 방법(Leitch, AE, Lucas, CD, Marwick, JA, Duffin, R, Haslett, C, and Rossi, AG (2012). Cyclin-dependent kinases 7 and 9 specifically regulate neutrophil transcription and their inhibition drives apoptosis to promote resolution of inflammation. Cell Death Differ 19: 1950-1961.)으로 수행하였고, 그 결과를 도 2b에 나타내었다.
그 결과 도 2에 나타낸 바와 같이, 인간 제대 중간엽줄기세포를 신경 세포와 공동 배양을 하였을 때, AgRP라는 단백질을 분비한다는 것을 확인하였다.
2. 동정된 단백질의 활성 분석
2.1. 동정된 단백질이 신경 세포에서 유비퀴틴 프로테아좀 활성에 미치는 영향 분석
상기 실시예 1.2.에서 동정된 단백질인 AgRP가 신경 세포에서 유비퀴틴 프로테아좀 활성에 미치는 영향을 분석하기 위해 하기와 같이 실험을 수행하였다.
구체적으로, 신경 세포 SH-SY5Y (ATCC, USA)세포가 90% confluency가 되었을 때, 혈청이 첨가되지 않은 Minimal Essential Medium alpha 1X (MEMα1x; Gibco-Invitrogen, USA)에 1, 10, 20, 50ng/mL AgRP (R&D, USA) 를 처리하고, 24시간 동안, 37 ℃, 5% CO2 환경에서 배양을 하였다. 대조군으로는 1, 10, 20, 50ng/mL의 PBS 중 0.1% BSA를 사용하였다.
프로테아좀의 활성은 상기 실시예 1.1과 동일한 방법으로 키모트립신 유사 활성을 확인하였고, 그 결과를 도 3a에 나타내었다. 또한, 상기 실시예 1.1과 동일한 방법으로 유비퀴틴 컨쥬게이트 단백질의 양을 확인하였고, 그 결과를 도 3b 및 3c에 나타내었다.
신경 세포 SH-SY5Y (ATCC, USA)세포가 90% confluency가 되었을 때, serum이 첨가 되지 않은 Minimal Essential Medium alpha 1X (MEMα1x; Gibco-Invitrogen, USA)에 농도별 (0-50 ng/mL)로 AgRP를 처리하고, 24시간 동안, 37 ℃, 5% CO2 환경에서 배양을 하였다. 대조군으로는 PBS 중 0.1% 소태아 혈청(BSA)을 동일한 농도로 SH-SY5Y에 처리 하였다.
추가적으로, 무혈청 DMEM (Biowest, USA)배지에서 생쥐 해마 유래 세포(HT22; 백남인 교수님, 경희대학교)에 프로테아좀 활성 저해제인 MG-132(Calbiochem, USA)를 1 μM의 농도로 처리하였고, AgRP를 농도 별로 (1,10,20,50 ng/mL) 같이 처리하고 24시간 동안, 37 ℃, 5% CO2 환경에서 배양을 하였다. 이후에, 상기 실시예 1.1.과 동일한 방법으로 유비퀴틴 컨쥬게이트 단백질의 양을 확인하였고, 그 결과를 도 4에 나타내었다
그 결과, 도 3에 나타낸 바와 같이, AgRP를 처리한 신경세포가 대조군인 정상 세포에 비해 키모트립신 유사 활성(Suc-Leu-Leu-Val-Tyr-AMC:SUC)이 증가하였고, 유비퀴틴 컨쥬게이트 단백질의 발현 양이 AgRP 농도 의존적으로 감소된 것을 확인하였다.
또한, 도 4에 나타낸 바와 같이, 프로테아좀 저해제인 MG-132를 AgRP와 같이 처리하여 주었을 때, AgRP가 농도 의존적으로 유비퀴킨 컨쥬게이트 단백질의 발현 양이 감소시키는 것을 확인하였다.
상기의 결과로, AgRP가 신경 세포에서 유비퀴틴 프로테아좀의 활성을 증가시킨다는 것을 알 수 있었다.
2.2. 동정된 단백질이 동물 모델에서 프로테아좀 활성에 미치는 영향 분석
AgRP가 동물 모델에서 프로테아좀 활성에 미치는 영향을 분석하기 위해 하기의 실험을 수행하였다. 동물 모델로서는 정상 마우스(C57BL6/J; 10-13개월; Jackson Laboratories, USA) 및 10-13 개월령의 알츠하이머병 동물 모델 5XFAD (Jackson Laboratories, USA)를 사용하였다.
먼저, 정상 마우스의 해마에 2 X 105 세포수(2 X 105/3μl)의 인간 제대 중간엽줄기세포를, 그리고 AgRP (1000ng/kg)을 stereotaxic기계 (Harvard apparatus, USA) 를 이용하여, bregma 좌표를 잡은 후 0.5 μl/min 속도로 해마에 투여 하였다 (A/P -2.3mm, M/L -1.3mm, and D/V -2.0 mm). 대조군으로는 PBS만 투여한 마우스를 사용하였다. 투여 1주일 후에, 정상 마우스를 관동맥 관류로 안락사 시킨 후, 해마를 제거하였다. 제거된 해마를 약사발로 갈은 다음, 상기 실시예 1.1과 동일한 방법으로 단백질을 추출하였다. 프로테아좀 활성은 상기 실시예 1.1.과 동일한 방법으로 측정하였고, 그 결과를 도 5a에 나타내었다.
그 결과, 인간 제대 중간엽줄기세포 와 AgRP를 투여한 군들이 PBS만 투여를 한 군에 비해 프로테아좀 활성이 증가한 것을 확인하였다.
이러한 정상 마우스에서의 실험 결과를 바탕으로 알츠하이머병 동물 모델인 5XFAD에서의 실험을 진행하였다. 실험은 도 5b에 나타낸 바와 같은 좌표를 이용하여 대조군으로 인산완충식염수를 3μl의 용량을 투여하였고, 실험군으로서는 인간 제대 중간엽줄기세포 및/또는 AgRP를 각각 2X105/3μl, 1000ng/kg의 용량으로 상기와 동일한 방법으로 동물 모델의 해마에 투여하였다. 투여 1주일 후에, 상기 정상 마우스에서의 실험과 동일한 방법으로 단백질을 추출하였다. 프로테아좀 활성은 상기 실시예 1.1.과 동일한 방법으로 키모트립신 유사 활성을 측정하였고, 그 결과를 도 5c에 나타내었다.
기존에 보고된 실험들과 동일하게 같은 개월 수 정상 쥐에 비해, 알츠하이머병 동물 마우스 모델 (5XFAD)에서 프로테아좀 활성이 줄어든 것을 확인하고자 예비 실험을 진행 하였다. 6개월 된 정상과 5XFAD 쥐를 안락사 시킨후, 해마를 적출하여 상기와 동일한 방법으로 단백질을 추출하여 프로테아좀 활성은 상기 실시예 1.1.과 동일한 방법으로 유비퀴틴 컨쥬게이트 단백질의 양을 측정하였고, 그 결과를 도 5d에 나타내었다.
그 결과, 인간 제대 중간엽줄기세포 와 AgRP를 투여한 군들은 대조군에 비하여 SUC 활성이 증가하였고, 인간 제대 중간엽줄기세포 및 AgRP를 모두 투여해준 군들의 유비퀴틴 컨쥬게이트 단백질의 양이 대조군에 비해 감소한 것을 확인하였다. 또한 6개월 정상쥐 해마의 비해, 6개월 5XFAD 해마에서 SUC 활성이 낮고 유비퀴틴 컨쥬게이트 단백질의 양이 높다는 것을 관찰하였다.
또한, 5XFAD 마우스 모델 외에 다른 알츠하이머병 동물 마우스 모델인 P301S (이하 “Tau” 마우스 라고 함) 마우스에서 인간 제대 중간엽 줄기세포 그리고 동정된 단백질이 유비퀴틴 프로테아좀 활성을 조절하는 효능을 확인하고자 추가 실험을 진행 하였다. 10개월된 Tau 마우스, 총 4 그룹에, 상기와 동일한 방법으로 인간 제대 중간엽줄기세포와 AgRP를 해마에 투여하였다. 4가지 그룹은 다음과 같았다:
(1) 인간 제대 중간엽줄기세포 (2 X 105/3μl)
(2) AgRP (Low): 20 ng / 3μl
(3) AgRP (High) : 50 ng / 3μl
(4) Vehicle : PBS
투여 1주일 후에, 단기 그리고 장기 기억 변화를 확인하고자 수동적 회피 행동 테스트(passive avoidance behavior test)를 시행하였고, 그 결과를 도 6에 나타내었다.
그 결과, 비어클 대조군 마우스군에 비해 AgRP/MSC 투여 군에서 트레이닝 및 테스트 시도(training and test trial) 에서 모두 인지기능이 향상된 것을 확인하였다.
행동 실험을 진행 한 다음, Tau 마우스를 관동맥 관류로 안락사 시킨 후, 해마를 제거하였다. 제거된 해마를 약사발로 갈은 다음, 상기 실시예 1.1 과 동일한 방법으로 단백질을 추출하였다. 유비퀴틴 컨쥬게이트 단백질의 양의 측정은 상기 실시예 1.1.과 동일한 방법으로 측정하였다. 추가로 인산화된 tau 단백질들의 양을 웨스턴 블랏팅을 통해 측정하고자 다음과 같은 항체들을 사용하였고: Tau 5, AT8, T231, 및 3p-tau, 그 결과를 도 7에 나타내었다.
그 결과, 알츠하이머병의 병인으로 예상되는 고인산화(hyperphosphorylated) tau (Tau 5, AT8, T231로 측정) 그리고 유비퀴틴 컨쥬게이트 단백질의 양이 정상 마우스 와 비교하였을 때 Tau 마우스에서 더 높게 발현되는 반면, AgRP/MSC 투여 군에서는 비정상적인 단백질 응집(abnormal protein aggregate)이 줄어드는 것을 웨스턴 블랏팅을 통해 관찰하였다. 이에 AgRP/MSC가 Ub 컨쥬게이트와 비정상적인 타우를 제거하는 효능이 있음을 확인하였다.
상기의 결과로, 일 구체예에 따른 방법을 사용하여 뇌의 유비퀴틴 프로테아좀의 활성을 조절하는 물질을 스크리닝 할 수 있으며, 또한 이에 의해 스크리닝된 물질은 뇌의 유비퀴틴 프로테아좀의 활성을 증가시켜 퇴행성 뇌질환의 치료에 유용하게 사용할 수 있음을 할 수 있다.
