Abstract
본 발명은 MED30을 포함하는 신규한 암 진단용 바이오 마커, 암 진단용 조성물, 암 진단용 키트 및 이를 이용한 암 진단 방법에 관한 것이다. 또한, 본 발명은 MED30의 억제제를 유효성분을 포함하는 암 예방 또는 치료용 조성물에 관한 것이다.
본 발명에 따른, MED30은 암 조직에서 유의적으로 발현하므로, 암의 진단에 효과적으로 이용될 수 있으며, 또한, MED30 억제제는 암의 증식, 이동 및 침윤을 억제할 수 있는 바, 신규한 항암제로서 활용될 수 있을 것으로 기대된다.
본 발명에 따른, MED30은 암 조직에서 유의적으로 발현하므로, 암의 진단에 효과적으로 이용될 수 있으며, 또한, MED30 억제제는 암의 증식, 이동 및 침윤을 억제할 수 있는 바, 신규한 항암제로서 활용될 수 있을 것으로 기대된다.
Description
본 발명은 MED30을 포함하는 신규한 암 진단용 바이오 마커, 암 진단용 조성물, 암 진단용 키트 및 이를 이용한 암 진단 방법에 관한 것이다.
또한, 본 발명은 MED30의 억제제를 유효성분을 포함하는 암 예방 또는 치료용 조성물에 관한 것이다.
2005년에 암으로 사망한 사람은 총 65,479명으로 전체 사망자의 26.7%가 암으로 사망하였다. 사망이 가장 많은 암종은 폐암으로 인구 10만명당 28.4명(21.1%)이었으며, 다음으로는 위암 22.6명(16.8%), 간암 22.5명 (16.7%), 대장암 12.5명(9.3%)의 순이었다. 1996년에서 2006년 위암 사망률의 추이를 분석한 결과, 지난 10년간 위암이 차지하는 암 사망률은 24%에서 16%로 점점 줄어들고 있으나 여전히 위암은 한국인의 대표적인 3대 암의 하나로서 이는 간과할 수는 없는 수치이다.
또한, 한국이나 일본 남성의 16% 이상이 위암으로 사망하고 있는 것으로 나타나고 있다. 한국, 일본 등 아시아에 위암이 많은 것은 민족이나 인종의 차이로 보기는 어렵고, 암 발생에 제일 중요한 생활환경의 차이, 특히 식생활의 차이에서 오는 것으로 보고되고 있다. 한국인의 경우 하루 소금 섭취량은 약 20 g으로 서양인보다 두 배 가까이 많이 섭취하고 있으며, 특히 소금에 절인 생선을 먹는 습관이 있는 한국, 일본, 핀란드, 아이슬란드 등에서 위암의 발생률이 높은 것으로 보고되고 있다. 또한 위암의 발병원인으로 식습관만큼이나 유전적인 원인도 중요시 대두되고 있다. 위암 환자의 1세대 자손들에게 위암의 발생률이 높고 A형 혈액형을 가진 사람이 위암의 발생률이 높은 것으로 보고되고 있다. 세 번째 원인으로는 위암 의 발생에 헬리코박터 파일로리 (Helicobacter Pylory. H.P)의 감염 여부이다. 아직까지 헬리코박터 파일로리의 감염과 위암의 인과관계가 결정적인 것이라고 생각하기는 섣부른 판단일 수 있으나, 한국인의 소화기계 질환 및 위암 환자의 40~60 %에서 헬리코박터 파일로리가 검출되는 것으로 볼 때, 감염자는 비감염자와 비교할 때 상대적으로 위암에 걸릴 가능성이 높은 것만은 분명하다. 따라서 헬리코박터 파일로리의 제거는 위염 및 위암 예방의 한 방법으로 대두되고 있다.
한편, 전사의 조절은 세포의 특정, 생장, 분화 및 발달에 중요한 단계이다. MED (Human mediator) 복합체는 30개 이내의 단백질을 포함하며, RNA 폴리머라아제 II (Pol II)-전사된 유전자의 발현에 주된 공활성자/활성자이다. MED 복합체는 Pol II와 유전자 특이적 전사인자 (transcription factors, TFs), 예를 들어 TFIIA, TFIIB, TFIID, TFIIE, TFIIF, 및 TFIIH와의 상호작용에 의하여 PIC (pre-initiation complex) 어셈블리를 가능하게 한다. MED 복합체는 3개의 구분되는 구조적 서브모듈 (submodules) (해드, 미들, 및 테일)로 이루어진다. 헤드 모듈은 Pol II과 직접적으로 상호작용하는 반면, 테일 모듈은 유전자-특이적 조절 단백질과 상호작용하고, 미들 모듈은 후-결합 단계 (post-binding stage)에서 조절 신호 전달에 작용한다.
MED의 작용 메커니즘은 완전히 밝혀진 바 없으며, 다만, MED 복합체가 Pol II에 강하게 결합하여, 이의 구조를 변화시켜 전사 개시 단계에 영향을 미치는 것으로 밝혀졌다. MED 복합체가 전사 과정에 있어서 필구 구성요소에 해당하는 바, MED의 서브유닛의 대부분은 배야 성장 및 세포 생존에 필요한 것으로 알려져있다.
암의 게놈 시퀀스 연구가 MED 서브유닛를 포함하는 전사 관련 요소의 돌연변이 또는 변이 등을 보고하였으며, MED 서브유닛, (예를 들어, MED1, MED12, MED19, MED23, MED28, CDK8, 및 cyclin C)의 변화가 다양한 암에 있어서의 암의 진행에 관련되어 있음이 보고된 바 있다. 그러나, 이의 메커니즘은 아직 밝혀진 바가 없다. 더욱이, MED 서브유닛 중 MED30과 암과의 관계는 종래에 알려진 바가 없다.
본 발명자들은 MED 서브유닛인 MED30과 암의 증식, 이동 및 침윤과의 관련성을 확인하여, 신규한 암의 진단용 바이오마커를 개발하였으며, 나아가 MED30의 억제가 암의 생장을 유의적으로 감소시킨다는 것을 확인하고 본 발명을 완성하기에 이르렀다.
이에 따라, 본 발명의 일 양상은 신규한 암 진단용 바이오 마커에 관한 것이다.
또한, 본 발명의 다른 양상은 암 진단용 조성물, 진단용 키트 및 암의 진단 방법에 관한 것이다.
또한, 본 발명은 암의 예방 또는 치료용 조성물에 관한 것이다.
본 발명의 일 양상은 MED30을 포함하는, 암 진단용 바이오 마커를 제공한다.
이하 본 발명을 상세히 설명한다.
본 발명에서 용어, "진단"은 병리 상태의 존재 또는 특징을 확인하는 것을 의미한다. 상기 진단은 발병 여부뿐 만 아니라 예후, 암의 경과, 병기 등을 확인하는 것을 모두 포함하는 의미이다. 본 발명의 목적상, 진단은 암의 발명, 예후, 경과, 병기 상태 또는 특징을 확인하는 것을 의미한다.
본 발명에서 용어, "진단용 마커, 바이오 마커 또는 진단 마커(diagnosis marker)"란 암 세포를 정상 세포와 구분하여 진단할 수 있는 물질로, 정상 세포에 비하여 암세포에서 증가 또는 감소를 보이는 폴리펩타이드 또는 핵산(예: mRNA 등), 지질, 당지질, 당단백질 또는 당(단당류, 이당류, 올리고당류 등) 등과 같은 유기 생체 분자 등을 포함한다.
본 발명의 목적상, 본 발명의 암 진단 마커는 정상 조직의 세포에 비하여, 암 세포에서 특이적으로 높은 수준의 발현을 보이는 MED30 및 이를 코딩하는 유전자를 의미한다.
MED30 (mediator complex subunit 30) (Gene ID: 90390)은 MED30 유전자에 의하여 코딩되는 단백질로서, MED (mediator) 복합체를 구성하는 서브유닛으로, 유전자 후생동물 (metazoan) 특이적이다.
또한, 본 발명의 일 양상은 MED30 또는 코딩하는 유전자의 발현 또는 활성을 측정할 수 있는 제제를 포함하는, 암 진단용 조성물을 제공한다.
본 발명에서 용어, "MED30 또는 코딩하는 유전자의 발현 또는 활성을 측정할 수 있는 제제”란 상기와 같이 암 세포포에서 발현이 증가하는 마커인 MED30의 단백질 또는 이를 코딩하는 mRNA에 특이적으로 결합하여, 이의 발현 또는 활성 수준을 확인함으로써 마커의 검출에 사용될 수 있는 분자를 의미한다.
상기 MED30의 발현 또는 활성을 측정할 수 있는 제제는 상기 단백질에 특이적으로 결합하는 올리고펩타이드, 모노클로날 항체, 폴리클로날 항체, 키메릭(chimeric) 항체, 리간드, PNA(Peptide nucleic acid) 또는 앱타머(aptamer)일 수 있다.
본 발명에서 "단백질 발현 또는 활성 수준 측정"이란 암을 진단하기 위하여 생물학적 시료에서의 암 마커 유전자에서 발현된 단백질의 존재 여부와 발현 정도를 확인하는 과정으로, 상기 단백질에 대하여 특이적으로 결합하는 분자를 이용하여 단백질의 양을 확인하는 것이다.
이를 위한 분석 방법으로는 웨스턴 블랏, ELISA(enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역 확산법, 로케트(rocket) 면역전기영동, 조직 면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), FACS 및 단백질 칩(protein chip) 등이 있으나 이로 제한되는 것은 아니다.
본 발명에서 용어, "항체"란 당해 분야에서 공지된 용어로서 항원성 부위에 대해서 지시되는 특이적인 단백질 분자를 의미한다. 본 발명의 목적상, 항체는 본 발명의 마커인 MED30에 대해 특이적으로 결합하는 항체를 의미하며, 이러한 항체는, 각 유전자를 통상적인 방법에 따라 발현벡터에 클로닝하여 상기 마커 유전자에 의해 코딩되는 단백질을 얻고, 얻어진 단백질로부터 통상적인 방법에 의해 제조될 수 있다. 여기에는 상기 단백질 에서 만들어질 수 있는 부분 펩티드도 포함되며, 본 발명의 부분 펩티드로는, 최소한 7개 아미노산, 바람직하게는 9개 아미노산, 더욱 바람직하게는 12개 이상의 아미노산을 포함한다. 본 발명의 항체의 형태는 특별히 제한되지 않으며 폴리클로날 항체, 모노클로날 항체 또는 항원 결합성을 갖는 것이면 그것의 일부도 본 발명의 항체에 포함되고 모든 면역 글로불린 항체가 포함된다. 나아가, 본 발명의 항체에는 인간화 항체 등의 특수 항체도 포함된다.
본 발명의 암 진단 마커의 검출에 사용되는 항체는 2개의 전체 길이의 경쇄 및 2개의 전체 길이의 중쇄를 가지는 완전한 형태뿐만 아니라 항체 분자의 기능적인 단편을 포함한다. 항체 분자의 기능적인 단편이란 적어도 항원 결합 기능을 보유하고 있는 단편을 뜻하며 Fab, F(ab'), F(ab') 2 및 Fv 등이 있다.
또한, MED30를 코딩하는 유전자의 발현 또는 활성을 측정하는 제제는 상기 유전자의 mRNA에 특이적으로 결합하는 안티센스 올리고뉴클레오티드, 프라이머 쌍 또는 프로브일 수 있다.
본 발명에서 "mRNA 발현수준 측정"이란 암을 진단하기 위하여 생물학적 시료에서 암 진당용 마커 유전자의 mRNA 존재 여부와 발현 정도를 확인하는 과정으로, mRNA의 양을 측정함으로써 알 수 있다. 이를 위한 분석 방법으로는 RT-PCR, 경쟁적 RT-PCR(Competitive RT-PCR), 실시간 RT-PCR(Real-time RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(Northern blotting) 및 DNA 칩 등이 있으나 이로 제한되는 것은 아니다.
유전자의 mRNA 수준을 측정하는 제제는 바람직하게는 프라이머 쌍 또는 프로브이며, MED30 유전자의 핵산 서열은 공지된 바 있으므로 당업자는 상기 서열을 바탕으로 이들 유전자의 특정 영역을 특이적으로 증폭하는 프라이머 또는 프로브를 디자인할 수 있다.
본 발명에서 용어, "프라이머"는 짧은 자유 3말단 수산화기(free 3' hydroxyl group)를 가지는 핵산 서열로 상보적인 템플레이트(template)와 염기쌍(base pair)을 형성할 수 있고 템플레이트 가닥 복사를 위한 시작지점으로 기능을 하는 짧은 핵산 서열을 의미한다. 프라이머는 적절한 완충용액 및 온도에서 중합반응(즉, DNA 폴리머레이즈 또는 역전사효소)을 위한 시약 및 상이한 4가지 뉴클레오사이드 트리포스페이트의 존재하에서 DNA 합성이 개시할 수 있다. 본 발명에서는 MED30 폴리뉴클레오티드의 센스 및 안티센스 프라이머를 이용하여 PCR 증폭을 실시하여 원하는 생성물의 생성 여부를 통해 암을 진단할 수 있다. PCR 조건, 센스 및 안티센스 프라이머의 길이는 당업계에 공지된 것을 기초로 변형할 수 있다.
본 발명에서 용어, "프로브" 란 mRNA와 특이적 결합을 이룰 수 있는 짧게는 수 염기 내지 길게는 수백 염기에 해당하는 RNA 또는 DNA 등의 핵산 단편을 의미하며 라벨링 되어 있어서 특정 mRNA의 존재 유무를 확인할 수 있다. 프로브는 올리고뉴클로타이드(oligonucleotide) 프로브, 단쇄 DNA(single stranded DNA) 프로브, 이중쇄DNA(double stranded DNA) 프로브, RNA 프로브 등의 형태로 제작될 수 있다. 본 발명에서는 MED30 폴리뉴클레오티드와 상보적인 프로브를 이용하여 혼성화를 실시하여, 혼성화 여부를 통해 암을 진단할 수 있다. 적당한 프로브의 선택 및 혼성화 조건은 당업계에 공지된 것을 기초로 변형할 수 있다.
본 발명의 프라이머 또는 프로브는 포스포르아미다이트 고체 지지체 방법, 또는 기타 널리 공지된 방법을 사용하여 화학적으로 합성할 수 있다. 이러한 핵산 서열은 또한 당해 분야에 공지된 많은 수단을 이용하여 변형시킬 수 있다. 이러한 변형의 비-제한적인 예로는 메틸화, "캡화", 천연 뉴클레오타이드 하나 이상의 동족체로의 치환, 및 뉴클레오타이드 간의 변형, 예를 들면, 하전되지 않은 연결체(예: 메틸 포스포네이트, 포스포트리에스테르, 포스포로아미데이트, 카바메이트 등) 또는 하전된 연결체(예: 포스포로티오에이트, 포스포로디티오에이트 등)로의 변형이 있다.
본 발명에 따른 조성물이 진단 할 수 있는 암의 경우, 그 종류의 제한은 없으나 췌장암, 담도암, 신경내분비종양, 폐암, 유방암, 난소암, 간암, 기관지암, 비인두암, 후두암, 위암, 방광암, 대장암, 결장암, 자궁경부암, 뇌암, 전립선암, 골암, 두경부암, 피부암, 갑상선암, 부갑상선암 또는 요관암일 수 있으며, 바람직하게는 위암일 수 있다.
또한, 본 발명의 일 양상은 MED30 또는 코딩하는 유전자의 발현 또는 활성을 측정할 수 있는 제제를 포함하는 암 진단용 키트를 제공한다.
본 발명의 키트는 암 진단 마커인 MED30 의 발현 수준을 mRNA 또는 단백질의 발현 수준을 확인함으로써 마커를 검출할 수 있다. 본 발명의 마커 검출용 키트에는 암 진단 마커의 발현 수준을 측정하기 위한 프라이머, 프로브 또는 선택적으로 마커를 인지하는 항체뿐만 아니라 분석 방법에 적합한 한 종류 또는 그 이상의 다른 구성 성분 조성물, 용액 또는 장치가 포함될 수 있다.
또 하나의 양태로서, 본 발명은 인간의 생물학적 시료 내의 MED30 또는 이를 코딩하는 유전자의 발현 또는 활성을 검출하는 단계를 포함하는, 암의 진단에 필요한 정보를 제공하는 방법을 제공한다.
MED30 발현 수준을 mRNA 수준 또는 단백질 수준에서 검출할 수 있고, 생물학적 시료에서 mRNA 또는 단백질의 분리는 공지의 공정을 이용하여 수행할 수 있다.
본 발명에서 용어 환자의 시료란 암 진단용 마커 유전자인 MED30의 발현 수준이 차이나는 조직, 세포, 전혈, 혈청, 혈장, 타액, 객담, 뇌척수액 또는 뇨와 같은 시료 등을 포함하나, 이에 제한되지 않는다.
상기 검출 방법들을 통하여, 정상 대조군에서의 유전자 발현 수준을 암 의심환자에서의 유전자 발현 수준과 비교함으로써 암 의심 환자의 실제 암 환자 여부를 진단할 수 있다. 즉, 암으로 추정되는 세포로부터 본 발명의 마커의 발현 수준을 측정하고, 정상 세포로부터 본 발명의 마커의 발현 수준을 측정하여 양자를 비교한 후, 본 발명의 마커의 발현 수준이 정상 세포의 것보다 암으로 추정되는 세포 유래에서 더 많이 발현되면 암으로 추정되는 세포를 암세포인 것으로 예측할 수 있는 것이다.
mRNA 수준을 측정하기 위한 분석 방법으로는 역전사효소 중합효소반응, 경쟁적 역전사효소 중합효소반응, 실시간 역전사효소 중합효소반응, RNase 보호 분석법, 노던 블랏팅, DNA 칩 등이 있으나 이로 제한되는 것은 아니다. 상기 검출 방법들을 통하여, 정상 대조군에서의 mRNA 발현량과 암 의심환자에서의 mRNA 발현량을 비교할 수 있고, 암 진단용 마커 유전자에서 mRNA로의 유의한 발현량의 증가여부를 판단하여 암 의심 환자의 실제 발병 여부를 진단할 수 있다.
mRNA 발현수준 측정은 바람직하게는, 암 진단용 마커로 사용되는 유전자에 특이적인 프라이머를 이용하는 역전사효소 중합효소반응법 또는 DNA 칩을 이용하는 것이다.
상기의 역전사효소 중합효소반응은 반응 후 전기영동하여 밴드 패턴과 밴드의 두께를 확인함으로써 암 진단 마커로 사용되는 유전자의 mRNA 발현 여부와 정도를 확인 가능하고 이를 대조군과 비교함으로써, 암 발생 여부를 간편하게 진단할 수 있다.
한편, DNA 칩은 상기 암 진단용 마커 유전자 또는 그 단편에 해당하는 핵산이 유리 같은 기판에 고밀도로 부착되어 있는 DNA 칩을 이용하는 것으로서, 시료에서 mRNA를 분리하고, 그 말단 또는 내부를 형광물질로 표지된 cDNA 프로브를 조제하여, DNA 칩에 혼성화시킨 다음 암 발병 여부를 판독할 수있다.
단백질 수준을 측정하기 위한 분석 방법으로는, 웨스턴 블랏, ELISA, 방사선면역분석, 방사 면역 확산법, 오우크테로니 면역 확산법, 로케트 면역전기영동, 조직면역 염색, 면역침전 분석법, 보체 고정 분석법, FACS, 단백질 칩 등이 있으나 이로 제한되는 것은 아니다. 상기 분석 방법들을 통하여, 정상 대조군에서의 항원 -항체 복합체의 형성량과 암 의심환자에서의 항원-항체 복합체의 형성량을 비교할 수 있고, 암 진단용 마커 유전자에서 단백질로의 유의한 발현량의 증가여부를 판단하여, 암 의심 환자의 실제 발병 여부를 진단할 수 있다.
또한, 본 발명은 MED30 또는 이를 코딩하는 유전자의 발현 또는 활성을 억제하는 제제를 포함하는, 암의 예방 또는 치료용 조성물을 제공한다.
상기 조성물은 약학적 및 식품 조성물을 모두 포함하는 개념이다.
본 발명의 구체적인 실시예에서는 MED30를 위암세포내에서 억제한 결과 암세포의 증식, 이동 및 침윤이 유의적으로 감소함을 확인하였다. 따라서, 본 발명의 MED30는 암의 진단 뿐만 아니라 암의 발병 및 발달에도 관여함을 확인하였으며, MED30의 억제제가 항암제로 이용될 수 있음을 제시하였다.
MED30의 발현 또는 활성을 억제하는 제제는 그 종류에 제한이 없지만 상기 단백질에 특이적으로 결합하는 올리고펩타이드, 모노클로날 항체, 폴리클로날 항체, 키메릭(chimeric) 항체, 리간드, PNA(Peptide nucleic acid), 앱타머(aptamer), 천연추출물 또는 화합물일 수 있다.
또한, MED30를 코딩하는 유전자의 발현 또는 활성을 억제하는 제제는 상기 유전자의 mRNA에 특이적으로 결합하는 siRNA(small interference RNA), shRNA(short hairpin RNA), miRNA(microRNA), 안티센스 올리고뉴클레오티드, 프로브, 천연추출물 또는 화합물일 수 있다.
본 발명에서 사용되는 용어, "siRNA"는 특정 mRNA의 절단(cleavage)을 통하여 RNAi(RNA interference) 현상을 유도할 수 있는 짧은 이중사슬 RNA를 의미한다. 표적 유전자의 mRNA와 상동인 서열을 가지는 센스 RNA 가닥과 이와 상보적인 서열을 가지는 안티센스 RNA 가닥으로 구성된다. siRNA는 표적 유전자의 발현을 억제할 수 있기 때문에 효율적인 유전자 넉다운 방법으로서 또는 유전자 치료의 방법으로 제공될 수 있다.
siRNA는 RNA끼리 짝을 이루는 이중사슬 RNA 부분이 완전히 쌍을 이루는 것에 한정되지 않고 미스매치 (대응하는 염기가 상보적이지 않음), 벌지(일방의 사슬에 대응하는 염기가 없음) 등에 의하여 쌍을 이루지 않는 부분이 포함될 수 있다. 전체 길이는 10 내지 100 염기, 바람직하게는 15 내지 80 염기, 더욱 바람직하게는 20 내지 70염기이다. siRNA 말단 구조는 표적 유전자의 발현을 RNAi 효과에 의해 억제할 수 있는 것이면 평활 (blunt) 말단 또는 점착(cohesive) 말단 모두 가능하다. 점착 말단 구조는 3' 말단 돌출한 구조와 5' 말단 쪽이 돌출한 구조 모두 가능하다. 돌출하는 염기 수는 한정되지 않는다. 또한 siRNA는 표적 유전자의 발현 억제 효과를 유지할 수 있는 범위에서 예를 들어, 한쪽 말단의 돌출 부분에 저분자 RNA(예를 들어, tRNA, rRNA, 바이러스 RNA와 같은 천연의 RNA 분자 또는 인공의 RNA 분자)를 포함할 수 있다. siRNA 말단 구조는 양측 모두 절단 구조를 가질 필요는 없고, 이중 사슬 RNA 일반의 말단 부위가 링커 RNA에 의하여 접속된 스텝 루프형 구조일 수도 있다.
본 발명에서 사용되는 siRNA는 그 자체로 폴리뉴클레오타이드 페어링을 갖는 완전한 형태, 즉 시험관에서 siRNA를 직접 합성한 두 형질전환 과정을 거쳐 세포 안으로 도입되는 형태이거나, 생체 내에 투여된 후 이러한 형태를 갖도록 하나의 단일쇄 올리고뉴클레오타이드 단편과 이의 역방향(reverse) 상보물이 스페이서에 의해분리된 단일쇄 폴리뉴클레오타이드로부터 유도될 수 있는 형태, 예를 들어 siRNA가 세포 안에서 발현되도록 제조된 siRNA 발현 벡터 또는 PCR-유도된 siRNA 발현 카세트를 형질전환 또는 감염(infection) 과정을 거쳐 세포안으로 도입되는 형태일 수 있다. siRNA를 제조하고 세포 또는 동물로 도입하는 방법의 결정은 목적 및 표적 유전자 산물의 세포 생물학적 기능에 따라 달라질 수 있다. 일양태로서, 본 발명에서는 siRNA로서 5'-CGA GCA ACU UAU UCC AUA U(dTdT)-3', 5'-GCU GCC AAA UGG UGU CAC U(dTdT)-3' 및 5'-CGA GAA AUU GCU GAA GUA A(dTdT)-3'을 사용하였다.
본 발명에서 사용되는 용어, "shRNA" 는 siRNA의 고가의 생합성 비용, 낮은 세포 형질감염 효율로 인한 RNA 간섭 효과의 단시간 유지 등의 단점을 극복하기 위한 것으로 RNA 중합효소 Ⅲ의 프로모터로부터 아데노 바이러스, 렌티 바이러스 및 플라스미드 발현 벡터 시스템을 이용하여 이를 세포 내로 도입하여 발현시킬 수 있으며, 이러한 shRNA는 세포 내에 존재하는 siRNA 프로세싱 효소(Dicer 또는 Rnase Ⅲ)에 의해 정확한 구조를 갖는 siRNA로 전환되어 목적 유전자의 사일런싱을 유도함이 널리 알려져 있다.
본 발명에서 사용되는 용어, "안티센스 올리고뉴클레오티드"란 특정 mRNA 의 서열에 상보적인 핵산 서열을 함유하고 있는 DNA 또는RNA 또는 이들의 유도체를 의미하고, mRNA 내의 상보적인 서열에 결합하여 mRNA의 단백질로의 번역을 저해하는 특징이 있다. 본 발명의 안티센스 서열은 CDCA5 mRNA에 상보적이고 CDCA5 mRNA에 결합할 수 있는 DNA 또는 RNA 서열을 의미하고, CDCA5 mRNA의 번역, 세포질 내로의 전위(translocation), 성숙 (maturation) 또는 다른 모든 전체적인 생물학적 기능에 대한 필수적인 활성을 저해할 수 있다. 안티센스 올리고뉴클레오티드의 길이는 6 내지 100 염기이고, 바람직하게는 8 내지 60 염기이고, 보다 바람직하게는 10 내지 40 염기이다.
안티센스 RNA의 경우 통상의 방법으로 시험관에서 합성되어 생체 내로 투여하거나, 생체 내에서 안티센스 RNA가 합성되도록 할 수 있다. 시험관에서 안티센스 RNA를 합성하는 한 예는 RNA 폴리머라제Ⅰ를 이용하는 것이다. 생체 내에서 안티센스 RNA가 합성되도록 하는 한가지 예는 인식부위(MCS)의 기원이 반대 방향에 있는 벡터를 사용하여 안티센스 RNA가 전사되도록 하는 것이다. 이런 안티센스 RNA는 서열 내에 번역 중지 코돈이 존재하도록 하여 펩타이드 서열로 번역되지 않도록 하는 것이 바람직하다.
본 발명의 조성물은 MED30의 억제제를 포함하는 외에, 환자의 암 세포의 증식을 억제시키는 추가의 물질을 포함할 수 있으며, 또는 siRNA 또는 안티센스 올리고뉴클레오티드 분자의 유입을 촉진시키는 제제, 예를 들어 리포좀(미국 특허 제4,897,355호, 제4,394,448호, 제4,23,871호, 제4,231,877호, 제4,224,179호, 제4,753,788호, 제4,673,567호, 제4,247,411호, 제4,814,270호)을 이용하거나 콜레스테롤, 콜레이트 및 데옥시콜산을 비롯한 다수의 스테롤류 중 1종의 친유성 담체와 함께 배합할 수도 있다. 또한 안티센스 올리고뉴클레오티드는 세포에 의해 흡수되는 펩티드에 접합시킬 수도 있다. 유용한 펩타이드의 예로는 펩타이드 호르몬, 항원 또는 항체 및 펩타이드 독소 등이 있다.
본 발명의 조성물은 단독으로 사용할 수 있지만, 치료 효율을 증가시키기 위해 방사선 요법 또는 화학 요법(세포 성장 정지 또는 세포 독성 물질, 항생 물질형 물질, 알킬화제, 항대사성 물질, 호르몬제, 면역제, 인터페론형 물질, 사이클로옥시게나제 억제제, 메탈로매트릭스프로테아제 억제제, 텔로머라제 억제제, 티로신 키나제 억제제, 항성장인자 수용체 물질, 항-HER 물질, 항-EGFR 물질, 항-혈관생성 물질, 파르네실 트랜스퍼라제 억제제, ras-raf 시그날 전도 경로 억제제, 세포 주기 억제제, 기타 cdk 억제제, 튜불린 결합체, 토포이소머라제Ⅰ 억제제, 토포이소머라제 Ⅱ 억제제 등)과 같은 다른 항암 치료법과 병용하여 사용할 수 있다.
본 발명의 조성물은 약제학적으로 허용되는 담체와 함께 투여될 수 있고, 경구 투여시에는 결합체, 활택제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소 투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭시르, 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조될 수 있다.
본 발명에서 사용되는 용어, 투여는 어떠한 적절한 방법으로 환자에게 본 발명의 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 경구 투여, 복강 내 투여, 정맥 내 투여, 근육 내 투여, 피하 투여, 피내 투여, 비내 투여, 폐내 투여, 직장내 투여, 강내 투여, 복강 내 투여, 경막 내 투여가 이루어질 수 있으나, 이에 제한되지는 않는다.
본 발명의 조성물의 유효 투여량의 범위는 성별, 체표면적, 질환의 종류 및 중증도, 연령, 약물에 대한 민감도, 투여경로 및 배출비율, 투여시간, 치료기간, 표적세포, 발현 수준 등 기타 의학 분야에 잘 알려진 다양한 요인에 따라 달라질 수 있으며, 당 분야의 전문가들에 의해 용이하게 결정될 수 있다.
또한, 본 발명은 MED30 억제제는 암의 예방 및 개선에 효과적인 약제, 식품 및 음료 등에 다양하게 이용될 수 있다. 본 발명의 MED30 억제제를 첨가할 수 있는 식품으로는, 예를 들어, 각종 식품류, 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등이 있고, 분말, 과립, 정제, 캡슐 또는 음료인 형태로 사용할 수 있다.
본 발명의 MED30 억제제는 암의 예방 및 개선을 목적으로 식품 또는 음료에 첨가될 수 있다. 이때, 식품 또는 음료 중의 상기 화합물의 양은 일반적으로 전체 식품 중량의 0.01 내지 15 중량%로 가할 수 있으며, 건강음료 조성물은 100 ㎖를 기준으로 0.02 내지 10 g, 바람직하게는 0.3 내지 1 g의 비율로 가할 수 있다.
본 발명에 따른, MED30은 암 조직에서 유의적으로 발현하므로, 암의 진단에 효과적으로 이용될 수 있으며, 또한, MED30 억제제는 암의 증식, 이동 및 침윤을 억제할 수 있는 바, 신규한 항암제로서 활용될 수 있을 것으로 기대된다.
도 1은 MED30가 위암 조직에서 과발현된 것을 확인한 도이다.

도 2는 MED30가 위암세포의 증식을 조절한다는 것을 나타낸 결과이다. 위암세포 (SNU216 및 SNU638)에 MED30 siRNA (100 nM) 또는 SCR (scrambled) siRNA를 형질감염 시켰다. MED30 녹다운 효율을 형질감염 48시간 후 웨스턴 블롯 (A) 및 실시간 PCR (C)으로 확인하였다. MED30의 발현을 MED30 과발현 (MED30-over) 및 비어있는 대조군 벡터로 형질전환된 (Mock) 세포에서 확인하였다.

1: 세포 배양 및 형질 감염
위암세포주 (SNU1, SNU16, SNU216, SNU620, SNU638, 및 N87)를 한국 세포주은행 (서울)로부터 구매하였다. 세포를 25 mM HEPES, 10% FBS, 및 100 ㎍/ml 페니시린/스트렙토마이신 (1 × P/S) 이 보충된 RPMI1640 내에서, 5%CO2/95%공기 하에서 37°C 에서 배양하였다. DharmaFECT reagent 1 또는 3 (Dharmacon, Lafayette, CO)를 이용하여 제조사의 지시에 따라 siRNA로 세포를 형질감염시켰다. 본 발명에서 사용한 siRNA의 서열은 하기와 같다: MED30 siRNA (Bioneer, Daejeon, Korea), 5'-CGA GCA ACU UAU UCC AUA U(dTdT)-3', 5'-GCU GCC AAA UGG UGU CAC U(dTdT)-3' 및 5'-CGA GAA AUU GCU GAA GUA A(dTdT)-3'; SCR (scrambled) siRNA (Dharmacon, Lafayette, CO), 5' GAU CCG CAA AAG AGC GAA A(dTdT)-3'.

MED30
의 과발현
MED30-과발현 벡터를 설계하기 위하여, pLenti6.3-V5/DEST 렌티바이러스 벡터 (Invitrogen, Carlsbad, CA)를 이용하였다. In vivo 재조합-기반 게이트웨이 클로닝 시스템 (Invitrogen)을 이용하여 MED30 cDNA를 pLenti6.3-V5/DEST 벡터 내로 클로닝 하였다. MED30 cDNA를 포함하는 도너 벡터 (pDONR221)를 UltimateTM ORF Clones (Invitrogen)로부터 구매하고, LR 클로나아제 효소 혼합물 (Invitrogen)를 이용하여 게이트웨이 데스트네이션 (Gateway destination) 벡터 pLenti6.3-V5/DEST의 계수기-선별가능한 (counter-selectable) ccdB 유전자와 재조합하였다. 빈 벡터인 pLenti6.3/V5-DEST를 위대조군 (mock control)로 사용하였다. 재조합된 렌티바이러스를 293FT 세포에서 생산하고, SNU638에 제조사의 지시에 따라 (ViraPower Lentiviral Expression System; Invitrogen) 형질감염 시켰다. 안정적인 세포주를 blasticidin (7.5 ㎍/ml) (Invitrogen)를 이용하여 선별하였다.

PCR
위암 조직을 부산대학교 병원에서 외과적 절제술을 받은 환자의 동의하에 수득하였다. 실험은 PNUH-IRB (Pusan National University Hospital- Institutional Review Board) 및 PNUYH-IRB (Pusan National University Yangsan Hospital-Institutional Review Board)의 승인 하에서 수행되었다.
총 RNA를 조직 또는 세포로부터 Trizol reagent (Invitrogen) 또는 RNeasy Mini kit (Qiagen, Valencia, CA)를 이용하여 추출하였다. cDNA를 MMLV 역전사효소 (Promega, Madison, WI), dNTP, 및 oligo-dT 프라이머를 이용하여 합성하였다. 사용한 프라이머의 서열정보는 하기와 같다: MED30, 5'- ACC GGT TAA CAA AGC TAC AGG A -3' (sense) 및 5'- TAA GTT GCT CGA CTG GAA TGG G -3' (antisense); CDH1, 5'- TGG GCC AGG AAA TCA CAT CC -3' 및 5'- CTC AGC CCG AGT GGA AAT GG -3' CDH2, 5'- CAC TGT GGA GCC TGA TGC CA -3' 및 5'- TCC CCA ATG TCT CCA GGG TG -3' CDH3, 5'- CCC CCA GAA GTA CGA GGC CC -3' 및 5'- ACG CCA CGC TGG TGA GTT GG -3' TWIST1, 5'- CGG GAG TCC GCA GTC TTA -3' 및 5'- TGG ATC TTG CTC AGC TTG TC -3' TWIST2, 5'- CTT ATG TTT GGG GGG AGG TT -3' 및 5'- TAG CCA AGC AAT CAC GGA GA -3' VIM, 5'- TGA GTA CCG GAG ACA GGT GCA G -3' 및 5'- TAG CAG CTT CAA CGG CAA AGT TC -3' SNAI1, 5'- GAG GCG GTG GCA GAC TAG -3' 및 5'- GAC ACA TCG GTC AGA CCA G -3' SNAI2, 5'- TAG GAA GAG ATC TGC CAG AC -3' 및 5'- CCC CAA GGC ACA TAC TGT TA -3' GAPDH, 5'- GCA GCC TCC CGC TTC GCT CT -3' 및 5'-TGG TGA CCA GGC GCC CAA TAC G -3'.
실시간 PCR은 LightCyclerTM 96 Real-time PCR system (Roche, Nutley, NJ, USA) 및 FastStart Essential DNA Green Master (Roche)를 이용하여 수행하였다. GAPDH를 인터널 컨트롤 (internal control)로 사용하였다.

분석
세포를 RIPA 버퍼로 용혈하고, 프로테아제 억제제 칵테일를 첨가하고, Bio-Rad protein assay kit (Bio-Rad, Hercules, CA)를 이용하여 단백질 농도를 결정하였다. 샘플 (80 ㎍/well)에 대하여 SDS-PAGE를 수행하고 PVDF 막에 이동시켰다. Anti-MED30 (1:500, Protein Tech #16787-1-AP, Chicago, IL), anti-E-cadherin (1:1000, BD Bioscience #610181, San Jose, CA), anti-N-cadherin (1:1000, BD Bioscience #610920), anti-P-cadherin (1:1000, BD Bioscience #610227), anti-Twist1/2 (1:750, Santa Cruz Biotechnology #sc-15393, Santa Cruz, CA), anti-vimentin (1:1000, DAKO #M7020, Carpentaria, CA) 및 anti-β-actin (1:2000, Santa Cruz Biotechnology #sc-47778)를 5%의 스킴 밀크에 희석하고, 막과 함께 밤새 4 ℃에서 인큐베이션 하였다. 적합한 이차항체 (1:2000, HRP (horseradish peroxidase)-접합된 anti-mouse 또는 anti-rabbit)를 실온에서 1시간동안 적용하였다. Chemiluminescence detection (GE Health Care, Little Chalfont, United Kingdom)를 이용하여 MED30, E-cadherin, N-cadherin, P-cadherin, Twist1, vimentin, 및 β-actin를 시각화 하였다. 웨스턴 블롯은 3회 실시하였다.
실시예
5: 면역염색화학
환자로부터 수득한 위암조직을 포함하는 조직 마이크로어레이 절편을 SuperBiochip (Seoul)로부터 수득하였다. 부산대학교병원 조직병리학과가 조직병리학적 진단을 수행하였으며, 임상병리학적 병기설정 (staging)을 TNM classification (AJCC, 7th ed.)를 이용하여 수행하였다. 모든 환자는 조직학적으로 위암임을 확정받았으며, 샘플의 80% 이상에서 종양조직이 포함되어 있음을 확인하였다.
면역염색화학을 위하여, 슬라이이드를 탈파라핀화 (deparaffinized)하고 재수화 (rehydrated) 하고, 0.3% 과산화수소를 30분간 처리하여, 내인적 과산화수소 (endogenous peroxidase) 활성을 퀀칭 (quench) 하고, 1 × PBS (phosphate buffered saline) 중 10% NDS (normal donkey serum) 및 1% BSA 로 블로킹하였다. 뒤이어 슬라이드를 일차 항체 (anti-human MED30; 1:50, Proteintech)를 포함하는 블로킹 버퍼 중에서 4℃에서 밤새 인큐베이션하였다. 이차 항체 (HRP-conjugated) 결합을 RT에서 2시간동안 블로킹 버퍼 내에 1:200로 희석하여 수행하였다. 뒤이어 슬라이드를 HRP (Vector Laboratories)에 대하여 염색하고, 1분 간 헤마톡실린 (hematoxylin) 염색 버퍼 (Sigma-Aldrich, St. Louis, MO)로 대비염색하였다. 절편에서 MED30에 대하여 염색된 세포의 퍼센트를 결정하고, 뒤이어 절편을 1-4 scale (1, <24%; 2, 25-49%; 3, 50-74%; 4, 75-100%)로 등급화하였다. 종양 세포 염색의 강도를 0, 1, 2, 또는 3로 등급화하였으며, 이는 각각 기적, 약함, 중간, 및 강함을 의미한다. 뒤이어 전체 염색을 종합점수로 나타내었으며, 이는 상기 두개의 등급의 곱셈으로 계산하였다. 이에 따라, 전체 염색은 0부터 12까지로 등급화 하였다.

6: 세포 증식
어세이
위암 세포의 증식에 대한 siRNA의 효과를 확인하기 위하여, siRNA 형질감염 1일 후 배양배지를 1% FBS 배지로 교환하였다. 형질감염 5일 후, 10 ㎕의 Ez-Cytox (ITSBIO, Seoul)를 첨가하고, 세포를 정상 배양 조건하에서 0.5 내지 2 시간 동안 인큐베이션하였다. 그 다음 흡광도를 ELISA reader (TECAN, Mannedorf, Switzerland)를 이용하여 450 mm에서 측정하였다. MED30 과발현의 효과를 측정하기 위하여, 세포를 1 % FBS 배지에 시딩하였다. 시딩 3일 후, 10 ㎕의 Ez-Cytox를 첨가하였다.

chamber assay)
트렌스 챔버의 바닥 챔버를 10% FBS를 포함하는 배지로 채웠다. SCR (scrambled) 또는 MED30 siRNA를 형질감염 시킨 후 1일 뒤, 위암세포를 상위 챔버내로 50㎕ 무혈청배지내로 5x105 cells/ml의 밀도로 시딩하였다.
MED 30의 과발현 효과를 확인하기 위하여, 세포 (mock 및 MED30-over)를 1x 105 cells/ml 밀도로 시딩하였다. 증식 연관 효과를 제거하기 위하여, 미토마이신 C (0.01 ㎍/ml, Sigma-Aldrich)를 첨가하였다. 세포를 4시간 또는 6시간 이동 (migrate) 하도록 두었다. 그 다음 막을 고정하고, Diff-quik solution (Sysmex, Kobe, Japan)를 이용하여 염색하고, 증류수로 세척하였다. 10개의 무작위로 선별한 필드 (fields)에서 세포 수를 광학 현미경을 이용하여 계수하였다.

위암세포의 침윤 효과를 확인하기 위하여, 24-well BioCoatTM MatrigelTM 챔버 삽입물(BD Bioscience, San Jose, CA, USA)를 사용하였다. 침윤 챔버내의 삽입물 최상위 표면을 0.5 mg/ml 성장인자-감소된 마트리겔 (BD Bioscience)로 코팅하고, 바닥 표면은 0.5 mg/ml의 피브로넥틴 (Sigma-Aldrich)으로 코팅하였다. SCR 또는 MED30 siRNA로 형질전환시킨 1일 후, 8-μm 다공성 BioCoat Matrigel 챔버 삽입물 내로, 무혈청 배지에 5X104 cells/ml의 밀도로 세포를 시딩하였다. 뒤이어, 화학주성인자 (chemoattractant)로서 10% FBS를 첨가한 750 ㎕의 배지로 채운 웰 내로 위치시켰다.
MED30 과발현의 효과를 확인하기 위하여, 세포 (mock 및 MED30-과발현 세포)를 1x 104 cells/ml의 밀도로 시딩하였다. 증식-연관 효과를 제거하기 위하여, 미토마이신 C (0.01 ㎍/ml, Sigma-Aldrich)를 첨가하였다. 24시간 동안의 인큐베이션 후, 막의 상위 표면 상의 비침윰 세포를 스크래핑으로 제거하였다. 바닥표면의 침윤된 세포를 Diff-quik 용액으로 고정하고 염색하였다. 실험은 3회 실시하였다.

SNU638 세포를 SCR 또는 MED30 siRNA으로 형질감염 시켰다. 2일 후, 세포를 트립신화하여 수집하여, PBS로 2회 세척하고, SCID (severe combined immunodeficiency) 마우스의 피하로 (100 ㎕ PBS 내로 1 × 106 cells) 주입하였다. 종양의 부피를 하기의 식에 따라 주입 후 3주 에서 7주 동안 매주 측정하였다: 종양 부피 (mm3) = (a × b2)/2, 여기서 a = mm 단위의 길이, 및 b = mm 단위의 넓이.
7주에서, 마우스를 희생시키고, 종양 부피 및 무게를 기록하였다. 이 실험은 동물실험윤리위원회에 따라 진행하였으며, PNUIACUC (Pusan National University Institutional Animal Care and Use Committee)에 의하여 승인되었다.

10: 통계적 분석
결과를 평균±SDs로 나타내었다. 비모수통계적 (nonparametric) Mann-Whitney U-test 또는 Student's t-test를 이용하여 두 군의 평균값 간의 차이의 유의성을 판단하였다. ANOVA (one-way analysis of variance) 뒤이어 Tukeys multiple comparisons를 이용하여 3개 이상의 군을 분석하였다. P 값은 <0.05이다. 생존 시간은 처리 후 사멸간의 경과시간, 또는 처리 후 마지막 정보가 관찰된 시점까지의 경과시간으로 정의하였다. Kaplan-Meier 방법을 사용하여 생존 및 MED30의 발현간의 관게를 결정하였다. 95%의 유의성 수준에서의 log-rank test를 이용하여 곡선을 비교하였다. P 값<0.05를 통계학적으로 유의적인 것으로 간주하였다. 데이터를 SPSS software version 12.0 (SPSS Inc., Chicago, IL, USA)을 이용하여 분석하였다.

11: 위암 조직에서의
MED30
의 과발현 확인
위암에서의 MED30의 역할을 확인하기 위하여, 먼저 23 개의 위암 세포 환자의 종양 조직에서의 MED30 발현 상태를 확인하였다. MED30 단백질은 암조직 부위에 광범위하게 과발현되고 있음을 확인하였으며, (도 1A-1D), 침윤된 위암세포 (도 1C) 및 림프절에 영향을 준 전이성 암세포 (도 1D)에 과발현이 명백하게 확인되었다.
또한, 위암세포주에서의 MED30의 발현 패턴을 확인하기 위하여, 실시간 PCR를 수행하였으며, 정상 위조직에 대비하여 실험된 5개의 위암 세포주 (SNU1 제외)에서 과발현을 확인하였다 (도 1E).
임상적 특징과 MED30의 발현과의 상관관계를 조사하기 위하여, 41개의 위암 조직 샘플을 이용하여 면역염색화학을 수행하였다 (표 1). 흥미롭게도 MED30은 N 병기와 양성적으로 관련성이 있었다 (도 1F).

12: 위암세포의 증식, 이동 및 침윤에서의
MED30
의 역할의 확인
위암의 발생에서 MED30의 역할을 확인하기 위하여, 3개의 위암 세포주 (SNU1, SNUI16, SNU216, SNU620, SNU638, 및 NCI-N87 세포)의 증식, 이동 및 침윤에서의 MED30의 녹아웃 또는 과발현의 효과를 확인하였다. 실시간 PCR 및 웨스턴 블롯을 이용하여 SNU216 및 SNU638 세포 (도 2A-2C)에서의 MED30의 녹아웃 및 과발현을 분석하였다.
이 세포들을 MED30 siRNA(100 nM)로 형질감염시킨 후, 2일 후 SCR siRNA에 비하여 약 80%의 단백질 및 mRNA수준에서의 MED30 발현의 감소를 확인하였다. 또한, cDNA의 형질감염에 의한 MED30의 과발현은 mRNA 및 단백질 수준에서 빈 (empty) 대조군 백터-형질감염된 세포 (mock)에 비하여 3배 이상인 것을 확인할 수 있었다.
뒤이어, 암세포의 증식에 대한 MED30의 역할을 조사하였다. 증식 어세이를 SCR 또는 MED30 siRNA의 형질감염 후 5일 뒤 수행하였따. MED30의 녹아웃은 처리된 모든 위암세포주 (SNU16, SNU216, SNU620, 및 SNU638)의 증식을 억제하였으며, SCR 처리군에 비하여 각각 18%, 68%, 98%, 및 42%로 억제하였다 (도 2D). 형질전환 후 2일 또는 3일 후 수행한 증식 어세이에서도 유사한 결과를 수득하였다. 일관되게, MED30의 과발현은 SNU216 및 SNU638세포에서 각각 mock 세포에 비하여 1.9배 및 2.2 배 성장을 증가시켰다.
위암세포의 이동에 대한 MED30의 역할을 확인하기 위하여, 보이덴 챔버 어세이 (Boyden chamber assay를 수행하였다. MED30의 녹아웃은 SNU216 및 SNU638 세포에서 각각 SCR 형질감염된 세포에 비하여 90% 및 52% FBS-유도된 이동을 감소시켰다 (도 3A 및 3C).
더욱이, MED30 과발현은 SNU216 및 SNU638세포에서 각각 mock 세포에 비하여 3.2배 및 2.8배 FBS-유도된 이동을 증가시켰다(도 3B 및 3C).
이러한 결과에 따라 위암세포주의 침윤에 대한 MED30의 역할을 확인하였다.
마크리겔 침윤 어세이 (Matrigel invasion assay)에서, MED30 siRNA는 SNU216 및 SNU638 세포에서 각각 SCR siRNA에 비하여 64% 및 47% FBS-유도된 침윤을 억제하였다 (도 4A 및 4C). 또한, MED30의 과발현은 SNU216 및 SNU638 세포에서 mock 세포에 비하여 각각 2.4배 및 2.2 배 FBS-유도된 침윤을 가속화하였다 (도 4B 및 4C).

녹다운의 효과
종양 성장에 대한 MED30의 효과를 확인하기 위하여, SCID 마우스의 피하에 SCR 또는 MED30 siRNA로 형질감염된 SNU638 세포를 주사하고, 종양의 성장을 7주간 관찰하였다 (도 5A).
SCR 대조군에서는 3주에 감지할 수 있는 (Palpable) 종양을 확인하였다. 그러나, MED30 siRNA로 형질감염된 종양 세포에서는 성장이 더 느리도록 억제되었다. 주사 7주 후, 마우스를 희생시키고 종양 부피 및 질량을 측정하였다 (도 5B 및 5C). 이 결과는 MED30 녹다운이 종양의 부피와 무게를 유의적으로 감소시킴을 나타낸다.

MED30
에 의한
EMT
경로의 조절
MED30에 의한 위암 발생의 촉진의 메커니즘을 확인하기 위하여, EMT (epithelial-mesenchymal transition)에 관여하는 유전자의 발현 패턴을 실시간 PCR로 확인하였다. EMT는 암의 전이 및 침윤의 주요 과정이다.
MED30 녹다운은 SCR 형질전환에 비하여 SNU638 세포에서 E-cadherin (CDH1) mRNA의 수준을 약간 증가시켰으며, 반면 N-cadherin (CDH2), P-cadherin (CDH3) 및 vimentin (VIM)의 발현을 각각 42%, 36%, 및 41% 감소시켰다 (도 6A). 일관되게, MED30의 과발현은 mock 세포에 비하여, CDH1 mRNA 수준을 35% 증가시켰으며, N-cadherin (CDH2), P-cadherin (CDH3), twist family bHLH transcription factor 1 및 2 (TWIST1/2), 및 vimentin (VIM)의 발현을 각각 2.9, 3.3, 3.4, 2.4, 2.2, 및 4.2 배 증가시켰다 (도 6A). snail family zinc finger 1 및 2 (SNAI1/2)의 mRNA 수준은 변화하지 않았다. 또한, MED30 과발현이 E-cadherin, N-cadherin, P-cadherin, Twist1, 및 vimentin의 단백질 수준을 증가시킨다는 것을 확인하였다 (도 6B). SNU638 세포의 형태학적 관찰에서는 MED30 과발현이 자갈 (cobblestone)-유사한 형태에서 길쭉한 섬유아세포-유사 형태로 변이하는 것을 유발하는 것을 확인하였다 (도 6C). 다만, 이에 반하여 MED30 녹다운은 형태학적 변화를 야기하지 않았다. 이러한 결과는 MED30가 양성적으로 EMT를 조절하는 것을 나타낸다.
