Abstract
본 발명에 따르면, 돼지의 육질 특성에 관여하는 1차 마이크로 RNA 영역에서 새로운 SNP 부위를 발견하고 이러한 SNP를 이용한 표지인자를 제공하여, 육질이 향상된 개체를 용이하게 선별할 수 있다.
Description
도 2는 돼지 마이크로 RNA-206 전구체 주위 영역에서 발견된 SNP의 위치를 나타내는 모식도이다.
도 3은 돼지 마이크로 RNA-206 전구체 주위 영역에서 발견된 SNP들이 완전연관되어 치환되는 것을 나타내는 표이다.
도 4는 본 발명에 따른 서열번호 2와 서열번호 3으로 표시된 프라이머를 이용하여 PCR-RFLP를 실시한 결과이다. 마이크로 RNA-206 전구체 주위 영역에서 발견된 서열번호 1의 염기서열에서 207번째 염기서열의 제한효소인 PstI 위치와 420번째 염기서열의 제한효소인 SacII 위치를 증폭시켜 줄 수 있게 제작되었다. 제한효소 PstI을 이용한 RFLP 방법에 의해 유전자형은 각각 TT, TC, CC 세 가지로 나타나며, T allele은 528 bp 및 210 bp, C allele는 738 bp의 밴드가 나타난다. 또한 SacII을 이용한 RFLP 방법에 의해 유전자형은 각각 GG, GA, AA 세 가지로 나타나며, G allele은 421 bp 및 317 bp, A allele는 738 bp의 밴드가 나타난다.
도 5는 본 발명에 따른 DNA 표지인자의 일례로서, 서열번호 5와 서열번호 6으로 표시된 프라이머를 이용한 직접염기서열분석에 사용된 DNA 단편을 나타낸다. 갈색으로 표시된 부분이 마이크로 RNA-206 전구체의 서열이다.
도 6a 및 도 6b는 본 발명에 따른 SNP에 대한 PCR 및 직접염기서열분석(direct sequencing)을 실시하여 염기에 따른 peak의 차이를 이용하여 유전자형을 분석하는 방법을 나타낸다.
Claims (9)
- 서열번호 1의 염기서열에서, 207번째 T 염기, 299번째 T 염기, 300번째 G 염기, 또는 420번째 G 염기를 포함하는 서열번호 1 내의 15 내지 700개의 연속서열로 구성되는 돼지 마이크로 RNA-206 전구체 주위 영역에서의 육질 향상 여부 관련 SNP(Single nucleotide polymorphism)를 탐색하기 위한 돼지의 육질 향상 여부 확인용 DNA 표지인자.
- 제1항에 있어서, 상기 DNA 표지인자는 돼지의 근육 내 근섬유 수 및 단면적 조성 중 근섬유 타입 IIa의 비율을 증가시키고, 근섬유 타입 IIb의 비율을 감소시키며, 육질 관련 형질인 유리육즙량(drip loss) 또는 육색 명도(lightness)를 감소시킴으로써 육질을 향상시키는 것과 관련된 것임을 특징으로 하는 DNA 표지인자.
- 서열번호 2와 서열번호 3으로 표시된 올리고뉴클레오티드로 구성되는, 서열번호 1의 염기서열에서 제한효소 PstI 위치인 207번째 염기와 서열번호 1의 염기서열에서 제한효소 SacII 위치인 420번째 염기를 포함하는 부위를 증폭시켜 돼지 마이크로 RNA-206 전구체 주위 영역에서의 육질 향상 여부 관련 SNP(Single nucleotide polymorphism)를 탐색하기 위한 PCR용 프라이머 세트.
- 제3항 기재의 프라이머 세트와, PCR 반응혼합물, 및 RFLP 분석용 제한효소인 PstI 또는 SacII를 포함하는 돼지의 육질 향상 여부 관련 SNP 분석용 키트.
- 서열번호 5와 서열번호 6으로 표시된 올리고뉴클레오티드로 구성되는, 서열번호 1의 염기서열에서 299번째 또는 300번째 위치의 염기구성을 확인하여 돼지 마이크로 RNA-206 전구체 주위 영역에서의 돼지의 육질 향상 여부 관련 SNP를 탐색하기 위한 PCR용 프라이머 세트.
- 제5항 기재의 프라이머 세트에 의해 증폭된 서열번호 4로 표시되는 올리고뉴클레오티드로 구성되는 DNA 표지인자에 상보적으로 결합하는 프로브를 포함하는, 돼지 마이크로 RNA-206 전구체 주위 영역에서의 돼지의 육질 향상 여부 관련 SNP 분석용 키트.
- 서열번호 1의 염기서열에서 207번째, 299번째, 300번째, 및 420번째 자리로 이루어진 군에서 선택되는, 돼지 마이크로 RNA-206 전구체 주위 영역에서의 하나 이상의 SNP를 이용하여 돼지의 육질 향상 여부를 확인하는 방법.
- 하기 단계들을 포함하는, 돼지 마이크로 RNA-206 전구체 주위 영역에서의 육질 향상 여부 관련 SNP(Single nucleotide polymorphism)를 탐색하여 돼지의 육질 향상 여부를 확인하는 방법:
a) 돼지로부터 게놈 DNA(genomic DNA)를 분리하는 단계;
b) 상기 a)단계의 DNA를 주형으로 하여 서열번호 1의 207번째, 299번째, 300번째, 또는 420번째 염기를 포함하는 특정 부위를 증폭시키는 단계;
c) 상기 b)단계에서 증폭된 DNA를 제한효소 PstI 혹은 SacII로 처리하는 단계; 및
d) 상기 c)단계에서 얻어진 유전자 단편들에서 다형성(RFLP; restriction fragment length polymorphism)을 비교하는 단계. - 하기 단계들을 포함하는, 돼지 마이크로 RNA-206 전구체 주위 영역에서의 육질 향상 여부 관련 SNP(Single nucleotide polymorphism)를 탐색하여 돼지의 육질 향상 여부를 확인하는 방법:
a) 돼지로부터 게놈 DNA를 분리하는 단계;
b) 상기 a)단계의 DNA를 주형으로 하여 서열번호 1의 207번째, 299번째, 300번째, 또는 420번째 염기를 포함하는 특정 부위를 증폭시키는 단계;
c) 상기 b)단계에서 증폭된 DNA의 염기서열을 분석하는 단계; 및
d) 상기 c)단계의 염기서열 분석 결과로부터 서열번호 1의 염기서열의 207번째 T 염기가 C 염기로 치환, 서열번호 1의 염기서열의 299번째 T 염기가 C 염기로, 서열번호 1의 염기서열의 300번째 G 염기가 A로, 또는 서열번호 1의 염기서열의 420번째 G 염기가 A로 치환된 여부를 확인하는 단계.
