Abstract
Description
이러한 목적을 달성하기 위하여 상기 미국특허를 시도하기 위한 유사한 연구결과가 한국 국내공개특허 제10-2009-28983호로 개시된 바 있다. 이는 고지혈증 치료용 합성의약물질 스타틴약물의 부작용을 막기 위해 합성의약물질 코엔자임10을 동시에 함유시킨 경구투여용 복합치료 조성물로서 물에 불용성인 lactone형의 합성스타틴의 용해성을 증가시키기 위해 폴리에틸렌글리콜과 같은 용해제, 계면활성제, 에멀전을 위한 오일류, 안정성 확보를 위한 나트륨 또는 칼슘계의 항산화제, pH조절제 등을 다량 함유시킨 약학조성물이 개시된 바 있다.
그러나, 상기 물질의 주성분인 스타틴계 약물과 CoQ10은 인공 합성한 화합물로서 수불용성이어서 장내 용해도에도 문제점이 상존하고 특히 이들 물질의 pH를 6.7로 조정한 칼슘염제는 용해도가 대단히 낮다는 결함이 있다.
그러나, 만일 천연발효공법에 의해 pH 6.5~7.0수준의 acid form의 천연스타틴화합물을 생합성할 수만 있다면 이러한 문제점을 한꺼번에 개선할 수 있는 데 ㅂ본 발명은 이러한 점을 감안하여 안출한 것이다.
여기에 더하여, 본원발명은 수용성인 에시드형 천연 스타틴물질 뿐만 아니라, 이 약물의 장기적인 사용으로 인한 근육통 등 합병증 발생원인 억제물질인 CoQ9, CoQ10의 천연 유비퀴논 화합물을 동시에 생합성하여 제공한다는 장점을 가진다(표 3). 더우기, 고지혈증 예방 또는 치료제로 사용시 별도의 용해제나 pH 안정화제의 사용없이도 직접 분리정제된 천연물질을 경구투여할 수 있도록 약학 조성물로 제제화할 수 있는 장점도 있다.
Claims (12)
- 모나스커스속 홍국균을 분자구조중에 이소프렌단위 또는 벤조퀴논핵을 가지는 천연물 소재분말 하나 이상과 쌀가루, 글루코스, 펩톤, 글라이신, 고기추출물 및 정제수로 이루어진 액체배지에 접종하고 30℃에서 진탕배양하여 천연 메비놀린과 유비퀴논 화합물을 동시에 생합성시키는 것을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 삭제
- 제1항에 있어서, 상기 천연물 소재분말은 인삼분말, 적색피망분말, 버섯분말, 토마토분말 중 하나 이상 첨가된 것임을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제1항에 있어서, 상기 홍국균은 모나스커스 앵카(M. anka), 모나스커스 퍼푸레우스(M. purpureus), 모나스커스 필로수스(M. pilosus), 모나스커스 루버(M. ruber), 모나스커스 아라네오수스(M. araneosus), 모나스커스 카올리앙 ATCC 595(M. kaoliang ATCC 595), 모나스커스 sp. IFO 301(M. vitreus sp. IFO 301)속중에서 하나 이상 선택되는 것을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제4항에 있어서, 상기 홍국균은 모나스커스 필로수스 sp. IFO 202, 및 모나스커스 필로수스 sp. IFO 480 중 어느 하나의 우량균주임을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제1항에 있어서, 상기 액상배양물은 PDA배지에서 배양한 홍국균의 종균을 접종하여 배양하는 것을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제1항에 있어서, 상기 액체배지는 쌀가루 2~10, 펩톤 0.5~2.0, 글라이신 1~5, 포도당 6~20, 고기추출물 0.1~1.0, 인삼가루 0.5~1.5, 적색피망가루 0.5~1.5, 표고버섯가루 0.5~1.5%(w/v)와 나머지 정제수로 이루어지고 초기 pH 6.0±0.5로 조정된 배지임을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제7항에 있어서, 상기 액체배지에 토마토가루 0.5~1.5%(w/v)를 더 추가함을 특징으로 하는 홍국균의 액상배양물의 제조방법.
- 제1항의 방법으로 제조되어 천연 메비놀린과 유비퀴논 화합물을 동시에 강화시킨 것이 특징인 홍국균의 액상배양물.
- 삭제
- 제9항 기재의 홍국균의 액상배양물 10%(v/w)를 종배양액으로 하고 멥쌀, 보리, 소맥, 수수, 조 중 하나 또는 그 이상의 혼합곡류로 조성된 멸균 고체배지에 무균적으로 접종하여 30℃에서 발효함을 특징으로 하는 천연 스타틴과 유비퀴논 화합물을 동시에 강화시킨 것이 특징인 홍국균의 고상발효물.
