먼저 마우스 유래 악성 멜라닌 종양 세포주 B16F1 (KCLB 8007)를 페니실린(100IU/㎖), 스트렙토마이신(100g/㎖), 10% FBS(fetal bovine serum)를 함유하는 DMEM(Dulbecco's Modified Eagle's Medium) 배지에 넣고 37℃를 유지하며 5% 이산화탄소를 포함하는 배양기 내에서 배양하였다. 수득된 멜라닌 종양 세포를 24웰 플레이트(well plate)에 1 × 104 세포/웰 (cells/well)에 접종하여 하루 동안 배양하였다. 배양 후 배지를 제거한 다음, 멜라닌 합성을 유도하기 위하여, 비처리군을 제외하고 α-MSH(melanocyte stimulating hormone; 멜라노사이트 자극 호르몬) 5nM을 첨가하여 멜라닌 합성을 유도하였고, 동시에 복합발효 추출액 및 코지산(양성 대조군) 800μM를 가한 DMEM 배지를 첨가하여 3일간 배양하였다. 각 처리군의 멜라닌 종양세포가 분비한 합성된 멜라닌 양을 육안으로 확인하고 그 변화를 디지털 카메라로 촬영하였고, 각 처리군의 세포를 수거하여 10% DMSO(Dimethyl sulfoxide)와 1N NaOH 100㎕를 각각 첨가하여 멜라닌을 용해하였고, 405㎚에서의 흡광도를 측정하여 멜라닌 양을 측정하였다.
