체중 180 내지 190g의 SD계 숫 흰쥐를 실험동물 사육실에서 2주 이상 사육하여 적응시켰다. 사육장은 인공 조명에 의하여 조명시간을 아침 8시부터 저녁 8시까지 12시간으로 조절하였으며, 실내 온도는 24±2℃로 유지하였다. 체중 200±5g 숫 흰쥐 6마리를 한 군으로 하였으며, Niemegeer 등의 방법에 따라 염증 유발물질인 카라기난 주사 직전에 각 군 실험동물의 오른쪽 뒷다리 부위(발목의 일정한 부위에 표시)의 부피를 측정한 다음, 생리식염수에 용해한 1% 카라기난(Sigma Chem) 50㎕를 오른쪽 뒷다리의 발바닥 중간 부위에 주사하고, 30분 및 3시간 경과 후에 다시 실험동물의 오른쪽 뒷다리 부피를 측정하였다. 카라기난을 피하에 주사하기 30분 전에 소나무잔나비버섯 열수 추출물 1군(WE1: 100mg/kg), 열수 추출물 2군 (WE2: 200mg/kg), 열수 추출물 3군(WE3: 400mg/kg) 및 대조군(Control: 동일량의 생리식염수 투여)을 경구투여하였다. 부종 증가율은 약물 투여 전 및 후의 부피에 대한 비율(%)을 하기 수학식 1과 같이 계산하였다. 상기 실시예 2-1의 방법으로 실시한 숫 흰쥐의 부종 증가율을 측정하고, 카라기난-유도성 부종 증가율에 대한 소나무잔나비버섯 열수 추출물의 효과를 살펴보았다. 각 실험군 간의 측정값에 대한 통계처리는 student's t-test를 이용하였다(참조: 표 1).The edema growth rate of male rats carried out by the method of Example 2-1 was measured, and the effects of the hot-spring extract of pine needles on the carrageenan-induced edema growth rate were examined. 상기 표 1의 결과를 토대로, 각 실험군의 부종 억제율을 하기 수학식 2와 같이 계산하였다. 부종 억제율은 대조군의 부종 증가율에 대한 약물 투여군의 부종 증가율을 비율로 계산하였다. 부종 억제율을 하기 표 2에 나타내었다. 상기 표 2에서 보듯이, 소나무잔나비버섯 열수 추출물을 투여하였을 때, 카라기난 주사 30분 후에 관찰된 부종 증가율은 대조군과 소나무잔나비버섯 열수 추출물 투여 실험군 간에는 유의한 차이를 보이지 않았으나, 3시간 후에 관찰된 부종 증가율은 염증 대조군에 비하여 현저히 감소하였으며, 특히, 소나무잔나비버섯 열수 추출물 200mg/kg 및 400mg/kg을 투여한 군에서 대조군에 비하여 각각 약 38.2% 및 32.7%의 억제율을 보였다. 일반적으로 염증 유발 모델 약물인 카라기난에 의해 발생하는 부종은 히스타민이나 세로토닌에 의해 나타나는 제 1상과 키닌(kinin)류, 프로스타글란딘류 및 slow reactive substance-A(SRS-A) 등에 의해 유도되는 제 2상으로 분류하고 있으며, 제 1상 부종은 카라기난 주사 직후 발생하여 30분 정도에서 피크에 도달한 다음 소실되나, 제 2상 부종은 카라기난 주사 1시간 이후에 발생하여 장시간 지속되는 것으로 알려져 있다. 상기 실험의 결과, 각종 소나무잔나비버섯 추출물은 히스타민이나 세로토닌에 의해 유도되는 제 1상 부종에는 별다른 영향을 미치지 못하였으나, 프로스타글란딘류 등에 의해 유도되는 제 2상 부종을 억제하는 항염증 작용이 있는 것으로 확인되었다.
