본 발명에서 사용한 잔류농약 제거효과에 대한 분석법은 다음의 순서에 따라 가스 크로마토그래피(GC)로 실시하였다. 실험에 사용한 농작물은 오이, 포도, 사과를 사용하였으며 분석을 실시한 농약은 유기인계 농약으로 널리 사용되고 있는 다이아지논을 사용했다. 먼저 농작물을 깨끗이 세척한 후 자연건조시킨 후 농약 사용의 희석배율에 맞추어 30초 동안 침적시킨 다음 하루밤 지연건조시켜 동일량의 농약을 농작물에 부착시킨 다음 본 발명의 과일·야채 세정제를 분무시킨후 흐르는 물로 5초 동안 헹구고 시판중인 주방세제는 표준사용 농도의 수용액을 사용하여 10분간 진탕하여 세정하고 역시 5초 동안 흐르는 물로 헹구는 방법을 사용하였다. 헹굼이 끝난 과일·야채는 150g 씩 분취하여 쥬스기로 갈은 후 클로포름과 노르말 헥산으로 농약을 추출해 냈다. 본 발명에서 실시한 세포독성 시험은 물질 및 제품의 동물세포에 대한 독성을 평가하는 동물시험 대체시험법으로 이미 FDA, CTFA, SDA 등에서 적극적으로 활용되고 있는 실정이다. 본 실험에서 사용한 세포독성 시험은 마우스 섬유아세포를 사용했다. 96웰 플레이트에 일정 농도의 마우스 섬유아세포를 배양한 후 세정제로 처리하면 시험물질의 독성에 의해 세포가 성장을 하지 못하거나 죽게되어 플레이트 바닥으로부터 떨어져 나가게 된다. 일정시간 경과후 배지를 제거하면 각 웰에는 생존한 세포들만이 웰 플레이트 바닥에 남게 되는데, 이 세포들을 NaOH로 용해시켜 단백질을 방출시키고 그 양을 흡광도에 의해 측정함으로써 세정제의 독성을 평가했다. 세포 독성의 평가는 반수 치사농도(H50)로 실시하였으며 세정 조제 10%용액을 배지로 희석한 후 세포 배양시켰다.Cytotoxicity test carried out in the present invention is an alternative to the animal test for assessing the toxicity of the material and the animal cells to the animal, the situation is actively used in FDA, CTFA, SDA and the like.
