Abstract
Description
なお、本出願は2017年3月31日に出願された日本国特許出願第2017−070758号に基づく優先権を主張しており、当該日本国出願の全内容は本明細書中に参照として援用されている。
例えば、免疫グロブリンスーパーファミリーに属する膜タンパク質であるPD−1に対する抗体(抗PD−1抗体)、或いは、腫瘍細胞を含む種々の細胞において日常的に発現しているPD−L1(Programmed cell death-1 ligand-1:B7−H1ともいう。)と呼ばれるPD−1のリガンドに対する抗体(抗PD−L1抗体)を、免疫チェックポイント阻害薬として患者に投与することにより、免疫逃避機構を遮断して免疫系細胞による抗腫瘍免疫活性を向上させ得ることが、一部の腫瘍に対して確認されている(例えば特許文献1参照)。
そこで本発明は、高価な抗PD−1抗体や抗PD−L1抗体のような抗体を使用する抗腫瘍剤とは異なる構成の新たな抗腫瘍性(抗がん性ともいえる。)の合成ペプチドを提供することを課題(目的)として創出されたものである。
(1)膜タンパク質であるPD−1(Programmed cell death-1)のシグナルペプチドを構成するアミノ酸配列、または、該アミノ酸配列について1個、2個又は3個のアミノ酸残基が欠失、置換又は付加された改変アミノ酸配列;および
(2)細胞膜透過性ペプチド(CPP)として機能するアミノ酸配列;
をともに備えることを特徴とする。
好ましい一態様では、ここで開示される抗腫瘍ペプチドは、総アミノ酸残基数が100以下である。製造コスト、合成のし易さ、取扱い性の観点からは、総アミノ酸残基数が80以下(例えば70以下)であるものがさらに好ましい。
或いは、上記(1)に示すアミノ酸配列と(2)に示すアミノ酸配列とを合わせたアミノ酸残基数が、抗腫瘍ペプチド全体のアミノ酸残基数の80%以上(より好ましくは90%以上、例えば100%)を占めるような合成ペプチドは、ここで開示される抗腫瘍ペプチドのうちの特に好適な一態様である。
また、ここで開示される抗腫瘍ペプチドの好適な他の一態様では、上記CPPとして機能するアミノ酸配列が、ポリアルギニン(特に限定しないが、典型的には、5個以上9個以下のアルギニン残基から構成される)、または、配列番号8〜25のいずれかに示すアミノ酸配列、または、該アミノ酸配列について1個、2個又は3個のアミノ酸残基が欠失、置換又は付加されたCPPとして機能する改変アミノ酸配列であることを特徴とする。
例えば、
(i)配列番号1〜7のうちのいずれかに示すアミノ酸配列、または、該アミノ酸配列について1個または複数個(例えば2個又は3個)のアミノ酸残基が欠失、置換又は付加された改変アミノ酸配列;および
(ii)ポリアルギニン、または、配列番号8〜25のうちのいずれかに示すアミノ酸配列、または、該アミノ酸配列について1個または複数個(例えば2個又は3個)のアミノ酸残基が欠失、置換又は付加されたCPPとして機能する改変アミノ酸配列;
をともに備える合成ペプチドが好適例として挙げられる。
かかる組成物は、ここで開示される抗腫瘍ペプチドを含むことにより、抗腫瘍剤(抗がん剤を包含する。以下同じ。)としての利用、或いは新たな抗腫瘍剤の開発のための材料として利用することができる。
かかる構成の方法では、ここで開示される抗腫瘍ペプチドを腫瘍細胞に供給することによって、該腫瘍細胞の増殖(ひいては腫瘍、癌組織の増大)を阻止若しくは抑制することができる。
本明細書中で引用されている全ての文献の全ての内容は本明細書中に参照として組み入れられている。
また、本明細書において「アミノ酸残基」とは、特に言及する場合を除いて、ペプチド鎖のN末端アミノ酸及びC末端アミノ酸を包含する用語である。
なお、本明細書中に記載されるアミノ酸配列は、常に左側がN末端側であり右側がC末端側である。
(1)PD−1のシグナルペプチドを構成するアミノ酸配列、または、該アミノ酸配列について1個、2個又は3個のアミノ酸残基が欠失、置換又は付加されたが抗腫瘍活性を失っていない改変アミノ酸配列、および、
(2)CPPとして機能するアミノ酸配列、
をともに備えることで特徴付けられるペプチドである。
PD−1は、典型的には270〜290程度のアミノ酸残基からなる免疫グロブリンスーパーファミリーに属する膜タンパク質であり、免疫チェックポイントタンパク質、換言すれば、生体内における免疫反応の負の制御に関与するタンパク質である。
しかしながら、PD−1のシグナルペプチド領域が、抗腫瘍活性を有することは見出されておらず、かかるシグナルペプチド領域のアミノ酸配列を合成し、該配列にCPPを付加することにより、人為的に合成された抗腫瘍ペプチドが得られることは、本願出願当時、全く想定されていないことであった。
配列番号1は、ヒト由来のPD−1に含まれる計20アミノ酸残基からなるシグナル配列である。
配列番号2は、マウス由来のPD−1に含まれる計20アミノ酸残基からなるシグナル配列である。
配列番号3は、ウシ由来のPD−1に含まれる計29アミノ酸残基からなるシグナル配列である。
配列番号4は、水牛由来のPD−1に含まれる計29アミノ酸残基からなるシグナル配列である。
配列番号5は、ネコ由来のPD−1に含まれる計24アミノ酸残基からなるシグナル配列である。
配列番号6は、サル(チンパンジー等)由来のPD−1に含まれる計20アミノ酸残基からなるシグナル配列である。
配列番号7は、ラット由来のPD−1に含まれる計20アミノ酸残基からなるシグナル配列である。
配列番号8のアミノ酸配列は、FGF2(塩基性線維芽細胞増殖因子)由来の合計14アミノ酸残基から成るNoLS(核小体局在シグナル:Nucleolar localization signal)に対応する。
配列番号9のアミノ酸配列は、核小体タンパク質の1種(ApLLP)由来の合計19アミノ酸残基から成るNoLSに対応する。
配列番号10のアミノ酸配列は、HSV−1(単純ヘルペスウイルス タイプ1)のタンパク質(γ(1)34.5)由来の合計16アミノ酸残基から成るNoLSに対応する。
配列番号11のアミノ酸配列は、HIC(human I-mfa domain-containing protein)のp40タンパク質由来の合計19アミノ酸残基から成るNoLSに対応する。
配列番号12のアミノ酸配列は、MDV(Marek病ウイルス)のMEQタンパク質由来の合計16アミノ酸残基から成るNoLSに対応する。
配列番号13のアミノ酸配列は、アポトーシスを抑制するタンパク質であるSurvivin- deltaEx3由来の合計17アミノ酸残基から成るNoLSに対応する。
配列番号14のアミノ酸配列は、血管増殖因子であるAngiogenin由来の合計7アミノ酸残基から成るNoLSに対応する。
配列番号15のアミノ酸配列は、核リンタンパク質であってp53腫瘍抑制タンパク質と複合体を形成するMDM2由来の合計8アミノ酸残基から成るNoLSに対応する。
配列番号16のアミノ酸配列は、ベータノダウイルスのタンパク質であるGGNNVα由来の合計9アミノ酸残基から成るNoLSに対応する。
配列番号17のアミノ酸配列は、NF−κB誘導性キナーゼ(NIK)由来の合計7アミノ酸残基から成るNoLSに対応する。
配列番号18のアミノ酸配列は、Nuclear VCP-like protein由来の合計15アミノ酸残基から成るNoLSに対応する。
配列番号19のアミノ酸配列は、核小体タンパク質であるp120由来の合計18アミノ酸残基から成るNoLSに対応する。
配列番号20のアミノ酸配列は、HVS(ヘルペスウイルスsaimiri)のORF57タンパク質由来の合計14アミノ酸残基から成るNoLSに対応する。
配列番号21のアミノ酸配列は、細胞内情報伝達に関与するプロテインキナーゼの1種であるヒト内皮細胞に存在するLIMキナーゼ2(LIM Kinase 2)の第491番目のアミノ酸残基から第503番目のアミノ酸残基までの合計13アミノ酸残基から成るNoLSに対応する。
配列番号22のアミノ酸配列は、IBV(トリ伝染性気管支炎ウイルス:avian infectious bronchitis virus)のNタンパク質(nucleocapsid protein)に含まれる合計8アミノ酸残基から成るNoLSに対応する。
配列番号23のアミノ酸配列は、HIV(ヒト免疫不全ウイルス:Human Immunodeficiency Virus)のTATに含まれるタンパク質導入ドメイン由来の合計9アミノ酸配列から成る膜透過性モチーフに対応する。
配列番号24のアミノ酸配列は、上記TATを改変したタンパク質導入ドメイン(PTD4)の合計11アミノ酸配列から成る膜透過性モチーフに対応する。
配列番号25のアミノ酸配列は、ショウジョウバエ(Drosophila)の変異体であるAntennapediaのANT由来の合計18アミノ酸配列から成る膜透過性モチーフに対応する。
これらのうち、特にNoLSやTATに関連するアミノ酸配列(又はその改変アミノ酸配列)が好ましい。例えば、配列番号21や配列番号22に示すようなNoLS関連のCPP配列、或いは配列番号23〜25のTATやANT関連のCPP配列は、ここで開示される抗腫瘍ペプチドを構築するために好適に用いることができる。
(2)CPPとして機能するアミノ酸配列(以下「CPP関連配列」ともいう。)を備えておればよく、PD−1SP関連配列とCPP関連配列のいずれが相対的にN末端側(C末端側)に配置されていてもよい。
PD−1SP関連配列とCPP関連配列とが、実質的に隣接して配置されていることが好ましい。具体的には、PD−1SP関連配列とCPP関連配列との間に、両配列部分に包含されないアミノ酸残基が存在しない、或いは、存在していても該アミノ酸残基数は10個以下(より好ましくは5個以下、例えば1個か2個のアミノ酸残基)程度が好ましい。
ここで開示される抗腫瘍ペプチドは、ペプチド鎖を構成する全アミノ酸残基数が100以下が適当であり、80以下が好ましく、70以下(例えば30前後から50前後程度のペプチド鎖)が好ましい。このような鎖長の短いペプチドは、化学合成が容易であり、容易に抗腫瘍ペプチドを提供することができる。特に限定されるものではないが、免疫原(抗原)になり難いという観点から直鎖状又はへリックス状のものが好ましい。このような形状のペプチドはエピトープを構成し難い。
ここで開示される抗腫瘍ペプチドは、市販のペプチド合成機を用いた固相合成法により、所望するアミノ酸配列、修飾(C末端アミド化等)部分を有するペプチド鎖を合成することができる。
一般的な技法によって、この組換えベクターを所定の宿主細胞(例えばイースト、昆虫細胞、植物細胞)に導入し、所定の条件で当該宿主細胞又は該細胞を含む組織や個体を培養する。このことにより、目的とするペプチドを細胞内で発現、生産させることができる。そして、宿主細胞(分泌された場合は培地中)からペプチドを単離し、必要に応じてリフォールディング、精製等を行うことによって、目的の抗腫瘍ペプチドを得ることができる。
なお、組換えベクターの構築方法及び構築した組換えベクターの宿主細胞への導入方法等は、当該分野で従来から行われている方法をそのまま採用すればよく、かかる方法自体は特に本発明を特徴付けるものではないため、詳細な説明は省略する。
こうして得られるポリヌクレオチドは、上述のように、種々の宿主細胞中で又は無細胞タンパク質合成システムにて、抗腫瘍ペプチド生産のための組換え遺伝子(発現カセット)を構築するための材料として使用することができる。
ここで開示される抗腫瘍組成物の用途や形態に応じて適宜異なり得るが、典型的には、水、生理学的緩衝液、種々の有機溶媒が挙げられる。適当な濃度のアルコール(エタノール等)水溶液、グリセロール、オリーブ油のような不乾性油であり得る。或いはリポソームであってもよい。また、抗腫瘍組成物に含有させ得る副次的成分としては、種々の充填剤、増量剤、結合剤、付湿剤、表面活性剤、色素、香料等が挙げられる。
抗腫瘍組成物(抗腫瘍剤)の典型的な形態として、液剤、懸濁剤、乳剤、エアロゾル、泡沫剤、顆粒剤、粉末剤、錠剤、カプセル、軟膏、水性ジェル剤等が挙げられる。また、注射等に用いるため、使用直前に生理食塩水又は適当な緩衝液(例えばPBS)等に溶解して薬液を調製するための凍結乾燥物、造粒物とすることもできる。
なお、抗腫瘍ペプチド(主成分)及び種々の担体(副成分)を材料にして種々の形態の組成物(薬剤)を調製するプロセス自体は従来公知の方法に準じればよく、かかる製法自体は本発明を特徴付けるものでもないため詳細な説明は省略する。処方に関する詳細な情報源として、例えばComprehensive Medicinal Chemistry, Corwin Hansch監修,Pergamon Press刊(1990)が挙げられる。この書籍の全内容は本明細書中に参照として援用されている。
或いは、生体外(インビトロ)において培養している腫瘍細胞(生体から摘出された細胞塊又は組織又は器官である場合を包含する。)に対し、ここで開示される抗腫瘍組成物の適当量(即ち抗腫瘍ペプチドの適当量)を、少なくとも1回、対象とする培養細胞(組織等)の培地に供給するとよい。1回当たりの供給量及び供給回数は、培養する腫瘍細胞の種類、細胞密度(培養開始時の細胞密度)、継代数、培養条件、培地の種類、等の条件によって異なり得るため特に限定されないが、培地中の抗腫瘍ペプチド濃度が概ね5μM以上100μM以下の範囲内、好ましくは10μM以上50μM以下(例えば12.5μM以上25μM以下)の範囲内となるように、1回〜複数回添加することが好ましい。
表1に示す計5種のペプチドを市販のペプチド合成機を用いて製造した。具体的には次のとおりである。
サンプル1は、一実施例として設計されたものであり、ヒトのPD−1のシグナル配列(配列番号1)のC末端側に、CPP関連配列として配列番号21のアミノ酸配列(LIMキナーゼ2のNoLS)を含む合成ペプチドである。
サンプル2は、一実施例として設計されたものであり、ヒトのPD−1のシグナル配列(配列番号1)のC末端側に、CPP関連配列として配列番号23のアミノ酸配列(HIVのTAT)を含む合成ペプチドである。
サンプル3は、比較対象として設計されたものであり、ヒトのPD−L1のシグナル配列のC末端側に、CPP関連配列として配列番号21のアミノ酸配列(LIMキナーゼ2のNoLS)を含む合成ペプチドである。
サンプル4は、比較対象として設計されたものであり、ヒトのPD−1のシグナル配列をばらばらにしてアミノ酸配列を並び換えたもののC末端側に、CPP関連配列として配列番号21のアミノ酸配列(LIMキナーゼ2のNoLS)を含む合成ペプチドである。
サンプル5は、比較対象として設計されたものであり、CPP関連配列である配列番号21のアミノ酸配列(LIMキナーゼ2のNoLS)のみからなる合成ペプチドである。
合成した各サンプルのペプチドは、DMSO(ジメチルスルホキシド)に溶かし、各サンプルペプチドのストック液(濃度2.5mM)を調製した。
上記試験例1で合成した各サンプルペプチドの幾つかについて、数種類の培養腫瘍細胞を対象として抗腫瘍活性を評価した。
具体的には、供試腫瘍細胞として現在市場において入手可能な、ヒト非小細胞肺がん細胞株(NCI−H2444)、ヒト非小細胞肺がん細胞株(HCC827)、ヒト肺胞基底上皮腺がん細胞株(A549)、ヒト小細胞肺がん細胞株(NCI−H446)、ヒトメラノーマ細胞株(A2058)、ヒト乳線がん細胞株(MDA−MB−231)を使用した。また、比較対象として、市販される正常ヒト乳腺上皮細胞の培養株(MCF−12F)を使用した。
各細胞株の培養には、以下の培地を使用した。即ち、
(1)ヒト非小細胞肺がん細胞株(NCI−H2444)、ヒト非小細胞肺がん細胞株(HCC827)、ヒト肺胞基底上皮腺がん細胞株(A549)およびヒト小細胞肺がん細胞株(NCI−H446):
2mMのL−グルタミン、1mMのピルビン酸ナトリウム、10mMのHEPES、4500mg/mLのグルコース、50ユニット/mLのペニシリン、50μg/mLのストレプトマイシン、および10%のウシ胎児血清(FBS)を含むRPMI−1640培地(和光純薬(株)製品)。
(2)ヒトメラノーマ細胞株(A2058)およびヒト乳線がん細胞株(MDA−MB−231):
2mMのL−グルタミン、0.1mMの非必須アミノ酸(non-essential amino acids)、50ユニット/mLのペニシリン、50μg/mLのストレプトマイシン、および10%のFBSを含むDMEM培地(和光純薬(株)製品)。
(3)正常ヒト乳腺上皮細胞の培養株(MCF−12F):
20ng/mLのリコンビナントEGF、10μg/mLのインスリン、0.5μg/mLのヒドロコルチゾン、および10%のFBSを含むDMEM/F12培地(和光純薬(株)製品)。
試験の詳細は以下のとおりである。
次いで、当該96穴(ウェル)プレートを、CO2インキュベータ内に配置し、37℃、5%CO2条件下で約1日間(21時間〜24時間)のプレインキュベーションを実施した。
その後、評価対象とするいずれかのサンプルペプチドの濃度が12.5μMおよび25μMのいずれかとなるように、濃度別にペプチド含有試験培地をそれぞれ調製し、1ウェルあたり90μLとなるように評価対象とする細胞が培養されているウェル(即ち、上記プレインキュベーション後のウェル)に供給した。そして、当該96穴(ウェル)プレートを、CO2インキュベータ内に戻し、37℃、5%CO2条件下で48時間のインキュベーションを実施した。
なお、各ペプチド添加試験区の各ペプチド濃度における試験ウェル数(n)は、いずれも3に設定した。従って、以下の表に示す結果の値は、試験ウェル数3のそれぞれで得た結果の平均値である。細胞生存率(%)は以下のように決定した。
インキュベーション終了後、上記試薬を添加した細胞培養液を回収するとともにテトラゾリウム塩の還元に基づく波長450nmの吸光度(波長650nmの吸光度で補正した値:A450−A650)を測定する比色法により、細胞生存率(%)を評価した。具体的には、ペプチドを含有しない培地のみで上記48時間のインキュベーションを行った比較試験区の測定値(測定吸光度)を細胞生存率100%とした相対値で、各試験細胞株の細胞生存率(%)を測定吸光度から算出した。結果を表2に示す。
なお、詳細なデータは示していないが、PD−1SP関連配列として、ヒト以外の数種哺乳類(ウシ、マウス等)由来のPD−1SP関連配列をCPP関連配列とともに含む合成ペプチドについてもプライマリーなインビトロ培養試験において同様の抗腫瘍性が認められており、PD−1SP関連配列およびCPP関連配列を両方備える合成ペプチドの抗腫瘍性ペプチドとしての有用性が示された。
Claims (6)
- 少なくとも一種の腫瘍細胞の増殖を抑制する合成ペプチドであって、
以下の(1)および(2)に示すアミノ酸配列:
(1)膜タンパク質であるPD−1(Programmed cell death-1)のシグナルペプチドを構成するアミノ酸配列、または、該アミノ酸配列について1個、2個又は3個のアミノ酸残基が欠失、置換又は付加された改変アミノ酸配列;および
(2)細胞膜透過性ペプチド(CPP)として機能するアミノ酸配列;
をともに備え、
総アミノ酸残基数が100以下である、合成ペプチド。 - 前記PD−1のシグナルペプチドを構成するアミノ酸配列が、配列番号1〜7のいずれかに示すアミノ酸配列である、請求項1に記載の合成ペプチド。
- 前記CPPとして機能するアミノ酸配列が、ポリアルギニン、または、配列番号8〜25のいずれかに示すアミノ酸配列、または、該アミノ酸配列について1個、2個又は3個のアミノ酸残基が欠失、置換又は付加されたCPPとして機能する改変アミノ酸配列である、請求項1又は2に記載の合成ペプチド。
- 配列番号26または配列番号27に示すアミノ酸配列を有する、請求項1に記載の合成ペプチド。
- 少なくとも一種の腫瘍細胞の増殖を抑制する抗腫瘍組成物であって、
請求項1〜4のいずれか一項に記載の合成ペプチドと、
薬学上許容され得る少なくとも一種の担体と、
を備える、抗腫瘍組成物。 - 少なくとも一種の腫瘍細胞の増殖を抑制する方法であって、
インビトロにおいて対象とする腫瘍細胞に対して請求項1〜4のいずれか一項に記載の合成ペプチドを少なくとも1回供給することを包含する、腫瘍細胞の増殖抑制方法。
