Abstract
Description
本発明者らは、これらの知見に基づいてさらに検討を重ねた結果、本発明を完成するに至った。
[1]IL-33をコードするDNAを皮膚において特異的に発現可能な状態で保持するトランスジェニック非ヒト哺乳動物であって、SPF(specific pathogen free)の飼育条件下で、対応する非トランスジェニック非ヒト哺乳動物と比較して
(1)皮膚炎を自然発症する、
(2)炎症細胞数が増加している、
(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度が増加している、および
(4)掻破行動時間が増加している
からなる群から選択される特徴を1つ以上有する、非ヒト哺乳動物;
[2]IL-33をコードするDNAがケラチンプロモーターの制御下にある、[1]に記載の非ヒト哺乳動物;
[3]ケラチンプロモーターがヒトケラチン14プロモーターである、[2]に記載の非ヒト哺乳動物;
[4]IL-33が配列番号:2で表されるアミノ酸配列と同一または実質的に同一のアミノ酸配列を有する[1]〜[3]のいずれか1つに記載の非ヒト哺乳動物;
[5]炎症細胞が好酸球、肥満細胞および2型自然リンパ球からなる群から選択される細胞である、[1]〜[4]のいずれか1つに記載の非ヒト哺乳動物;
[6]サイトカインがIL-4、IL-5またはIL-13である、[1]〜[5]のいずれか1つに記載の非ヒト哺乳動物;
[7]ケモカインがCCL2、CCL4、CCL5またはCCL11である、[1]〜[6]のいずれか1つに記載の非ヒト哺乳動物;
[8]掻破行動時間または皮膚炎の症状が抗ヒスタミン薬またはステロイド薬によって低減することを特徴とする、[1]〜[7]のいずれか1つに記載の非ヒト哺乳動物;
[9]非ヒト哺乳動物がマウスである、[1]〜[8]のいずれか1つに記載の非ヒト哺乳動物;
[10]SPF(specific pathogen free)の飼育条件下で、[1]〜[9]のいずれか1つに記載の非ヒト哺乳動物に試験化合物を適用し、(1)皮疹スコア、(2)炎症細胞数、(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度、および(4)掻破行動時間からなる群から選択される項目を1つ以上測定し、測定された前記項目を試験化合物非投与の場合と比べて改善させる試験物質を選択することを含む、アトピー性皮膚炎治療薬のスクリーニング方法;
[11]SPF(specific pathogen free)の飼育条件下で、[1]〜[9]のいずれか1つに記載の非ヒト哺乳動物にアトピー性皮膚炎の予防・治療薬を適用し、(1)皮疹スコア、(2)炎症細胞数、(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度、および(4)掻破行動時間からなる群から選択される項目を1つ以上測定し、測定された前記項目をアトピー性皮膚炎の予防・治療薬非投与の場合と比べる、アトピー性皮膚炎の予防・治療薬の効果の評価方法;
を提供する。
本発明のTg動物の生体の一部としては、皮膚や、皮膚由来の組織片(例えば、表皮)および細胞(例えば、表皮角化細胞)などが好ましく例示される。
また、哺乳動物以外にもニワトリなどの鳥類が本発明で対象とする「非ヒト哺乳動物」と同様の目的に用いることができる。
「実質的に同一のアミノ酸配列を有する蛋白質」としては、例えばマウスIL-33の場合、配列番号:2で表わされるアミノ酸配列と実質的に同一のアミノ酸配列を含み、配列番号:2で表わされるアミノ酸配列からなる蛋白質と実質的に同質の活性を有する蛋白質が好ましい。「実質的に同質の活性」としては、例えば、(1)皮膚炎誘導活性、(2)真皮層への炎症細胞の遊走誘導活性、(3)血清中の総IgE増大活性、(4)血漿中のヒスタミン増大活性、(5)サイトカイン、ケモカイン増大活性および(6)掻破行動惹起活性などが挙げられる。実質的に同質とは、それらの活性が定性的に同等であることを示す。したがって、該活性が同等であることが好ましいが、これらの活性の程度(例、約0.01〜100倍、好ましくは約0.5〜20倍、より好ましくは約0.5〜2倍)は異なっていてもよい。該活性の測定は、自体公知の方法に準じて行なうことができる。また、後述する実施例の通りに測定することもできる。
用いる非ヒト哺乳動物の齢や飼育条件等は動物種によってそれぞれ異なるが、例えばマウス(好ましくはC57BL/6J(B6)などの近交系マウス、B6と他の近交系とのF1など)を用いる場合は、雌が約4〜約6週齢、雄が約2〜約8ヶ月齢程度のものが好ましく、また、約12時間明期条件(例えば7:00-19:00)で約1週間飼育したものが好ましい。
体内受精は自然交配によってもよいが、性周期の調節と1個体から多数の初期胚を得ることを目的として、雌非ヒト哺乳動物に性腺刺激ホルモンを投与して過剰排卵を誘起した後、雄非ヒト哺乳動物と交配させる方法が好ましい。雌非ヒト哺乳動物の排卵誘発法としては、例えば初めに卵胞刺激ホルモン(妊馬血清性性腺刺激ホルモン、一般にPMSGと略する)、次いで黄体形成ホルモン(ヒト絨毛性性腺刺激ホルモン、一般にhCGと略する)を、例えば腹腔内注射などにより投与する方法が好ましいが、好ましいホルモンの投与量、投与間隔は非ヒト哺乳動物の種類によりそれぞれ異なる。例えば、非ヒト哺乳動物がマウス(好ましくはC57BL/6J(B6)などの近交系マウス、B6と他の近交系とのF1など)の場合は、通常、卵胞刺激ホルモン投与後、約48時間後に黄体形成ホルモンを投与し、直ちに雄マウスと交配させることにより受精卵を得る方法が好ましく、卵胞刺激ホルモンの投与量は約20〜約50IU/個体、好ましくは約30IU/個体、黄体形成ホルモンの投与量は約0〜約10IU/個体、好ましくは約5IU/個体である。
一定時間経過後、膣栓の検査等により交配を確認した雌非ヒト哺乳動物の腹腔を開き、卵管から受精卵を取り出して胚培養用培地(例:M16培地、修正Whitten培地、BWW培地、M2培地、WM-HEPES培地、BWW-HEPES培地等)中で洗って卵丘細胞を除き、微小滴培養法等により5%炭酸ガス/95%大気下でDNA顕微注入まで培養する。直ちに顕微注入を行わない場合、採取した受精卵を緩慢法または超急速法等で凍結保存することも可能である。
受胚用雌は自然排卵のものを用いてもよいし、あるいは精管切除(結紮)雄との交配に先立って、黄体形成ホルモン放出ホルモン(一般にLHRHと略する)もしくはその類縁体を投与し、受精能を誘起させたものを用いてもよい。LHRH類縁体としては、例えば、[3,5-DiI-Tyr5]-LH-RH、[Gln8]-LH-RH、[D-Ala6]-LH-RH、[des-Gly10]-LH-RH、[D-His(Bzl)6]-LH-RHおよびそれらのEthylamideなどが挙げられる。LHRHもしくはその類縁体の投与量、ならびにその投与後に雄非ヒト哺乳動物と交配させる時期は、非ヒト哺乳動物の種類によりそれぞれ異なる。例えば、非ヒト哺乳動物がマウス(好ましくはICR系のマウスなど)の場合には、通常、LHRHもしくはその類縁体を投与した後、約4日目に雄マウスと交配させることが好ましく、LHRHあるいはその類縁体の投与量は、通常、約10〜60μg/個体、好ましくは約40μg/個体である。
通常、F0動物は相同染色体の一方にのみ導入DNAを有するヘテロ接合体として得られる。また、個々のF0個体は相同組換えによらない限り異なる染色体上にランダムに挿入される。従って、相同染色体の両方にIL-33をコードするDNAを有するホモ接合体を得たい場合には、F0動物と非トランスジェニック動物とを交雑してF1動物を作製し、相同染色体の一方にのみ導入DNAを有するヘテロ接合体の兄妹同士を交雑すればよい。1遺伝子座にのみ導入DNAが組み込まれていれば、得られるF2動物の1/4がホモ接合体となる。
また、第二次セレクションとして、例えば、G-バンディング法による染色体数の確認等により行うことができる。得られるES細胞の染色体数は正常数の100%が望ましいが、細胞株樹立の際の物理的操作等の関係上困難な場合は、ES細胞への遺伝子導入の後、正常細胞(例えば、マウスでは染色体数が2n=40である細胞)に再びクローニングすることが望ましい。
ES細胞は、適当な条件により、高密度に至るまで単層培養するか、または細胞集塊を形成するまで浮遊培養することにより、頭頂筋、内臓筋、心筋などの種々のタイプの細胞に分化させることが可能であり〔M. J. Evans及びM. H. Kaufman, ネイチャー(Nature)第292巻、154頁、1981年;G. R. Martin, プロシーディングズ・オブ・ナショナル・アカデミー・オブ・サイエンシーズ・ユーエスエー(Proc. Natl. Acad. Sci. U.S.A.)第78巻、7634頁、1981年;T. C. Doetschmanら, ジャーナル・オブ・エンブリオロジー・アンド・エクスペリメンタル・モルフォロジー、第87巻、27頁、1985年〕、本発明のIL-33をコードするDNAを導入されたES細胞を分化させて得られるIL-33を皮膚において特異的に発現する非ヒト哺乳動物細胞は、生体におけるIL-33の細胞生物学的検討において有用である。
また、生殖系列キメラ形成能はES細胞と宿主胚との組み合わせに大きく依存するので、生殖系列キメラ形成能の高い組み合わせを選択することがより好ましい。例えばマウスの場合、129系統由来のES細胞に対してはC57BL/6系統由来の宿主胚等を用いることが好ましく、C57BL/6系統由来のES細胞に対してはBALB/c系統由来の宿主胚等が好ましい。
採卵用雌マウスは約4〜約6週齢程度が好ましく、交配用の雄マウスとしては約2〜約8ヶ月齢程度の同系統のものが好ましい。交配は自然交配によってもよいが、好ましくは性腺刺激ホルモン(卵胞刺激ホルモン、次いで黄体形成ホルモン)を投与して過剰排卵を誘起した後に行なわれる。
共培養法による場合は、8細胞期胚および桑実胚(例えばマウスの場合、交配後約2.5日)を採卵用雌の卵管および子宮から採取して(あるいは8細胞期以前の初期胚を卵管から採取した後、上述の胚培養用培地中で8細胞期または桑実胚期まで培養してもよい)酸性タイロード液中で透明帯を溶解した後、ミネラルオイルを重層した胚培養用培地の微小滴中にIL-33をコードするDNAが導入されたES細胞塊(細胞数約10〜約15個)を入れ、さらに上記8細胞期胚または桑実胚(好ましくは2個)を入れて一晩共培養する。得られた桑実胚または胚盤胞を上記と同様にして受胚用雌非ヒト哺乳動物の子宮内に移植する。
生殖系列キメラの選択は、まずES細胞の雌雄が予め判別されている場合はES細胞と同じ性別のキメラマウスを選択し(通常は雄性ES細胞が使用されるので、雄キメラマウスが選択される)、次いで毛色等の表現型からES細胞の寄与率が高いキメラマウス(例えば、50%以上)を選択する。例えば、129系マウス由来の雄性ES細胞であるD3細胞とC57BL/6マウス由来の宿主胚とのキメラ胚から得られるキメラマウスの場合、アグーチの毛色の占める割合の高い雄マウスを選択するのが好ましい。選択されたキメラ非ヒト哺乳動物が生殖系列キメラであるか否かの確認は、適当な系統の同種動物との交雑により得られるF1動物の表現型に基づいて行なうことができる。例えば、上記キメラマウスの場合、アグーチはブラックに対して優性であるので、雌C57BL/6マウスと交雑すると、選択された雄マウスが生殖系列キメラであれば得られるF1の毛色はアグーチとなる。
さらに、プロシーディングズ・オヴ・ナショナル・アカデミー・オヴ・サイエンシーズ・ユーエスエー(Proc. Natl. Acad. Sci. USA)第98巻, 第13090-13095頁, 2001年に記載されるように、雄非ヒト哺乳動物から採取した精原細胞をSTOフィーダー細胞と共培養する間にウイルスベクターに感染させた後、雄性不妊非ヒト哺乳動物の精細管に注入して雌非ヒト哺乳動物と交配させることにより、効率よくIL-33へテロTg(+/-)産仔を得ることができる。
(1)皮膚炎を自然発症する、
(2)炎症細胞数が増加している、
(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度が増加している、および
(4)掻破行動時間が増加している
からなる群から選択される特徴を1つ以上有することを特徴とする。
アトピー性皮膚炎患者は、表皮角化細胞において健常者の該細胞よりも、IL-33の発現強度が高い(非特許文献3)。また、ヒトのアトピー性皮膚炎患者の皮疹部においてIL-5遺伝子発現が亢進しており(Corren J, Discov Med 13:305-12, 2012)、血中の好酸球増多がみられない患者でも皮疹部には好酸球浸潤が認められる(Kiehl P et al., Br J Dermatol. 145:720-9, 2001)。さらに、アトピー性皮膚炎患者においては、肥満細胞の活性化に伴い、ヒスタミン濃度、IgE濃度の増加が見られる。
一方、上記の本発明のTg動物も、皮膚において顕著に高いレベルのIL-33発現を確認することができると共に、その表現型も、アトピー性皮膚炎患者の有する特徴と一致する。後述する実施例では、本発明のTg動物がTgマウスであった場合、SPFの飼育条件下で6週齢から一部のTgマウスが皮膚炎を発症し、8週齢以降では全てTgマウスが皮膚炎を自然発症した。また、炎症細胞(特に好酸球、肥満細胞、2型自然リンパ球(type 2 innate lymphoid cells))について、皮膚、末梢血またはリンパ節において有意な増加を認めた。さらに、血清中の総IgE濃度についても、野生型マウスと比較して有意に増加し、また、血漿中のヒスタミン濃度についても、野生型マウスと比較して有意に増加した。サイトカインとして、IL-4、IL-13が皮膚中で、IL-5が、皮膚、血清中で野生型マウスと比較して有意に増加した。また、ケモカインについてCCL5、CCL11が皮膚中で、CCL2、CCL4が皮膚、血清中で野生型マウスと比較して有意に増加した。加えて、野生型マウスと比較して掻破行動時間が有意に増加し、抗ヒスタミン薬(ジフェンヒドラミン塩酸塩)やステロイド薬(ベタメタゾン酪酸エステルプロピオン酸エステル)によって増加した掻破行動時間や皮疹スコアを低減させることができた。
より具体的には、本発明のTgマウスの前記特徴は、皮膚炎の皮疹スコアとしては約10〜15であった。また、炎症細胞数としては野生型マウスと比較して好酸球は皮膚では約7〜8倍、末梢血では約4〜5倍であり、肥満細胞は皮膚では約2〜3倍であり、2型自然リンパ球は皮膚では約12倍、末梢血では約10倍、リンパ節では約20倍であった。また、総IgE濃度としては野生型マウスと比較して約20〜30倍であり、ヒスタミン濃度としては野生型マウスと比較して約8〜9倍であった。また、サイトカイン濃度としては野生型マウスと比較してIL-4、IL-13が皮膚ではそれぞれ約50倍、約3倍、IL-5が皮膚、血清中ではそれぞれ約50倍、約1500倍以上(IL-5は血清中において、野生型では検出限界(0.3pg/mL)以下であったが、Tgマウスでは約500pg/mLであった)に増加した。また、CCL5、CCL11が皮膚中ではそれぞれ約4倍、約5倍、CCL2が皮膚、血清中それぞれ約7倍、約14倍、CCL4が皮膚、血清中それぞれ約8倍、約100倍に増加した。
以上の特徴から、本発明のTg動物では、ヒトケラチン14プロモーターの制御の下IL-33をコードするDNAが、皮膚特異的に高発現され、好酸球、肥満細胞、2型自然リンパ球の活性化を介して、炎症反応を惹起し、アトピー性皮膚炎を発症していると考えられる。
pBluescript II KS(-)ファージミドベクター(Agilent technologies, La Jolla, CA)のEcoRIとHindIII部位の間に、2.0 kbのヒトケラチン14プロモーターを含むDNA配列(GenBank AC105208.15の40251-42254の塩基配列)、0.64 kb ウサギβグロビンイントロン 2を含むDNA配列(GenBank V00878.1の551-1195)、0.49 kbヒトケラチン14 poly Aシグナルを含むDNA配列(GenBank AC105208.15の46730- 47216)をもつK14発現ベクターを作製した。このベクターは、ウサギβグロビンイントロン 2とヒトケラチン14 poly Aシグナルの間にBamHI 部位を配し、BamHI部位に挿入したcDNAがケラチン14プロモーターの制御下に、表皮などの重層扁平上皮で安定して発現する構築である。0.8kb完全長マウスIL-33バリアント1をコードするcDNA(GenBank AK163464.1の61-861の塩基配列)はfantomTM clone B230120N24(ダナフォーム、横浜)に含まれるプラスミドDNAを鋳型にして、

のPCRプライマーで増幅し、BamHI部位にIn-Fusion HD cloning kit(タカラバイオ, 大津)を用いて挿入した。得られたマウスIL-33 cDNA発現ベクターをXmaI、XhoI、XmnI の制限酵素で消化し、ヒトケラチン14プロモーター、ウサギβグロビンイントロン2、マウスIL-33 variant 1 cDNA、ヒトケラチン14 poly Aシグナルを含む4.0 kbの線状DNA を単離、精製した(図1)。
マイクロインジェクションによるトランスジェニックマウス作製はHoganらの方法(Manipulating the mouse embryo, Cold Spring Harbor Laboratory Press. 1994)に従って行った。実施例1にて作製したマウスIL-33 variant 1 cDNAを含む前記DNA断片をC57BL/6J系統由来受精卵400個の前核にマイクロインジェクションした。インジェクションした受精卵を偽妊娠雌マウスの卵管内に移植し、飼育した。得られた産仔122匹を4週齢で離乳させた。6週齢に達した時点で、生育した109匹のマウス(雄68匹、雌41匹)の尾からDNAを採取した。取得したDNAに対して、

の配列を有するプライマーセットを用いてPCRを行い、12匹のマウスがトランスジーンを有することを確認した。
実施例2において取得された12匹のIL-33遺伝子トランスジェニックマウスについて、耳生検組織からRNAを抽出した。得られたRNAをもとにして、TaqManプローブMm00505403_m1 (Life Technologies Corp, Carlsbad, CA) を用いて定量的リアルタイムRT-PCRを実施し、マウスIL-33遺伝子(Il33)を高発現するマウスを選別した。12匹のIL-33遺伝子トランスジェニックマウスのうち4匹は耳組織において、野生型マウスに比べてIL-33遺伝子が11〜41倍高発現していることを確認した(図2)。このうち#89マウスをファウンダーとして用いることによって、生育とともに皮膚炎が安定的に生じ、生殖細胞系列へのトランスジーンの伝播が可能なマウス系統hK14mIL33tg #1 (HCM-0374)(以下、hK14mIL33tg)を樹立した。以後の解析は、hK14mIL33tgをC57BL/6Jマウスと交配して得た子孫のマウスを用いて行った。
マウスの耳介を4%パラホルムアルデヒドで固定後、パラフィンに包埋し、4μmの切片を作製した。切片を抗IL-33ポリクローナル抗体と反応させ、Alexa594で蛍光染色した。3回の実験のなかで、代表的な結果を図に示した(図3)。蛍光強度は黒色で示した。図の実線は角層表面を、点線は表皮真皮境界部を示す。野生型マウス(WT)に比較して、hK14mIL33tgマウス(Tg)皮膚の表皮の角化細胞の核にIL-33の発現増強を認めた。
野生型マウスおよびhK14mIL33tgマウスの各臓器におけるIL-33遺伝子の発現をqPCRで比較した。データは2回の実験の代表的な結果を示した(図4)。hK14mIL33tgマウスでは、各臓器に比較してIL-33遺伝子の発現が皮膚において選択的に増強していることが示唆された。
hK14mIL33tgマウスにおいて、痂皮、びらん、あるいは落屑を伴う浸潤性紅斑が顔面、とくに眼周囲、鼻周囲、その他、頚部、耳介、手足、尾部等、毛が少なく外界からの刺激を受けやすい部位に生じた(図5)。写真は24-28週齢のマウスの代表的な所見を示す。
hK14mIL33tgマウスは正常に出生し、6週齢までは正常に発育した。野生型マウス(WT)(n = 7)と比較し、hK14mIL33tgマウス(Tg)(n = 4)では6週齢から8週齢にかけて皮膚病変が発症した。8週齢以降は全てのhK14mIL33tgマウスに皮膚病変が見られた(図6)。
野生型マウス(WT)の皮膚組織とhK14mIL33tgマウス(Tg)の皮膚病変組織(いずれも眼囲)をヘマトキシリン・エオシン染色した。3匹のマウスを用いた2回の実験の代表的な組織像を示した(図7)。hK14mIL33tgマウスでは、表皮の肥厚と真皮内に好酸球を含む炎症細胞浸潤がみられた。
野生型マウス(WT)およびhK14mIL33tgマウス(Tg)の皮膚に浸潤する好酸球のフローサイトメトリー解析を行った。4匹のマウスを用いた2回の実験のうちの代表的データを示した(図8)。好酸球の比率は、皮膚細胞のB220-CD3-CD45+、即ち、T細胞、B細胞を除いた白血球(好中球、好酸球、好塩基球、肥満細胞、2型自然リンパ球などが相当)の細胞数から計算した。皮膚では、CCR3+Siglec-F+の好酸球浸潤がhK14mIL33tgマウスでは野生型マウスに比較して平均値で7.4倍と、著しい増加が見られた(図9)。
また同様に、野生型マウス(WT)およびhK14mIL33tgマウス(Tg)の末梢血細胞のフローサイトメトリー解析を行った。4匹のマウスを用いた2回の実験のうちの代表的データを示した(図10、11)。末梢血でも、同様にhK14mIL33tgマウスでは野生型マウスに比較して平均値で4.5倍と有意な好酸球増多が認められた。
さらに、眼囲皮膚から全RNAを調製し、好酸球マーカーである好酸球ペルオキシダーゼ(Epx)および好酸球顆粒主要塩基性タンパク質遺伝子(Prg2)の発現を定量的リアルタイムRT-PCRで解析した。その結果、EpxとPrg2の遺伝子発現はhK14mIL33tgマウス(Tg)において野生型マウス(WT)よりも、それぞれ、平均値で26.9倍、45.7倍と、有意に高値であった(図12)。
野生型マウス(WT)の皮膚組織とhK14mIL33tgマウス(Tg)の皮膚病変組織(いずれも眼囲)をトルイジンブルー染色した。3匹のマウスを用いた2回の実験の代表的な組織像を示した(図13)。トルイジンブルーによる染色の結果、hK14mIL33tgマウスに生じた皮膚炎病変部組織では、真皮の肥満細胞が増加していることが判明した。
野生型マウス(WT)あるいはhK14mIL33tgマウス(Tg)の皮膚に存在する肥満細胞のフローサイトメトリー解析を行った(図14)。肥満細胞の比率は、皮膚のB220-CD3-細胞のフローサイトメトリー解析から計算した(図15)。hK14mIL33tgマウス病変部皮膚では、c-kit+IgE+である肥満細胞が野生型マウスの正常皮膚に比較して、有意に増加していることが明らかになった。
野生型マウス(WT)およびhK14mIL33tgマウス(Tg)から血漿を採取し、ヒスタミン濃度をELISAで測定した。また、野生型マウスおよびhK14mIL33tgマウスから血清を採取し、総IgE濃度をELISAで測定した。血漿中のヒスタミン濃度は、野生型マウスでは平均 0.18 μMであったが、hK14mIL33tgマウスでは平均1.46 μMと、著しい高値であり、hK14mIL33tgマウスにおける肥満細胞の活性化が示唆された(図16)。また、血清中の総IgE値は、野生型マウスでは平均15 ng/mLに対し、hK14mIL33tgマウスでは平均386 ng/mLと、20倍以上の高値を示した(図17)。
野生型マウス(WT)およびhK14mIL33tgマウス(Tg)について、任意の20分間における皮膚掻破時間(sec/min)を測定した。野生型マウスに比較してhK14mIL33tgマウスでは有意な掻破時間の増加が見られ、皮膚の掻痒(かゆみ)が強いことが示唆された(図18)。さらに、掻破行動に対する抗ヒスタミン薬の効果を検証した。ジフェンヒドラミン塩酸塩2mgをhK14mIL33tgマウスの腹腔内に投与した。それにより、投与6分後から20分間に観察されるマウスの掻破行動の時間が平均4.6分から平均0.2分に減少した(図19)。hK14mIL33tgマウスの皮膚炎に伴う掻痒の原因として、肥満細胞が関与している可能性が示唆される。
好酸球の誘導はIL-5、IL-13などのTh2サイトカインや、好酸球が発現する受容体CCR3に結合するリガンドであるRANTES/CCL5やEotaxin-1/CCL11などのケモカインで制御されることが知られている。そこで、野生型マウス(WT)とhK14mIL33tgマウス(Tg)の血清および眼囲皮膚抽出液中のサイトカインとケモカイン濃度を網羅的に測定した。野生型マウスに比較して、hK14mIL33tgマウスでは、IL-5は血清と皮膚の両方で高値を示した(図20、21)。またhK14mIL33tgマウスにおいて、CCL2、CCL4は血清と皮膚の両方で増加し、CCL5とCCL11は皮膚で増加した(図20、21)。これらの結果から、hK14mIL33tgマウスにおける好酸球性炎症にこれらのサイトカインが重要な役割を担う可能性が示唆された。一方、hK14mIL33tgマウスにおいてTNF-αやIFN-γなどのTh1サイトカイン、IL-12p40には変化が無かった。他のアレルギー性炎症を引き起こす上皮細胞由来サイトカインであるIL-18、TSLP、IL-25については、皮膚病変部におけるIL-18の増加はわずかであり、他の皮膚炎モデルマウスで見られるような血清中のIL-18の増加は見られなかった。また、TSLPやIL-25には有意な増加は見られなかった。
IL-33を誘導する肺の寄生虫感染の実験(Yasuda K et al., Proc Natl Acad Sci U S A,109:3451-62012, 2012)では、IL-33に依存してIL-5を産生する細胞として2型自然リンパ球が想定されている。実施例14において、hK14mIL33tgマウスにおいてIL-5やIL-13の産生増強が見られたため、これらのサイトカインを産生する2型自然リンパ球が増加しているかどうかを検討するため皮膚、末梢血、リンパ節由来細胞のフローサイトメトリー解析を行った。細胞は、Linマーカー陰性の細胞分画にゲートをかけた。3匹のマウスを用いた2回の実験の内の代表的なデータである(図22)。野生型マウス(WT)と比べて、hK14mIL33tgマウス(Tg)ではLin- ST2+ Sca-1+2型自然リンパ球が皮膚局所、末梢血、リンパ節で増加していることが判明した。即ち、IL-33は2型自然リンパ球の誘導に関与することが示唆された。次に、リンパ節のLin- ST2+ 細胞におけるIL-5、IL-13 の発現を解析した。IL-5やIL-13を産生する細胞を同定するため、細胞内染色を行った。細胞は、Linマーカー陰性の細胞分画にゲートをかけた。3匹のマウスを用いた実験の代表的なデータである(図23)。さらに、Lin-ST2+IL-5+またはLin-ST2+IL-13+細胞の比率を、Lin-細胞のフローサイトメトリー解析から計算した(図24)。野生型マウス(WT)に比較してhK14mIL33tgマウス(Tg)のリンパ節では、IL-5あるいはIL-13を産生する2型自然リンパ球の増加が認められた。
hK14mIL33tgマウスにおける好酸球増多にIL-5が必要かどうかを明らかにする目的で、対照のIgG1抗体(20μg/匹)あるいはIL-5中和抗体(20μg/匹)をhK14mIL33tgマウスの腹腔内に2日おきに2週間投与した。その後、hK14mIL33tgマウスの末梢血細胞のフローサイトメトリー解析を行った。細胞は、B220-CD3-CD45+分画にゲートをかけた。4匹のマウスで行った実験の代表的なデータを示す(図25)。得られた好酸球の比率はB220-CD3-CD45+細胞の末梢血細胞のフローサイトメトリー解析から計算した(図26)。IL-5中和抗体投与群のhK14mIL33tgマウスでは、対照群に比して末梢血中の好酸球増多が有意に改善した。さらに、hK14mIL33tgマウスの眼囲皮膚組織のヘマトキシリン・エオシン染色を行った。4匹のマウスで行った実験の代表的な図を示す(図27)。皮膚炎の病変部では、表皮肥厚の改善、浸潤細胞の減少がみられ、好酸球の浸潤も明らかに改善した。また、皮膚病変部組織におけるIL-33と好酸球顆粒主要塩基性タンパク質遺伝子(Prg2)の発現を解析した(図28)。hK14mIL33tgマウスにおいて、IL-5中和抗体投与により、皮膚のIL-33遺伝子発現誘導には変化が無かったが、好酸球のマーカーであるPrg2の発現は有意に低下した。従って、IL-5中和抗体投与によってhK14mIL33tgマウスにおける皮膚への好酸球浸潤が抑制された。以上から、hK14mIL33tgマウスの好酸球増多には2型自然リンパ球などから産生されるIL-5が重要であることが示唆された。
hK14mIL33tgマウスの掻破行動に対するステロイド薬の効果を検証した。hK14mIL33tgマウスの皮疹部に、対照として白色ワセリン、ステロイド薬としてベタメタゾン酪酸エステルプロピオン酸エステルを2日おきに5回塗布した。皮疹スコアとして掻痒、紅斑/出血、浮腫、表皮剥離/糜爛、落屑/乾燥の5項目について、無し(0点)、軽度(1点)、中等度(2点)、重症(3点)で評価を行った(Matsuda H et al., Int. Immunol, 9(3):461-466, 1997)。その結果、皮疹スコアの合計点が平均12.3点から塗布後に4.8点にまで減少し、有意な改善が確認できた(図29)。
本出願は、日本で出願された特願2013−096637(出願日:平成25年5月1日)を基礎としており、その内容はすべて本明細書に包含されるものとする。
Claims (11)
- IL-33をコードするDNAを皮膚において特異的に発現可能な状態で保持するトランスジェニック非ヒト哺乳動物であって、SPF(specific pathogen free)の飼育条件下で、対応する非トランスジェニック非ヒト哺乳動物と比較して
(1)皮膚炎を自然発症する、
(2)炎症細胞数が増加している、
(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度が増加している、および
(4)掻破行動時間が増加している
からなる群から選択される特徴を1つ以上有する、非ヒト哺乳動物。 - IL-33をコードするDNAがケラチンプロモーターの制御下にある、請求項1に記載の非ヒト哺乳動物。
- ケラチンプロモーターがヒトケラチン14プロモーターである、請求項2記載の非ヒト哺乳動物。
- IL-33が配列番号:2で表されるアミノ酸配列と同一または実質的に同一のアミノ酸配列を有する請求項1〜3のいずれか1項に記載の非ヒト哺乳動物。
- 炎症細胞が好酸球、肥満細胞および2型自然リンパ球からなる群から選択される細胞である、請求項1〜4のいずれか1項に記載の非ヒト哺乳動物。
- サイトカインがIL-4、IL-5またはIL-13である、請求項1〜5のいずれか1項に記載の非ヒト哺乳動物。
- ケモカインがCCL2、CCL4、CCL5またはCCL11である、請求項1〜6のいずれか1項に記載の非ヒト哺乳動物。
- 掻破行動時間または皮膚炎の症状が抗ヒスタミン薬またはステロイド薬によって低減することを特徴とする、請求項1〜7のいずれか1項に記載の非ヒト哺乳動物。
- 非ヒト哺乳動物がマウスである、請求項1〜8のいずれか1項に記載の非ヒト哺乳動物。
- SPF(specific pathogen free)の飼育条件下で、請求項1〜9のいずれか1項に記載の非ヒト哺乳動物に試験化合物を適用し、(1)皮疹スコア、(2)炎症細胞数、(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度、および(4)掻破行動時間からなる群から選択される項目を1つ以上測定し、測定された前記項目を試験化合物非投与の場合と比べて改善させる試験物質を選択することを含む、アトピー性皮膚炎治療薬のスクリーニング方法。
- SPF(specific pathogen free)の飼育条件下で、請求項1〜9のいずれか1項に記載の非ヒト哺乳動物にアトピー性皮膚炎の予防・治療薬を適用し、(1)皮疹スコア、(2)炎症細胞数、(3)総IgE濃度、ヒスタミン濃度、サイトカイン濃度および/またはケモカイン濃度、および(4)掻破行動時間からなる群から選択される項目を1つ以上測定し、測定された前記項目をアトピー性皮膚炎の予防・治療薬非投与の場合と比べる、アトピー性皮膚炎の予防・治療薬の効果の評価方法。
