Abstract
Description
すなわち、本発明は、以下の発明を包含する。
(1) スフィンゴシン1−リン酸リアーゼの活性を阻害する化合物を同定、選択または試験する方法であって、(i)スフィンゴシン1−リン酸リアーゼを発現する細胞を播種して培養する工程、(ii)該細胞に、標識された脂質基質および被験化合物を混合して培養する工程、(iii)該細胞を溶解する工程、または該細胞の培養上清を分取する工程、(iv)リン酸化された脂質と結合するが、リン酸化されていない脂質とは結合しない担体材料に(iii)の細胞溶解物、または培養上清を接触させる工程、および(v)該担体に結合しているリン酸化されている脂質量を測定する工程、(vi)被験化合物を含まない場合と比較してリン酸化された脂質量が増加していることを指標としてスフィンゴシン1−リン酸リアーゼの活性が阻害されていると判断する工程、を含む方法。
(2) 脂質基質がスフィンゴシンまたはジヒドロスフィンゴシンであることを特徴とする(1)に記載の方法。
(3) 工程(i)および/または(ii)において、ビタミンB6類の濃度が通常の培地よりも低い培地を用いて細胞を培養することを特徴とする、(1)または(2)に記載の方法。
(4) ビタミンB6類の濃度が20μM以下であることを特徴とする、(3)に記載の方法。
(5) ビタミンB6類の濃度が1μM以下であることを特徴とする、(3)に記載の方法。
(6) ビタミンB6類の濃度が100nM以下であることを特徴とする、(3)に記載の方法。
(7) 被験化合物の添加の前に細胞がビタミンB6類を含まない培地で洗浄されており、工程(ii)において培地がビタミンB6類を実質的に含まないことを特徴とする、(3)に記載の方法。
(8) 担体がSPAビーズであることを特徴とする、(1)乃至(7)のいずれか一つに記載の方法。
(9) SPAビーズが、RNA結合ケイ酸イットリウムSPAビーズであることを特徴とする請求項8に記載の方法。
(10) 工程(i)および/または(ii)において、セラミドシンターゼ阻害剤が添加されることを特徴とする、(1)乃至(9)のいずれか一つに記載の方法。
(11) セラミドシンターゼ阻害剤がフモニシンB1であることを特徴とする、(10)に記載の方法。
(12) 炎症性腸疾患、自己免疫疾患、多発性硬化症もしくはアレルギー性疾患を予防または治療するための化合物を同定、選択または試験するための(1)乃至(11)のいずれか一つに記載の方法。
(13) 炎症性腸疾患が、潰瘍性大腸炎またはクローン病である、(12)に記載の方法。
(14) 自己免疫疾患が、関節リウマチ、全身性エリテマトーデス、抗リン脂質抗体症候群、多発性筋炎、皮膚筋炎、全身性皮膚硬化症、シェーグレン症候群、結節性多発動脈炎、顕微鏡的多発動脈炎、アレルギー性肉芽腫性血管炎、ウェゲナー肉芽腫症または混合性結合組織病である、(12)に記載の方法。
(15) アレルギー性疾患が、アトピー性皮膚炎、アレルギー性鼻炎、花粉症、アレルギー性結膜炎、アレルギー性胃腸炎、気管支喘息、小児喘息、食物アレルギー、薬物アレルギーまたは蕁麻疹である、(12)に記載の方法。
新たに構築したスクリーニング法の原理を図1に示す。なお、本明細書中におけるスクリーニング法とは、スフィンゴシン1−リン酸リアーゼの活性を阻害する化合物を同定、選択または試験する方法を意味する。培養細胞に標識された脂質基質、例えば3H標識スフィンゴシン(3H−Sph)または3H標識ジヒドロスフィンゴシン(3H−dhSph)を添加して細胞内に取り込ませた後、内在的に発現している、または強制発現させたスフィンゴシンキナーゼ(SPHKs)によって、リン酸化された脂質、例えば3H標識S1P(3H−S1P)または3H標識dhS1P(3H−dhS1P)に変換させる。これらのリン酸化脂質は、速やかにSPLによる非可逆的切断またはS1Pホスファターゼ(SPPs)による脱リン酸化を受けるが、このときSPL活性を阻害することによって分解速度を遅らせ、これらを細胞内に蓄積させる。すなわち、被験化合物の添加時において、被験化合物の非添加時と比較して3H−S1Pまたは3H−dhS1Pの細胞内における蓄積量が増加していれば、該被験化合物はSPL阻害活性を有すると判断することができる。また、ある種の細胞株では細胞内で生成されたS1PまたはdhS1Pは細胞外に分泌されることが示されており(Proc Natl Acad Sci USA.2006;103(44):16394−16399)、細胞内に蓄積された3H−S1Pまたは3H−dhS1Pの一部は細胞外に分泌されると考えられる。従って、培養上清中の3H−S1Pまたは3H−dhS1P量が増加していれば、同じく該被験化合物はSPL阻害活性を有すると判断することができる。なお、このときセラミドシンターゼ阻害剤(例えば、フモニシンB1)やSPPsの阻害剤を処理しておくことにより、シグナル/バックグラウンド比の向上や、SPL特異的な化合物の取得確率向上が期待できる。また、S1Pホスファターゼ遺伝子またはセラミドシンターゼ遺伝子をノックアウトまたはノックダウンすることによっても同様の効果が期待できる。S1Pホスファターゼ遺伝子としては、S1P phosphatase−1、S1P phosphatase−2を、またセラミドシンターゼ遺伝子としては、LAG1 homolog,ceramide synthase 1乃至6を挙げることができる。
2.本発明において使用されるアッセイ方法と担体材料
いくつかのアッセイ形式を、本発明の方法を実施するために使用することができるが、好ましいアッセイ形式は、シンチレーション近接アッセイ(SPA)などのシンチレーションアッセイである。SPA技術は、PPOなどの有機シンチラントを含有するシンチラントビーズの使用を伴う。アッセイは通常、水性環境ではそのエネルギーが容易に消失する低エネルギー放射線を放射する、3H、125I、14C、35Sまたは33Pなどの放射性同位体を使用して水性緩衝液中で行われる。例えば、3Hにより放射される電子は、6keVの平均エネルギーを有するにすぎず、水中では非常に短い経路長を有する。これらの同位体の1つで標識された分子が、直接的に、またはビーズに以前に結合させられた別の分子との相互作用を介してビーズ表面に結合した場合、放射される放射線により、シンチラントが活性化され、光が発生する。発生する光の量は、ビーズに結合している標識された分子の量に比例しており、液体シンチレーション(LS)カウンターで簡便に測定することができる。標識された分子がビーズに結合していない場合、その放射エネルギーは、ビーズに到達する前に周りの水性溶媒によって吸収され、光が発生しない。したがって、結合したリガンドはシンチレーションシグナルをもたらすが、遊離状態のリガンドは非常に低いバックグラウンドをもたらし、したがって、従来の放射性リガンド結合アッセイの特徴である時間のかかる分離工程の必要性がなくなる。アッセイにおいて必要とされる操作は数回の簡単なピペット操作工程に減らされ、これにより、より良好な正確性および再現性ならびにより大きいスループットが得られる。
3.SPL阻害剤を含有する医薬
本発明のスクリーニング方法によって見出される化合物またはその薬学的に許容され得る塩は、優れたS1Pリアーゼ阻害能を有するので、免疫系の活動を抑制することができる。従って、炎症性腸疾患(IBD)(例えば、潰瘍性大腸炎、クローン病など)、自己免疫疾患(例えば、関節リウマチ、全身性エリテマトーデス、抗リン脂質抗体症候群、多発性筋炎、皮膚筋炎、全身性皮膚硬化症、シェーグレン症候群、結節性多発動脈炎、顕微鏡的多発動脈炎、アレルギー性肉芽腫性血管炎、ウェゲナー肉芽腫症、混合性結合組織病など)、多発性硬化症(MS)、アレルギー性疾患(例えば、アトピー性皮膚炎、アレルギー性鼻炎(花粉症)、アレルギー性結膜炎、アレルギー性胃腸炎、気管支喘息、小児喘息、食物アレルギー、薬物アレルギー、蕁麻疹など)を予防または治療するために、あるいは、移植に対する拒絶反応を抑制するための医薬組成物の有効成分として、本発明のスクリーニング方法によって見出される化合物またはその薬学的に許容され得る塩を用いることができる。
実施例
以下、本発明を実施例により具体的に説明するが、本発明はこれらに限定されるものではない。
3H−dhS1P(60Ci/mmol、American Radiolabeled Chemicals、ART0618)または3H−dhSph(60Ci/mmol、American Radiolabeled Chemicals、ART0460)を細胞融解バッファー(Cisbio社製IP−One Tb Kitのコンポーネントである、IP−One Tb conjugate & lysis bufferとIP1 stimulation buffer(1×)の3:7混合液)で段階希釈した。
1)細胞培養および培地
マウス胸腺上皮由来細胞株(IT−79MTNC3細胞、ヒューマンサイエンス振興財団細胞バンク)は、10%ウシ胎児血清(FBS)を含むダルベッコ変法イーグル培地(DMEM,Invitrogen)(DMEM−10%FBS)を用いて37℃、5%CO2存在下で培養された。トランスフェクションやアッセイで使用する際には、0.05%トリプシン−EDTA(Invitrogen)を用いて剥離させたのち、遠心して細胞を回収した。化合物を処理したり3H−dhSphを取り込ませたりする際には、0.1%ウシ血清アルブミン(BSA,Sigma)を含むDMEM(DMEM−0.1%BSA)または0.1%BSAを含むビタミンB6類除去ダルベッコ変法イーグル培地(VB6−free DMEM、細胞科学研究所)(VB6−free DMEM−0.1%BSA)を使用した。
Nucleofector Solution L(Lonza)で1×106細胞/100μLに調製したIT−79MTNC3細胞に、マウスSPLに対する3種配列のsiRNA(Silencer select Pre−designed siRNA、Ambion、s73643〜s73645)またはネガティブコントロールsiRNA(QIAGEN)を200pmol添加し、Nucleofector II(Amaxa)を用いて、プログラムT−030でトランスフェクションした。
ノックダウン効率を調べるために、トランスフェクションした細胞をDMEM−10%FBSに懸濁し、細胞培養用6穴プレート(住友ベークライト)に2×105細胞/ウエルで播種して、37℃、5%CO2存在下で24時間または48時間培養した。培養後のIT−79MTNC3細胞からRNA抽出用キット(RNeasy Mini Kit、QIAGEN)を用いて全RNAを抽出し、さらにPrimeScript 1st strand cDNA Synthesis Kit(タカラバイオ)を用いてcDNAを調製した。これを鋳型として、QuantiTect SYBR Green PCR Kit(QIAGEN)と、マウスグリセルアルデヒド3リン酸脱水素酵素(GAPDH)遺伝子またはマウスSPL遺伝子に対する特異的プライマー(Perfect Real Time Primer、タカラバイオ)を用いて、リアルタイムPCRシステム(Mx4000 Multiplex Quantitative PCR System、Stratagene)により、各々のサンプルのSPL mRNAの相対定量解析を行った。結果は、ネガティブコントロールsiRNA導入細胞のSPL mRNA発現量に対する比として算出した。
SPL遺伝子をノックダウンしたIT−79MTNC3細胞のライセートを調製するために、トランスフェクションした細胞をDMEM−10% FBSに懸濁し、細胞培養用12穴プレート(住友ベークライト)に1.5×105細胞/ウエルで播種して、37℃、5%CO2存在下で24時間または48時間培養した。培養後のIT−79MTNC3細胞を回収し、120μLのホモジェナイズバッファー(50mM HEPES−NaOH(pH=7.4)、0.15M NaCl、10%Glycerol、1mM EDTA、1mM DTT、Complete protease inhibitor cocktail(Roche))を添加し、ソニケーター(HandySonic、トミー精工)で超音波処理を施した後、1,000G、4℃で3分間遠心して得た上清を細胞ライセートとした。このうち一部を用いて、Bradford法によるタンパク定量を実施した。
SPL遺伝子をノックダウンしたIT−79MTNC3細胞内の3H−dhS1Pの蓄積をSPAで検出するために、トランスフェクションした細胞を25μM フモニシンB1(Enzo Life Sciences)を含むDMEM−10%FBSに懸濁し、細胞培養用96穴プレート(住友ベークライト)に1×104細胞/ウエルで播種して、37℃、5%CO2存在下で24時間または48時間培養した。培養上清を一旦除去した後、8.3nM 3H−dhSphおよび1μM dhSph(Enzo Life Sciences)を含むDMEM−0.1%BSAを50μL添加して、37℃、5%CO2存在下で30分間培養した。培養上清を除去した後、70μLの細胞融解バッファーを加えて室温で2時間放置した。このうち50μLを、予め96穴白色プレートに20μLずつ分注したSPAビーズ(終濃度50%のグリセロールを含むように8倍希釈したもの)と混合し、透明なプレートシールで覆って室温、遮光下で一晩放置した。翌日、マイクロプレートシンチレーションカウンターで放射活性を測定した。
1)既知SPL阻害剤を用いたSPA
被験化合物の3H−dhS1P蓄積作用を評価するために、IT−79MTNC3細胞を25μMフモニシンB1を含むDMEM−10%FBSに懸濁し、細胞培養用96穴プレートに1×104細胞/ウエルで播種して、37℃、5%CO2存在下で一昼夜培養した。翌日、培養上清を一旦除去した後、段階希釈した被験化合物を含むDMEM−0.1%BSA(またはVB6−free DMEM−0.1%BSA)を40μL添加して37℃、5%CO2存在下で4.5時間培養した後、83nMの3H−dhSphおよび5μM dhSphを含むDMEM−0.1%BSA(またはVB6−free DMEM−0.1%BSA)を10μL添加して、37℃、5%CO2存在下で30分間培養した。培養上清を除去した後、70μLの細胞融解バッファーを加えて室温で2時間放置した。このうち50μLを、予め96穴白色プレートに20μLずつ分注したSPAビーズ(終濃度50%のグリセロールを含むように8倍希釈したもの)と混合し、透明なプレートシールで覆って室温、遮光下で一晩放置した。翌日、マイクロプレートシンチレーションカウンターで放射活性を測定した。なお、VB6−free DMEMを用いて実施する場合は、被験化合物を含む培地を添加する前に150μLのVB6−free DMEM−0.1% BSAで2回洗浄することにより、播種時に用いたDMEMに含まれるVB6類のキャリーオーバーを防止した。
被験化合物のS1P蓄積作用を評価するために、IT−79MTNC3細胞を25μMフモニシンB1を含むDMEM−10%FBSに懸濁し、細胞培養用24穴プレートに5×104細胞/ウエルで播種して、37℃、5%CO2存在下で一昼夜培養した。翌日、培養上清を一旦除去した後、段階希釈した被験化合物を含むDMEM−0.1%BSA(またはVB6−free DMEM−0.1%BSA)を450μL添加して37℃、5%CO2存在下で4.5時間培養した後、10μM Sph(Enzo Life Sciences)を含むDMEM−0.1%BSA(またはVB6−free DMEM−0.1%BSA)を50μL添加して、37℃、5%CO2存在下で30分間培養した。培養上清を除去し、PBSで洗浄した後に細胞を回収し、アセトニトリル:1M塩酸(4:1)混合液を150μL添加してソニケーターで超音波処理を施した。S1P量は、LC−MSにより三菱化学メディエンス株式会社で測定された。なお、VB6−free DMEMを用いて実施する場合は、被験化合物を含む培地を添加する前に600μLのVB6−free DMEM−0.1% BSAで2回洗浄することにより、播種時に用いたDMEMに含まれるVB6類のキャリーオーバーを防止した。
被験化合物を処理した細胞のライセートを調製するために、IT−79MTNC3細胞を25μMフモニシンB1を含むDMEM−10%FBSに懸濁し、細胞培養用12穴プレートに1×105細胞/ウエルで播種して、37℃,5%CO2存在下で一昼夜培養した。翌日、培養上清を一旦除去した後、段階希釈した被験化合物を含むDMEM−0.1% BSA(またはVB6−free DMEM−0.1% BSA)を500μL添加して37℃、5% CO2存在下で4.5時間培養した後、10μM dhSphを含むDMEM−0.1% BSA(またはVB6−free DMEM−0.1%BSA)を55μL添加して、37℃、5%CO2存在下で30分間培養した。PBSで洗浄した後にIT−79MTNC3細胞を回収し、150μLのホモジェナイズバッファーを添加してソニケーターで超音波処理を施した後、1,000G、4℃で3分間遠心して得た上清を細胞ライセートとした。このうち一部を用いて、Bradford法によるタンパク定量を実施した。なお、VB6−free DMEMを用いて実施する場合は、被験化合物を含む培地を添加する前に1mLのVB6−free DMEM−0.1% BSAで2回洗浄することにより、播種時に用いたDMEMに含まれるVB6類のキャリーオーバーを防止した。
フモニシンB1処理によるシグナル/バックグラウンド比の向上効果について検討するために、IT−79MTNC3細胞を0または25μMフモニシンB1を含むDMEM−10%FBSに懸濁し、細胞培養用96穴プレートに1×104細胞/ウエルで播種して、37℃、5%CO2存在下で一昼夜培養した。翌日、培養上清を一旦除去した後、DOPを含む(または含まない)DMEM−0.1%BSAを40μL添加して37℃、5%CO2存在下で4.5時間培養した後、42nMの3H−dhSphおよび5μM dhSphを含むDMEM−0.1%BSAを10μL添加して、37℃、5%CO2存在下で30分間培養した。培養上清を除去した後、70μLの細胞融解バッファーを加えて室温で2時間放置した。このうち50μLを、予め96穴白色プレートに20μLずつ分注したSPAビーズ(終濃度50%のグリセロールを含むように8倍希釈したもの)と混合し、透明なプレートシールで覆って室温、遮光下で一晩放置した。翌日、マイクロプレートシンチレーションカウンターで放射活性を測定した。
Claims (15)
- スフィンゴシン1−リン酸リアーゼの活性を阻害する化合物を同定、選択または試験する方法であって、(i)スフィンゴシン1−リン酸リアーゼを発現する細胞を播種して培養する工程、(ii)該細胞に、標識された脂質基質および被験化合物を混合して培養する工程、(iii)該細胞を溶解する工程、または該細胞の培養上清を分取する工程、(iv)リン酸化された脂質と結合するが、リン酸化されていない脂質とは結合しない担体材料に(iii)の細胞溶解物、または培養上清を接触させる工程、および(v)該担体に結合しているリン酸化されている脂質量を測定する工程、(vi)被験化合物を含まない場合と比較してリン酸化された脂質量が増加していることを指標としてスフィンゴシン1−リン酸リアーゼの活性が阻害されていると判断する工程、を含む方法。
- 脂質基質がスフィンゴシンまたはジヒドロスフィンゴシンであることを特徴とする請求項1に記載の方法。
- 工程(i)および/または(ii)において、ビタミンB6類の濃度が通常の培地よりも低い培地を用いて細胞を培養することを特徴とする、請求項1または2に記載の方法。
- ビタミンB6類の濃度が20μM以下であることを特徴とする、請求項3に記載の方法。
- ビタミンB6類の濃度が1μM以下であることを特徴とする、請求項3に記載の方法。
- ビタミンB6類の濃度が100nM以下であることを特徴とする、請求項3に記載の方法。
- 被験化合物の添加の前に細胞がビタミンB6類を含まない培地で洗浄されており、工程(ii)において培地がビタミンB6類を実質的に含まないことを特徴とする、請求項3に記載の方法。
- 担体がSPAビーズであることを特徴とする、請求項1乃至7のいずれか一つに記載の方法。
- SPAビーズが、RNA結合ケイ酸イットリウムSPAビーズであることを特徴とする請求項8に記載の方法。
- 工程(i)および/または(ii)において、セラミドシンターゼ阻害剤が添加されることを特徴とする、請求項1乃至9のいずれか一つに記載の方法。
- セラミドシンターゼ阻害剤がフモニシンB1であることを特徴とする、請求項10に記載の方法。
- 炎症性腸疾患、自己免疫疾患、多発性硬化症もしくはアレルギー性疾患を予防または治療するための化合物を同定、選択または試験するための請求項1乃至11のいずれか一つに記載の方法。
- 炎症性腸疾患が、潰瘍性大腸炎またはクローン病である、請求項12に記載の方法。
- 自己免疫疾患が、関節リウマチ、全身性エリテマトーデス、抗リン脂質抗体症候群、多発性筋炎、皮膚筋炎、全身性皮膚硬化症、シェーグレン症候群、結節性多発動脈炎、顕微鏡的多発動脈炎、アレルギー性肉芽腫性血管炎、ウェゲナー肉芽腫症または混合性結合組織病である、請求項12に記載の方法。
- アレルギー性疾患が、アトピー性皮膚炎、アレルギー性鼻炎、花粉症、アレルギー性結膜炎、アレルギー性胃腸炎、気管支喘息、小児喘息、食物アレルギー、薬物アレルギーまたは蕁麻疹である、請求項12に記載の方法。
