HepG2細胞(オーダー番号HB-8065、ロット58483209、ATCC, Manassas, USA)を、DMEM/F12中で、L−グルタミン、ならびに10%(v/v)FBS、1%(v/v)ペニシリン(10kU/ml)/ストレプトマイシン(10mg/ml)溶液、0.1%(v/v)ゲンタマイシン(50mg/ml)、および1mMのピルビン酸ナトリウムを補充した15mMのHepesと共に、37℃および5%CO2において、培養フラスコ中で培養した。実験に依存して、トリプシンを用いる細胞の剥離の後で、適切な数の細胞をプレート上に播種し、細胞を、37℃および5%CO2において、処置の前に24時間培養した。全ての実験を、4〜20回継代した細胞を用いて、3回行った。
・細胞の処置および用量の選択
細胞を、黒色のポリ−D−リジンコートされた96ウェルプレート(BD Biocoat, Franklin Lakes, USA)上に播種し(0.02×106細胞/ウェル)、接着のために24時間静置し、次いで、試験化合物で処置した。各化合物について、2倍連続希釈による7つの濃度を試験した。DMSO(1%(v/v))を、実験のためのビヒクル対照として利用した。処置の手順は、直接的遺伝毒性化合物と前駆型遺伝毒性化合物とで、異なった。直接的遺伝毒性化合物のための処置を続けたまま、48時間にわたって、1日1回繰り返した。前駆型遺伝毒性化合物は、代謝活性化システムと一緒に6時間インキュベートした(S9混合物の細胞傷害性効果を限定するために)。この期間の後で、細胞を細胞培地で洗浄し、48時間にわたり、1日18回繰り返した。用いられた代謝活性化システムは、以下の成分および濃度からなった:プレ混合物中で、8mMのMgCl2、32.8mMのKCl、12mMのNADPH、124mMのリン酸バッファーおよび2500pmol/mlのチトクロムP450(CYP)の含有物は、細胞培養培地での1:3.33希釈の後での最終濃度として、2.4mMのMgCl2、9.8mMのKCl、3.6mMのNADPH、37.2mMのリン酸バッファー、および750pmol/mlのCYPに対応した。

これらの5つのタンパク質は、薬物発見プロセスの初期における潜在的遺伝子毒の同定のための潜在的候補であった。
磁性ビーズキット(Milliplex MAP)を用いて、Luminexシステムシステムを用いる細胞ライセート中のリン酸化されたp53(Ser15)ならびにp21の合計タンパク質レベルの変化を検出した。
MILLIPLEX MAPは、Luminex xMAP技術に基づく。Luminexは、独自技術を用いて、マイクロスフェアの内部で2つの蛍光色素により色でコードする。これらの色素の正確な濃度を通して、100の区別し得るように色分けされたビーズセットを作製することができ、それらの各々は、特異的捕捉抗体によりコートされる。試験試料からの分析物をビーズにより捕捉した後、ビオチン化された検出抗体を導入する。反応混合物を次いで、レポーター分子であるストレプトアビジン−フィコエリトリン(SAPE)コンジュゲートと共にインキュベートし、各マイクロスフェアの表面上で反応を完了する。マイクロスフェアおよび内部色素の蛍光に照射し、特定のアッセイにおいて用いられたマイクロスフェアセットを標識する。第2の照射源は、レポーター分子上の蛍光色素であるPEを励起する。最終的に、高速デジタルシグナルプロセッサが、各々の個々のマイクロスフェアを同定し、蛍光レポーターシグナルに基づいて、そのバイオアッセイの結果を定量する。
アッセイの実行は、以下に記載されるように行い、試薬は、MILLIPLEX MAPキット(EMD Millipore カタログ#46-662および46-621)から用いた。
無菌の96ウェルの組織培養グレードのプレート中で生育された接着性または非接着性細胞は、同じプレート中で試験し、洗浄し、溶解することができるが、別の96ウェルフィルタープレート中で濾過されることを必要とした。プロトコルのステップは、以下のとおりである:非接着性細胞について、組織培養プレートを、2分間、500×gにおいて遠心分離して、細胞をペレット化した。接着性細胞を用いた場合、それは次のステップから開始した。吸引を介して培地を除去し、100μLの氷冷PBSまたはTBSを添加した。非接着性細胞について、第1のステップを繰り返した。吸引を介して洗浄液を除去した。それに、ホスファターゼ阻害剤(1mMのオルトバナジン酸ナトリウム(Na3VO4)を含む)および新たに調製したプロテアーゼ阻害剤を含有する35μL/ウェルの氷冷1×MILLIPLEX MAP溶解バッファーを添加した。プレートを、オービタルシェイカー(600〜800rpm)上で10〜15分間、4℃において置いた。ライセートを、−70℃で、使用のために準備が整うまで保存した。
濾過されたライセートを、少なくとも1:1で、MILLIPLEX(登録商標)MAPアッセイバッファー中で希釈した。提案されるアッセイのためのタンパク質濃度の機能的範囲は、合計タンパク質1〜25μg/ウェル(40〜1000μg/mLにおいて、25μL/ウェル)であった。50μLのアッセイバッファーを、プレートの各ウェル中に添加し、カバーして、プレートシェイカー上で10分間、室温(20〜25℃)において混合した。アッセイバッファーをデカントし、プレートを逆さまにして吸収性タオルの上に数回しっかりとタップすることにより、残りの量を全てのウェルから取り除いた。1×のビーズ懸濁液を10秒間ボルテックスし、25μLの1×のビーズ懸濁液を各ウェルに添加した。25μLのアッセイバッファー、再構成された対照細胞ライセートおよび試料ライセートを、適切なウェルに添加し、一晩(16〜20時間)、2〜8℃において、プレートシェイカー(600〜800rpm)上で、光から保護してインキュベートした。手で保持した磁性分離ブロックをプレートに接合して、試料および対照をデカントする前に、ビーズを60秒間定着させた。プレートを磁性分離ブロックから取り除き、ウェルあたり100μLのアッセイバッファーで洗浄した。それを、合計2回の洗浄のために繰り返した。25μL/ウェルの1×MILLIPLEX MAP検出抗体を添加した。プレートを密閉し、蓋でカバーして、プレートシェイカー上で撹拌しながら1時間、室温(20〜25℃)でインキュベートした。検出抗体をデカントする前に、磁性分離ブロックを接合して、60秒間待った。25μLの1×MILLIPLEX MAPストレプトアビジン−フィコエリトリン(SAPE)を添加した。プレートを密閉し、蓋でカバーして、プレートシェイカー上で撹拌しながら15分間、室温(20〜25℃)でインキュベートした。25μLのMILLIPLEX MAP増幅バッファーを各ウェルに添加した。プレートを密閉し、蓋でカバーして、プレートシェイカー上で撹拌しながら15分間、室温(20〜25℃)でインキュベートした。SAPE/増幅バッファーをデカントする前に、磁性分離ブロックを接合して、60秒間待った。ビーズを150μLのMILLIPLEX MAPアッセイバッファー中で懸濁し、プレートシェイカー上で5分間混合して、その後、Luminexシステムを用いて分析した。
処置された試料のビヒクル対照に対する調節の倍率を、各化合物および濃度について計算した。各パラメーターについての閾値を、対照値と比較して、対照値に標準偏差の3倍を加算したものに基づいて選択した。p-p53(Ser 15)およびp21について、閾値を1.5に設定した。タンパク質のうちの少なくとも1つがこの閾値より高い倍率での変化を示した場合、化合物を陽性として設定した。そうでない場合、化合物を陰性として設定した。
Claims (9)
- 遺伝毒性および/または前駆型遺伝毒性活性について、化合物をハイスループットスクリーニングするための方法であって、化合物と共にインキュベートされたシステムにおいて、少なくとも、タンパク質である、細胞腫瘍抗原p53(p-p53(Ser15))、サイクリン依存性キナーゼ阻害剤1(p21)、ヒストンH2A.X(p-H2A.X(Ser139))、セリン−タンパク質キナーゼ毛細血管拡張性運動失調症変異(p-ATM(Ser1981))およびセリン/スレオニン−タンパク質チェックポイントキナーゼ1(p-Chk1(Ser345))の活性形態の発現レベルを、前記化合物と共にインキュベートされていないシステムにおける発現レベルと比較して決定するステップを含み、ここで、前記化合物と共にインキュベートされたシステムにおける前記タンパク質の発現レベルの、前記化合物と共にインキュベートされていないシステムと比較した場合の少なくとも1.5倍の増大が、少なくとも80%の感度および/または特異度で前記活性を示す、前記方法。

- 化合物による処置に対する応答において、損傷の修復の調節解除により引き起こされるか、媒介されるか、および/または伝播される、癌、腫瘍、転移および/または血管新生の障害を発症する見込みをモニタリングするための方法であって、ここで、少なくとも、タンパク質である、p-p53(Ser15)、p21、p-H2A.X(Ser139)、p-ATM(Ser1981)およびp-Chk1(Ser345)の発現レベルが、哺乳動物に投与される前記化合物によるかかる処置を必要とする前記哺乳動物から回収された生物学的試料において決定され、ここで、前記タンパク質の発現レベルの少なくとも1.5倍の増大が、前記化合物が少なくとも80%の感度および/または特異度で遺伝毒性および/または前駆型遺伝毒性活性を有すること、ならびに前記見込みが増大することを示す、前記方法。
- 薬物による治療的処置に対する応答において患者が腫瘍を罹患する見込みを予測するためのin-vitroの方法であって、以下のステップ:
(i)前記患者の組織または血漿からの生検試料において、少なくとも、タンパク質である、p-p53(Ser15)、p21、p-H2A.X(Ser139)、p-ATM(Ser1981)およびp-Chk1(Ser345)の発現レベルを測定すること、
(ii)前記患者の組織または血漿からの試料を、前記薬物にex-vivoで暴露すること、ならびに、
(iii)ステップ(ii)の前記暴露された試料において、ステップ(i)において特定された前記タンパク質の発現レベルを測定し、ステップ(i)および(iii)において測定される発現レベルの差異を計算することに加えて、ここで、このステップ(iii)において得られた前記タンパク質の発現レベルの、ステップ(i)と比較した少なくとも1.5倍の増大が、前記薬物が少なくとも80%の感度および/または特異度で遺伝毒性および/または前駆型遺伝毒性活性を有すること、ならびに前記見込みが増大することを示す、

- 遺伝毒性および/または前駆型遺伝毒性活性の検出における使用のためのキットであって、各々がp-p53(Ser15)、p21、p-H2A.X(Ser139)、p-ATM(Ser1981)、p-Chk1(Ser345)、p-ATR(Ser428)、p-cdc2(Thr14/Tyr15)、Gadd45aおよびp-Chk2(Thr68)の群から選択される異なるタンパク質に対する特異的結合を有する、9つの抗体を含む、前記キット。
