器官再生研究と実現可能な医療的、経済的効果に関する調査(2006年10月)
次いで、MDU1細胞株を、プラスティックディッシュから、トリプシン−EDTA処理(GIBCO)により剥がして回収して、プラスチックフィルターを通してバラバラにし、次いで、低接着性の96ウェルのプラスティックプレート(Nunc社)に播種して胚様体様構造(embryoid body−like sphere)を作製した。作製した胚様体様構造をおよそ1mm×1mm×1mmに細断したコラーゲン担体(Collagen Sponge HoneycombTM、株式会社高研:コラーゲン スキャフォルド(collagen scaffold)、(CS))とともに5−12日間培養した。細胞をすべてCSに接着するのに5日間かかり、12日までに毛髪が産生しない場合は、3週間以上培養しても毛髪が産生しなかった。この結果、本発明の実施形態では、5−12日間の培養が好適であることが認識できる。胚様体様構造を接着したCSの一部を、そのまま固定して、パラフィン包埋試料として観察した。残りの試料は、6週齢の成体マウス(ddy)の頭蓋上皮下および背筋筋膜下に常套手段で移植して15日後に取り出し光学顕微鏡で観察した。図3に以上に述べた本発明の実施形態のプロトコルを示す。
[試験管内での検証]
はじめに、試験管内で毛髪の形成状況を検証した(図4)。胚様体様構造を接着したCS上に黒色毛が成長していた(典型的な例を矢印で示した)。すなわち、この培養系で、毛髪が形成できるだけでなく、メラノサイトも形成でき、さらに、メラノサイトが産生したメラノソームを含む毛髪が形成できることが確認された。MDU1細胞株は、白ネズミ由来なので、この系でメラノサイトが新たに形成されたことが検証された。成長因子を含む培養液を使い、同様にしてMDU1細胞株を培養したところ、インスリン成長因子1(insulin growth factor−1,IGF−1;TOYOBO)、繊維芽細胞成長因子2(fibroblast growth factor−2,FGF−2;TOYOBO)、トランスフォーミング増殖因子β1(transforming growth factor−β1,TGF−β1;TOYOBO)が有意に毛髪の成長を促進した。また、MDU1細胞株を、単独でなく、マウス由来の胚性肝細胞(ES細胞)と混合して、同様に培養する(ここで、MDU1とES細胞を混合した培養体をキメラ胚様体と呼ぶ)と、これも有意に毛髪の成長を促進した。さらに、MDU1細胞株及び上記キメラ胚様体ではなく、胚性肝細胞(ES細胞)単体で、上述した実施例と同様に培養あるいは、誘導因子を含めずに同様に培養しても毛髪を形成したことが確認された(図5)。
次に、培養したキメラ胚様体を吸着したコラーゲン担体をマウスの頭蓋上皮下及び背筋筋膜下に常套手段で移植して15日後に取り出し光学顕微鏡で観察した。この結果、マウスの生体内で毛髪が形成されることを検証した(図6)。ここで形成された毛髪は、試験管内で形成された毛髪と同じく黒い毛髪だった。また、毛髪を産生する器官である毛包もまた、頻繁に見出され、確かに、この方法で、毛髪が新しく形成されたことが検証された。
付記(1):哺乳類の未分化細胞を培養して胚様体を作製するステップと、該胚様体をさらに培養するステップとよりなる毛髪の形成方法であって、前記培養ステップは、前記胚様体を三次元マトリクスにて5乃至12日間培養することを特徴とする毛髪の形成方法。

- 哺乳類の未分化細胞を培養して胚様体を作製するステップと、該胚様体をさらに培養するステップとを含む毛髪の形成方法であって、
前記培養ステップは、前記胚様体を三次元マトリクスにて5乃至12日間培養する、毛髪の形成方法。 - 前記胚様体を培養する三次元マトリクスは、培養又は移植用の担体である、請求項1に記載の毛髪の形成方法。
- 前記担体は、タイプ I コラーゲン スキャフォルドである、請求項2に記載の毛髪の形成方法。
- 前記未分化細胞は、さらに、誘導因子を含む培養液で培養する、請求項1に記載の毛髪の形成方法。
- 前記誘導因子は、インスリン成長因子1、繊維芽細胞成長因子2及びトランスフォーミング増殖因子β1のすべてを含む、請求項4に記載の毛髪の形成方法。
