Abstract
本発明の目的は、TLRシグナルにおけるIKKαの役割を解明し、インターフェロン−αの制御剤及び制御方法を提供することである。本発明によれば、IKKαの阻害剤を含む、インターフェロン−α産生抑制剤が提供される。
Description
本発明は、インターフェロン−αの制御剤及び制御方法に関する。より詳細には、IKKαの阻害剤を用いたインターフェロン−α産生の抑制剤又は抑制方法、並びにIKKα、又はIKKαの発現を増大させる物質を用いたインターフェロン−α産生の誘導剤又は誘導方法に関する。
Toll様受容体(Toll-like receptors; TLRs)は、樹状細胞(DC)等の抗原提示細胞上に発現しており、多様な微生物由来の分子構造の認識に関与している。TLRリガンドとしては、細菌及びウイルスに由来する脂質、タンパク質及び核酸成分が挙げられる。TLRの多くは、IL-6などの炎症性サイトカインの誘導、共刺激分子の発現などを含む、TLRに共通の樹状細胞活性化作用を有しているが、各TLRは各TLRに特有の機能を示す。
TLR7及びTLR9はそれぞれ一本鎖RNA及び非メチル化CpG DNAを認識できる。これらの核酸を認識するTLRは、抗ウイルス免疫に必要なIFN−α及びIFN−β等のI型インターフェロン(IFN)を誘導できることを特徴とする(Wagner, H, Trends Immunol 25, 381-6 (2004))。TLR7及びTLR9は、細胞質内アダプター分子であるMyD88と会合する。MyD88は、樹状細胞がTLRリガンドに応答して、I型インターフェロンのみならず炎症性サイトカインを産生するのに必要である。転写因子IRF−7も、TLR−7/9により誘導されるI型インターフェロンの産生に重要である(Honda, K et al., Nature, 434, 772-7 (2005))。IRF7はMyD88と腫瘍壊死因子(TNF)受容体関連因子6(TRAF6)に会合して、IFN−α遺伝子を誘導する(Kawai, T. et al., Nat Immunol 5, 1061-8 (2004);及びHonda, K. et al., Proc Natl Acad Sci USA 101, 15416-21 (2004)
)。さらに、IRAK−1が、IRF−7との会合を通じてこの分子中に含まれ、TLR7/9シグナル伝達によるIFN−α産生に必要であることが分かっている(Uematsu, S. et al., J Exp Med 201, 915-23 (2005))。

二本鎖RNA及びLPSをそれぞれ認識するTLR3及びTLR4は、IFN−β遺伝子の誘導のためのシグナルを仲介できる。この誘導には、2つのIKKファミリーの因子TBK−1及びIKKι/IKKεが必要である(Hemmi, H. et al. J Exp Med 199, 1641-50 (2004);及びPerry, A.K., et al., J Exp Med 199, 1651-8 (2004))。これらのキナーゼはMyd88関連アダプター分子TRIFと会合し、TLR3/4誘導IFN−β遺伝子発現に関与している(Yamamoto, M. et al. Science 301, 640-3 (2003);及びFitzgerald, K.A. et al. Nat Immunol 4, 491-6 (2003))。しかし、TLR9誘導IFN−α産生は、TBK−1又はIKKι/IKKε欠損マウスで損なわれないので、別のIFN−α誘導経路の関与が示唆されている(Kawai, T. et al., Nat Immunol 5, 1061-8 (2004))。
IKKα及びIKKβは、IKKファミリーの中で最初に同定された因子である(Hayden, M.S. et al., Genes Dev 18, 2195-224 (2004);及びBonizzi, G. et al., Trends Immunol 25, 280-8 (2004))。IKKβは、TLR誘導のみならずサイトカイン誘導のNF−κB活性化の正規の経路に必須であり、炎症性サイトカインの産生を導く。IKKαはこの経路には必須ではないが、LT−α、B細胞活性化因子(BAFF)、あるいはCD40の下流のNF−κB活性化の正規でない経路に必須である。IKKαは、ケラチノサイトの分化のみならず、B細胞の成熟やパイエル板の形成にも必須である。しかしながら、TLRシグナルにおけるIKKαの役割は依然として不明であった。
本発明は、TLRシグナルにおけるIKKαの役割を解明し、インターフェロン−αの制御剤及び制御方法を提供することを解決すべき課題とした。
本発明者らは、IKKα欠損マウスを分析して、IKKαがTLRにより誘導される作用にいかに関与するのかを調べた結果、TLR7,9刺激による樹状細胞からのIFN-α産生誘導にIKKαが必要であることを見出し、本発明を完成するに至った。
即ち、本発明によれば、IKKαの阻害剤を含む、インターフェロン−α産生抑制剤が提供される。
好ましくは、IKKαの阻害剤は、IKKαの発現を阻害する物質、又はキナーゼ活性を欠損するIKKα変異体である。

本発明の別の側面によれば、細胞内のIKKα活性を阻害することを含む、細胞からのインターフェロン−αの産生を抑制する方法が提供される。
本発明のさらに別の側面によれば、IKKα、又はIKKαの発現を増大させる物質を含む、インターフェロン−α産生誘導剤が提供される。
本発明のさらに別の側面によれば、細胞にIKKα、又はIKKαの発現を増大させる物質を投与することを含む、細胞からのインターフェロン−αの産生を誘導する方法が提供される。
以下、本発明の実施の形態について説明する。
本発明は、IKKαの阻害剤を含むインターフェロン−α産生抑制剤、並びにIKKα、又はIKKαの発現を増大させる物質を含むインターフェロン−α産生誘導剤に関するものである。本明細書中では、インターフェロン−α産生抑制剤及びインターフェロン−α産生誘導剤を総称して、インターフェロン−α制御剤と称する場合がある。
本発明は、IKKαの阻害剤を含むインターフェロン−α産生抑制剤、並びにIKKα、又はIKKαの発現を増大させる物質を含むインターフェロン−α産生誘導剤に関するものである。本明細書中では、インターフェロン−α産生抑制剤及びインターフェロン−α産生誘導剤を総称して、インターフェロン−α制御剤と称する場合がある。
IKKαは、キナーゼの1種であり、その遺伝子の塩基配列はすでに報告されている(Accession numberは、NM#007700;Connelly MA, Marcu KB. CHUK, a new member of the helix-loop-helix and leucine zipper families of interacting proteins, contains a serine-threonine kinase catalytic domain. Cell Mol Biol Res. 1995; 41: 537-49)。
本発明で用いるIKKαの阻害剤としては、IKKαの発現を阻害する物質、キナーゼ活性を欠損するIKKα変異体、又はIKKαに作用してIKKαの活性及び機能を阻害する物質等が含まれる。本明細書において、「阻害」との用語は、抑制又は低減との意味を含む。
IKKαの発現を阻害する物質としては、RNAi、アンチセンス法又はリボザイム法を利用した物質等が挙げられ、特に限定されないが、RNAiを利用したsiRNAsが好ましい。キナーゼ活性を欠損するIKKα変異体の具体例としては、44番目のリジン残基をアラニン残基に置換した変異体などが挙げられる。また、IKKαに作用してIKKαの活性及び機能を阻害する物質としては、低分子化合物及び抗体等が挙げられる。
抗体としては、例えば、IKKαの全長又は部分配列を有するペプチドを免疫源として作製した抗体を用いることができる。全長のIKKαとしては、例えば組み換えIKKα等を用いることができる。抗体の作製は慣用の方法に従って行えばよい。抗体はモノクローナル抗体が好ましい。ペプチドとしては、例えば、IKKαの部分配列を有するペプチド等が挙げられる。
RNAi(RNA interference)は、細胞に導入された2本鎖RNAが、同じ配列を持つ遺伝子の発現を抑制する現象を言う。RNAiによりIKKαの発現を阻害する物質の具体例としては、下記に説明するようなsiRNA又はshRNA等が挙げられる。
siRNA とはshort interfering RNAの略称であり、約21〜23塩基程度の長さの二本鎖RNAをいう。siRNAはRNAiを引き起こすことができる限り、どのような形態のものでもよく、例えば、化学合成もしくは生化学的合成、又は生物体内の合成で得られたsiRNA、あるいは約40塩基以上の二本鎖RNAが体内で分解されてできた10塩基対以上の短鎖二本鎖RNA等であればよい。siRNA の配列と、IKKαのmRNAの部分配列とは100%一致することが好ましいが、必ずしも100%一致していなくてもよい。
siRNAの塩基配列と、IKKα遺伝子の塩基配列との間で相同性のある領域は、IKKα遺伝子の翻訳開始領域を含まないことが好ましい。相同性を有する配列は、IKKα遺伝子の翻訳開始領域から20塩基離れていることが好ましく、70塩基離れていることがより好ましい。相同性を有する配列としては、例えば、IKKα遺伝子の3'末端付近の配列でもよい。
RNAiによりIKKαの発現を阻害する物質としては、siRNA を生成する約40塩基以上のdsRNA等を用いてもよい。例えば、IKKα遺伝子の核酸配列の一部に対して約70%以上、好ましくは75%以上、より好ましくは80%以上、より好ましくは85%以上、さらに好ましくは90%以上、特に好ましくは95%以上、最も好ましくは100%の相同性を有する配列を含む、二本鎖部分を含むRNA又はその改変体を使用することができる。相同性を有する配列部分は、通常は、少なくとも15ヌクレオチド以上であり、好ましくは約19ヌクレオチド以上であり、より好ましくは少なくとも20ヌクレオチド以上であり、さらに好ましくは21ヌクレオチド以上である。
RNAiによりIKKαの発現を阻害する物質としては、3'末端に突出部を有する短いヘアピン構造から成るshRNA(short hairpin RNA)を使用することもできる。shRNAとは、一本鎖RNAで部分的に回文状の塩基配列を含むことにより、分子内で二本鎖構造をとり、ヘアピンのような構造となる約20塩基対以上の分子のことである。また、shRNAとしては3'突出末端を有するのが好ましい。二本鎖部分の長さは特に限定されないが、好ましくは10ヌクレオチド以上であり、より好ましくは20ヌクレオチド以上である。ここで、3'突出末端は、好ましくはDNAであり、より好ましくは少なくとも2ヌクレオチド以上のDNAであり、さらに好ましくは2〜4ヌクレオチドのDNAである。
RNAiによりIKKαの発現を阻害する物質は、人工的に化学合成してもよいし、センス鎖及びアンチセンス鎖のDNA配列を逆向きに連結したヘアピン構造のDNAをT7 RNAポリメラーゼによってインビトロでRNAを合成することによって作製してもよい。インビトロで合成する場合は、T7 RNAポリメラーゼ及びT7プロモーターを用いて、鋳型DNAからアンチセンス及びセンスのRNAを合成することができる。これらをインビトロでアニーリングした後、細胞に導入すると、RNAiが引き起こされ、IKKαの発現が抑制される。細胞への導入は、例えば、リン酸カルシウム法、又は各種のトランスフェクション試薬(例えば、oligofectamine、Lipofectamine及びlipofection等)を用いた方法等により行うことができる。
RNAiによりIKKαの発現を阻害する物質としては上述のsiRNA又はshRNAをコードする核酸配列を含む発現ベクターを用いてもよい。さらに該発現ベクターを含む細胞を用いてもよい。上記した発現ベクターや細胞の種類は特に限定されないが、既に医薬として用いられている発現ベクターや細胞が好ましい。
本発明では、IKKα、又はIKKαの発現を増大させる物質をインターフェロン−α産生誘導剤として用いることができる。IKKαの発現を増大させる物質としては、転写因子等のタンパク質や低分子化合物などが挙げられる。
本発明のインターフェロン−α制御剤の投与経路は特に限定されず、経口投与又は非経口投与(例えば、静脈内投与、筋肉内投与、皮下投与、皮内投与、粘膜投与、直腸内投与、膣内投与、患部への局所投与、皮膚投与等)のいずれの投与経路により投与してもよい。経口投与に適する製剤形態としては、固形又は液体の形態が挙げられ、非経口投与に適する製剤形態としては、注射剤、点滴剤、坐剤、外用剤、点眼剤、点鼻剤等の形態が挙げられる。本発明のインターフェロン−α制御剤は徐放剤の製剤形態であってもよい。本発明のインターフェロン−α制御剤は、その製剤形態により必要に応じて薬学的に許容可能な添加剤が加えられていてもよい。薬学的に許容可能な添加剤の具体例としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、抗酸化剤、保存剤、安定化剤、等張化剤、着色剤、矯味剤、希釈剤、乳化剤、懸濁化剤、溶媒、フィラー、増量剤、緩衝剤、送達ビヒクル、希釈剤、キャリア、賦形剤及び/又は薬学的アジュバント等が挙げられる。
経口用の固形製剤形態の本発明のインターフェロン−α制御剤は、例えば、有効成分であるIKKαの阻害剤、IKKα、又はIKKαの発現を増大させる物質に賦形剤を加え、さらに必要に応じて結合剤、崩壊剤、滑沢剤、着色剤、又は矯味剤などの製剤用添加物を加えた後、常法により錠剤、顆粒剤、散剤、カプセル剤として調製することができる。経口用の液体製剤形態の本発明のインターフェロン−α制御剤は、有効成分であるIKKαの阻害剤、IKKα、又はIKKαの発現を増大させる物質に矯味剤、安定化剤、又は保存剤など製剤用添加物の1種又は2種以上を加え、常法により内服液剤、シロップ剤、エリキシル剤等として調製することができる。
本発明のインターフェロン−α制御剤を液体製剤として処方するために使用される溶媒は、水性又は非水性のいずれでもよい。液体製剤は当該分野において周知の方法により調製することができる。例えば、注射剤は、生理食塩水、PBSのような緩衝液、滅菌水等の溶剤に溶解した後、フィルター等で濾過滅菌し、次いで無菌容器(例えば、アンプル等)に充填することにより調製することができる。この注射剤には、必要に応じて、慣用の薬学的キャリアを含めてもよい。また、非侵襲的なカテーテルを用いる投与方法を用いてもよい。本発明で用いることができるキャリアとしては、中性緩衝化生理食塩水、又は血清アルブミンを含む生理食塩水等が挙げられる。
IKKαのsiRNAまたはsiRNA発現ベクターなど遺伝子送達の種類に関しては、適用される細胞内でIKKαのsiRNAをコードするRNAまたはsiRNA発現ベクターの発現を得る限り特に方法は限定されるものではなく、例えば、ウイルスベクター、リポソームを用いた遺伝子導入を用いる事が可能である。ウイルスベクターとしては、例えば、レトロウイルス、ワクシニアウイルス、アデノウイルス、シンリンセムリキウイルス等の動物ウイルスが挙げられる。
RNAiによりIKKαの発現を阻害する物質は、細胞に直接注入してもよい。
本発明のインターフェロン−α制御剤の投与量は、使用目的、疾患の重篤度、患者の年齢、体重、性別、既往歴、又は有効成分として用いる物質の種類等を考慮して、当業者が決定することができる。本発明のインターフェロン−α制御剤の投与量は、例えば、有効成分量として、成人一人当たり、約0.1 ng〜約100 mg/kg、好ましくは約1 ng〜約10 mgであり、ウイルスベクター又は非ウイルスベクターとして投与される場合は、通常、0.0001〜100 mg、好ましくは0.001〜10 mg、より好ましくは0.01〜1 mgである。
本発明のインターフェロン−α制御剤の投与頻度は、例えば、一日一回〜数ヶ月に1回であればよい。RNAiによりIKKαの発現を阻害する物質を用いる場合は一般に投与後1〜3日間効果が見られるので、毎日〜3日に1回の頻度で投与することが好ましい。発現ベクターを用いる場合には1週間に1回程度の投与が適する場合もある。
本発明のインターフェロン−α制御剤(インターフェロン−α産生抑制剤、又はインターフェロン−α産生誘導剤)については、インターフェロンの過剰産生が関与している疾患、又は体内においてインターフェロン産生を誘導することにより治療又は予防を期待することができる疾患の治療又は予防のために使用することができる。
このような疾患の具体例としては、種々の炎症状態、膠原病、自己免疫疾患、種々の免疫疾患、特に関節および筋肉における炎症および疼痛(慢性関節リウマチ、リウマチ様脊椎炎、骨関節症、尿酸性関節炎等)、皮膚の炎症性状態(湿疹等)、眼の炎症性状(結膜炎等)、炎症を伴う肺の障害(喘息、気管支炎等)、炎症を伴う消化器の状態(アフタ性潰瘍、クローン病、萎縮性胃炎、いぼ状胃炎、潰瘍性大腸炎、脂肪便症、限局性回腸炎、過敏性腸症候群等)、歯肉炎、(手術または障害後の炎症、疼痛、腫脹)、炎症に関連した発熱、疼痛、その他の状態、移植による拒絶、全身性エリテマトーデス、強皮症、多発性筋炎、多発性軟骨炎、結節性動脈周囲炎、懐死性血管炎、反応性脊椎関節症、強直性脊椎炎、炎症性慢性腎状態(糸球体腎炎、ループス腎炎、膜性腎炎等)、リウマチ熱、シェーグレン症候群、ベーチェット病、甲状腺炎、I型糖尿病、皮膚筋炎、慢性活動性肝炎、重症筋無力症、グレーヴス病、多発性硬化症、原発性胆汁性肝硬変、自己免疫性血液疾患(溶血性貧血、真性赤血球性貧血、特発性血小板減少症、再生不良性貧血等)、橋本病、ぶどう膜炎、接触皮膚炎、乾癬、川崎病、I型アレルギー反応が関与する疾患(アレルギー性喘息、アトピー性皮膚炎、蕁麻疹、アレルギー性結膜炎、花粉症、湿疹、食物過敏症、アレルギー性鼻炎等)、ショック(敗血性ショック、アナフィラキシー性ショック、成人型呼吸窮迫症候群等)、サルコイドーシス、ウェゲナー肉芽腫症、ホジキン病、癌(肺癌、胃癌、結腸癌、肝癌等)が例示される。また、種々の微生物感染症、特に各種ウイルスによる急性(たとえばインフルエンザウイルス、単純ヘルペスウイルス、水疱性口内炎ウイルスなど)、慢性(たとえばB型肝炎ウイルス、C型肝炎肝炎ウイルスなど)感染症、各種細菌感染症、各種真菌感染症、各種寄生虫感染症等も例示される。
以下の実施例により本発明をさらに具体的に説明するが、本発明は実施例によって限定されるものではない。
実施例1:TLR7,9刺激による骨髄由来in vitro樹状細胞からのIFN-α産生誘導におけるIKKαの必要性
IKKα+/-マウス同士を交配させることにより、胎生13.5-15.5日の胎児を得た。この胎児の中から、IKKα+/+,-/-の遺伝形質を有するマウスをPCRにて選別したのち、胎児肝細胞を採取した。さらに、胎児肝細胞を放射線照射野生型C57BL6-CD45.1マウスに静注したのち6-10週後にIKKα+/+あるいはIKKα-/-骨髄キメラマウスとして使用した。
IKKα+/-マウス同士を交配させることにより、胎生13.5-15.5日の胎児を得た。この胎児の中から、IKKα+/+,-/-の遺伝形質を有するマウスをPCRにて選別したのち、胎児肝細胞を採取した。さらに、胎児肝細胞を放射線照射野生型C57BL6-CD45.1マウスに静注したのち6-10週後にIKKα+/+あるいはIKKα-/-骨髄キメラマウスとして使用した。
このキメラマウス骨髄細胞を5x106個/mlで、 10%FCS, 100ng/ml human Flt3ligand (Peprotech)を含むRPMI1640培地で6〜8日間培養することにより、Flt3L誘導骨髄樹状細胞(Flt3L BM DC)を調整した。このFlt3L BM DCを非刺激(med)あるいは100ng/ml Bacterial lipopeptide (BLP, Invivogen), 50μg/ml polyinosinic-polycytidylic acid (poly (I:C), Amersham)(但し、図1cでは、lipofectamine 2000(Invitrogen)と混合した最終濃度25μg/ml poly(I:C)を使用), 100ng/ml Salmonella Minnesota Re595由来LPS (Sigma), 100nM R848 (Invivogen), 合成DNA (1μM 1668:5'-tccatgacgttcctgatgct-3'(配列番号1), 3μM D19:5'-ggTGCATCGATGCAgggggG-3' (配列番号2)、小文字はphosphorothioate DNA, 大文字はphosphodiester DNA), Lipofectamine 2000 (Invitrogen)と混合した最終濃度20μg/ml polyuridylic acid (PolyU)により20-24時間刺激し、培養上清中のIL-12p40、TNF-αの量をGenzyme Techne社のELISAにて、IFN-αの量をPBL社のELISAにて測定を行った (図1a、b及びc)。
野生型Flt3L誘導骨髄樹状細胞(Flt3L BM DC)は、細菌リポペプチド(BLP, TLR2リガンド)、ポリ(I:C)(TLR3リガンド)、LPS(TLR4リガンド)、R848(TLR7リガンド)及びODN1668(TLR9リガンド)に応答してIL-12p40を産生した。IKKα-/- Flt3L BM DCもIL-12p40を産生することによってこれらのTLRアゴニストに応答した。TLR7又はTLR9シグナル伝達によるIL-12p40の誘導量は僅かに減少したが、刺激後は有意に増加した(図1a)。TLR7又はTLR9シグナル伝達によるTNF-αの誘導量は野生型及びIKKα-/- Flt3L BM DCにおいて有意差はみられなかった(図1b)。また、D/A型CpG DNA(D19)は、野生型Flt3L BM DCにおけるIFN-α産生を誘導できたが、この誘導は、IKKα-/- Flt3L BM DCでは大きく障害されていた(図1c)。Poly U誘導IFN-α産生もIKKα-/- Flt3L BM DCでは大きく障害されていた(図1c)。一方、poly (I:C)刺激によるIFN-α産生は正常であった(図1c)。
また、CD40とB220の発現をFACSにより解析を行った (図1d)。B220+(PDC)及びB220-(通常のDC)の2種類の樹状細胞の割合は正常であり、各々のDCのTLR7及びTLR9によるCD40の発現誘導は、IKKα+/+及びIKKα-/- Flt3L BM DCの間で有意な差を認めなかった(図1d)。
Flt3L BM DCにおけるIFN-α, IFN-β, IL-12p40遺伝子の発現を解析するため、Flt3L BM DCをLPS、D19またはPolyUにより刺激したのち、RNAを調製し、IFN-α, IFN-β, IL-12p40遺伝子をプローブとしてNorthern blot analysisを行った (図1e)。TLR7又はTLR9シグナル伝達によるIFN-α遺伝子誘導は、IKKα-/- Flt3L BM DCでは大きく損なわれたが、IFN-β及びIL-12p40遺伝子のTLR7又はTLR9誘導発現上昇は、野生型及びIKKα-/- Flt3L BM DCの間で同等であった(図1e)。この結果より、IKKαが、in vitro樹状細胞においてTLR7又はTLR9に仲介されたIFN-α誘導に必須であることが示された。
実施例2:TLR7,9刺激によるin vivo樹状細胞からのIFN-α産生誘導におけるIKKαの必要性
キメラマウスの骨髄細胞におけるCD11c, B220の発現をFACSにて解析した(図2a)。その結果、IKKα+/+ 及びIKKα-/- キメラは共に、CD11c+B220+細胞を骨髄細胞中に含んでいた。

また、骨髄細胞を1x106個/mlで、10%FCS を含むRPMI1640培地で非刺激、あるいは100nM R848, 1μM 1668, 3μM D19, Lipofectamine 2000 (Invitrogen)と混合した最終濃度25μg/ml polyUの刺激後20〜24時間の培養上清中のIL-12p40の量をGenzyme Techne社のELISAにて、IFN-αの量をPBL社のELISAにて測定を行った (図2b)。TLR7又はTLR9誘導IFN-α産生は、IKKα-/- キメラ骨髄細胞において大きく減少していた。一方、IL-12p40誘導は軽度減少していた(図2b)。
また、キメラマウスの脾臓細胞から、磁気ビーズを用いたソーティング(MACS)によりCD11c陽性細胞を調製し、そのCD11c, B220の発現をFACSにて解析した(図2c)。CD11c陽性細胞におけるB220陽性細胞の数は、IKKα+/+ 及びIKKα-/- キメラ脾臓細胞の間で同等であった(図2c)。
また、このCD11c陽性脾細胞を骨髄細胞と同様の条件で刺激し、IL-12p40,IFN−αの産生量をELISAにより測定した(図2d)。IKKα-/- CD11c陽性脾細胞は、IKKα+/+ CD11c陽性脾細胞よりもIL-12p40の産生量が軽度減少していたが、IKKα-/- CD11c陽性脾細胞からのTLR7/9で誘導されるIFN−αの産生は大きく低下していた(図2d)。
キメラマウスに1匹あたり50nmolのR848を静注し、静注後1,3,6時間に採血を行い、血清中のIFN-α濃度をELISAにより測定した(図2e)。血清中のIFN-α濃度のTLR-7リガンド(R848)によって誘導される上昇は、PDCからの産生に依存しているが、IKKα-/- キメラで大きく低下していた(図2e)。
実施例3:MyD88依存性経路によるIFN-αプロモーター活性化におけるIKKαの機能
(1)プラスミドの調製
pUNO-MyD88, pUNO-TRAF6: Invivogenより購入した。
pUNO: pUNO-TRAF6をAgeI+NheIで切断し、インサートを除去後、self annealingを行い、コントロールベクターとして使用した。
pEF-BOS-FLAG-mIRF-7: プライマー5'-GGGTCGACATGGACTACAAAGACGATGACGACAAGGCTGAAGTGAGGGGGGTCCAGCGA-3' (配列番号3)と5'-GGGCGGCCGCTCAAGGCCACTGACCCAGGTCCATGAG-3'(配列番号4)によるPCRにて、GM-CSFによって誘導されたマウス骨髄由来樹状細胞のcDNAライブラリーから、IRF7 cDNAを増幅し、プラスミドpEF-BOS (S. Mizushima, S. Nagata. Nucleic Acids Res. 18:5322,1990.)のSalI-NotI部位に挿入した。
(1)プラスミドの調製
pUNO-MyD88, pUNO-TRAF6: Invivogenより購入した。

Mizushima, S. Nagata. Nucleic Acids Res. 18:5322,1990.)のSalI-NotI部位に挿入した。
pSRα-6myc-mIKKα: 徳島大学松本満先生より供与を受けた。野生型IKKα(IKKα-WT)の発現ベクターとして使用した。
pSRα-6myc-mIKKα(K44A): pSRα-6myc-mIKKαを基に、44番目のリジン残基をアラニン残基に変換するようにin vitro mutagenesisを行い、キナーゼ活性欠損IKKα(IKKα−KD)の発現ベクターとして使用した。
pSRα-6myc: pSRα-6myc-mIKKαからmIKKαをEcoRI+XhoIにより切断、末端を修復し、self annealingを行った。

(2)ルシフェラーゼアッセイ(図3a,b,f)
ヒト腎臓由来細胞株293Tを2x105個/mlの濃度で0.35ml/well, 24穴プレート上で10%FCSを含むDMEM培地で培養した。24時間後、pUNO、pUNO-MyD88(Invivogen), pUNO-TRAF6, pEF-BOS-Flag-mIRF-7, pSRα-6myc, pSRα-6myc-mIKKα, pSRα-6myc-mIKKα(K44A)を種々の組み合わせで、レポーター遺伝子pIFN-α4-luc (図3a及びb), pELAM1-luc (図3f)あるいはpIFN-β-luc (図3f)、およびpRL-TK(Promega)と共に添加し、lipofectamine 2000(Invivogen)を用いて、遺伝子導入を行った。18-24時間後、細胞抽出物を調製し、Dual-Luciferase Reporter Assay System(Promega), 20/20nLuminometer (Turner Biosystem)を用いてluciferase活性を測定した。遺伝子導入効率を補正するために、得られたFirefly luciferase 活性をRenilla luciferase活性で除することにより、相対活性値とした。最終的に実験データは空ベクターのみで遺伝子導入した相対活性値を1として、fold inductionで示した。
ヒト腎臓由来細胞株293Tを2x105個/mlの濃度で0.35ml/well, 24穴プレート上で10%FCSを含むDMEM培地で培養した。24時間後、pUNO、pUNO-MyD88(Invivogen), pUNO-TRAF6, pEF-BOS-Flag-mIRF-7, pSRα-6myc, pSRα-6myc-mIKKα, pSRα-6myc-mIKKα(K44A)を種々の組み合わせで、レポーター遺伝子pIFN-α4-luc (図3a及びb), pELAM1-luc (図3f)あるいはpIFN-β-luc (図3f)、およびpRL-TK(Promega)と共に添加し、lipofectamine 2000(Invivogen)を用いて、遺伝子導入を行った。18-24時間後、細胞抽出物を調製し、Dual-Luciferase Reporter Assay System(Promega), 20/20nLuminometer (Turner Biosystem)を用いてluciferase活性を測定した。遺伝子導入効率を補正するために、得られたFirefly luciferase 活性をRenilla luciferase活性で除することにより、相対活性値とした。最終的に実験データは空ベクターのみで遺伝子導入した相対活性値を1として、fold inductionで示した。
TLRアダプターMyD88だけの発現ではIFN-αプロモーターを活性化できないが、プロモーターのIRF-7誘導活性化を相乗的に増大することができる(図3a)。IKKα-KDの発現は、IRF-7のみによる活性化に影響しないが(図3a)、IRF-7共存下で認められるMyD88による相乗効果を阻害できた(図3a)。一方、IKKα-WTの発現は、MyD88、IRF-7の相乗効果をわずかながら増強した(図3a)。MyD88と同様、別のアダプター分子であるTRAF6も、IRF-7と相乗的にIFN-αプロモーターを活性化することができる(図3b)。IKKα-KDは、このTRAF6による誘導相乗効果も阻害できた。MyD88もNF-κB又はIFN-βプロモーターを活性化できるが、この活性はIKKα-KDの発現では阻害されなかった(図3f)。従って、IKKαは、そのキナーゼ活性を介して、MyD88又はTRAF6と相乗してIFN-αプロモーターを活性化するIRF7の能力に関与していることが示された。
(3)IKKαとIRF7との会合 (図3c,d,e)
(3−1)293T細胞におけるIKKαとIRF7の会合(図3c)
60mm dish1枚あたり2x106個の293Tを10%FCSを含むDMEM培地で一晩培養した。次に示されているプラスミドを全量5μgとして、 lipofectamine 2000(Invivogen)を用いて遺伝子導入を行った。遺伝子導入20時間後、細胞を回収し、溶解液(1% Nonidet P-40, 150mM NaCl, 5mM EDTA, 1mM PMSF, 20μg/ml aprotinin, 10mM NaF, 10mM β-glycerophosphate, 1mM Na3VO4, 及び20mM Tris-HCl, pH8.0)にて細胞抽出を行った。protein G-Sepharose4FF beadsと1時間混合した後、上清をさらに6μg/ml anti-Flag M2 mAb (Sigma-Aldrich) あるいは5μg/ml anti-Myc-tag clone PL14 mAb (MBL, Japan)を結合させたprotein G-Sepharose4FF beadsと4℃で12時間混合を行った。ビーズを3回洗浄した後、Laemmli液にて100℃5分で溶出したものを10%SDSポリアクリルアミドゲルにて電気泳動し、PVDFメンブレンに転写した。次にメンブレンをビオチン標識anti-Flag M2 mAb (Sigma-Aldrich) あるいは ビオチン標識anti-Myc-tag clone PL14 mAb (MBL, Japan)と室温で1時間混合した。TBS-Tバッファー (0.1%Tween20, 137mM NaCl, 2.6mM KCl, 25mM Tris-HCl, pH7.0)で洗浄後、さらにstreptavidin-HRP conjugate (Amersham bioscience) にて室温30分混合した。さらに洗浄後、ECL system (PerkinElmer Life sciences)を用いて、バンドの検出を行った。
(3−1)293T細胞におけるIKKαとIRF7の会合(図3c)

その結果、IKKαはIRF-7と一緒に免疫沈降することが確認された(図3c)。MyD88又はTRAF6の共発現は、IRF7及びIKKαの相互作用を有意に増強できた(図3c)。
(3−2)Flt3L 誘導BM DCにおけるIKKaとIRF-7の会合(図3d)
野生型Flt3L BM DCを調製し、ウサギ抗IRF-7抗体(United States Biological)、そのコントロールとして正常ウサギ抗体IgG(Santa Cruz Biotechnology)、抗IKKα抗体(M-280, Santa Cruz Biotechnology)を用いて、図3cと同様に、細胞抽出物の作成、免疫沈降およびWestern blotを行った。IRF-7のバンドの検出にはHRP標識ロバ抗ウサギIgG抗体(Amersham Bioscience)を用いた。
野生型Flt3L BM DCを調製し、ウサギ抗IRF-7抗体(United States Biological)、そのコントロールとして正常ウサギ抗体IgG(Santa Cruz Biotechnology)、抗IKKα抗体(M-280, Santa Cruz Biotechnology)を用いて、図3cと同様に、細胞抽出物の作成、免疫沈降およびWestern blotを行った。IRF-7のバンドの検出にはHRP標識ロバ抗ウサギIgG抗体(Amersham Bioscience)を用いた。
その結果、IKKαとIRF-7の会合が検出された(図3d)。
(3−3)GM-CSF誘導BMDCにおけるIKKαとIRF7の会合(図3e)
IKKα+/+あるいはIKKα-/-キメラマウスの骨髄細胞を1x106個/mlで、10%FCS, 10ng/ml GM-CSF (R&D)を含むRPMI1640培地で6〜8日間培養することにより、GM-CSF誘導骨髄樹状細胞(GM-CSF BM DC)を調整した。さらに、レンチウイルスベクターpCSII-EF-FLAG-mIRF7-IRES2-Venus を用いて、GM-CSF BM DCにIRF7を強制発現させた。pCSII-EF-FLAG-mIRF7-IRES2-Venusは、理化学研究所三好浩之先生、宮脇敦史先生より供与されたレンチウイルスベクターpCSII-EF-MCS-IRES2-Venusに、実施例3(1)で作成したIRF7 cDNAを挿入することにより作成した。このGM-CSF-DCを、Anti-FLAG M2 mAb、そのコントロールとしてanti-myc-tag clone PL14 mAb、抗IRF7抗体、及び抗IKKα抗体(M-280, Santa Cruz Biotechnology)を用いて、図3cと同様に、細胞抽出物の作成、免疫沈降およびWestern blotを行った。
IKKα+/+あるいはIKKα-/-キメラマウスの骨髄細胞を1x106個/mlで、10%FCS, 10ng/ml GM-CSF (R&D)を含むRPMI1640培地で6〜8日間培養することにより、GM-CSF誘導骨髄樹状細胞(GM-CSF BM DC)を調整した。さらに、レンチウイルスベクターpCSII-EF-FLAG-mIRF7-IRES2-Venus を用いて、GM-CSF BM DCにIRF7を強制発現させた。pCSII-EF-FLAG-mIRF7-IRES2-Venusは、理化学研究所三好浩之先生、宮脇敦史先生より供与されたレンチウイルスベクターpCSII-EF-MCS-IRES2-Venusに、実施例3(1)で作成したIRF7 cDNAを挿入することにより作成した。このGM-CSF-DCを、Anti-FLAG M2 mAb、そのコントロールとしてanti-myc-tag clone PL14 mAb、抗IRF7抗体、及び抗IKKα抗体(M-280, Santa Cruz Biotechnology)を用いて、図3cと同様に、細胞抽出物の作成、免疫沈降およびWestern blotを行った。
その結果、IRF7とIKKαの会合が検出された(図3e)。
実施例4:IKKαによるGST-IRF7のリン酸化(図4)
(1)プラスミドの調製
pGST-IRF7: pEF-BOS-FLAG-mIRF-7の422-457番目の アミノ酸に相当する領域をPCRにて増幅し、pGEX-5X-1 (Amersham Biosciences)にサブクローニングを行った。
pGST-IκBα: マウスIκBαの5-55番目のアミノ酸に相当する領域をPCRにて増幅し、pGEX-5X-1 (Amersham Biosciences)にサブクローニングを行った。
(1)プラスミドの調製
pGST-IRF7: pEF-BOS-FLAG-mIRF-7の422-457番目の アミノ酸に相当する領域をPCRにて増幅し、pGEX-5X-1 (Amersham Biosciences)にサブクローニングを行った。
pGST-IκBα: マウスIκBαの5-55番目のアミノ酸に相当する領域をPCRにて増幅し、pGEX-5X-1 (Amersham Biosciences)にサブクローニングを行った。
(2)リン酸化アッセイ(図4)
pGEX-5X-1, pGST-IRF7およびpGST-IκBαを大腸菌BL21に導入し、Glutathione sepharose 4B affinity chromatography (Amersham Biosciences)を用いて、GSTタンパク(コントロール)、GST融合IRF7およびGST融合pGST-IκBαを精製し、基質として使用した。293T細胞にpSRα-6myc(コントロールベクター)、pSRα-6myc-mIKKα、pSRα-6myc-mIKKα(K44A)を導入し、細胞抽出物を作成し、PL14を用いて免疫沈降を行った。この免疫沈降物と基質2μgを混合し、20mM HEPES-NaOH(pH7.6),10mM MgCl2, 2mM MnCl2, 50mM NaCl, 0.1mM Na3VO4, 10mM β-glycerophosphate, 10mM PNPP, 1mM DTT, 10mM NaF, 50μM ATP, 370kBq[γ-32P]ATPの存在下で37度30分反応させた(図4a)。反応後、10%SDSポリアクリルアミドゲルにて電気泳動、引き続きオートラジオグラフィーを行った。また、組み換えヒトIKKα (Lake Placid) 100ng (60units/mg protein)を用いて同様の反応を行った(図4b)。
pGEX-5X-1, pGST-IRF7およびpGST-IκBαを大腸菌BL21に導入し、Glutathione sepharose 4B affinity chromatography (Amersham Biosciences)を用いて、GSTタンパク(コントロール)、GST融合IRF7およびGST融合pGST-IκBαを精製し、基質として使用した。293T細胞にpSRα-6myc(コントロールベクター)、pSRα-6myc-mIKKα、pSRα-6myc-mIKKα(K44A)を導入し、細胞抽出物を作成し、PL14を用いて免疫沈降を行った。この免疫沈降物と基質2μgを混合し、20mM HEPES-NaOH(pH7.6),10mM MgCl2, 2mM MnCl2, 50mM NaCl, 0.1mM Na3VO4, 10mM β-glycerophosphate, 10mM PNPP, 1mM DTT, 10mM NaF, 50μM ATP, 370kBq[γ-32P]ATPの存在下で37度30分反応させた(図4a)。反応後、10%SDSポリアクリルアミドゲルにて電気泳動、引き続きオートラジオグラフィーを行った。また、組み換えヒトIKKα (Lake Placid) 100ng (60units/mg protein)を用いて同様の反応を行った(図4b)。
その結果、IRF7は、IKKαによりリン酸化されることが分かった(図4a)。
本発明により、インターフェロン−αの産生を制御することが可能になった。本発明のインターフェロン−α制御剤は、インターフェロン−αの産生の抑制又は誘導により治療や予防に有効となる各種疾患の治療や予防のために用いることができる。
Claims (5)
- IKKαの阻害剤を含む、インターフェロン−α産生抑制剤。
- IKKαの阻害剤が、IKKαの発現を阻害する物質、又はキナーゼ活性を欠損するIKKα変異体である、請求項1に記載のインターフェロン−α産生抑制剤。
- 細胞内のIKKα活性を阻害することを含む、細胞からのインターフェロン−αの産生を抑制する方法。
- IKKα、又はIKKαの発現を増大させる物質を含む、インターフェロン−α産生誘導剤。
- 細胞にIKKα、又はIKKαの発現を増大させる物質を投与することを含む、細胞からのインターフェロン−αの産生を誘導する方法。
